Quality Properties and Antioxidant Activities of Korean Traditional Rice-Based Wine, Makgeolli Added with Sweet Pumpkin (단호박을 첨가하여 제조한 막걸리의 품질특성 및 항산화 활성)

  • Sweet pumpkin is rich in minerals such as calcium, phosphorus and fiber, and also contains a large amount of β-carotene, which has antioxidant effects. In this study, Makgeolli containing steaming sweet pumpkin (SP) was manufactured to enhance the antioxidant activity. To prepare the Makgeolli, SP was added in amounts of 5, 10, 20, and 30% (w/w), and the chemical (pH, total acidity, alcohol), microbiological (lactic acid bacteria, yeast) properties, and antioxidant activities (DPPH, ABTS) were examined during fermentation and storage for 9 days. The pH of SP Makgeolli was 4.00-4.23 at day 1 of fermentation, and then the pH gradually decreased as fermentation progressed, showing the lowest results at day 9 (3.28-3.52). At day 1, the total acidity was significantly increased (P<0.05) as the amount of SP (0-30%; 0.09-0.55%) and the total acidity in SP Makgeolli (1.01-1.20) was also rapidly increased by 5 days of fermentation. Alcohol content was significantly increased (P<0.05) as the amount of SP (0-30%; 4.59-5.77%) increased at day 9. The counts of lactic acid bacteria and yeast in SP Makgeolli were 8.0-8.1 and 7.9-7.8.0 CFU/mL, respectively, which was higher than SP 0% at day 9. DPPH and ABTS radical scavenging activities were significantly increased (P<0.05) as the amount of SP (0-30%; 48.29-78.97% for DPPH, 62.12-86.68% for ABTS) increased at day 9. This study suggests that Makgeolli added with SP could be potentially and commercially developed due to its superior microbiological and chemical properties, including high antioxidant activities.

  • To evaluate hepatoprotective effects, the antioxidant capacities of matcha green tea extract (Camellia sinenesis) were compared to those of green leaf tea and the anti-inflammatory activities in HepG2 cells were investigated. Evaluation of the total phenolic and total flavonoid content, 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and inhibitory effect on lipid peroxidation indicated that the aqueous extract of matcha green tea presented significant catechin content and antioxidant capacity compared to those of green leaf tea. In addition, the extract had considerable inhibitory effects on α-glucosidase, α-amylase, and advanced glycation end-products. The matcha green tea extract significantly increased cell viability and reduced reactive oxygen species in H2O2- and high-glucose-treated HepG2 cells. Furthermore, in response to oleic acid-induced HepG2 cell injury, treatment with matcha green tea aqueous extract inhibited lipid accumulation and regulated the expression of inflammatory proteins such as p-JNK, p-Akt, p-GSK-3β, caspase-3, COX-2, iNOS, and TNF-α. Matcha green tea could be used as a functional material to ameliorate hepatic lipid accumulation and inflammation.

Effects of Light Intensity and Electrical Conductivity Level on Photosynthesis, Growth and Functional Material Contents of Lactuca indica L. 'Sunhyang' in Hydroponics (수경재배에서 광도와 양액 농도가 베이비 산채 왕고들빼기 '선향' 광합성과 생육 및 기능성 물질 함량에 미치는 영향)

  • This study was conducted to examine the changes of photosynthesis, growth, chlorophyll contents and functional material contents in light intensity and EC concentration of wild baby leaf vegetable, Indian lettuce (Lactuca indica L. cv. 'Sunhyang') in DFT hydroponics. The cultivation environment was 25±1℃ of temperature and 60±5% of relative humidity in growth system. At 14 days after sowing, combination effect of light intensity (Photosynthetic Photon Flux Density (PPFD 100, 250, 500 µmol·m-2·s-1) and EC level (EC 0.8, 1.4, 2.0 dS·m-1) of nutrient solution was determined at the baby leaf stage. The photosynthesis rate, stomatal conductance, transpiration rate and water use efficiency of Indian lettuce increased as the light intensity increased. The photosynthesis rate and water use efficiency were highest in PPFD 500-EC 1.4 and PPFD 500-EC 2.0 treatment. The chlorophyll content decreased as the light intensity increased, but chlorophyll a/b ratio increased. Leaf water content and specific leaf area decreased as light intensity increased and a negative correlation (p < 0.001) was recognized. Plant height was the longest in PPFD 100-EC 0.8 and leaf number, fresh weight and dry weight were the highest in PPFD 500-EC 2.0. Anthocyanin and total phenolic compounds were the highest in PPFD 500-EC 1.4 and 2.0 treatment, and antioxidant scavenging ability (DPPH) was high in PPFD 250 and 500 treatments. Considering the growth and functional material contents, the proper light intensity and EC level for hydroponic cultivation of Indian lettuce is PPFD 500-EC 2.0, and PPFD 100 and 250, which are low light conditions, EC 0.8 is suitable for growth.

Evaluation of antioxidant properties and oxidative stability of oregano seed solvent fraction (추출용매에 따른 오레가노 종자 분획물의 항산화 및 유지산화안정성 평가)

  • The in vitro antioxidant activity of oregano seed fractions, fractionizing with 80% ethanol, n-hexane, ethyl acetate, n-butanol, and water, was evaluated, and their effects on edible oils were determined in corn oil at 180℃. The ethyl acetate fraction had the highest radical-scavenging activity. The ferric reducing antioxidant power activity and total phenol content of the ethyl acetate fraction were determined as 6,130 µmol ascorbic acid equivalents/g extract and 770 µmol tannic acid equivalents/g extract, respectively, which were significantly higher than those of the other fractions (p<0.05). Primary and secondary oxidation products in corn oil added with the ethyl acetate fraction of oregano seed significantly decreased by 1.5 and 1.26 times, respectively, compared with those in the control groups. The major volatile ingredients in the ethyl acetate fraction of oregano seeds were determined to be carvacrol, thymoquinone, and 3-cyclopentylcyl-cyclopentan-1-one. Ethyl acetate is a suitable solvent for extracting antioxidant compounds from oregano seeds and can be used as a natural antioxidant.

Whitening and Antioxidant Effects of Extracts from Angelica gigas Nakai Thin Root (참당귀 세미 추출물의 미백 및 항산화 효과)

  • Angelica gigas Nakai (AGN) is a perennial plant belonging to the family Apiaceae. Its root has been utilized as a traditional medicine especially in Korea. This study was carried out to evaluate the potential use of extracts from AGN root parts as a cosmetic material. The dried AGN roots are divided into body (B), thick root (TkR), medium root(MR) and thin root (TnR) according to their diameter before cutting into medicine. B, TkR and MR of AGN are combined and used as medicinal herbs (MH). The extracts from AGN each root part (B, TkR, MR, TnR, MH) were used to test the effect on cell viability using MTS assay and to examine inhibitory effect on melanin accumulation in B16F10 melanoma cells. All extracts (50 - 200 ㎍/mL) from the each root part did not affect the cell viability. And inhibitory effect of all root extracts (200 ㎍/mL) on melanin accumulation was 12-19%. Especially, TnR showed similar inhibitory effect on melanin accumulation to MH. In addition, DPPH and ABTS free radical scavenging activity were higher in the TnR extract compared to MH. This study showed that the TnR extract exhibit high inhibitory effect on melanin accumulation and antioxidant activity compared to MH, suggesting that TnR extract has potential as a cosmetic ingredient.

Development and Validation of Analytical Method and Antioxidant Effect for Berberine and Palmatine in P.amurense (황백의 지표성분 berberine과 palmatine의 분석법 개발과 검증 및 항산화 효능 평가)

  • The aim of this study was to develop and validate a simultaneous analytical method for berberine and palmatine, which are representative substances of Phellodendron amurense, and to evaluate the antioxidant activity. We evaluated the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification (LOQ) of analytical methods for berberine and palmatine using high-performance liquid chromatography. Our result showed that the correlation coefficients of the calibration curve for berberine and palmatine exhibited 0.9999. The LODs for berberine and palmatine were 0.32 to 0.35 µg/mL and the LOQs were 0.97 to 1.06 µg/mL, respectively. The inter-day and intra-day precision values for berberine and palmatine were from 0.12 to 1.93 and 0.19 to 2.89%, respectively. The inter-day and intra-day accuracies were 98.43-101.45% and 92.39-100.60%, respectively. In addition, the simultaneous analytical method was validated for the detection of berberine and palmatine. Moreover, we conducted FRAP and NaNO2 scavenging activity assays to measure the antioxidant activities of berberine and palmatine, and both showed antioxidant activity. These results suggest that P.amurense could be a potential natural resource for antioxidant activity and that the efficacy can be confirmed by investigating the content of the berberine and palmatine.

Manufacturing and Quality Characteristics of Puffed Black Bean Fermented by Lactobacillus plantarum Strains Isolated from Kimchi (김치 유래 Lactobacillus plantarum을 이용한 팽화 검은콩 발효물의 제조 및 품질특성)

  • The purpose of this study was to optimize the fermentation condition of black bean by lactic acid bacteria (LAB) and to evaluate the quality characteristics of fermented black bean. Lactobacillus plantarum SU22 isolated from kimchi was selected as a starter for the fermentation of black bean because the strain exhibited strong antimicrobial activity against pathogenic bacteria and did not produce biogenic amines or a carcinogenic enzyme, β-glucuronidase. Fermentation was performed with broth containing puffed black bean (PBB) inoculated with 1% (v/v) of L. plantarum SU22 at 37℃ for 48h. The viable cell count of LAB was over 9 Log CFU/mL in PBB (20%) broth fermented with L. plantarum SU22. Fermentation of alcalase-treated PBB (20%) broth with L. plantarum SU22 was found to be the optimal condition, increasing viable cell count of LAB up to 10.30 Log CFU/mL. Under the optimal condition, the total polyphenol content (94.02 mg GAE/g) and DPPH radical scavenging activity (92.50%) were significantly increased, compared to non-fermented control (87.74 mg GAE/g, 83.14%).

A Comparative Study on the Chemical Characteristics and Antioxidant Effects of Sea Mustards Sourced from Different Areas in Taejongdae (태종대산 5종 돌미역의 화학성분 및 항산화활성 비교)

  • This study compared the nutritional characteristics and antioxidant effects of sea mustards sourced from five different areas (Barammaegi, Gultongmeori, Chanmulgae, Johongtaek, and Goraedeung) in Taejongdae, Youngdo, Busan. The contents of total flavonoids and phenols and fatty acid composition were measured. To evaluate their antioxidant effects, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays were used. Acetone/methylene chloride (A+M) extracts from all the sea mustards contained higher amounts of total flavonoids and phenols than methanol (MeOH) extracts. Among the sea mustards obtained from the different areas, the total flavonoid and total phenolic content of the A+M extract of the sea mustard from Gultongmeori was 1.44±0.04 mg/g and 1.72±0.06 mg/g, respectively. In terms of the fatty acid composition, the Gultongmeori sea mustard had higher percentages of total n-6, total n-3, eicosapentaenoic acid (EPA, 20:5n-3), and docosahexaenoic acid (DHA, 22:6n-3) than the sea mustards from the other areas. The A+M extract of the sea mustard from Gultongmeori was more effective in terms of scavenging free radicals as compared with that of the other sea mustards, as assessed by the DPPH and ABTS assays (p<0.05). In a 120-minute reactive oxygen species (ROS) production assay, all the extracts tested decreased cellular ROS production induced by H2O2 compared to that produced by exposure to an extract-free control (p<0.05). The extracts from Barammaegi and Gultongmeori had a greater inhibitory effect on cellular ROS production. These results indicated that the antioxidant effects of sea mustards might be associated with a higher amount of flavonoids and phenols. This study suggests that food-processed products from sea mustard can be developed as functional foods for promoting health in the local population.

Quality characteristics of Halal chicken sausages prepared with biji powder (비지 분말 첨가 Halal 계육 소시지의 조직감 및 항산화특성)

  • To meet the needs of Muslim consumers, sausages were prepared using Halal-certified chicken thighs and different amounts of biji powder (0, 20, 30, 40, and 50%), and then the properties of the sausages were compared. As the biji powder levels increased, both the moisture content and the pH of the sausages significantly decreased, whereas their fiber content increased. As the biji powder levels increased, the free radical scavenging effect (DPPH, ABTS) and water holding capacity also increased, and the textural properties also improved. When Muslim consumers evaluated the sensory attributes of the sausages prepared in this experiment and those currently on the market, the ones prepared in this experiment were preferred over the market products. Based on the above results, chicken sausage for Muslim consumers could be successfully produced, and their overall quality and antioxidant effects could be improved by the addition of biji powder (up to 30%).

Quality Characteristics and Antioxidant Activity of Black Soybean Sunsik Product with Functional Food Ingredients and Functional Labeling System (기능성 원료를 첨가하여 기능성 표시제도를 적용한 검은콩 선식 제품의 품질 특성 및 항산화활성)

  • This study was performed to investigate the quality characteristics and antioxidant activity of black soybean Sunsik product with functional food ingredients and functional labeling system. We prepared black soybean Sunsik (BS) containing black beans, cereals, and vegetables. Black soybean Sunsik with nondigestible maltodextrin and calcium lactate (BSN) was prepared by adding non-digestible maltodextrin and calcium lactate to the base recipe to apply a functional labeling system. The particle size in BS was 118.00 ㎛, whereas BSN was 127.00 ㎛. The respective L, a, and b color values of BS were 73.25, 2.36, and 14.21. The respective L, a, and b values of BSN were 73.21, 2.36, and 14.31. The respective water retention capacities of BS and BSN were 241.67% and 216.33%. No significant difference was observed between BS and BSN in the three physicochemical properties described above. However, the pH of BSN was 5.45, which was significantly lower than that of BS. The total respective phenolic contents of BS and BSN were 1.75 mg GAE/g and 1.61 mg GAE/g, and total respective flavonoid contents of BS and BSN were 6.36 mg RE/g and 5.95 mg RE/g. The antioxidant capacities of BS and BSN were compared via assays of DPPH and ABTS radical scavenging activities, FRAP, and reducing power. The antioxidant activities of BS and BSN increased in a dose-dependant manner. No significant difference between BS and BSN was observed in any measure of antioxidant capacity. These results suggested that the addition of functional food ingredients (non-digestible maltodextrin and calcium lactate) did not affect the quality characteristics and antioxidant activity of black soybean Sunsik.
