일본도시근교낙농장(日本都市近郊酪農場) 유우(乳牛)의 우백혈병감염(牛白血病感染)에 대한 혈청학적(血淸學的) 조사연구(調査硏究) (A serological survey of bovine leukemia virus infection in dairy cattle in the suburban farming area of Japan)

  • 1983년(年)부터 1985년도(年度)까지 사이에 일본(日本) Saitama현(縣) 각소도시근교(各小都市近郊)의 낙농장(酪農場)에서 사육(飼育)되는 2,407두(頭)의 유우(乳牛)를 대상(對象)으로 BLV(bovine leukemia virus) 감염여부(感染與否)를 혈청학적검사(血淸學的檢査)(agar-gel immunodiffusion test)를 실시(實施)한 결과(結果) 평균양성율(平均陽性率)은 4.9%이었고, 연령(年齡)이 증가(增加)함에 따라 2.6%에서 부터 9.8%까지 양성율(陽性率)이 있으며, 동일지역(同一地域)에서도 조사낙농장별(調査酪農場別)로 양성율(陽性率)은 큰 차(差)가 있었다. 본조사연구(本調査硏究)의 결과(結果)를 BLV감염(感染)에 대한 수의역학적(獸醫疫學的) 기초자료(基礎資料)로 제시(提示)하는 바이다.

소 브루셀라병 동시 다두 발생 농장의 발생 요인 조사 (Investigation of occurrence factors on brucellosis-outbreak farm in Korea)

  • From the end of July 2012, several cases of abortion have been happened at the Korean indigenous cattle farm with 124 heads in Chungbuk province, Korea. Serological tests such as Rose-bengal test (RBT) and standard tube agglutination test (STAT) have been performed according to the standard official protocols of bovine brucellosis and 41 cattle turned out to be brucellosis-positive simultaneously. To find out the main factors of brucellosis outbreaks and spreads, additional serological, etiological and molecular investigation were applied. Totally, 11 B. abortus were isolated from 10 cattle's specimens including lymph-nodes and/or testis, and drinking water in cowhouse. In genotyping by multi-locus VNTR assay (MLVA) using 17 loci markers, the present B. abortus isolates were shown all the same pattern, D1 genotype, which has been reported in Gyeonggi and Gangwon province, Korea. These results suggest that the input of brucellosis might come from neighboring farms directly or indirectly, even if by unknown factor and expansion within farm would accelerate by materials related with aborting cows.

우리나라 벼 흰빛잎마름병균 (Xanthomonas campestris pv. oryzae)의 혈청학적 분류 및 진단 (Serovars of Xanthomonas campetris pv. oryzae Collected from Korea and Serological Diagnosis of Bacterial leaf Blight)

  • 벼 흰빛잎마름병균주 Q7472와 Q7502의 항혈청을 사용하여 우리나라에서 수집한 벼 흰빛잎마름병균의 혈청형을 한천gel내 확산법에 의해 분류한 결과, 71균주중 65균주가 혈정형 A형, 5균주가 B-I, 1균주가 B-II형에 속하였다. 혈청형A에 속하는 균주는 $0.1-1\%$$CaCl_2$용액에서 자기의집이 거의 일어나지 않았으나 혈청형 B-I과 B-II에 속하는 균주는 심한 자기의집이 일어나 혈청형과 자기의집 사이에는 밀접한 관계가 있었다. 흰빛잎마름병균은 혈청학적 방법에 의해 쉽게 검색될 뿐만 아니라 한천gel내 확산법에 의해 벼 세균성줄무늬병균인 X.campestris pv. olyzicola의 부생세균인 E. herbicola와도 쉽게 구별되었다. 흰빛잎마름병균 검출은 병반을 NaCl용액에 넣은 후에 착압하거나 $100^{\circ}C$에서 1시간 열처리 후 착압한 즙액을 한천gel내 확산법에 의해 항혈청과 반응시킴으로써 가능하였으며, 병반이 적을때는 PSA배지에서 배양한 후 사용하면 효과적이었다.

  • 류마티스 관절염은 주로 관절의 활막 세포의 증식, 병리학적 면역 반응과 관절의 진행성 파괴를 특징으로 하는 만성 염증성 질환이다. 이 질환은 중요한 사회적 건강 문제로 대두된다. 이 논문을 통해 분자수준에서 변화와 류마티스 관절염의 진단 또는 질병의 진행에 대한 발병 기전을 새로운 관점에서 발병기전을 제공한다. 또한 조기 진단과 류마티스 관절염의 예후에 도움이 될 것으로 보인다. 새로운 혈청학적 및 면역학적 바이오마커에 초점을 맞추고 있다. 따라서 넓게 질병 진행의 위험 환자를 식별하는 혈청학적, 생체 면역학적 요인 등을 규명하고 적용시키는 것을 도출할 수 있다. 진단 검사 의학을 기반으로 하는 증거는 환자에게 최선의 결과를 제공할 수 있다. 마지막으로. 최근의 연구 데이터는 이 접근을 통해서 치료를 최적화하기 위해 조기 진단 및 치료에 도움이 되는 새로운 접근 방식으로 궁극적인 유용성을 정립하는데 도움이 될 것으로 사료된다.

일반시설에서 사육되는 마우스의 품질향상을 위한 기초조사 연구 (Survey on environmental condition and health state of laboratory mouse in conventional facility)

  • For the improvement of quality control of laboratory mouse, we investigated the environmental condition, histopathological findings and serological test using ELISA to mouse hepatitis virus(MHV), Mycoplasma pulmonis(MP), Clostridium piliforme(TZ) and Sendai virus (HVJ) of ICR, C57BL/6, CBA and C3H/He mice that were supplied from conventional laboratory animal facility. 1. The ammonia concentration of facility was below the recommended concentration, 15ppm, by the KNIH, and the room temperature($21{\sim}23^{\circ}C$) and relative humidity(40~60%) was optimum range recommend by the Ministry of Health and Welfare, respectively. 2. The incidence rate of inapparent disease was 86.6% and the major findings in the liver were vacuolar degeneration with nucleic pleomorphism. The lung was shown the thickening of alveolar wall and interstitial pneumonia with congestion. The kidney and spleen were observed the mild congestion and extramedullary hematopoiesis, respectively. 3. The positive reaction rates against MHV and MP in serological test was 97.9% and 37.5%, respectively but HVJ and TZ were negative. These results suggest that laboratory mice could be infected with MHV and MP under conventional environments. Therefore we recommend to select thoroughly inapparent infected mice and to convert conventional system into SPF facility as soon as possible.

추백리-가금티푸스의 혈청학적 모니터링 계획수립을 위한 표본크기 (Determination of sample size to serological surveillance plan for pullorum disease and fowl typhoid)

  • The objective of this study was to determine appropriate sample size that simulated different assumptions for diagnostic test characteristics and true prevalences when designing serological surveillance plan for pullorum disease and fowl typhoid in domestic poultry production. The number of flocks and total number of chickens to be sampled was obtained to provide 95% confidence of detecting at least one infected flock, taking imperfect diagnostic tests into account. Due to lack of reliable data, within infected flock prevalence (WFP) was assumed to follow minimum 1%, most likely 5% and maximum 9% and true flock prevalence of 0.1%, 0.5% and 1% in order. Sensitivity were modeled using the Pert distribution: minimum 75%, most likely 80% and maximum 90% for plate agglutination test and 80%, 85%, and 90% for ELISA test. Similarly, the specificity was modeled 85%, 90%, 95% for plate agglutination test and 90%, 95%, 99% for ELISA test. In accordance with the current regulation, flock-level test characteristics calculated assuming that 30 samples are taken from per flock. The model showed that the current 112,000 annual number of testing plan which is based on random selection of flocks is far beyond the sample size estimated in this study. The sample size was further reduced with increased sensitivity and specificity of the test and decreased WFP. The effect of increasing samples per flock on total sample size to be sampled and optimal combination of sensitivity and specificity of the test for the purpose of the surveillance is discussed regarding cost.

Surveillance and molecular epidemiology of avian influenza viruses from birds in zoos, backyard flocks and live bird markets in Korea

  • The circulation and infection of avian influenza virus (AIV) in zoos and backyard flocks has not been systematically investigated. In the present study, we surveyed the birds including those in live bird markets (LBMs) and evaluated co-circulation of AIVs among them. Overall, 26 H9N2 AIVs and one H6N2 AIV were isolated from backyard flocks and LBMs, but no AIVs were isolated from zoo birds. Genetic analysis of the HA and NA genes indicated that most of the H9N2 AIVs showed higher similarities to AIVs circulating in domestic poultry than to those in wild birds, while the H6N2 AIV isolate from an LBM did to AIVs circulating in migratory wild birds. In serological tests, 15% (391/2619) of the collected sera tested positive for AIVs by competitive-ELISA. Among them, 34% (131/391) of the sera tested positive for AIV H9 antigen by HI test, but only one zoo sample was H9 positive. Although AIVs were not isolated from zoo birds, the serological results indicated that infection of AIVs might occur in zoos. It was also confirmed that H9N2 AIVs continue to circulate and evolve between backyard flocks and LBMs. Therefore, continuous surveillance and monitoring of these flocks should be conducted to control further epidemics.

가잠에서 분리된 새로운 미포자충 K79의 병리학적 연구 I. 미포자충 포자의 정제 및 식별 (Pathological Studies on the New Microsporidia K79 Isolated from the Silkworm, Bombyx mori L. I. Purification and Serological Discrimination of Microsporidian Spores.)

  • 미포자충 포자의 정제법을 개발하고 포자표면의 혈청학적 반응을 구명하기 위하여 수행한 시험결과를 요약하면 다음과 같다. 1. 미포자충 포자의 분리정제는 예비원심-percoll 평형밀도원심-세척의 3단계법이 우수하였고, 분리정제된 포자는 극사추출에 나쁜 영향도 받지 않았다. 2. 포자의 형태는 K79 포자가 장타원형(장단경비 2.76)이고 N. bombycis는 단타원형(장단경비 2.07)으로 양포자가 상이하였다. 3. 형광항체 간접법에 의하여 항 N. bombycis 포자혈청에 대하여는 N. bomvycis 포자만이 강한 형광을 발현하였으나 항 K79 포자의 혈청에 대하여는 10배희석구에서는 양포자 모두 강하게, 20배 이상 희석구에서는 약하게 형광을 발현하였고 K79 포자의 특이항체가 포자외각에 인정되었다.

Effect of a Gluten Free Diet on Hepatitis B Surface Antibody Concentration in Previously Immunized Pediatric Celiac Patients

  • Purpose: To evaluate the effect of gluten-free diet (GFD) on hepatitis B surface antibody (HBsAb) concentrations among previously immunized pediatric celiac disease (CD) subjects. Methods: We retrospectively evaluated pediatric CD subjects in serological remission who were previously immunized for hepatitis B virus as infants. The temporal relationship between HBsAb concentration, the amount of time on a GFD, and age were evaluated. Results: Overall, 373 CD subjects were analyzed: 156 with HBsAb sampled prior to GFD initiation and 217 after initiation of a GFD and in serological remission. Median age at HBsAb concentration measurement for those before and after GFD initiation was 5.3 years (interquartile range [IQR], 3.1-9.2 years) and 7.6 years (IQR, 5.4-10.9 years), respectively (p<0.001). There was no sex difference between the groups. The median time of HBsAb measurement was 2 months (IQR, 0-5.7 months) before and 12.8 months (IQR, 5.3-30.3 months) after initiation of GFD. The HBsAb concentration was low in 79 (50.6%) and 121 (55.7%) subjects before and after GFD initiation, respectively (p=0.350). Age was inversely associated with low HBsAb concentrations. Neither being on a GFD nor sex was associated with low HBsAb concentrations. Conclusion: Adherence to a GFD does not affect HBsAb concentration in children with CD. Age is inversely associated with HBsAb concentration.

International travel of Korean children and Dengue fever: A single institutional analysis

  • Purpose: Dengue fever occurs in many popular tourist destinations and is increasingly imported by returning travelers in Korea. Since Korea is not an endemic country for dengue fever, pediatricians do not usually suspect dengue fever in febrile children even with typical presentation and exposure history. This study was performed to describe the international travel experiences and dengue fever in Korean children. Methods: Travel histories were collected based on questionnaires completed by all patients' guardians who visited the pediatric infectious diseases clinic at Samsung Medical Center from January 2008 to December 2008. For patients who were suspected of dengue fever, a serological test was performed. Results: Five hundred and seventeen children visited the pediatric infectious diseases clinic for the first time during this period. About 30% of patients who responded to the questionnaire (101/339) had experienced international travel within the last 2 years. Four patients were diagnosed with dengue fever by serological test. Conclusion: Increasing numbers of Korean children visit dengue endemic areas and they may return home with dengue fever. Dengue fever should be suspected in patients who have a travel history to endemic areas.
