  • This paper is on a study for discrimination on relative sequence as a most actively discussed topic in forensic document fields. This paper describes the application of the visual spectral comparator and infinite focus microscope as observation methods for overlapping region of printing and writing lines. As a result, we could categorize overlapping region images and identify the sequence of printing and writing lines by various inks.

  • This paper presents the rapid and accurate algorithm for fault discrimination in transmission lines. When faults occur in transmission lines, fault discrimination is very important. If high impedance faults occur in transmission lines, it cannot be detected by overcurrent relays. The method using current and voltage cannot discriminate high impedance fault. Because of this reason this paper uses voltage and zero sequence current, and the proposed algorithm uses fuzzy logic method. This algorithm uses voltage and zero sequence current per period in case of faults. Single line ground fault and three-phase fault can be detective using voltage. Two-line ground fault and line to line fault and high impedance can be detected using zero sequence current. To prove the performance of the algorithm, it test algorithm with signal obtained from ATPDraw simulation.

  • According to the use of distribution automation systems, the function to find or to search a fault phase is necessary for automatic switches in a distribution substation. In this paper, two algorithms are developed to fine the fault circuit and the fault phase for the automatic switches in substation with ungrounded power system. One is the fault circuit searching method using the zero sequence voltage at the bus and zero sequence current of circuit current and the other is to find the fault phase using the line voltage and zero sequence current. The developed algorithms are tested in the case study simulations. An ungrounded power system is modeled by EMTP as a case study system. The developed algorithms are tested in the case study simulations and each shows correct results.

  • Objectives : The application of polymerase chain reaction (PCR) for the discrimination of the herbal medicine Leonuri Herba (Leonurus japonicus) was evaluated by the comparison of the DNA sequence with Lamiaceae herbal medicine. Method : Genetic analysis showed that phylogenetic tree and comparing sequences through the DNA analysis of rbcL (ribulose-1, 5-bisphosphatecarboxylase) region and trnL-F (tRNA-Leu, trnL-trnF intergeni cspacer, and tRNA-Phe) region of chloroplast DNA from six Lamiaceae sold in market. And we developed IMCF and IMCR primers in order to distinction Leonuri Herba in six Lamiaceae using rbcL and trnL-F sequences. Results : Genetic analysis showed that six Lamiaceae showed individual group on phylogenetic tree. PCR amplification product of Leonuri Herba and another five Lamiaceae were developed for amplification of a 281 bp sequence and the specific PCR amplification of a 460 bp sequence that was exclusive to Leonuri Herba was designed using IMCF and IMCR primers. Conclusion : PCR analysis based on the chloroplast DNA sequences allows the discrimination of Leonuri Herba-based medicine.

Transfer RNA Acceptor Stem Determinants for Specific Aminoacylation by Class II Aminoacyl-tRNA Synthetases

  • A critical step in the faithful translation of genetic information is specific tRNA recognition by aminoacyl-tRNA synthetases. These enzymes catalyze the covalent attachment of particular amino acids to the terminal adenosine of cognate tRNA substrates. In general, there is one synthetase for each of the twenty amino acids and each enzyme must discriminate against all of the cellular tRNAs that are specific for the nineteen noncognate amino acids. Primary sequence information combined with structural data have resulted in the division of the twenty synthetases into two classes. In recent years, several high-resolution co-crystal structures along with biochemical data have led to an increased understanding of tRNA recognition by synthetases of both classes. The anticodon sequence and the amino acid acceptor stem are the most common locations for critical recognition elements. This review will focus on acceptor stem discrimination by class II synthetases. In particular, the results of in vitro aminoacylation assays and site-directed and atomic group mutagenesis studies will be discussed. These studies have revealed that even subtle atomic determinants can provide signals for specific tRNA aminoacylation.

Analysis of Genetic Polymorphism of Korean Ginseng Cultivars and Foreign Accessions using SSR Markers (SSR 마커를 이용한 국내산 인삼 품종 및 국외 수집종의 유전적 다양성 분석)

  • In this study, simple sequence repeat (SSR) analyses were utilized for evaluation of genetic diversity and discrimination of 17 accessions. Five cultivars, which were developed from Korea, and 12 foreign accessions, which were collected from China, Japan, Russia and USA, were evaluated by nine markers out of 22 SSR markers. A total of 39 alleles were detected, ranging from 2 to 8, with an average of 4.3 alleles per locus. The expected heterozygosity and PIC values were 0.627 and 0.553, with a range from 0.21 (GB-PG-078) to 0.76 (GB-PG-142) and from 0.19 (GB-PG-078) to 0.70 (GB-PG-142), respectively. Four makers out of nine SSR markers, GB-PG-026, GB-PG-043, GB-PG-142 and GB-PG-177, were selected as key factors for discrimination of Korean ginseng cultivars and foreign accessions. All of Korean ginseng cultivars and foreign accessions were individually by the combination of four SSR markers. Consequently, the SSR markers developed in this study may prove useful for the evaluation of genetic diversity and discrimination of Korean ginseng cultivars and foreign accessions.

Discrimination of Korean rice varieties as revealed by DNA profiling and its relationship with genetic diversity (한국 육성 벼 품종의 DNA profiling에 의한 유전적 다양성 분석 및 품종 판별)

  • This study is to establish the varietal discrimination based on DNA profiling of different varieties of rice. We examined the genetic distance among Korean rice varieties using allele frequencies and a genetic diversity analysis with Simple Sequence Repeats (SSRs) markers. The analysis of the genetic diversity and genetic relationships of 243 Korean rice varieties was varied out using 20 SSRs markers. A total of 268 alleles were detected, ranging from 6 to 32, with an average of 13.45 alleles per locus, and and average of gene diversity (GD) of 0.556. Seven SSR markers were selected as key markers for discrimination among the Korean rice varieties. Concerning the results, 243 varieties (100%) were discriminated among by using acrylamide gel and fragment analyzer-based markers. In conclusion, this study provides useful basic data that can be utilized concerning Korean rice varieties breeding and development. In addition, we will have to manage and conserve as a valuable genetic resource, without losing the diversity of Korean rice varieties.

Determination and Application of Combined Genotype of Simple Sequence Repeats (SSR) DNA Marker for Cultivars of Cymbidium goeringii (춘란(Cymbidium goeringii) 품종에 대한 Simple Sequence Repeats (SSR) DNA 마커의 복합 유전자형 결정과 적용)

  • Cymbidium goeringii is one of the most important and popular species in the orchid family in north-east Asia. In the present study, we prepared multiplex PCR system, and used it for the genotyping of eight simple sequence repeats (SSR) markers (CG409, CG415, CG709, CG722, CG787, CG1023, CG1210, and CG1281) in subject with 40 samples of cultivated varieties. All the analyzed samples showed different combined genotypes. The average combined power of discrimination was very high value of $7.14{\times}10^{-10}$, and observed heterozygosity (Ho = 0.466) was similar with two wild populations of C. goeringii, which may indicate no or little occurrence of genetic change after collection from wild habitats. The present study also developed a two-dimensional barcode to express information of genotype results of eight SSR markers (SSR DNA ID). The discrimination power of DNA ID between two individuals will be statistically more than 99.999999%. The SSR DNA ID and two-dimensional barcode may be very usefully applied for the discrimination and maintence of cultivars of C. goeringii.

This paper presents the rapid and accurate algorithm for fault discrimination and fault location in underground transmission lines.
