Enterococcus faecalis 2B4-1 세포벽 성분 중 Polysaccharide Fraction의 종양세포 증식억제 효과 (Growth Inhibition of Polysaccharide Fraction in Cell Wall Components from Enterococcus faecalis 2B4-1 against Tumor Cell Lines)

  • This study was developed to evaluate the growth inhibition effects of cell wall components of Enterococcus faecalis 2B4-1 obtained from feces of neonates against tumor cell lines. Polysaccharide fraction (PS) shown sensitive growth inhibition effect in the cell wall components was isolated and characterized. In growth inhibition effects, residue fractin of whole cell was shown sensitive level of percent survival about 30% when administrated at ehe concentration of 100${\mu}$g/ml, and that was more effective than that of supernatant fraction against the tumor cell lines, SNU-1, 3LL, FARROW and HEC-1-B. Sensitive growth inhibition effects against SNU-1, FARROW and HEC-1-B were performed by whole cell (WC) fraction from Ent. faecalis 2B4-1. Cytoplasm fractin (CP) of WC was shown non-inhibition effect, however, the other part of WC, precipitate of disrupted cell (PD), was sensitive against the tumor cell line mentioned above. Followed by separation to peptidoglycan fraction (PG) and polysaccharide fraction (PS) were all sensitive which the latter was shown more sensitive percent survival than the former. Composed sugars of polysaccharide fraction were determined to D-glucose, L-rhamnose and D-glucosamine, and the rate fo composition was calculated to about 1:1:1 by the data of elemental analysis, IR, TLC and HPLC.

폴리이미드 용량형 습도센서의 전극 패턴에 따른 감습 특성 (Humidity Sensitive Characterization by Electrode Pattern on the Capacitive Humidity Sensor Using Polyimide)

  • Electrode pattern effects on the capacitive humidity sensor were investigated. The fabrication of the capacitive humidity sensor was formed with three steps. The bottom electrode was formed on the silicon substrate with Pt/Ti thin layer by using shadow mask and e-beam evaporator. The photo sensitive polyimide was formed on the bottom electrode by using photolithography process as a humidity sensitive thin film. The upper electrode was formed on the polyimide thin film with Pt/Ti thin layer by using e-beam evaporator and lift-off method. Three electrode patterns, such as circle, square, and triangle pattern, were used and changed the sizes to investigate the effects. The capacitances of the sensors were decreased 622 to 584 pF with the area decreament of patterns 250,000 to $196,250{\mu}m^2$. From these results, a capacitive humidity sensor with photo sensitive polyimide is expected to be applied to a high sensitive humidity sensor.

평지방막에 융합된 골격근의 single ATP-sensitive K+ channel의 이온투과성에 대한 연구 (Permeability properties of skeletal muscle ATP-sensitive K+ channels reconstituted into planar lipid bilayer)

  • Properties of unitary ATP-sensitive $K^+$ channels were studied using planar lipid bilayer technique. Vesicles were prepared from bullfrog (Rana catesbeiana) skeletal muscle. ATP-sensitive $K^+$ (K (ATP)) channels were identified by their unitary conductance and sensitivity to ATP. In the symmetrical solution containing 200mM KCI, 10mM Hepes, 1mM EGTA and pH 7.2, single K (ATP) channels showed a linear current-voltage relations with slight inward rectification. Slope conductance at reversal potential was $60.1{\pm}0.43$ pS(n=3)). Micromolar ATP reversibly inhibited the channel activity when applied to the cytoplasmic side. In the range of -50~+50 mV, the channel activity was not voltage-dependent, but the channel gating within a burst was more frequent at negative voltage range. Varying the concentrations of external/internal KCl(mM) to 40/200, 200/200, 200/100 and 200/40 shifted reversal potentials to $-30.8{\pm}2.9$(n=3), $-1.1{\pm}2.7$(n=3), 10.5 and 30.6(mV), respecrivety. These reversal potentials were close to the expected values by the Nernst equation, indicating nearly ideal selectivity for $K^+$ over $Cl^-$. Under bi-ionic conditions of 200mM external test ions and 200mM internal $K^+$, the reversal potentials for each test ion/K pair were measured. The measured reversal potentials were used for the calculation of the releative permeability of alkali cations to $K^+$ ions using the Goldman-Hodgkin-Katz equation. The permeability sequence of 5 cations relative to $K^+$ was $K^+$(1), $Rb^+$(0.49), $Cs^+$(0.27), $Na^+$(0.027) and $Li^+$(0.021). This sequence was recognized as Eisenman's selectivity sequence IV. In addition, modelling the permeation of $K^+$ ion through ATP-sensitive $K^+$ channel revealed that a 3-barrier 2-site multiple occupancy model can reasonably predict the observed current-voltage relations.

  • As an efficient controlled release system for rhBMP-2, a functional nanoparticle-hydrogel complex, incorporated with matrix metalloproteinase(MMP) sensitive peptide cross-linker, was developed and used as a bone transplant. In vivo bone formation was evaluated by soft x-ray, histology, alkaline phosphatase(ALP) activity and mineral contents analysis, based on the rat calvarial critical size defect(8mm in diameter) model. Significantly, effective bone regeneration was achieved with the rhBMP-2 loaded MMP sensitive hyaluronic acid(HA) based hydrogel-Nanoparticles(NP) complex, as compared to only MMP HA, the MMP HA-NP without rhBMP-2, or even with the rhBMP-2. These improvements included the formation pattern of bone and functional marrow, the degree of calcium quantification, and the ALP activity. These results indicate that the MMP sensitive HA with nano-particle complex can be a promising candidate for a new bone defect replacement matrix, and an enhanced rhBMP-2 scaffold.

Molecular Characterization of Pseudomonas aeruginosa Isolates Resistant to All Antimicrobial Agents, but Susceptible to Colistin, in Daegu, Korea

  • Multi-drug resistant Pseudomonas aeruginosa has been implicated in a variety of serious therapeutic problems in clinical environments. Among the 968 P. aeruginosa isolates obtained from two hospitals in Daegu, Korea, we acquired 17 isolates that were resistant to all available tested antimicrobial agents, with the exception of colistin (colistin-only sensitive). We characterized the antimicrobial susceptibilities, $metallo-{\beta}-lactamases$, and epidemiological relatedness among the colistin-only sensitive P. aeruginosa isolates. All colistin-only sensitive isolates were positive in the modified Hodge test and imipenem-EDTA synergy test, thereby indicating the production of $metallo-{\beta}-lactamases$. 11 isolates from the secondary hospital and six isolates from the tertiary teaching hospital harbored $bla_{VIM-2}$ and $bla_{IMP-1}$, respectively. The pulsed-field gel electrophoretic analysis of the SpeI-digested DNA from P. aeruginosa isolates indicated that two different clones of colistin-only sensitive P. aeruginosa originated from each hospital, and had spread within the hospital environment. Overall, colistin-only sensitive P. aeruginosa was detected in Korea for the first time, but no pan-drug resistant bacteria were identified. Nationwide surveillance is required in order to monitor the emergence of colistin-only sensitive or pan-drug resistant bacteria.

캐시를 고려한 T-트리 인덱스 구조 (Cache Sensitive T-tree Index Structure)

  • 지난 10년간 CPU의 속도는 메모리의 속도에 비해 급속한 속도로 발전하였다 그 결과 데이터 베이스 시스템을 포함한 다른 컴퓨터 응용분야에서 메모리의 접근이 병목현상을 일으키게 되었다. 메모리의 접근 속도를 줄이기 위해 캐시 메모리가 도입되었다 하지만 캐시 메모리는 원하는 데이타가 캐시에 옮겨져 있어야 메모리 접근 속도를 줄일 수 있다. 때문에 응용프로그램에서 데이타를 어떤 순서로 액세스 하느냐에 따라 캐시의 활용도가 달라지고 응용프로그램의 성능이 달라지게 된다. 이 시점에서 현재 컴퓨터에서 B+-트리가 T-트리보다 더 빠르다는 사실이 알려졌다. B+-트리가 T-트리보다 캐시를 더 효율적으로 사용하기 때문이다. 또한 B+-트리를 개선하여 캐시를 더욱 효율적으로 사용하는 CSB+-트리(Cache Sensitive B+-tree)가 제안되기도 하였다 본 논문의 목표는 T-트리가 캐시를 효율적으로 사용하도록 새로운 T-트리 구조를 개발하는 것이다. CSB+-트리와 같이 시스템의 L2 캐시를 최대한 활용하며 기존 T-트리가 가지는 장점을 가지는 새로운 CST-트리(Cache Sensitive T-트리)를 설계 개발하고, 실험을 통해 기타 다른 인덱스 구조에 비교하여 CST-트리의 우수성을 보인다.

A Novel Mobility Management Scheme for Time Sensitive Communications in 5G-TSN

  • 본 논문에서는 5G 시스템과 IEEE 시간 민감형 네트워킹(TSN)에 대해서 소개 분석하고 초저지연 네트워크를 지원하기 위하여 5G-TSN에서 시간 민감형 통신을 위한 새로운 이동성 관리 방안을 제안한다. 시간 민감형 네트워킹(TSN)은 이더넷 기반의 저지연, 높은 신뢰성, 결정형 통신을 제공할 수 있는 핵심 요소 기술로써 산업 자동화와 산업용 사물인터넷(IIoT) 분야의 유망한 미래 기술이다. TSN 지원 단말이 TSN 비서비스 영역에서 TSN 서비스 영역으로 이동했을 때, 단말은 관련 이동성 관리가 적절히 수행되지 못하기 때문에 신속하게 TSN 서비스를 받을 수 없다. TSN 서비스 영역의 이동성 상황의 경우, 제안하는 방안은 기존 5G 이동통신망에서의 방안과 비교하여 TSN 서비스 및 초저지연 통신을 즉시 제공받기 위해서 TSN 서비스 사용 가능성을 네트워크에 알리고, 초기 등록을 동작시킨다.

CNG 용기용밸브의 압력방출장치 성능에 관한 연구 (A Study on Performance of Pressure Relief Devices of CNG Cylinder Valves)

  • 이 연구는 천연가스버스에 장착된 용기가 국부화염에 집중적으로 노출되었을 때나 외기온도가 높은 여름철 충전과정에서 과 충전되었을 때 용기파열을 방지하기 위한 방안으로 압력방출장치의 성능을 확인하고 확대 적용할 목적으로 의도되었다. 열사이클링 실험결과, $-40^{\circ}C{\sim}82^{\circ}C$ 인증기준에서 가스누출요건을 모두 충족하였지만 $-45^{\circ}C{\sim}135^{\circ}C$의 가속조건에서는 3개사 시험편중 2개사 시험편의 오링이 손상되었다. 또한 온도감응형 PRD의 평균작동시간은 액체봉입식이 1분 39초가 걸렸고, 가용합금식이 2분 31초가 소요되어 액체봉입식이 가용합금식보다 약 1분 정도 빠름을 나타내었다. 또한 압력감응형 PRD의 작동압력은 가속조건에서 약 32.1 MPa을 나타내었고 인증기준에서의 압력감응형 PRD의 파열압력은 $30.7{\sim}32.1\;MPa$를 기록하였다. 이상의 실험을 통해서 압력방출장치의 성능은 온도감응형 PRD의 경우, 액체봉입식이 가용합금식보다 화염에 더 효과적이며, 압력 감응형 PRD의 파열압력은 외부온도 및 반복가압이 파열판의 작동압력에 크게 영향을 미치지 않음을 확인할 수 있었다.

Eco-physiological Responses of Two Populus deltoides Clones to Ozone

  • One-year-old cottonwood (Populus deltoides Bartr.) clones, which were classified as sensitive or tolerant, were exposed to 150 n1/1 ozone (O$_3$) over 8 days for 8 hours each day under glass chamber conditions with natural sunlight. The leaves of the sensitive clone had black stipple and bifacial necrosis after $O_3$ treatment. Photosynthesis and stomatal conductance were measured before, during, and after the $O_3$ treatment. The photosynthetic rates due to $O_3$ treatment were decreased 51 percent and 34 percent on the sensitive and tolerant clone, respectively. The stomatal conductance of the sensitive clone was more than 40 percent higher than that of the tolerant clone regardless of the $O_3$ treatment. As light intensity increased, the $O_3$ effect on photosynthesis was clear. Compared to the previous growth chamber studies, our natural light exposure system was able to maintain a stable photosynthetic responses of the control treatment throughout the fumigation period. In addition, changes in assimilation versus intercellular $CO_2$ concentration (A/C curves) showed that $O_3$ decreased the slope and asymptote of the curves for the sensitive clone. This indicates that $O_3$ decreases the biochemical capacity of photosynthesis on the sensitive clone. Chlorophyll contents and fluorescence of the two clones were analyzed to examine the $O_3$ effects on photosystem 11, but $O_3$ did not impact these variables on either clone. Although the tolerant clone did not show any foliar injury, we could not find any ecophysiological defensive responses to $O_3$ treated. Stomatal conductance of the tolerant clone was originally much lower than that of the sensitive one. Thus, the mechanisms of the tolerant clone in this system are to narrowly open stomata and efficiently maintain photosynthesis with a more durable biochemical apparatus of photosynthesis under $O_3$ stress. The sensitive clone has higher photosynthetic capacity and more efficient light reaction activity than the tolerant one under charcoal filtered condition, but is not as resilient under stress.

Lactococcus sp. CU216이 생산하는 박테리오신을 함유한 pH Sensitive Liposome의 응용 (Use of Bacteriocin Produced by Lactococcus sp. CU216 with pH Sensitive Liposome Entrapment)

  • 발효식품의 후산발효를 조절하기 위하여 pH sensitive liposome의 이용 가능을 검토하였다. Lactococcm sp. CU216이 생산하는 박테리오신은 L.. acidophilus를 제외하고는 대부분의 유산균주들에 생육억제 효과가 있는 것으로 확인되었으며, 이를 Octyl-Sepharose column으로 분획하여 정제하였다. 정제된 박테리오신을 dipalmitoyl phosphocholine, dipalmitoyl phosphoethanolamine, dioleoyl phosphocholine 및 콜레스테롤를 각각 4:2:1:4(㏖ ratio)의 비율로 섞은 liposome을 제조하여 최종적으로 pH sensitive bacteriocin-liposome을 제조하였다. 이렇게 제조된 liposome은 pH 4부근에서 대부분 유리되는 것으로 확인되었으나 pH 6이상에서는 일어나지 않았다. 또한 pH sensitive bacteriocin-liposome 한국의 전통 발효식품인 김치에 적용하여 저장시 pH의 저하를 억제시키는 것으로 확인되었다. 본 실험 결과, pH sensitive bacteriocin-liposome은 발효식품의 후산발효 억제에 적용될 수 있으며 추후에 요구르트를 포함한 다른 발효식품에 대한 추가적인 실험이 필요한 것으로 생각되었다.
