Comparison of Developmental Efficiency of Murine Somatic Cell Nuclear Transfer Protocol

  • The Somatic cell nuclear transfer (SCNT) method can be applied to various fields such as species conservation, regenerative medicine, farming industries and drug production. However, the efficiency using SCNT is very low for many reasons. One of the troubles of SCNT is that it is highly dependent on the researcher's competence. For that reason, four somatic cell nuclear injection methods were compared to evaluate the effect of hole-sealing process and existence of cytochalasin B (CB) on efficiency of murine SCNT protocol. As a results, the microinjection with the hole-sealing process, the oocyte plasma membrane is inhaled with injection pipette, in HCZB with CB was presented to be the most efficient for the reconstructed in SCNT process. In addition, we demonstrated that the oocytes manipulated in Hepes-CZB medium (HCZB) with CB does not affect the developmental rate and the morphology of the blastocyst during the pre-implantation stage. For this reason, we suggest the microinjection involving hole-sealing in HCZB with CB could improve SCNT process efficiency.

Isolation Performance of the Single-Sided air Curtain in Air-Conditioned Space (공조공간에서 수평토출형 에어커튼의 차단 성능)

  • Air curtains are widely used in commercial and public buildings to replace solid doors where traffic of people is predicted. At doorways where the solid door is open continuously or intermittently, an air curtain may be installed in order to reduce the flow of heat and moisture from the enclosed space to the outside. The purpose of this paper is to predict isolation performance of the single-sided air curtain when the wind is blowing. For the study, a numerical simulation is used to find the influence of various jet velocities on the efficiency of the single-sided air curtain device which is mounted at the side of the doorway. The isolation performance of the single-sided air curtain is evaluated by sealing efficiency which provides the assessment of the infiltration air ratio. According to the result of this study, the single-sided air curtain has lower sealing efficiency than downward-blowing air curtain. Therefore, for energy conservation in heating space, the single-sided air curtain is not recommended because of its low effectiveness.

The Analysis of the Discharging Characteristics on the Base Vacuum Level in a Vacuum In-line Sealing Process for High-efficiency PDP (고효율 PDP 제작을 위한 진공 인라인 실장에서의 초기 진공도에 따른 방전특성 분석)

  • We have examined the electrical and optical characteristics of the plasma display panel(PDP) produced by vacuum in-line sealing technology. We found that the luminous efficiency was decreased as the base vacuum level was increased. For the base vacuum level of $1\times10^{-3}$ Torr, the firing voltage was 235V at the discharge gas pressure of 400 Torr and the luminous efficiency was 0.8 lm/W at 180V sustaining pulse. However, for the base vacuum level of $1\times10^{-6}$ Torr, the firing voltage was reduced to 215V and the luminous efficiency was improved to 2.5lm/w. Finally, we demonstrated successfully the operation of tip-less PDP fabricated using vacuum in-line sealing method.

  • The one of the major problems in the development of PEMFC was regarding to the assurance of sealing on stack. The failure on the sealing creates the problems of fuel leakage, mixing, internal combustion, damage on parts and can be a direct reason for the degrading the efficiency of fuel cell. This paper studies on the analytical approach for improving the contacting pressure distribution on the gasket at the evaluation on the sealing of fuel cell stack. So, the assembly analysis on multi layered fuel cell stack was performed. The research on the simplification of finite element model was performed for three dimensional analysis at the multi layered state. The improved contact pressure distribution was obtained through the case studies on gasket for better sealing. In addition, the number of the cell was determined for the effective analysis and the structural characteristics were evaluated based on this research.

  • AC PDP with MgO protective layer coated with the optimum evaporation rate of $5{\AA}/s$ can generate more enhanced efficiency through the vacuum in-line sealing process. However, the optimized process conditions still require the optimum driving scheme on the ramp-up and ramp-down slope of the reset waveform for enhancing the efficiency. In this paper, for the in-situ vacuum sealed PDP with the optimum evaporation rate of MgO protective layer, the address delay time was investigated with various slopes of ramp waveform during a reset ramp-up and ramp-down period. In this study, the minimum statistical delay time was obtained at the ramp-up rate of $6.0 V/{\mu}s$ and the ramp-down rate of $0.7 V/{\mu}s$ of the reset waveform.

  • This study proposes a multi-robot system, using multiple autonomous robots, to explore concrete structures and assist in their maintenance by sealing any cracks present in the structure. The proposed system employed a new self-localization method that is essential for autonomous robots, along with a visualization system to recognize the external environment and to detect and explore cracks efficiently. Moreover, more efficient crack search in an unknown environment became possible by arranging the robots into search areas divided depending on the surrounding situations. Operations with increased efficiency were also realized by overcoming the disadvantages of the infeasible logical behavioral model design with only six basic behavioral strategies based on distributed control-one of the methods to control swarm robots. Finally, this study investigated the efficiency of the proposed multi-robot system via basic sensor testing and simulation.

Characterization of PDP Performance Prepared by Vacuum In-line Sealing Technology

  • By using vacuum in-line driving and photoluminescence measuring system, we have examined the electrical and optical characteristics of plasma display panel produced by vacuum in-line sealing technology. In addition, the relationship between luminous efficiency and base vacuum level before filling discharge gas was analyzed. In the case of base vacuum level of $1.33{\times}10^{-1}$ Pa, the firing voltage of a 2-inch diagonal PDP panel was ranged from 312 to 343 V depending on the discharge gas pressure of $2.667{\times}10^4$ to $4{\times}10^4$ Pa at room temperature, Whereas in the case of $1.33{\times}10^{-4}$ Pa, the firing voltage was reduced by 40 V and luminescence was improved slightly.

Numerical Study on Variations in the Sealing Performance of Air Curtains in Large-Scale Factory Opening Considering Various Design Factors (대형 공장 개구부용 에어커튼의 설계 인자에 따른 차단 성능 변화에 대한 전산해석 연구)

  • In large-scale factories, there are usually openings in the building to increase the efficiency of workers. However, if the factory is heated during winter, openings significantly increase the heating load. Therefore, there is a need for air curtains to be installed at the top of openings in factories to reduce the heating load due to the cold air entering from outside. The main design variables of these air curtains are the discharge angle, speed, and temperature, etc. While there have already been many studies focusing on these design variables, the distance from the opening and the width of the discharge have not been studied even though they also affect the sealing performance. As a result, when the distance from the opening decreases and the width of the discharge increases, we realize an optimum air curtain performance. However, if the distance from the opening is about 1.5 m, by adjusting the discharge angle and the distance from the opening, the sealing performance of the air curtain is improved by 13.7%.

Enhancing the Reproducibility of a Photoacoustic Signal Using a Minimum-volume Cell (최소 부피 광음향 셀의 광음향 신호 재현성 향상에 관한 연구)

  • Reproducibility obtained by a minimum-volume cell for photoacoustic (PA) signals is strongly dependent upon the sealing materials and sealing efficiency. Therefore, we have proposed, designed, and constructed an apparatus to enhance the sealing efficiency, and we have examined the reproducibility of amplitude and phase of PA signals with a minimum-volume cell. In particular, we have measured the PA signal as a function of modulation frequency for various rubber O-rings by using our apparatus. The results show that a silicon rubber O-ring can improve the reproducibility of the PA signal and phase by up to 53.4% and 80.4%, respectively, compared to that obtained by using the conventional sealing material, vacuum grease.

Development of an Optimal Trajectory Planning Algorithm for Automated Pavement Crack Sealer (도로면 크랙실링 자동화 장비의 최적 경로계획 알고리즘 개발)

  • During the last two decades, several tele-operated and machine-vision-assisted systems have been developed in construction and maintenance area such as pavement crack sealing, sewer pipe rehabilitation, and excavation. In developing such tele-operated and machine-vision-assisted systems, trajectory plans are very important tasks for optimal motions of robots whether their environments are structured or unstructured. This paper presents an optimal trajectory planning algorithm used for a machine-vision-assisted automatic pavement crack sealing system. In this paper, the performance of the proposed optimal trajectory planning algorithm is compared with the greedy trajectory plans which are used in previously developed pavement crack sealing systems. The comparison is based on computational cost versus overall gains in crack sealing efficiency. Finally, it is concluded that the proposed algorithm plays an important role in productivity improvement of the automatic pavement crack sealing system developed.
