Effect of Cl2 on Electrodeposition Behavior in Electrowinning Process

  • Pyroprocessing at KAERI (Korea Atomic Energy Research Institute) consists of pretreatment, electroreduction, electrorefining and electrowinning. SFR (Sodium Fast Reactor) fuel is prepared from the electrowinning process which is composed of LCC (Liquid Cadmium Process) and Cd distillation et al. LCC is an electrochemical process to obtain actinides from spent fuel. In order to recover actinides inert anodes such as carbon material are used, where chlorine gas ($Cl_2$) evolves on the surface of the carbon material. And, stainless steel (SUS) crucible should be installed in large-scale electrowinning system. Therefore, the effect of chlorine on the SUS material needs to be studied. LiCl-KCl-$UCl_3$-$NdCl_3$-$CeCl_3$-$LaCl_3$-$YCl_3$ salt was contained in 2 kinds of electrolytic crucible having an inner diameter of 5cm, made of an insulated alumina and an SUS, respectively. And, three kinds of electrodes such as cathode, anode, reference were used for the electrochemical experiments. Both solid tungsten (W) and LCC were used as cathodes. Cd of 45 g as the cathode material was contained in alumina crucibles for the deposition experiments, where the crucible has an inner diameter of 3 cm. Glassy carbon rod with the diameter of 0.3 cm was employed as an anode, where shroud was not used for the anode. A pyrex tube containing LiCl-KCl-1mol% AgCl and silver (Ag) wire having a diameter of 0.1cm was used as a reference electrode. Electrodeposition experiments were conducted at $500^{\circ}C$ at the current densities of $50{\sim}100mA/cm^2$. In conclusion, Fe ions were produced in the salt during the electrodeposition by the reaction of chlorine evolved from the anode and Fe of the SUS crucible and thereby LCC system using SUS crucible showed very low current efficiencies compared with the system using the insulated alumina crucible. Anode shroud needs to be installed around the glassy carbon not to influence surrounding SUS material.

Growth Regulators Prolong Bract Longevity of Potted Bougainvillea

  • When bougainvilleas are subjected to indoor low-light conditions, flower bracts regularly abscise. This study elucidates the effects of plant growth regulators on bract longevity of potted bougainvillea. Potted 'Taipei Red' bougainvillea in four different bract development stages were treated with 1-MCP (1-methylcyclopropene), NAA (1-naphthaleneacetic acid), SNA (sodium salt of naphthaleneacetic acid), IBA (indolebutyric acid), BA (6-benzylaminopurine), $KH_2PO_4$ (potassium dihydrogen phosphate), Put (diamine putrescine), SA (salicylic acid), or STS (silver thiosulfate) and were moved to indoor low-light conditions after treatments. Experimental results indicate that 1-MCP, NAA, SNA, BA, Put, and SA prolonged bract longevity, and this effect increased as bract stage increased. The effect of STS was significant in early bract stages and decreased as bract stages increased. Additionally, 1-MCP, NAA, SNA, BA, Put, SA, and STS treatment significantly reduced endogenous ACC (1-aminocyclopropene-1-carboxylate) content and ACC oxidase activity, suggesting that the inhibition of ethylene production was achieved via physiological metabolism. However, treatment with IBA or $KH_2PO_4$ had no effect on the bract longevity at any stage. In the combined chemical treatments, NAA + STS or NAA + SA were effectively for prolonging bract longevity and contained less protein or chlorophyll degradation, decrease ACC oxidase or ethylene production than the control. In conclusion, we propose that combined chemical treatment significantly prolonged the bract longevity and more effectively than single chemical treatment at any stage.

  • Most reinforcements in concrete are constructed by steel. Corrosion of reinforcement is the main cause of damage and early failure of reinforced concrete structures. The corrosion is mainly professed by the chloride ingress. In general, chloride in concrete can be discriminated by two components, total chloride and fire chloride. This paper provides a testing method on the coefficient of chloride diffusion in concrete and the relationship between total chloride and free chloride in concrete for the composition of predicting model on diffusion rate of chloride. In order to complete this predicting model, this study will use chloride penetration characteristic, diffusion coefficient and experiment of color change on silver nitrate solution. This predicting model is going to help that grasp special quality on salt content inclusion of concrete structure that is exposed in chloride environment. Accurate predicting model can be effectively used not only in selecting of repair time but also in preventing from various deteriorations.

Determination of Ag(Ⅰ) Ion at a Modified Carbon Paste Electrode Containing N,N'-Diphenyl Oxamide

  • New approach for the determination of Ag(I) ion was performed by using a carbon paste electrode (CPE) containing N,N'-Diphenyl oxamide (DPO) with anodic stripping voltammetry. The CMEs have been prepared by making carbon paste mixtures containing an appropriate amount of DPO salt coated onto graphite particles to analyze trace metal ions via complexation followed by stripping voltammetry. Various experimental parameters affecting the response, such as pH, deposition time, temperature, and electrode composition, were carefully optimized. Using differential pulse anodic stripping voltammetry, the logarithmic linear response range for the Ag(I) ion was 1.0 × $10^{-7}$ - 5.0 × $10^{-9}$ M at the deposition time of 10 min, with the detection limit was 7.0 × $10^{-10}$ M. The detection limit adopted from anodic stripping differential pulse voltammetry was 7.0 × $10^{-10}$ M for silver and the relative standard deviation was ± 3.2% at a 5.0 × $10^{-8}$ M of Ag(I) ion (n = 7). The proposed electrode shows a very good selectivity for Ag(I) in a standard solution containing several metals at optimized conditions.

  • In order to secure competitiveness of the home meal replacement (HMR) industry from a long-term perspective, development of packaging and processing technologies must be achieved. The development of technology that can secure freshness, nutrition, and taste but secures the shelf life is the key to the future growth of HMR. The future of HMR can be considered in terms of nutrition, environment and safety. From the nutritional point of view, it is expected that development of healthy HMR such as low-salt, low-sugar, low-fat, and high fiber and premium HMR with functional ingredient enhanced, and personalized HMR for silver, infant and patients will be done. And it is expected that development of HMR utilizing environmentally friendly food or local food, development of energy reduction and environmentally friendly disinfection technology, development of environmentally friendly packaging material, and providing information on HMR preparation using QR code and RFID from the environmental and safe point of view.

Effect of Additives on Preparation of Silver Chloride Nanoparticles using AOT-Based W/O Microemulsions (AOT W/O 마이크로에멀젼을 이용한 AgCl 나노입자 제조에서 첨가제의 영향)

  • Effect of additives such as NP series nonionic surfactant and cosurfactant on AgCl nanoparticles was investigated where nanoparticles were prepared using two different types of water-in-oil (W/O) microemulsions containing silver nitrate and sodium chloride, respectively. Phase behavior experiments showed that the region of one phase W/O microemulsion was found to be broadened with an increase in the ethylene oxide length of a nonionic surfactant mainly due to an increase in hydrophilic nature of a surfactant. Photomicrographs obtained by transmission electron microscopy indicated that an increase in ethylene oxide length of a nonionic surfactant results in both increases in particle size and size distribution. Phase behavior experiments for the systems containing AOT surfactant, isooctane and aqueous solution of an inorganic salt showed that addition of a cosurfactant caused a shrinkage in phase region of one phase W/O microemulsion, especially water contents contained in W/O microemulsion with an increase in the chain length or the concentration of a cosurfactant used. Photomicrographs obtained by transmission electron microscopy indicated that characteristics of AgCl nanoparticles produced were dependent both on the radius of spontaneous curvature and film rigidity of a microemulsion.

Expression and Promoter Analyses of Pepper CaCDPK4 (Capsicum annuum calcium dependent protein kinase 4) during Plant Defense Response to Incompatible Pathogen

  • CaCDPK4, a full-length cDNA clone encoding Capsicum annuum calcium-dependent protein kinase 4, was isolated from chili pepper (Capsicum annuum L.). Deduced amino acid sequence of CaCDPK4 shares the highest homology with tobacco NpCDPK8 and chickpea CaCDPK2 with 79% identity. Genomic blot analyses revealed that CaCDPK4 is present as a single copy in pepper genome, but it belongs to a multigene family. CaCDPK4 was highly induced when pepper plants were inoculated with an incompatible bacterial pathogen. Induced levels of CaCDPK4 transcripts were also detected in pepper leaves by the treatment of ethephon, an ethylene-inducing agent, and high-salt stress condition. The bacterial-expressed GST-CaCDPK4 protein showed to retain the autophosphorylation activity in vitro. GUS expression driven by CaCDPK4 promoter was examined in transgenic Arabidopsis containing transcriptional fusion of CaCDPK4 promoter. GUS expression under CaCDPK4 promoter was strong in the root and veins of the seedlings. GW (-1965) and D3 (-1377) promoters conferred on GUS expression in response to inoculation of an incompatible bacterial pathogen, but D4-GUS (-913) and DS-GUS (-833) did not. Taken together, our results suggest that CaCDPK4 can be implicated on signal transduction pathway of defense response against an incompatible bacterial pathogen in pepper.

  • Aspergillus sp. MK-24 was cultured at 3$0^{\circ}C$ in the medium consisting of 2% glucose, 0.2% NaNO$_3$, 0.02% $K_2$HPO$_4$, 0.02% MgSO$_4$ 7$H_2O$, 0.02% KCl, and at initial pH of 5.0. The production of the inhibitor on venom proteinase reached to the maximum in 7 days. Sodium nitrate or potassium nitrate as a nitrogen source was favorable. The production of inhibitor was not affected by the addition of most of the inorganic salt used but depressed by lead, zinc, cobalt, mercuric or silver salts.

Do Paneth Cells Regulate the Zinc Body Burden? (Zinc 대사와 관련된 Paneth 세포활성의 변화에 관한 조직화학적 연구)

  • Paneth cells have been suggested to contribute to the elimination of excess metals into the intestinal lumen. The purpose of this study wat to investigate the changes of the zinc pools in rats subjected to functional loading with zinc salt by mean of both light and electron microscopical autometallography (AMG). Wistar rats 4 were administrated with zinc chloride (20 mg/kg body weight) intraperitoneally dissolved in 1 ml distilled water. The control group received 1 ml saline IP. After further one hour the animals were transcardially perfused with 0.4% sodium sulphide dissolved in 0.1 M PB fellowed by 3% glutaraldehyde solution for 10 minutes. Pieces of ileum were frozen with solid $CO_2$ and sectioned on a cryostat. The sections $(20{\mu}m)$ were autometallographically developed. Sections selected for EM were reembedded on top of a blank Epon block, from which ultrathin sections (100 nm) were cut. The ultrathin sections were double stained with uranyl acetate (30 min) and lead citrate (5 min), then examined under electron microscope. Studies of comparable sections from control and zinc loaded animals with the AMG selenium method gave quite different results. The control animals demonstrated a weakly positive staining in the cytoplasm of the Paneth cells. In the electron microscope the AMG silver grains were found to be located in the cytoplasm, while the electron dense secretary granules and other cell organelles were void of staining. Few AMG grains were located at the apical surface of the Paneth cells. In sections from zinc loaded rats, the AMG grains were seen in abundance in the lumen of the Lieberkuhn crypts at light microscopic levels. At EM levels the zinc revealing silver grains were located in the cytoplasm as in the controls, but much more AMG grains were shifted into the secretary granules. Furthermore, profound AMG grains were found in the lumen of the crypts and surrounding vessels. And a few grains were seen in the endothelium. The AMG technique demonstrated a pattern of AMG grains in the Paneth cells that strongly suggests a transport of zinc ions through these cells.

Comparison of Statistical Methods for Optimization of Salts in Medium for Production of Carboxymethylcellulase of Bacillus amyloliquefaciens DL-3 by a Recombinant E. coli JM109/DL-3 (Bacillus amyloliquefaciens DL-3의 carboxymethylcellulase를 재조합 균주 E. coli JM109/DL-3에서 생산하는 배지의 염 농도를 최적화하기 위한 통계학적 실험 방법의 비교)

  • The optimal concentrations of salts in medium for cell growth and the production of carboxymethylcellulase (CMCase) by a recombinant E. coli JM109/DL-3 were established using two statistical methods: orthogonal array method (OAM) and response surface method (RSM). The analysis of variance (ANOVA) of data based on OAM indicated that $K_2HPO_4$ gave maximum sum of square (S) and percentage contribution (P) for cell growth as well as production of CMCase. The optimal concentrations of $K_2HPO_4$, NaCl, $MgSO_4{\cdot}7H_2O$, and $(NH_4)_2SO_4$ in medium for cell growth extracted by Qualitek-4 (W32b) Software were 10.0, 1.0, 0.2, and 0.6 g/l, respectively, whereas those for the production of CMCase by E. coli JM109/DL-3 were 5.0, 1.0, 0.4, and 0.6 g/l. The analysis of variance (ANOVA) resulting from RSM indicated that a highly significant salt for cell growth was $K_2HPO_4$ ("probe>F" less than 0.0001), whereas $K_2HPO_4$ and $MgSO_4{\cdot}7H_2O$ were significant for the production of CMCase. The optimal concentrations of $K_2HPO_4$, NaCl, $MgSO_4{\cdot}7H_2O$, and $(NH_4)_2SO_4$ for cell growth extracted by Design Expert Software were 7.44, 1.08, 0.22, and 0.88 g/l, respectively, whereas those for production of CMCase were 5.84, 0.69, 0.28, and 0.54 g/l. The optimal concentrations of salts and their influences on cell growth and production of CMCase extracted by OAM were almost the same as those by RSM. Production of CMCase by a recombinant E. coli JM109/DL-3 under optimized concentration of salts was 1.93 times higher than that by Bacillus amyloliquifaciens DL-3.
