Antigastritic and Antiulcerative Activities of Water Extracts Derived from Scutellaria baicalensis

  • Gastritis and gastric ulcer were known to be induced by gastic acid, stress, ethanol, Helicobacter pylori and free radical, etc. This study was performed for the development of a new drug or nutraceutical from medicinal plants or natural products with anti-gastritis, anti-ulcerative and gastroprotective activities. The water extract of Scutellaria baicalensis was exhibited potent inhibition in HCl ethanol-induced gastric lesion, acetic acid-induced and Shay ulcers, indicating the effects on gastric lesion and ulcer in rats. The water extract of Scutellaria baicalensis significantly inhibited HCl ethanol-induced gastric lesions at the oral dose of 300, 500 mg/kg. In pylorus ligated rats, the treatments of the water extract from Scutellaria baicalensis showed decrease in the volume of gastric secretion and acid output and increase pH at oral dose of 300, 500 mg/kg. And significantly reduced acetic acid-induced ulcer at the oral dose of 500 mg/kg for 12 days. In this study, we have found that the water extract from Scutellaria baicalensis had significant improvement in acute gastritis and ulcer at the dose of 300, 500 mg/kg and in chronic gastritis and ulcer at the dose of 500 mg/kg. Also we evaluated the antibacterial activity against Helicobacter pylori treated with Scutellaria baicalensis. Scutellaria baicalensis had a equivalent antibacterial activity with ampicilin against H. pylori at the dose of $100\;{\mu}/ml$. In histological examination, the water extract of Scutellaria baicalensis drastically restored gastric damages induced by HCl ethanol solution, pylorus- ligature and acetic acid. Therefore, we may use the water extract from Scutellaria baicalensis as antigastritic and antiulcerative agent for the purpose of the improvement or treatment of gastritis and gastric ulcer.

The inhibitory effect of Scutellaria baicalensis on type 1 interferon production in Raw 264.7 cells (LPS로 자극한 Raw 264.7 cell에서 황금(黃芩)의 type 1 interferon 억제 효과)

  • Objective : The present study was designed to investigate whether the water extract of the root of Scutellaria baicalensis could regulate lipopolysaccharide (LPS)-induced type 1 interferon. Methods : To evaluate of type 1 interferon inhibitory effect of the root of Scutellaria baicalensis, we examined type 1 interferon in LPS-stimulated RAW264.7 cells. Furthermore, Interleukin (IL)-10 and interferon regulatory factor (IRF) - 1, 7 expression level were examined to study the inhibition mechanisms. Results 1. Extract from the root of Scutellaria baicalensis didn't have any cytotoxity itelf. 2. Extract from the root of Scutellaria baicalensis inhibited interferon-a,b in dose dependant- and type 1 interferon production in time dependant manner. 3. Extract from the root of Scutellaria baicalensis reduced IL-10 and IRF-1, 7 production in LPS-stimulated RAW 264.7 cells. Conclusion : The extract from the root of Scutellaria baicalensis down-regulated LPS-induced type 1 interferon through suppression of IL-10 and IRF-1, 7 expression. This results suggested that the extract from the root of Scutellaria baicalensis may be a beneficial drug against inflammatory diseases.

Antimicrobial Activities of Scutellaria baicalensis and Phellodendron amurense Against MRSA and Candida (황금과 황백 추출물의 MRSA와 Candida에 대한 항균활성)

  • Scutellaria baicalensis and Phellodendron amurense, which have been used traditionally in treatment of many kinds of illness including infectious diseases, were extracted with hexane, dichloromethane, methanol and water, respectively and serially. Antimicrobial activities of the extracts were examined by disk diffusion method. Methanol extract from Scutellaria baicalensis revealed high antimicrobial activities against MRSA, Gm-bacteria and Candida albicans, dichloromethnane extract from Phellodendron amurense showed lower activity than the extract of Scutellaria baicalensis. Results suggest that methanol extract from Scutellaria baicalensis could be one of the candidates for new antimicrobial agent against the antibiotic-resistant microorganisms by further steps for the purification, determination of structure and synthesis.

Effect of Anti-Inflammation through Creation of Skin Fat Barrier on Scutellaria baicalensis extract (황금추출물의 지방장벽 생성을 통한 염증완화 효과)

  • Objectives The purpose of this study is to confirm the effect of alleviating inflammation through creating skin fat barrier from Scutellaria baicalensis extract. Methods Four-week-old Balb/C mice were divided into four groups: control group (Ctrl), lipid barrier eliminated group (LBEG), dexamethasone (DM) treated group after lipid barrier elimination (DMTG), and Scutellaria baicalensis (SB) treated group after lipid barrier elimination (SBTG). Scutellaria baicalensis extract were administered for 5 days after removal of the fat barrier. Changes in skin condition, improvement of the fat barrier, and relief of inflammation were observed in each group. Results Compared to LBEG and DMTG, pathological skin damage and tissue changes were less in SBTG, and transepidermal water loss (TEWL) and pH were also significantly reduced. Filaggrin was also significantly increased in SBTG. KLK7, PAR-2, and TSLP in SBTG also showed significant reduction compared to the LBEG and DMTG. Conclusions Scutellaria baicalensis extract restores skin barrier and relieves inflammation through the creation of skin fat barrier. This means that the Scutellaria baicalensis extract can regulate Th2 differentiation through the creation of the epithelial fat barrier.

The Effect of Medicinal Herb Extract on Antimicrobial Activity against Helicobacter pylori and Antioxidant Activity (약용식물 추출물의 Helicobacter pylori에 대한 항균 활성과 항산화 활성에 미치는 효과)

  • This study was designed to investigate the effect of medicinal herb extract on the antioxidant and antimicrobial activities against Helicobacter pylori, which is known as a ulcerogenic pathogen. The concentration of total phenolic compound of Scutellaria baicalensis(13.14%) was highest among water extracts and that of Ulmus parvifolia(15.12%) was highest among the ethanol extracts. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 91.00% and 65.03%, respectively, in DPPH assay. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 32.90% and 27.70%, respectively, in SOD assay. The antioxidant activity of the water extract of Ulmus parvifolia and of the ethanol extract of Glycyrrhiza uralensis Fischer was 2.15 and 2.17, respectively, in TBARS assay. In disc method, Scutellaria baicalensis showed the highest anti-microbial activity against H. pylori, followed by Glycyrrhiza uralensis Fischer among the water extracts and Glycyrrhiza uralensis Fischer showed the highest anti-microbial activity followed by Radix puerariae among ethanol extract.

The Use Possibility as a Dyestuff of Scutellaria Baicalensis Extract (황금(黃芩)의 천연염재로의 이용가능성에 관한 연구)

  • The use possibility as a dyestuff of Scutellaria baicalensis was investigated. The optimum conditions were evaluated when silk fabrics were dyed by Scutellaria baicalensis extract. The colorants of Scutellaria baicalensis were extracted better in water than methanol. The optical dyeing concentration was 80%(v/v), dyeing temperature was $80^{\circ}C$, dyeing time was 60minites, dyeing repetition was 2 turns. A Post-mordant method showed higher K/S value than premordant one, especially post-Fe appeared the highest value and post-Cr was second. Surface color was all Y. The color fastness was very good at the second repetition of no mordanted silk fabrics, all mordant fabrics were not improved the colorfastness. The antibacterial activity showed at Staphylococcus aureus tested specimen that bacterial reduction rate was 99.7%.

Effect of Scutellaria baicalensis Water Extract on Antioxidative Activity and Epidermal Thickness in DNCB-induced Allergic Contact Dermatitis Animal Model (황금열수추출물이 DNCB로 유도된 알레르기성 접촉피부염 흰쥐의 항산화능 및 표피회복능에 미치는 영향)

  • After allergic contact dermatitis (ACD) elicited by application of 1 mL of 2.5% 1-chloro-2,4-dinitrochloro-benzene (DNCB) onto dorsal skin, Sprague Dawley male rats fed Scutellaria baicalensis water extract were observed to investigate the mitigative effect of Scutellaria baicalensis water extract on ACD. The Scutellaria baicalensis water extract (100 mg/kg/day) was daily administered to rats suffered from ACD. Concentrations of TBARS in plasma and liver were increased in rats with ACD. The concentrations of TBARS in liver were significantly decreased in Scutellaria baicalensis-fed group (SBFG) compared to control group (CG), but not in plasma. Liver SOD activity was significantly increased in SBFG compared to that of normal group and CG. Serum Ig E level was significantly increased in CG compared to normal group, while that of SBFG was significantly decreased. The epidermal thickness of CG was significantly increased compared to that of normal group, while that of SBFG was significantly decreased compared to that of CG. These results indicated that the Scutellaria baicalensis water extract administration was effective for antioxidative activity in liver, serum Ig E level and epidermal thickness of rats with ACD.

Antioxidant Activity of Extract from Scutellaria baicalensis Georgi (황금 열수추출물의 항산화 작용에 대한 연구)

  • The purpose of this study was to investigate the antioxidant activity of the water extract from Scutellaria baicalensis Georgi. using linoleic acid as lipids. The extract exhibited antioxidative effect at all concentrations tested. With metal ion(Fe3+) present, the extract showed strong antioxidative effect. These result indicated that the extract can be used for antioxidative agents in oriental medicine.

The Effective Preparation of Flavonoids from Scutellaria baicalensis GEORGI by Diaion HP-20 Resin

  • Scutellaria baicalensis $G_{EORGI}$ (Scutellariae Radix) has been used to clear heat and to dry dampness in the stomach or intestines, which manifests as diarrhea or dysenteric disorder. In this study, we investigated the effective preparation of active components in Scutellariae Radix using the methods of solvent extraction and absorption fractionation for the development of new functional food or pharmaceuticals. The marker substances, baicalin, baicalein, wogonoside, and wogonin were directly isolated from the Scutellariae Radix. There chemical structures were elucidated by spectroscopic analysis. The Scutellariae Radix was extracted with hot water. To enhance yield of flavonoids in Scutellariae Radix, the hot water extract was dissolved in ethanol with concentration dependent manner. The precipitates were separated using centrifugal techniques at 10,000 rpm. Supernatant liquid was applied to the HPLC for quantification of major compounds. Separately, the hot water extract was absorbed on Diaion HP-20 resin. And then, the absorbed fraction was eluted with methanol for HPLC. The contents of baicalin, baicalein, wogonoside and wogonin in different treatment methods were analyzed by HPLC. Total amount of four major components were 16.9% in 50% ethanol extract, 21.7% in 70% ethanol extract, 20.5% in 90% ethanol extract, and 39.3% in absorbed fraction of Diaion HP-20 resin. In these results, we found that resin absorption method is suitable for the extraction of enriched flavonoids from Scutellariae Radix.

Anti-inflammatory activity of Scutellaria Baicalensis root extract in lipopolysaccharide-induced RAW 264.7 cells (LPS로 유도된 RAW264.7세포주에서 황금뿌리 물추출물의 항염증활성)

  • Scutellaria baicalensis has been used as a traditional medicine for diarrhea, dysentery, hypertension, hemorrhaging, insomnia, inflammation, and respiratory infections. This study examined the anti-inflammatory effect of Scutellaria baicalensis water extract (SWE) in lipopolysaccharide (LPS)-induced RAW 264.7 cells. To evaluate the anti-inflammatory effect of SWE, RAW 264.7 macrophages were stimulated with LPS to induce the production of inflammation-related factors, which were measured by western blotting. In RAW 264.7 cells, SWE inhibited the production of nitric oxide (NO) without causing cell toxicity. SWE also reduced the expression of inducible NO synthase and cyclooxygenase-2 protein, as well as the production of pro-inflammatory cytokines (such as tumor necrosis factor-α). The phosphorylation levels of the mitogen-activated protein kinase family members, such as JNK and p38, were also reduced by SWE. Thus, SWE could be used as a potential anti-inflammatory agent.
