Food Component Characteristics of Seafood Cooking Drips (수산 자숙액의 식품성분 특성)

  • This study was conducted to investigate on food component characteristics of seafood cooking drips (skipjack tuna cooking drip, octopus cooking drip and oyster cooking drip) as a source of functional seasoning. Heavy metal contents of seafood cooking drips were below food safety level. Among seafood cooking drips concentrated to 5 folds, the crude protein content was the highest (18.1%) in skipjack tuna cooking drip (SCD). The free amino acid content and taste value were higher in SCD than in other seafood cooking drips, and the major free amino acids were glutamic acid and aspartic acid. Total amino acid content of SCD was 16.2 g/100 mL and the major amino acids were glutamic acid (11.9%), proline (9.2%), glycine (9.1%) and histidine (11.5%). SCD in comparison with other seafood cooking drips showed the highest angiotensin I converting enzyme (ACE) inhibitory activity at $IC_{50}$ of 14.1 mg/mL. These results suggested that SCD could be used as a source of functional seasoning.

Inhibitory effect of Allium macrostemon extracts on adipogenesis of 3T3-L1 preadipocytes (산달래 추출물의 3T3-L1 지방전구세포 분화 억제 효능)

  • The aim of this study was to compare the biological activities of whole-plant (WAE), bulb (BAE), and leaf (LAE) extracts of Allium macrostemon. The antioxidant activities, total polyphenol contents, and anti-adipogenic activities of WAE and LAE were superior to those of BAE, whereas the biological effects of WAE and LAE were similar. Therefore, the effect of LAE on adipogenesis was further investigated. Treatment of preadipocytes with LAE at 100 g/mL resulted in the inhibition of intracellular lipid accumulation by 49.64%. Consistent with this result, quantitative reverse transcription-PCR showed that LAE treatment decreased the gene expressions of CCAAT/enhancer-binding protein beta (C/EBPβ), peroxisome proliferator-activated receptor gamma (PPARγ), C/EBPα and stearoyl-CoA desaturase 1 (SCD1). Thus, LAE attenuates the adipogenesis of preadipocytes by suppressing the expression of adipogenic and lipogenic genes. These results suggest that LAE can be potentially useful as a functional ingredient to prevent obesity in the food industry.

Effects of Geranium wilfordii Maxim. Ethanol Extract of on Adipogenesis and Lipogenesis (세잎쥐손이풀(Geranium wilfordii Maxim.) 에탄올 추출물이 지방생성 및 지방합성에 미치는 영향)

  • In this study, the anti-obesity effect of Geranium wilfordii Maxim. extract was studied using 3T3-L1 cells. Geranium wilfordii Maxim. was extracted with water (NG-GT-T1L), 10% ethanol (NG-GT-T2L), 30% ethanol (NG-GT-T3L), 50% ethanol (NG-GT-T4L), 70% ethanol (NG-GT-T5L), and the effects on cell viability, lipid accumulation, triglyceride content, and protein expression in 3T3-L1 cells were confirmed. It was confirmed that NG-GT-T3L extract was superior to other extract conditions in reducing lipid accumulation and triglyceride content in the concentration range that did not show cytotoxicity. In addition, it was confirmed to suppress adipogenesis and lipogenesis by reducing the expression of peroxisome proliferator-activated receptor-gamma (PPARγ) and CCAAT/enhancer binding protein-α(C/EBPα) proteins that regulate adipogenesis, decreasing the expression of fatty acid synthetase (FAS) and stearoyl CoA desaturase-1 (SCD-1) proteins that regulate lipogenesis, and increasing the expression of AMP-activated protein kinase (AMPK) protein. From these research results, Geranium wilfordii Maxim. NG-GT-T3L extract is believed to have anti-obesity reduction effects through suppressing lipid accumulation and triglyceride accumulation and regulating adipogenesis and lipogenesis-related proteins.

  • Flocculation-spectrophotometry and streaming current detector( SCD ) method were investigated and compared in order to determine the optimum dosages of synthetic cationic polymers of different charge density and molecular mass for the removal of humic acid. The optimum dosage for each of the polymers was determined with the dosage at which the lowest absorbance of humic acid was shown for the formal. and was determined with the dosage required during charge neutralization of humic acid for the latter It was in good agreement between both methods and there is a strong inverse correlation between the optimum dosage and charge density of the polymers, with highly charged polymer giving the lowest optimum dosage, pointing out the importance the charge neutralization. By flocculation-spectrophotometry, it was found that the absorbance of humid acid with the amount of each of the polymers dosed, changes sharply for polymers of high charge density, but changes rather broadly for polymers of low and middle charge density, Both methods showed that a stoichiometric correlation exists between the optimum dosage of each of the cationic polymers and the negatively charged humic acid.

Restoration of the adipogenic gene expression by naringenin and naringin in 3T3-L1 adipocytes

  • Background: Naringenin and its glycoside naringin are well known citrus flavonoids with several therapeutic benefits. Although the anti-adipogenic effects of naringenin and naringin have been reported previously, the detailed mechanism underlying their anti-adipogenesis effects is poorly understood. Objectives: This study examined the anti-adipogenic effects of naringenin and naringin by determining differential gene expression patterns in these flavonoids-treated 3T3-L1 adipocytes. Methods: Lipid accumulation and triglyceride (TG) content were determined by Oil red O staining and TG assay. Glucose uptake was measured using a 2-[N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2-deoxy-d-glucose fluorescent d-glucose analog. The phosphorylation levels of AMP-activated protein kinase (AMPK) and acetyl Co-A carboxylase (ACC) were observed via Western blot analysis. Differential gene expressions in 3T3-L1 adipocytes were evaluated via RNA sequencing analysis. Results: Naringenin and naringin inhibited both lipid accumulation and TG content, increased phosphorylation levels of both AMPK and ACC and decreased the expression level of 3-hydroxy-3-methylglutaryl CoA reductase (HMGCR) in 3T3-L1 adipocytes. RNA sequencing analysis revealed that 32 up-regulated (> 2-fold) and 17 down-regulated (< 0.6-fold) genes related to lipid metabolism, including Acaca, Fasn, Scd1, Mogat1, Dgat, Lipin1, Cpt1a, and Lepr, were normalized to the control level in naringenin-treated adipocytes. In addition, 25 up-regulated (> 2-fold) and 25 down-regulated (< 0.6-fold) genes related to lipid metabolism, including Acaca, Fasn, Fabp5, Scd1, Srebf1, Hmgcs1, Cpt1c, Lepr, and Lrp1, were normalized to the control level by naringin. Conclusions: The results indicate that naringenin and naringin have anti-adipogenic potentials that are achieved by normalizing the expression levels of lipid metabolism-related genes that were perturbed in differentiated 3T3-L1 cells.

AMPKα, C/EBPβ, CPT1β, GPR43, PPARγ, and SCD Gene Expression in Single- and Co-cultured Bovine Satellite Cells and Intramuscular Preadipocytes Treated with Palmitic, Stearic, Oleic, and Linoleic Acid

  • We previously demonstrated that bovine subcutaneous preadipocytes promote adipogenic gene expression in muscle satellite cells in a co-culture system. Herein we hypothesize that saturated fatty acids would promote adipogenic/lipogenic gene expression, whereas mono- and polyunsaturated fatty acids would have the opposite effect. Bovine semimembranosus satellite cells (BSC) and intramuscular preadipocytes (IPA) were isolated from crossbred steers and cultured with 10% fetal bovine serum (FBS)/Dulbecco's Modified Eagle Medium (DMEM) and 1% antibiotics during the 3-d proliferation period. After proliferation, cells were treated for 3 d with 3% horse serum/DMEM (BSC) or 5% FBS/DMEM (IPA) with antibiotics. Media also contained $10{\mu}g/mL$ insulin and $10{\mu}g/mL$ pioglitazone. Subsequently, differentiating BSC and IPA were cultured in their respective media with $40{\mu}M$ palmitic, stearic, oleic, or linoleic acid for 4 d. Finally, BSC and IPA were single- or co-cultured for an additional 2 h. All fatty acid treatments increased (p = 0.001) carnitine palmitoyltransferase-1 beta ($CPT1{\beta}$) gene expression, but the increase in $CPT1{\beta}$ gene expression was especially pronounced in IPA incubated with palmitic and stearic acid (6- to 17-fold increases). Oleic and linoleic acid decreased (p = 0.001) stearoyl-CoA desaturase (SCD) gene expression over 80% in both BSC and IPA. Conversely, palmitic and stearic acid increased SCD gene expression three fold in co-cultured in IPA, and stearic acid increased $AMPK{\alpha}$ gene expression in single- and co-cultured BSC and IPA. Consistent with our hypothesis, saturated fatty acids, especially stearic acid, promoted adipogenic and lipogenic gene expression, whereas unsaturated fatty acids decreased expression of those genes associated with fatty acid metabolism.

  • This study was performed to stabilize the physical properties of Sodium Carbonate Decahydrate that was selected as a highly concentrative thermal energy storage medium. The addition of ARS(additives to prevent supercooling) showed to prevent the supercooling of Na$_{2}$CO$_{3}$.10H$_{2}$O, and the supercooling was decreased below $1.5^{\circ}C$ with ARS of 3 wt% and the addition of PSC(phase separation controller) of 1.5 wt% controlled the phase separation of Na$_{2}$CO$_{3}$ .10H$_{2}$O with the phase change cycles increased from 0 to 1,500, the phase change temperature and the latent heat has changed in the range of 30$\pm$1.$0^{\circ}C$ and 54$\pm$2.0Kcal.kg$^{-1}$ respectively.

Effect of Jungmanbunso-hwan Extract on HepG2 Cell Model of Nonalcoholic Fatty Liver Disease Caused by Palmitate (중만분소환 추출물이 Palmitate로 유발된 비알코올성 지방간 HepG2 cell 모델에 미치는 영향)

  • Objectives: This study was performed to investigate the anti-lipogenic effect and the mechanism of Jungmanbunso-hwan extract (JMBSH) on a cellular model of non-alcoholic fatty liver disease (NAFLD) caused by palmitate in HepG2 cells.Methods: The JMBSH was prepared, andHepG2 cells were treated with various concentrations of JMBSH in order to perform an MTT assay. The HepG2 cells were cultivated in palmitate-containing media with or without extract of JMBSH. The intracellular lipid content in the HepG2 cells was examined. The effects of JMBSH on sterol regulatory element-binding transcription factor-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), stearoyl-CoA desaturase-1 (SCD-1), and AMP-activated protein kinase (AMPK) activation in HepG2 cells were measured.Results: JMBSH did not reduce HepG2 cell viability under 1,000 μg/mL. JMBSH considerably decreased intracellular lipid accumulation caused by palmitate in HepG2 cells. JMBSH repressed expression of SREBP-1c, which mediates the induction of lipogenic genes (ACC, FAS, and SCD-1). JMBSH also activated AMPK, which plays animportant role in the regulation of hepatic lipid metabolism.Conclusions: This study suggested that JMBSH relieves hepatic steatosis by repressing SREBP-1c, which mediates the induction of lipogenic genes. The anti-lipogenic effect of JMBSH may also be related to the activation of AMPK. Therefore, JMBSH could potentially be applied to NAFLD treatment after further clinical studies.
