Preparation of Talc-Silica Composites by Controlling Surface Charge Behavior (표면전하 거동 조절을 이용한 탈크-실리카 복합체의 제조)

  • A plate-type inorganic pigment complex was manufactured in a manner that treats the surface of the complex by adjusting zeta potential between talc, an inorganic pigment used as a material for color cosmetics, and hydrophobic silica. Talc, which is usually used in the prescription of color cosmetics, is a plate-type, white-colored inorganic substance with good application and spreadability to skin. Furthermore, it features excellent dispersibility and extensibility as well as outstanding heat tolerance, light stability, and chemical resistance. In general, silica contributes to durable makeup and stabilized formulation. This paper covers a process of manufacturing an inorganic pigment complex, where hydrophobic silica was applied to the surface of talc by using differences in zeta potential after the surface charges of talc and hydrophobic silica had been adjusted with cationic and anionic surfactants, respectively. The resulting inorganic pigment complex was composed of talc whose surface is coated hydrophobic silica to the thickness of $1{\mu}m$ or less, which developed an effective hydrophobic property. Zeta potential was measured to analyze the surface charge of an inorganic pigment, and FT-IR, used to check the functional group of a surfactant, was applied to treat the surface of the pigment. The surface of the inorganic pigment complex was observed employing SEM, EDS, and FIB, while its structure was confirmed with XRD and FT-IR.

Molecular Subtyping of Methicillin-Resistant Staphylococcus aureus Isolated from Patients' Nasal Cavity (환자의 비강으로부터 분리된 메티실린 내성 황색 포도알균의 분자 아형 분석)

  • Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterium that causes infections in different parts of the body and causes skin and soft tissue infections (SSTI). The present study examined the antimicrobial resistance patterns and molecular epidemiological characteristics of MRSA isolated from nasal swabs in clinical patients. SCCmec type of MRSA isolates from clinical patients were analyzed: 24 cases were SCCmec type-II; two cases were type-II/IVa; one case was type-II/V; one case was type-IVa; 11 cases were not-typeable. The mec complex type of MRSA isolates from clinical patients were analyzed: 29 cases were mec complex type A, and 10 cases were not-typeable, but type B was not found in the present study. In conclusion, SCCmec type-II and mec complex type A were the most dominant MRSA subtypes among the MRSA isolates from a nasal swab of patients, and the results were similar to other studies on hospital-acquired MRSA (HA-MRSA). These results can not only provide basic data for hospital infection management but also be a good guideline for MRSA infections in the Republic of Korea.

Evaluation of Thermoregulatory Properties of Thermal Underwear Named as 'Heating Underwear' using Thermal Manikin and Human Performance Test ('발열내복'이라 광고되는 시판 기능성 보온내복의 써멀 마네킹과 인체 착용 실험을 통한 체온조절 성능 평가)

  • This study evaluated the thermoregulatory properties of functional thermal underwear ('heating underwear') in markets using a thermal manikin and human wear trials. One ordinary thermal underwear (ORD) and two functional thermal underwear (HEAT1 and HEAT2; manufactured goods, HEAT1: moisture absorbing heat release mechanism, HEAT2: heat storage, release mechanism) were chosen. Thermo-physiological and subjective responses were evaluated at an air temperature of $5.0{\pm}0.5^{\circ}C$ and air humidity of $30{\pm}5%RH$ with five male subjects ($21.6{\pm}1.3yr$ in age, $178.0{\pm}5.9cm$ in height, $68.2{\pm}5.9kg$ in body mass). Experimental conditions consisted of four ensembles that included winter clothes (Control: no underwear, ORD, HEAT1, HEAT2). Water-vapor resistance was greater in fabric of HEAT1 than others. The results were: 1) Total thermal insulation (IT) using a thermal manikin were not greater for HEAT1 (0.860clo) and HEAT 2 (0.873clo) than for ORD (0.886clo). 2) There were no significant differences in rectal temperature, mean skin temperature, heart rate and total body mass loss between the four conditions. Microclimate clothing temperature on the back was greater for ORD than for HEAT1 and HEAT2. Subjects felt more comfortable with HEAT1 than for others at rest. HEAT2 was higher in microclimate humidity when compared to other conditions. The results suggest that thermoregulatory properties of 'heating underwear' in market did not differ from those of ordinary thermal underwear in terms of total thermal insulation and thermoregulatory responses in a cold environment.

RalA-binding Protein 1 is an Important Regulator of Tumor Angiogenesis (Tumor angiogenesis에 있어서 RLIP76의 중요성)

  • Tumor angiogenesis is important in tumorigenesis and therapeutic interventions in cancer. To know inhibitor and effector of tumor angiogenesis in cancer, the specific gene of tumor and angiogenesis may develop the mechanisms of cancer suppression and therapy. Recently, we described the role of RalA-binding protein 1 (RLIP76) in tumor angiogenesis. Tumor vascular volumes were diminished, and vessels were fewer in number, shorter, and narrower in RLIP76 knockout mice than in wild-type mice. Moreover, angiogenesis in basement membrane matrix plugs was blunted in the knockout mice in the absence of tumor cells, with endothelial cells isolated from the lungs of these animals exhibiting defects in migration, proliferation, and cord formation in vitro. RLIP76 is expressed in most human tissues and is overexpressed in many tumor types. In addition, the protein regulates tumorigenesis and angiogenesis in vivo and in vitro. As the export of chemotherapy agents is a prominent cellular function of RLIP76, it is a major factor in mechanisms of drug resistance. Moreover, RLIP76 acts as a selective effector of the small GTPase, R-Ras, and regulates R-Ras signaling, leading to cell spreading and migration. Furthermore, in skin carcinogenesis, RLIP76 knockout mice are resistant, with tumors that form showing diminished angiogenesis. Thus, RLIP76 is required for efficient endothelial cell function and angiogenesis in solid tumors.

Efficient Production of Cloned Bovine Embryos from Transformed Somatic Cells (형질전환 체세포로부터 소 복제수정란의 효율적인 생산)

  • Human thrombopoietin (hTPO) is a cytokine that plays a central role in megakaryopoiesis. To direct hTPO expression in the mammary gland, an expression vector was constructed by combining the promoter of bovine beta-casein gene, cDNA of hTPO and neomycin resistance gene (pBT-L neo). Fibroblast cells derived from cow's ear skin tissue were transfected with the expression vector (pBT-L neo) using Lipofectamine. Transfected cells resistant to G418 trea?nt were cultured to form the colonies for more than 2 weeks. The transformed colonies identified by PCR were further expanded prior to nuclear transfer. Reconstructed oocytes with transformed cells were electrofused, activated using calcium ionophore and 6-DMAP, and cultured in vitro for 7 days. Of 35 cell colonies analyzed by PCR, 29 colonies (82.9%) were positive for the hTPO gene. Cleavage and developmental rates to the blastocyst stage of reconstructed embryos with the transformed cells were 65.1% and 23.8%, respectively Of 29 blastocysts that developed from reconstructed embryos with the transformed cells, 27 embryos (93.1%) were transgenic. These results indicate that transgenic bovine embryos can be efficiently produced by somatic cell nuclear transfer using transformed cells.

  • A common method for treating oral mucosal diseases is taking medication by oral administration. The oral administration is the method of least resistance. Because large part of drugs is degraded by liver, it is necessary to take more drugs getting to appropriate level in blood stream. And there are so many side effects when patients take drugs by oral administration. In so many cases, the patients who suffer from oral mucosal problems have the other general diseases simultaneously. Willingly or not, some patients can't take the medicine by oral administration. Number of topical drugs for oral mucosal disease is less than that for skin diseases because the environment of oral mucosa prevents activity of medicine. In this paper, research on effects of topical type medication for treating oral mucosal diseases is conducted through investigating currently used medications and their effects. In addition, effects of dissolved oral medications with appropriate solvent are demonstrated if this medication is useful for patients clinically.

Tigecycline Treatment for Infections Caused by Multidrug-Resistant Pathogens (다약제내성 균주 감염에 대한 Tigecycline의 치료)

  • Background: Tigecycline (TIG), a new broad-spectrum glycylcycline with anti-multidrug-resistant-(MDR)-pathogen activity, was launched in March 2009 in South Korea, but there are insufficient clinical studies on its use in the country. As such, this study was performed to analyze cases of severe MDR-pathogen-caused infections treated with TIG. Methods: Patients treated with TIG within the period from May 2009 to June 2010 were enrolled in this study. Their clinical and microbiologic data were reviewed retrospectively. Results: Twenty-one patients were treated with TIG for complicated skin and soft-tissue infections (cSSTIs) (42.9%), complicated intra-abdominal infections (cIAIs) (38.1%), or pneumonia (19.1%) caused by MDR pathogens like carbapenem-resistant $Acinetobacter$ $baumannii$ (76.2%), methicillin-resistant $Staphylococcus$ $aureus$ (61.9%), extended-spectrum beta-lactamase-producing $Escherichia$ $coli$ and $Klebsiella$ $pneumoniae$ (38.1%), and penicillin-resistant $Enterococcus$ species (33.3%). Thirteen patients (61.9%) had successful clinical outcomes while five (23.8%) died within 30 days. The rate of clinical success was highest in cSSTI (77.8%), followed by cIAI (50%) and pneumonia (50%), and the mortality rate was highest in pneumonia (50%), followed by cIAI (25%) and cSSTI (11.1%), Conclusion: Tigecycline therapy can be an option for the treatment of severe MDR-pathogen-caused infections in South Korea, Due to its high risk of failure and mortality, however, prudence is required in its clinical use for the treatment of severe infections like nosocomial pneumonia.

Fabrication and Characterization of Aluminum Honeycomb Panel (경량 알루미늄 허니콤 판재의 제작 및 특성 평가)

  • A honeycomb panel is a plate made by attaching two surface plateson eitherside of a honeycomb core. The honeycomb plate hasexcellent specific strength and energy absorption and is suitable for use in regions where good impact resistance is required. Recently, with the increasing the need for a lightweight design to facilitate transportation, numerous studies have been conducted using aluminum honeycomb plates as body materials for vehicles such as automobiles and high-speed trains. In addition, honeycomb plates have excellent sound deadening properties, as well as excellent heat insulation and durability. Savings in weight using lightweight materials such as aluminum alloy for honeycomb panel's skin can lead to increase fuel economy and reduction in air pollution. In this study, in order to improve the design technology of the honeycomb plate material, the manufacturing technology of the aluminum honeycomb core and honeycomb plate material and various mechanical properties of the honeycomb plate were evaluated. From the results, it was found that the design of the manufacturing process of the aluminum honeycomb plate, as well as itsproduction and characteristics, were improved. The resulting excellent energy absorption capability of the honeycomb plate was due to the repetitive core buckling, indicating that the higher the compressive strength, the higher the strength per bonded area.

Isolation and Characterization of Bacterial Cellulose-Producing Bacteria for Silver Nanoparticle Synthesis (은 나노입자 합성을 위한 Bacterial Cellulose 생산 세균의 분리 및 특성)

  • As a basic study for environment-friendly production of bacterial cellulose (BC) dressing with antimicrobial activity, we isolated and identified acetic acid bacteria which are resistant to silver ions and can biosynthesize silver nanoparticles. Furthermore, conditions of BC production by selected strain were also investigated. Strain G7 isolated from decayed grape skin was able to grow in the presence of 0.1 mM $AgNO_3$ which was identified as Acetobacter intermedius based on 16S rRNA gene analysis. BC production was the highest in a medium containing 2% glucose as a carbon source, 2% yeast extract as a nitrogen source, and 0.115% acetic acid as a cosubstrate. Structural properties of BC produced in optimal medium were studied using Fourier-transform infrared spectroscopy and X-ray diffractometer, and it was found that BC produced was cellulose type I that was the same as a typical native cellulose. When strain G7 was cultured in an optimal medium containing 0.1 mM $AgNO_3$, the color of the culture broth turned into reddish brown, indicating that silver nanoparticles were formed. As a result of UV-Vis spectral analysis of the culture, it was found that a unique absorption spectrum of silver nanoparticles at 425 nm was also observed. Scanning electron microscopic observations showed that silver nanoparticles were formed on the surface and pores of BC membrane.

Studies on Transparent Sunscreen of Water-in-Oil Emulsion Type (유중수 타입의 외관이 투명한 자외선 차단 화장료에 관한 연구)

  • This study is related to the developing method of a transparent sunscreen cosmetic which has waterproofing property and no white turbidity when applied to skin. The transparent sunscreen is prepared by exploiting refractive index difference between oil-phase and water-phase of water-in-oil(W/O) emulsion. The sunscreen according to this study is prepared as a W/O type emulsion so that it is water-stable and water resistance. Also, the stability of W/O type emulsion is developed by adjusting the content of oil phase part and water phase part. As a result of this studying, the transparent W/O emulsion is prepared by adjusting the refractive index of oil-phase and water-phase within 0.004 and it is found that the stability of the transparent sunscreen is increasing when the water phase part is over 75% (w/w) of the W/O emulsion. Through clinical test of transparent sunscreen, the value of sun protection Factor(SPF) and Protection Factor of UVA(PFA) were determined. SPF and PFA values of transparent sunscreen were indicated $30.99{\pm}1.65$ and $3.01{\pm}0.30$.
