The antimutagenic activity of a methanol extract of roasted Cassia tora seed against aflatoxin $B_1(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using Salmonella typhimurium TA100 and or TA98. The MeOH extract was then sequentially partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. The $CH_2Cl_2$ and n-BuOH fractions possessed antimutagenic activity, but the EtOAc and $H_2O$ fractions were inactive. Both the MeOH extract and its fractions were capable of inhibiting the indirect-acting mutagen $AFB_1$, suggesting that these fractions may prevent the metabolic activation of $AFB_1$, or scavenge the electrophilic intermediate capable of inducing mutations. Column chromatography using silica gel yielded pure alaternin from the $CH_2Cl_2$ a fraction, and adenosine and isorubrofusarin gentiobioside from the n-BuOH fraction. Alaternin and isorubrofusarin gentiobioside demonstrated significant antimutagenic activities.
Peroxynitrite formation in vivo is implicated in numerous human diseases and there is considerable interest in the use of antioxidants and natural products for their treatment. The three components (1-3) isolated from Sargassum thunbergii as well as the organic solvent-soluble fractions and the aqueous layer of S. thunbergii were evaluated for their potential to scavenge authentic ONOO$\^$-/ and ONOO$\^$-/ derived from 3-morpholinosydnonimine (SIN-1). The antioxidant activity of the individual fractions was in the order of 85% aqueous (aq.) MeOH>n$\^$-/ BuOH>n-hexane>H$_2$O. The three known compounds, sargahydroquinoic acid (1), sargaquinoic acid (2) and sargachromenol (3) showed peroxynitrite-scavenging activities comparable to those of L-ascorbic acid and penicillamine. These results showed a possible antioxidant activity in major constituents of S. thunbergii.

We investigated the cytoprotective effect of phloroglucinol, which was isolated from Ecklonia cava (brown seaweed), against oxidative stress induced cell damage in Chinese hamster lung fibroblast(V79-4) cells. Phloroglucinol was found to scavenge intracellular reactive oxygen species (ROS) generated by $\gamma-ray$ radiation. In addition, Phloroglucinol inhibited cell damage induced by radiation through scavenging ROS. Phloroglucinol increased the superoxide dismutase and glutathione peroxidase activity, Taken together, the results suggest that phloroglucinol protectes V79-4 cells against oxidative damage by enhancing the cellular antioxidant enzymes activity.
We have studied the cytoprotective effect on H$_2$O$_2$ induced oxidative stress from leaves of Carpinus tschonoskii. The methanol extract of Carpinus tschonoskii was found to scavenge intracellular reactive oxygen species (ROS) using flow cytometry and confocal microscope. This extract prevented lipid peroxidation and thus reduced cell death of Chinese hamster lung fibroblast (V79-4) induced by H$_2$O$_2$ treatment. The extract increased catalase activity and phosphorylation of extracellular signal regulated kinase (ERK). Taken together, the results suggest that Carpinus tschonoskii protects V79-4 cells against oxidative damage by H$_2$O$_2$ through scavenging ROS.
Matrix metalloproteinases (MMPs) are known to play an important role in photoaging by mediating the degradation of extracellular matrix proteins. In this study, to develop a n ew anti-aging agent, we investigated the antioxidant activity and the inhibitory effect of Melothria heterophylla extract on expression of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The M.heterophylla extract was found to scavenge radicals and reactive oxygen species (ROS) with the $SC_{50}$ values of $13{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $20{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA-induced MMP-1 expression was reduced about 80% by $100{\mu}g/ml$ of the M.heterophylla extract but MMP-1 Mrna expression was not inhibited. Therefore, we conclude that the M.heterophylla extract significantly inhibits MMP-1 expression at the protein level. Also, the M.heterophylla extract inhibited MMP-1 activity in a dose dependent manner. From these results, we suggest that the M.heterophylla extract can be used as a new anti-aging agent by antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity.
This work aimed to assess some pharmacological activities of P. leptostachya var. asiatica Hara. The dried roots of P. leptostachya var. asiatica Hara were extracted with 70% ethanol to generate the powdered extract, named PLE. Anti-angiogenic activity was detected using chick chorioallantoic membrane (CAM) assay. In vitro anti-inflammatory activity was evaluated via analyzing nitric oxide (NO) content, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. Antioxidant activity was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and reactive oxygen species (ROS) level in the stimulated macrophage cells. Matrix metalloproteinase-9 (MMP-9) and -2 (MMP-2) activities in the culture media were detected using zymography. PLE exhibits an anti-angiogenic activity in the CAM assay, and displays an inhibitory action on the generation of NO in the LPS-stimulated macrophage cells. In the stimulated macrophage cells, it is able to diminish the enhanced ROS level. It can potently scavenge the stable DPPH free radical. It suppresses the induction of iNOS and COX-2 and the enhanced MMP-9 activity in the stimulated macrophage cells. Both monooxygenase and oxidase activities of tyrosinase were strongly inhibited by PLE. Taken together, the dried roots of P. leptostachya var. asiatica Hara possess anti-angiogenic, anti-inflammatory, antioxidant and skin whitening activities, which might partly provide its therapeutic efficacy in traditional medicine.
To obtain a natural antioxidant from a marine biomass, this study investigated the antioxidative activity of methanolic extracts from the marine brown alga, Ishige okamurae collected off Jeju Island. A potent free radical scavenging activity was detected in the ethyl acetate fraction containing polyphenolic compounds, and the potent antioxidant elucidated as a kind of phlorotannin, diphlorethohydroxycarmalol, by NMR and mass spectroscopic data. The free radical scavenging activities of the diphlorethohydroxycarmalol were investigated in relation to 1,1-diphenyl-2-picrylhydrazyl (DPPH), alkyl, and hydroxyl radicals using an electron spin resonance (ESR) system. The diphlorethohydroxycarmalol was found to scavenge DPPH ($IC_{50}=3.41{\mu}M$) and alkyl ($IC_{50}=4.92{\mu}M$) radicals more effectively than the commercial antioxidant, ascorbic acid. Therefore, these results present diphlorethohydroxycarmalol as a new phlorotannin with a potent antioxidative activity that could be useful in cosmetics, foods, and pharmaceuticals.

Aging is divided into intrinsic aging and photo-aging. Intrinsic aging is naturally occurred as the time passed and photo-aging is induced by the UV radiation of skin. The main reason of aging is the free radicals and the degeneration of the cellular materials by free radicals. In this paper, we checked the anti-aging effects of Mallotus japonicus bark extracts. It has the ability to scavenge free radicals and the SOD like activity. Also, it reduced the cell damage by hydrogen peroxide treatment. Mallotus japonicus bark extracts showed the excellent activity on inhibiting the UV induced cell damage and DNA damage. In conclusion, Mallotus japonicus bark extracts can be used as active ingredients for anti-aging cosmetics.
Aim: Developing antitumor drugs from natural products is receiving increasing interest worldwide due to limitations and side effects of therapy strategies for the second leading cause of disease related mortality, cancer. Methods: The antiproliferative activity of a methanolic extract from the aerial parts of Marrubium persicum extract was assessed with the MCF-7 breast cancer cell line using the MTT test for cell viability and cytotoxicity indices. In addition, antioxidant properties of the extract were evaluated by measuring its ability to scavenge free DPPH radicals. Moreover, the total phenolic and flavonoid content of the extract was determined based on Folin-Ciocalteu and colorimetric aluminum chloride methods. Results: The findings of the study for the antiproliferative activity of the methanolic extract of M. persicum showed that growth of MCF-7 cells was inhibited by the extract in a dose and time dependent manner, where a gradual increase of cytotoxicity effect has been achieved setting out on 200 ${\mu}g/mL$ concentration of the plant extract. The antioxidant assay revealed that the extract was a strong scavenger of DPPH radicals with an $RC_{50}$ value of 52 ${\mu}g/mL$. The total phenolic and flavonoids content of the plant extract was 409.3 mg gallic acid equivalent and 168.9 mg quercetin equivalent per 100g of dry plant material. Conclusion: Overall, M. persicum possesses potential antiproliferative and antioxidant activities on the malignant MCF-7 cell line that could be attributed to the high content of phenolics and flavonoids, and therefore warrants further exploration.
The purpose of this study was to investigate the antioxidant activities of water soluble browning reaction products (WS-BRPs) isolated 5\ulcorneron korea red ginseng. Antioxidant activities of WS-BRPs were examined with the various systems. All three WS-BRPs (L, S-1 and S-2) were found to have an ability to linoleic acid, Ox-brain autoxidant, Fe$^{2+}$ ADP/NAD system and cumene hydroperoxide system. Especially, S-2 was had the strongest activity of theses three WS-BRPs to scavenge free radicals such as more effective than S-1, L. MDA determination showed the antioxidant effect on linoleic acid oxidation inhibition ratio of 22.5%, 31.7%, 31.9% and 33.5%, respectivity Especially; Ox-brain autoxidant was strong inhibited activity by 49.52%,62,44,97.54% by addition of various concentration.
