This parer proposes the six color separation using th color difference and granularity. Conventional method using the color difference increases the graininess in the bright region due to the usage of the cyan or magenta. To reduce the graininess in the bright region, we proposed the six color separation minimizing the graininess within the tolerance of the co]or difference. Initially, granularity is calculated based on the standard deviation of the lightness value and chrominance of the SCIELAB space and is applied to the six color separation using the color difference. Proposed six color separation using the color difference and granularity reduces the graininess in the bright region and obtains the smooth tone.

This paper proposes an improved six-color separation reducing the graininess in a middle tone region based on the standard deviation of the lightness and chrominance in S-CIELAB space. Graininess is regarded as visual perception for the fluctuation of the lightness of the light cyan and cyan or light magenta and magenta. In the conventional methods, the granularity is extremely heuristic and inaccurate due to the use of the visual examination score. Accordingly, this paper proposes a method to calculate the objective granularity for six color separation. First, we use the lightness, redness-greenness, and yellowness-blueness of the S-CIELAB space reflecting the spatial-color sensitivity of the human and normalize the sum of the three standard deviations. Finally, we apply the proposed granularity to the six color separation after assigning the granularity to the lookup table and obtain the result reducing the graininess in a middle tone region.

This paper proposed a six-color separation using additional colorants and quantitative granularity metric to reduce color difference and graininess. In the conventional method, light magenta and light cyan are used in the bright region instead of magenta and cyan. However, the hue value of liBht magenta and light cyan is different from the one of magenta and cyan in CIELAB space, so that this makes the colorimetric reproduction more or less inaccurate. To improve this inaccuracy, the proposed method uses yellow and light magenta colorants as the additional colorants. In the bright region, magenta is replaced with light magenta and yellow, while cyan is replaced with light cyan and light magenta. This selection reduces hue difference because it creates the color of similar hue to magenta and cyan. In addition, smooth image can be simultaneously obtained by the less dot visibility of additional colorants. In the middle region, magenta is replaced with light magenta and magenta, while cyan is replaced with light cyan and cyan. The use of two colorants having a different concentration makes the dot Pattern coarse. To reflect this Phenomenon, quantitative granularity metric is used. In the dark region, only magenta and cyan colorant is used as usual. Through experiments, it is shown that the proposed method improves both colorimetric and smooth tone reproductions.

The Conventional methods of color separation in computer-based machine vision offer only weak performance because of environmental factors such as light source, camera sensitivity, and others. In this paper, we propose an improved color separation method using RGB, HLS, color coordination space, and fuzzy similarity measure. RGB consists of red, green and blue, the three primary colors of light. HLS includes hue, light and saturation, the human recognition elements of co]or. A fuzzy similarity measure was employed for evaluate the similarity among fuzzy colors with the six features of RGB and HLS.

This paper proposes a six-color separation method of reducing unnecessary usage of colorants based on the limitation of total colorant amount and dot visibility ordering. First, the CIELAB values of input RGB image are estimated through the color-mixing model and compared with pre-calculated CIELAB values corresponding to all combination of CMYKlclm colorants with a constraint of color difference, thereby selecting initial CMYKlclm candidates. Next, the limitation on total colorant amount Is imposed on initial CMYKlclm candidates to remove the excessive amounts of colorants, and then final CMYKlclm candidates are determined by minimizing the usage of light cyan and light magenta in the dark region based on the dot visibility ordering of C, M, Y, K, lc, and lm. Through the experiment, the proposed method is shown to reduce the excessive amount of colorants with preserving good image quality.
Activated carbons were used to examine their performance for the separation of undesirable colored materials from culture broth containing hyaluronic acid. Six local samples and a NORIT ROX 08 were tested, whereas the latter was mainly studied under batch and continuous modes. The optimal wavelength for the detection of colored materials was 330nm. The optimal choice of NORIT ROX 08 provided 30% colored residuals with 96% hyaluronic acid recovery of original broth in batch experiments. The nonlinear adsorption behavior of protein and colored materials with activated carbon (C) was correlated by a Langmuir equation to give 18C/24+C and 500C/892+C for protein and colored materials, respectively. It appears that colored materials were composed of 78% protein and 22% glucose residuals on the basis of clearance results. A microscopic study using a scanning electron microscope suggests that regeneration of used activated carbon with 0.1N NaOH and hot water was not satisfactory. The present study proposes that the continuous monitoring of colored materials during purification can be accomplished by Installation of a UV monitor commonly used for continuous detection of protein during the process, as resulted from the significant correlation of color (A330)=0.353protein(mg/ml)+0.1(R=99.7%).
In order to develop procedures for the rapid isolation of recombinant sugar transporter in functional form from away from the endogenous insect cell transporter, gene fusion techniques were exploited. Briefly, BamH1-digested human HepG2 type glucose transport protein cDNA was first cloned into a transfer vector pBlueBacHis, containing a tract of six histidine residues. Recombinant baculoviruses including the human cDNA were then generated by allelic exchange following transfection of insect cells with wild-type BaculoGold virus DNA and the recombinant transfer vector. Plaque assay was then performed to obtain and purify recombinant viruses expressing the human transport protein. All the cell samples that had been infected with viruses from the several blue plaques exhibited a positive reaction in the immnuassay, demonstrating expression of the glucose transport protein. In contrast, no color development in the immunoassay was observed for cells infected with the wild-type virus or no virus. Immunoblot analysis showed that a major immunoreactive band of apparent Mr 43,000~44,000 was evident in the lysate from cells infected with the recombinant baculovirus. Following expression of the recombinant fusion protein with the metal-binding domain and enterokinase cleavage site, the fusion protein was recovered by competition with imidizole using immobilized metal charged resin. The leader peptide was then removed from the fusion protein by cleavage with porcine enterokinase. Final separation of the recombinant protein of the interest was achieved by passage over $Ni^{2+}$-charged resin under binding conditions. The expressed transport protein bound cytochalasin B and demonstrated a functional similarity to its human counterpart.

This study aimed to isolate and identify freshwater algae from the organic agricultural ecosystems and investigate its biological characteristics to study the possibility of utilizing a biomass freshwater algae in organic farming. In the survey area, average water temperature was $12.4{\sim}28.2^{\circ}C$ and the pH ranges were from 6.1 to 8.5. The solid culture method is more suitable than liquid culture method for isolation of freshwater algae with lower contamination level and higher isolation frequency. A total of 115 strains were isolated from six freshwater algae habitats in nine regions in Korea. BGMM (BG11 Modified Medium) amended with NaNO3 and $KNO_3$ as a nitrogen, and $Na_2CO_3$ as carbon source was designed to isolate and culture freshwater algae. Absorbance of freshwater algae culture has increased dramatically to four days and decreased after eight days after inoculation. CHK008 of the seven isolates showed the highest absorbance in seven days after culturing in BGMM. The optimal pH of BGMM for culturing freshwater algae was pH 6-7. As light intensity increased, growth of freshwater algae increased. Among the five kinds of carbon sources, glucose and galactose promoted good growth of freshwater algae in BGMM. The colony color of purified 16 green algae isolates showed a separation of green, dark and light green, and of them, eleven algae strains showed a strong fluorescent light under fluorescence microscopy. Cell size of the green algae showed a wide range of variation depending on the species. General morphology of the green algae strains was spherical. Chlamydomonas sp. was elliptical, and Chlorella sorokiniana was ellipsoidal and cylindrical. All strains of the green algae except for Chlamydomonas sp. did not have flagella. One isolate of Chlamydomonas sp. and five isolates of C. sorokiniana secreted mucus. Sixteen isolates of 16 green algae were identified as two family and six species, Chlorella vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp. based on their morphological characteristics.
