The Glycopeptide, a Promoter of Thymidine Uptake, from Aloe Vera

  • As a part of search for new biologically active constituents from aloe, we have isolated a glycopeptide, called G1G1M1DI2, from the gel(G1) of Aloe vera. Chemical and spectroscopic evidence indicated that G1G1M1DI2 is a glycopeptide. The molecular weight of G1G1M1DI2 was about 5,500 daltons, and the carbohydrate and protein contents were 20.9% and 32.6%, respectively. Periodate oxidation and enzymic degradation gave peptide moiety and carbohydrate moiety, respectively. Carbohydrate moiety is composed of fucose, galactose, glucose and mannose in a molar ratio of 0.5:2.4;48.8:48.3. Peptide moiety is composed of fifteen amino acids, and glutamic acid and glycine were the major componants. The glycopeptide, G1G1M1DI2, stimulated thymidine uptake of SCC 13 cells about 6.5 times the control. This result suggests that this glycopeptide has a skin cell proliferating activity.

Antioxidant Activity and Inhibition of MMP-1 Expression of Schizandrae fructus (Schizandra chinensis) Extract (오미자 추출물의 항산화 활성 및 MMP-1 발현저해)

  • We invesitagat the effects of antioxidant and suppression of MMP-1 in UV-irradiated human dermal fibroblasts (HDF) by schizandrin isolated from Schisandra chinensis. Ultrasonic technique was used to extract schizandrin from S. chinensis. The higher contents of schizandrin (6.34 mg/ml) in 70% EtOH extract obtained by ultrasonic treatment at 20 MHz for 2 h during 10 days. Schizandrin had antioxidant activities. Schizandrin (1-10 ${\mu}g/ml$) induced a significant dose-dependent decrease in the expression fo MMP-1 protein. These results suggest that schizandrin is a potential candidate for the prevention and treatment of skin aging.

Effects of Various Polyols on Antiseptic System in Emulsions (에멀젼 제형에서 수종의 폴리올이 방부 시스템에 미치는 영향)

  • It is inevitable to use chemical germicidal agents like paraben, imidazolidinyl urea and phenoxyethanol to preserve the emulsions which is usually used in cosmetics. Although these chemical preservatives are good enough to reduce the microbiological contamination, they are irritative, allergenic to the skin. Several kinds of polyols are used in cosmetics as moisturizer and solvent. In this study, we evaluate the effects of polyols on anti-microbial activities, safety and resistant index. MIC(minimal inhibitory concentration) of polyols determined against 6 germs including Staphylococcus aureus. The order of MIC was PG $\cong$ DPG $\cong$ 1,3BG > HG > 1,2-PD > 1,2-HD $\cong$ 1,2-OD. The $2{\sim}3\;wt%$ of 1,2-HD(hexanediol) shows good anti-microbial effects in emulsions without allergenic response. Resistant index of 1,2-HD was less than 2 and this value was smaller than that of chemical preservatives. The mechanism of antimicrobilogical effect might be disturb the membrane of germs by investigating using electron microscope. Added to that, using this paradigm, low preservative contents, paraben-free system, and even preservative-free systems can be expected from these results.

The Effects of Fatty Acids Supplementation in Culture Medium on Proliferation and Lipid Peroxides Production of Fibroblast from Neonate Rats (신생흰쥐 피부섬유아세포의 배양액의 지방산의 종류와 양을 변화시켰을 때 세포의 증식과 지질과산화물 생성에 미치는 영향)

  • This study was performed to investigate the effects of concentration and degree of unsaturation of fatty acids on cellular proliferation and lipid peroxide production, using primary skin fibroblasts from neonate rats Fibroblasts (CPD : 2.8-5.4). Cells were cultured either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various kinds (stearic, oleic, linoleic, arachidonic, linolenic, eicosapentaenoic acid) and amounts (5, 10, 25, 50, 100, 150uM)of fatty acids. Cellular proliferation ratio and lipid peroxice production were measured and morphological changes were observed. Cellular proliferation was inhibited and morphological changes were observed. Cellular proliferation was inhibited and morphological changes were observed in cells grown in stearic containing media. Oleic, arachidonic, and eicosapentaenoic aicd tend to stimulate cellualar proliferation, and linolenic acid had no effects. Lipid peroxide concentrations in fibroblasts increased in proportion to the contents and unsaturation of fatty acids in media. Especially supplementation of arachidonic acid accelerated cellualr lipid peroxidation. Free radicals may cause severs damage to biological molecules, so lipid peroxidation probably contributes cellular membrane damages. However there were little relationship between lipid peroxide production and cellular proliferation in this study. (Korean J Nutrition 29(2) : 159~165, 1996)

A Study on Skin Care Effects of Rapeseed Meal Extract (유채박 추출물의 피부미용 효과에 관한 연구)

  • This research was to investigate the inhibitory effects of tyrosinase, elastase and collagenase using rapeseed meal extract for the functional cosmetics. Gamma-tocopherol and alpha-tocopherol contents were 304.9 and 212.2 mg/kg, respectively. In the case of delta-tocopherol and plastochromanol-8, they were 12.1 and 35.7 mg/kg, respectively. The total phenol content of methanol extract was the highest (49.6 mg/g) which was about 4.96 fold higher than that of water extract. The maximum nitrite scavenging activities of methanol and acetone extract at pH 1.2 were 85.2 and 80.1%, respectively, at 8.0 mg/mL. When the extract concentration of rapeseed meal increased upto 2.0 mg/mL, cell viabilities did not appear to have any significant cytotoxic effect, irrespective of extracts. Tyrosinase and elastase inhibition activities increased from 25.2 to 42.5% and 25.3 to 48.0%, respectively, as methanol extract concentration increased from 0.5 to 1.0 mg/mL. The collagenase inhibition activities of methanol and acetone extracts at 1.0 mg/mL were 67.2 and 68.0%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic agent.

Electrospinning Fabrication of Poly(vinyl alcohol)/TiO2 Nanofibers (전기방사를 이용한 Poly(vinyl alcohol)/TiO2 나노섬유 제조)

  • Poly(vinyl alcohol) (PVA)/titanium dioxide ($TiO_2$) composite nanofibers were produced at different $TiO_2$ concentrations (0.5, 1 and 1.5 wt.%) using the electrospinning method. The parameters of electrospinning including polymer contents, voltage and tip-to-collector distance (TCD) were optimized for fabrication process. The study showed that 7.5 wt.% PVA, 15 kV applied voltage and 15 cm TCD are the best condition to obtain uniform PVA/$TiO_2$ nanofibers. $TiO_2$ nanoparticles give significant effect in fiber morphology. $TiO_2$ content increases the diameter of the fibers and roughen the fiber surfaces. The PVA/$TiO_2$ nanofibers were characterized by scanning electron microscope (SEM), transmission electron microscope (TEM) and X-ray diffraction (XRD).

Quality Characteristics and Microbial Safety of Sunsik with Dandelion (Taraxacum platycarpum) Complex Extract Powder (AF-343) for Home Meal Replacement (간편가정식용 민들레복합추출물 (AF-343) 첨가 선식의 품질특성 및 미생물적 안전성)

  • We investigated the antioxidant and physicochemical qualities as well as the sensory characteristics, and microbial safety of sunsik containing varied amounts of AF-343, which can help add moisture to the skin and relieve the symptoms of atopic dermatitis. Samples did not show significant differences in pH measurements, but the pH had a tendency to increase with tendencies as increased amounts of AF-343. The total phenolic compound contents and DPPH radical scavenging activity, indicators of biologically active ingredients such as antioxidant, anticancer and antibacterial activity, significantly increased as the amounts of AF-343 increased (p<0.05). In an acceptance test, the samples did not show significant differences, however samples with the 750 mg AF-343 received the highest scores out of all the samples in overall acceptance. All samples were confirmed as microbially safe according to the food code applied to food manufacturers. Aerobic plate counts of the control group were 1.60 log CFU/g, while those of samples with 750 mg AF-343 were 1.70 log CFU/g. E. coli. Pathogenic microorganisms tests were either negative or not detected in all samples.

  • Schizonepeta tenuifolia Briq. are used in folk medicine as common cold, sore throat, headache and skin infection in Korea, China and Japan. To compare the contents of domestic and Chinese Schizonepeta tenuifolia quantitative HPLC analysis was performed using a Luna $C_{18}$ column $(4.6{\times}250\;mm,\;5{\mu}m)$, with 1 ml/min of flow rate of 0.01% formic acid in water : acetonitrile = 50 : 50 under UV 254 nm of detector. Pulegone was detected at retention time of about 17.14 min.

  • Acquired pigmentary skin diseases such as abnormal melanogenesis, vitiligo, chloasma and inflammatory pigmentation are related to regulate the melanin production. The aim of this study was to investigate the effect of Codonopsis lanceolata on the melanogenesis of HM3KO human melanoma cells biologically. The cells were treated for 5 days with Codonopsis lanceolata at several concentrations. Treatment with Codonopsis lanceolata suppressed melanin contents as a dose dependent manner without cytotoxicity and morphological change. And the extract of Codonopsis lanceolata also inhibited tyrosinase, a key enzyme forming melanin, in a dose-dependent manner. And it did not affect the DOPAchrome tautomerase(TRP-2) activity. These results suggest that Codonopsis lanceolata is a candidate for an efficient whitening agent which supresses melanogenesis by a Raper-Mason pathway.

Contents Analysis on the Dwellers' Medical Reports in High-Rise Mixed-Use Apartment (주상복합아파트 거주자의 질병자료에 관한 내용 분석)

  • This purpose of this study is to figure out the inter-relationship between the residence stories in high-rise mixed-use apartments and their residents' disease patterns throughout the dweller's medical reports in high-rise mixed-use apartments. Research basic data are obtained from medical fee request of National Health Insurance Corportion. Data are limited a housing complex to 'A' high-rise mixed-use apartment and a medical treatment time to 3 years(2004-2006). Analysis data of total 346,286 medical records, 43,159 disease records, and 8,999 persons are collected. By analyzing those data, findings are as follows: 1) Women is more medical treatments than men, 40-50 age group is more treated, and the residents of 6-25 stories are more received medical treatments. Diseases of the respiratory system and diseases of the eye and adnexa are relatively treated higher than other diseases. 2) The diseases of the respiratory system, the eye and adnexa, the skin and subcutaneous tissue, the ear and mastoid process), and the asthma have not relation to the high-storied residence through the data of disease records and personal records. But the analysis on the data of children, 7 ages and less, is showed a significant relation. And to conclude, there is no relationship between the residence of high-stories in that apartment and dwellers' disease patterns, but there is a little probable to the relationship in the pre-school child.
