Osteoporosis is characterized by bone loss and mobidity with osteoporotic fracture. This study was performed to evaluate the effect of on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporsis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with DBE, extract of Dioscorea batatas, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of DBE. Osteoporetic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns. DBE showed inhibitory effect on bone loss in osteoporetic condition, and reduced the increase of ALP activity and osteocalcin level in serum, and reduced the increase of OH-proline level in urine. But, DBE had no effect on cell proliferation and ALP activity in rat calvarial cell culture.
Purpose: Hypoestrogenism caused by ovariectomy, disease, or menopause is associated with increased obesity in women. Altered fat distribution and weight gain are consequences of menopausal hypoestrogenism, but the mechanisms responsible are not completely known. This study examined the effect of estrogen on obesity in ovariectomized rats. Method: The groups of female rats were 4 weeks post ovariectomy (OVX) or, 4 weeks post-sham operation (SHAM), and 2 weeks post ovariectomy followed by 2 weeks replacement with estradiol benzoate (ER-$16{\mu}g$/kg, subq, qd). Serum ghrelin level was measured by radioimmunoassay (RIA). The expression of adrenergic receptors in adipose tissue was measured by Western blotting assay. Result: OVX significantly increased body weight, serum cholesterol. Two weeks estrogen replacement reduced body weight accompanied by the increment of serum ghrelin and the reduction of the receptor ratio of adrenergic ${\alpha}_{2A}/{\beta}_1$, and ${\alpha}_{2A}/{\beta}_3$. Conclusion: We provide evidence that estrogen reduces obesity through the altered receptor ratio of adrenergic ${\alpha}_{2A}/{\beta}_1$, and ${\alpha}_{2A}/{\beta}_3$ in adipose tissue in ovariectomized rats.
Objectives: This study was conducted to assess the effects of Hwanggimacmundong-tang extract (HM) on obese rats with estrogen deficiency. Methods: The experiments were performed with ovariectomized rats as estrogen-deficient obesity model. Rats were grouped NC (Normal Control Group; sham operation group), OC (Obesity Control Group; estrogen-deficient group), HML (20 mg/kg/day), HMH (100 mg/kg/day). HM was administered orally for six weeks. Body weights and serum lipid level were measured, and real-time PCR was performed to estimate the effect of HM on gene expression in liver. Results: HM decreased the total cholesterol and triglyceride in serum. And HM increased leptin, CPT1 gene expression in liver tissue of obese rats with estrogen deficiency. However HM decreased $PPAR{\gamma}$, $PGC-1{\alpha}$, HMGCR, CYP8B1, LPL, ACAT1, ACAT2, ApoB, ACOX gene expression in liver tissue of obese rats with estrogen deficiency. Conclusions: It is concluded that HM reduced total cholesterol and triglyceride in serum, and regulate gene expression related to lipid metabolism. These results indicate Hwanggimacmundong-tang that might have potential for treatment of obesity and complications during the menopause caused by estrogen-deficiency.
본 연구는 흰쥐의 난소제거로 유발한 골다공증에 대한 흥화씨(Carhamus inctorius L)의 투여 효과를 알아보기 위하여 혈청내 호르몬과 골소주의 변화를 관찰하였다 실험동물은 4개월 령의 흰쥐를 난소절제를 실시하여 골다공증을 유발 시킨 후 실험에 이용하였으며 30일간 격일 간격으로 0.03g/kg의 용량을 투여 하였다. 혈청 내에서 Insulin-like Growth Factors, Insulin-like Growth Factor binding protein-3 (IGFBP-3), Estrogen, Bone-specific alkaline phosphotase, Calcium, and Phospotase를 매 10일 간격으로 측정하였으며 비골의 골간을 채취하여 조직 형태학적인 검사를 실시하였고 체중에 대한 대퇴골 무게를 측정하였다. 홍화씨 투여 10일과 20일에서는 혈청내 IGF-I, IGF-II그리고 IGFBP-3의 변화는 대조군에 비하여 유의성 있는 변화를 관찰할 수 없었으나 홍화씨 투여 30일에 있어서는 IGF-I, IGF-II그리고 IGFBP-3의 변화가 대조군에 비하여 현저히 높은 유의서 있는 변화를 관찰할 수 있었다(p<0.05). Bone alkaline phosphatase(BALP)에 있어서도 홍화씨 투여 30일 에 가장 많은 변화가 있었으나 estrogen과 체중에 대한 대퇴골의 무게에 있어서는 유의성 있는 변화를 관찰하지 못했다. 오히려 이시기에 난소를 절제하지 않는 대조군의 estrogen치가 높게 나타났다. 난소절제로 골다공증을 유발시킨 흰쥐에 있어서 흥화씨의 투여는 혈청내 IGFs, IGFBP-3 and BALP을 높임으로서 골다공증 치료에 효과가 있는 것으로 사료됩니다.
Objective : It is known that Pueriaria lobata has an anti-osteoporetic effect, anti-cancer effect, anti-pyretic effect, and anti-diabetic effect. The aim of this study was to evaluate anti-obesity effect of Pueriaria lobata extract (PLE), and elucidate the effect of it on gene expression related to lipid metabolism. Method : The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity model. They were grouped NC (normal control), OC (estrogen-deficient control), PLH (100mg/kg of PLE), PLL (20mg/kg). PLE was orally administered for 6 weeks. Body weights and serum lipid level were estimated, and real-time PCR was performed to investigate the effect of PLE on gene expression in liver. Results : PLE decreased the body weight and serum cholesterol and triglyceride, but increased HDL-cholesterol. And PLE increased leptin, CYP27, CPT1, CYP8B1, ACAT2, LDLR, and SCD1, but reduced $PPAR{\gamma}$, PGC1A, HMG-CoA-R, ACAT1, SCD1, and APoB gene expression in liver tissue of rat with estrogen-deficient obesity. Conclusion : It is concluded that Pueriaria lobata reduced body weight, and its effect was expressed by regulation of gene expression related to lipid metabolism in rats with estrogen-deficient obesity.
This study was intended to investigate the effects of nutritional, physical and environmental conditions on the bone status of 102 pre and post menopausal women living in Ulsan area. The results of this study are summarized as follows : when subjects were classified into two groups by bone status, there were significant differences in age, calcium index, serum estrogen and physical activity between two groups. Average daily calcium intake of subjects was 682.9 (149.2)mg, which is almost same as RDA, but 41.1% of the subjects consumed calcium below RDA. Calcium index(p<0.05), serum estrogen(p<0.001), physical activity(p<0.001) of poor bone group were significantly lower than those of good bone group. Although there was not a significant difference between bone status and other influential factors, such as family type, skip meal, alcohol drinking, smoking and BMI, each has been shown to have an effect, if slight, on the bone status.
The role of estrogen in calcium metabolism has not been well documented. To further investigate the effects of endogenous estrogen on the calcium regulating hormones in women, we correlated the changes of estrogen level with those of calcium regulating hormones during ovulation induction in 12 hypogonadal and 8 normally menstruating women. During ovulation induction, the serum leveles of estradiol rose from 40.0 to 831.0pg/ml. There were no significant changes in the serum leveles of total calcium, inorganic phosphorus, parathyroid hormone, calcitonin. However, 1, $25-(OH)_2D_3$ rose significantly from 31.0 to 47.2pg.ml as the endogenous $E_2$ increaed (p<0.005).

Background: Cholelithiasis is associated in 54%-98% of patients with carcinoma of the gallbladder, and a high incidence among females suggests a role of female hormones in the etiology of the disease. Cytochrome $P450C17{\alpha}$ (CYP-17) is a key enzyme involved in estrogen metabolism and polymorphisms in CYP-17 are associated with altered serum levels of estrogens. Thus, we investigated whether the CYP-17 MspA1 gene polymorphism might impact on risk of gall bladder cancers or gallstones, as well as to determine if this gene polymorphism might be linked with estrogen serum levels and lipid profile among the North Indian gall bladder cancer or gallstone patients. Materials and Methods: CYP-17 gene polymorphisms (MspA1) were genotyped with PCR-RFLP in cancer patients (n=96), stone patients (n=102), cancer + stone patients (n=52) and age/sex matched control subjects (n= 256). Lipid profile was estimated using a commercial kit and serum estrogen was measured using ELISA. Results: The majority of the patients in all groups were females. The lipid profile and estrogen level were significantly higher among the study as compared to control groups. The frequency of mutant allele A2 of CYP17 MspA1 gene polymorphism was higher among cancer (OR=5.13, 95% CI+3.10-8.51, p=0.0001), stone (OR=5.69, 95%CI=3.46-9.37, p=0.0001) and cancer + stone (OR=3.54, 95%CI=1.90-6.60, p=0.0001) when compared with the control group. However there was no significant association between genotypes of CYP17 MspA1 gene polymorphism and circulating serum level of estrogen and lipid profile. Conclusions: A higher frequency of mutant genotype A1A2 as well as mutant allele A2 of CYP-17 gene polymorphism is significantly associated with risk of gallbladder cancer and stones. Elevated levels of estrogen and an altered lipid profile can be used as predictors ofgall bladder stones and cancer in post menopausal females in India.
The purpose of this study was to investigate whether the extracts of Daucus carota L. could lead to a beneficial outcomes on serum lipid and antioxidative enzyme activity in ovariectomized rats. Sprague-Dawley rats were randomly assigned to one control and three diet groups; carrot seed, carrot root and estrogen after ovariectomy operation. Experimental diets were fed for 8 weeks. The GOT activity was decreased in the carrot extracts treated group than in the control group. The carrot seed extracts treated group showed the lowest SOD and catalase activities compare to other groups. Serum total cholesterol and LDL cholesterol decreased in the carrot seed extracts group than in the control group. From these results, it suggest that carrot seed extracts positively influenced on serum lipid and antioxidative enzyme activity.
This study was designed to investigate the effects of dietary calcium. serum estrogen level and physical activity on the bone status of 116 healthy elderly women living in urban area. Current calcium intake was assessed by convenient method(refered to as Ca intake) and calcium containing food frequency method(refered to as Ca index) Daily activity record was used for the estimation of physical activity level, and serum estrogen level was measured from fasting blood of subjects. The rate of bone resorption was evaluated by the determination of hydroxyproline(Hpr) in fasting urine with correction for creatinine excretion. The results of this study are summarized as follows : 1) Average daily Ca intake of subjects was 621.4$\pm$155.8mg, which is above the Korean recommended dietary allowances. However 44.8% of the subjects consumed Ca below RDA level. Ca index score was significantly correlated with the bone status(P<0.05), Ca intake did not show significant correlation with the bone status although a positive trend of influence was evident. 2) Average serum estrogen level of subjects was 18.7$\pm$9.8pg Contrary to our anticipation. estrogen level did not show any significant relation to age and bone status. 3) Daily physical activity was classified into four categories by activity intensity : sedentary. moderate, active and severe. The average physical activity of subjects belong to moderate level. and the bone status was significantly related to the physical activity(P<0.01) 4) Among other influential factors such as age, pocket-money. family type. drinking, smoking and BMI, there was a significant difference between bone status and BMI(P<0.05). 5) Multiple regression analysis of variables showed that physical activity has greater effect than other variables when the entire subjects were taken into account. However. eliminating the subjects whose bone status rated as excellent(Hpr/cr<0.009), Ca index showed higher correlation than physical activity. These results have demonstrated that dietary calcium intake is the primary important factor for keeping good bone health and that bone status of subjects with a sufficient calcium intake is affected by various factors such as physical activity, age, smoking. BMI and others.
