한번의 생약제로 쓰고 있는 황금(Scutellaria baicalensis Georgi)에서 얻은 추출물에 대하여 유지에 대한 항산화 효과를 검토하였다. 즉 황금 추출물을 linoleic acid에 소정량 첨가하고 5$0^{\circ}C$에 일정기간 저 장하여 linoleic acid의 산화 안정성을 비교 검토하였으며, 또한 유지 자동산화의 촉진인자인 금속이온 (Fe3+)을 첨가하여 황금 추출물의 금속이온 봉쇄효과를 조사하였다. 황금 추출물의 항산화 효과는 전 농도별에서 높은 항산화 효과를 나타내었다. 그리고 시간 경과에 따른 항산화 효과에 있어서도 정도의 차이는 있지만 전 농도에서 유도기의 연장효과가 상당히 우수한 것으로 나타났다. 황금 추출물의 금속 이온 봉쇄효과에 있어서는 linoleic acid에 황금 추출물과 Fe3+을 첨가한 구가 황금 추출물의 무첨가구보 다 항산화 효과가 아주 우수한 것으로 나타나 금속이온 봉쇄력이 강한 것으로 나타났다.

The feature of asthma are airway inflammation (AI), reversible airway obstruction, and an increased sensitivity to bronchoconstricting agents, elevated airway hyperresponsiveness (AHR), excess production of Th2 cytokines, and eosinophil accumulation in the lungs. This study was performed to investigate if oral administration of $Scutellaria$ $baicalensis$ Georgi water extracts (SBG) have the antiasthmatic potential for the treatment of asthma. Asthmatic HI and AHR were induced by systemic sensitization to ovalbumin (OVA) with intratracheal instillation with 0.1 mg/mL of diesel exhaust particles (DEP) suspension once a week for 10 weeks in BALB/c mice. SBG was orally administered with the concentraion of 200 mg/kg 5 days a week for 10 weeks. Long-term SBG treatment suppressed the eosinophil infiltration into airways from blood, the asthmatic AI and AHR by attenuating the production of cytokine IL-4, IL-5 and IL-13, histamine and OVA-specific IgE. Our data suggest that SBG has inhibitory effects on AI and AHR in a mouse model of asthma, may act as a potential Th2 cytokine antagonist, and may have a therapeutic effect on allergic asthma.

Acute toxicity of a combined preparation of the standardized extracts Scutellaria baicalensis GEORGI and Salvia miltiorrhiza BUNGE in a ratio of 3:1 was examined in male and female ICR mice. Mice were treated with the test substance intragastrically at a dose of 0 mg/kg, 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. No death or abnormal clinical sign was shown during the observation period. Also there were no difference in net body weight gain, organ weight, and gross pathological findings at the terminal sacrifice. The results suggested that acute oral toxicity of a combined preparation of the standardized extracts is very low at the conditions employed in this study.

GATA-binding protein-3 (GATA-3) and T-box expressed in T-cells (T-bet) are now considered as master transcription factors involving Th cell differentiation, but the roles of these factors are still uncertain in vivo. This study was conducted to investigate the expression of these transcription factors in the liver damage induced by carbon tetrachloride ($CCl_4$) in rats. In this study, liver damage were induced with Scutellaria baicalensis Georgi water extracts (SBW) and followed for 4 weeks. The expression of GATA-3 and T-bet protein in liver damage induced by $CCl_4$ and the serum levels of immunoglobulin A (IgA), IgE were studied after 4 weeks of treatment. We found that effect of SBW on IFN-$\gamma$, STAT1, pSTAT1 and T-bet was decreased in vivo. Several genes were demonstrated to be IL-4 inducible prior to the discovery of STAT6. $CCl_4$+SBW group was significantly lower than $CCl_4$ group in IL-4, STAT6, pSTAT6 and GATA-3. Our data indicate that cytokine protein production were increased in $CCl_4$ group and $CCl_4$+SBW group. From these results, water extracts obtained from Scutellaria baicalensis Georgi may have an immunoregulatory effect in the liver induced by $CCl_4$ of rats.

Scutellaria baicalensis Georgi is one of the most important medicinal herbs in traditional chinese medicine. The object of this study was to determine the effects of water extracts of Scutellaria baicalensis (SB) and Scutellaria baicalensis metabolite (SBM) on the anxiolytic-like activities in the elevated plus-maze (EPM) test. The water extracts of SB (100, 200, or 400 mg/kg), and SBM (100 mg/kg) were orally administered to male SD rats for 3 day. All rats were subjected to behavioral tests for the anxiolytic activity at 3 days. (omitted)

Scutellaria baicalensis Georgi (SBG) is traditionally used medicinal herb that has anti-oxidant, anticancer and anti-inflammatory effects. In this study, we investigated whether the extracts of SBG have the inhibitory activity in the osteoclast differentiation by using mouse monocytes RAW264.7 cells and primary mouse bone marrow-derived macrophages (BMMs). Methanol extract (ME) from SBG was successively fractionated into methylene chloride (MF), ethylacetate (EF) and n-butanol fraction (BF). The activity assay for tartrateresistant acid phosphatase (TRAP) and Western blot analysis were employed to evaluate the osteoclasts differentiation and the activation of mitogen-activated protein (MAP) kinases, respectively. ME, MF, EF and BF significantly and dose-dependently inhibited osteoclast differentiation without the decrease of cell viability at the concentrations used in this study. In addition, ME significantly inhibited the activation of c-jun-N-terminal kinase (JNK). In conclusion, this study firstly demonstrated that ME of SBG has the potential to inhibit the osteoclast differentiation through the suppression of JNK activation partially.

Scutellaria baicalensis Georgi (Scutellariae Radix)는 이뇨제, 고지혈증, 항박테리아, 항알레르기, 항염증제 및 항암제와 같은 건강보조식 및 전통 생약으로도 널리 사용되어 왔다. 본 연구에서 복잡한 S. baicalensis 추출물에서 지표 물질 또는 유효화합물들을 분리하는 것은 신원 확인 및 생리활성 평가를 위한 필수적인 단계다. 8개의 성분들로 구성된 타겟 분획물을 두개의 gradient elution를 사용하여 고성능 액체 크로마토 그래피에서 분석하였다. 중압 액체크로마토그래피 및 개방형칼럼으로 분취를 시뮬레이션함으로 예비실험에서 충분히 분리가 되도록 용리 조건을 결정할 수 있었다. 최적 분취방법으로 확보된 표준유효분획물로부터 8개의 지표성분들이 포함된 것을 확인하였다. 또한, 분획물은 UPLC-QTof-MS 비교 분석으로 MS, UV, HRESIMS 결과를 확인할 수 있었다. 따라서, 스케일 업 실험법은 S. baicalensis 추출물에 성공적으로 적용될 수 있었다.

An ethanolic extracts of Scutellaria Baicalensis GEORGI are used to treat cancer, infectious diseases, and inflammation. In the present study, we investigated the effects of an ESBG on the growth and survival of 5637 cells, a human bladder carcinoma cell line. Cells were treated with different concentrations of an ethanolic extract of Scutellaria Baicalensis GEORGI (ESBG), and cell death was assessed using a MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay. Analyses of the sub G1 peak, caspase-3 and -9 activities, and mitochondrial membrane depolarizations were conducted to confirm cell death by apoptosis. ESBG had a cytotoxic effect on 5637 cells, and increased the sub G1 peak, caspase-3 and -9 activities, and mitochondrial depolarization, indicating ESBG induced apoptosis. Furthermore, MAPK (mitogen-activated protein kinases) inhibitors suppressed this apoptosis. In an in vitro study, a combination of sub-optimal doses of ESBG and paclitaxel, 5-fluorouracil, or docetaxel noticeably suppressed tumor growth by 5637 cells. Our findings provide insight of the mechanisms underlying cellular apoptosis induced by ESBG, and suggest new therapeutic strategies for bladder cancer.

본 실험은 황금이 Allergy성(性) 접촉피부염의 면역과민반응 및 염증반응에 미치는 영향을 조사하기 위해서 시행되었다. 실험을 위해 BALB/C계 생쥐를 DNCB 처리군과 DNCB 처리 후 황금추출액을 경구 투여한 황금추출물투여군(HGET군)으로 나누어 2,4-dinitrochlorobenzene(DNCB)를 도포하여 Allergy성(性) 접촉피부염을 유발한 뒤, 시간의 경과에 따라 contact hypersensitivity assay, 피부의 일반적인 구조, 혈관생성, 림프구를 비롯한 염증세포, sulfated acid mucosubstance, 비만세포, IL-2 R, ICAM-1 그리고 CD11b의 변화를 관찰하였다. Contact hypersensitivity assay에서 HEGT군은 DNCB처리군에 비해 유의성(有意性)있는 ear swellig의 감소를 보였다. 피부표피의 변화에서 HEGT군에서 DNCB처리군에 비해 감소된 것으로 나타났다. 또한 혈관생성, 림프구의 침투 및 표피 기저층과 가시층 세포의 손상은 HEGT군에서 DNCB처리군에 비해 감소된 것으로 나타났다. 또한 혈관생성, 림프구를 비롯한 염증세포, sulfated acid mucosubstance와 비만세포, IL-2 R 양성반응세포, ICAM-1 양성반응세포 그리고 CD11b 양성반응은 Allergy성(性) 접촉피부염의 면역과민반응억제와 항염증작용에 효과가 있는 것으로 사료된다.
