Comparison in Productivity of Pleurotus ostreatus Sawdust Spawn Under Different Storage Conditions (저장기간에 따른 Pleurotus ostreatus 톱밥 종균의 생산성 비교)

  • The contamination rates of Pleurotus ostratus spawn after 120 days of storage at $5^{\circ}C\;and\;20^{\circ}C$ were 2.1% and 86.5%, respectively. Longer periods of storage resulted in longer culture periods at both temperatures. The yield of oyster mushroom produced from sawdust spawn stored at $5^{\circ}C$ was higher than at $20^{\circ}C$, and yields decreased with increasing storage periods.

The effect of different culture conditions of liquid spawn on the quality characteristics of shiitake mushroom (Lentinula edodes) (표고의 액체종균 배양 조건 및 품질 특성)

  • To improve the productivity of shiitake mushroom (Lentinula edodes), seven different types of media for liquid spawn (denoted as "A" to "G") were prepared with 0.3% soybean meal and varying sugar and glucose concentrations. During 14 days of incubation, the pH of the liquid culture gradually acidified with increasing incubation period. Additionally, there was a significant, but not prominent, difference in the degree of acidification depending on the sugar to glucose ratio. Liquid spawn culture "G," which had the highest sugar content was the most acidic on the last day of incubation. Mycelium dry weight increased significantly with increasing incubation period, and there was no significant difference in mycelium dry weight irrespective of the sugar to glucose ratio even after 14 days of culture. The inoculation of liquid spawn in sawdust medium with an inoculation volume ${\geq}45mL$ and incubation period of 15 to 18 days were the optimal culture conditions. Productivity of fruit bodies in sawdust medium and mushrooms treated with liquid spawn was significantly higher compared to solid spawn treatment. The mushrooms treated with liquid spawn had better chewiness, and the free amino acid content, which is associated with savory taste, was higher in these mushrooms compared to those treated with solid spawn.

  • Cauliflower mushroom(Sparassis crispa) contains much amount of ${\beta}$-glucan, which make lots of farmer want to cultivate the mushroom. Since a practical cultivation method is not provided yet, the mushroom is considered as a difficult crop to deal with. In this study, we tried to develope a simple method to cultivate the mushroom by use of coniferous sawdust-based medium with wheat flour and molasses. There was no significant differences between the sawdust spawn and the liquid spawn for the mycelial growth of the mushroom. The cold shock in $4^{\circ}C$ for a day was thought to be the best way to seduce primordium formation. The sawdust medium of Pseudotsuga menziesii mixed with wheat flour, corn chip, cottonseed meal and 10% molasses was showed the best yield with 41% followed by that of Larix leptolepis mixed with the same additives with 37% of yield. There was good relations between the fruit-body production and the weight loss of the sawdust substrate.

Mycelial growth and wood decaying enzymatic activity analysis by various addition rates of oak powder in the liquid spawn of Lentinula edodes (참나무분 첨가에 따른 표고 액체종균의 균체생산 및 효소 활성)

  • This study was carried out to establish a suitable method for liquid spawn production from Lentinula edodes. The optimum production of liquid spawn (OLS) was achieved using soybean meal medium (SMM) with 0.3% of 850 um oak powder and 10-day incubation period and 0.6 vvm aeration volume. OLS showed activities of laccase on ABTS agar plate and carboxymethyl cellulase (CM-cellulase) on CMC agar plate. In case of liquid spawn, fruiting-body development period was delayed approximately 1 day compared to that of sawdust spawn, however, the yield of 153 g per 1.2 kg polypropylene bag was similar to that of sawdust spawn.

Studies on Improvement of Artificial Cultivation and Antioxidative Activity of Poria cocos (복령의 인공 재배법 개선과 항산화활성에 관한 연구)

  • This study was carried out to improve an artificial culture techniques and antioxidative activity of the crude extract isolated from sclerotia of Poria cocos(Fr.) Wolf. In the test of different spawns and inoculation method, the sclerotia formation, number of sclerotia and production yield were excellent in the both sides inoculation method of log spawn, whereas the both side inoculation method of sawdust spawn was poor in sclerotia formation and yield. The optimal spawn and inoculation method for the quality and productivity of P. cocos was in the order of log spawn (both sides inoculation > log spawn(cutting section inoculation) > sclerotia (both sides inoculation) > sawdust spawn (both sides inoculation). The physiological activity substance, crude extract content of P. cocos NIAST 13007 was about 83%. As the concentration of crude extracts increased, the relative viscosity tended to be increased. However, as the concentration of sodium chloride increased, the relative viscosity did not affected. In antioxidative activities, electron donating ability (EDA) of P. cocos was about 10% of butylated hydroxytoluene (BHT). Thiobarbituric acid (TBA) value was similar to that of the vitamin C, however the peroxide value (POV) was lower than those of BHT and vitamin C.

Development of mushroom spawn production technology of low price by the superior bag spawn (저비용 우량봉지종균 생산기술 개발)

  • The word spawn is derived from an old French verb, espandre, meaning to spread out or expand. Spawn is also defined as "the mycelium of fungi, especially of mushrooms grown to be eaten, used for propagation". The effects of bag spawn to sawdust substrate on the growth of Pleurotus ostreatus were conducted. The duration of mycelial growth and days of pinhead formation of bag spawn(2.5kg) were 18~19days and 7~8days, whereas bottle spawn ($1,000m{\ell}$) was 18days and 6days, respectively. The yield of mushroom fruitbody was that bag spawn is 100~118g, bottle spawn is 95~115g. In economical analysis, bag spawn is increased to 50%, compared to bottle spawn in relative income.

  • Studies were made to optimize the media composition in bag culture and conducted to determine the possibility of artificial cultivation of Auricularia auricula. Sawdust spawn of media composition for optimal growth were found to be oak-sawdust 80%combination of 20% popla-sawdust were the best of the optimal combination. And optimal substrate combination were found to be cotton-seed meal combination of 10% wheat bran 5% mixed were the best of combination. The duration of spawn run period and primordial formation period on bag(1.2 kg) were 50 days and 7 days, respectively. The weight of fruiting body and the yield(100ea, 1.2 kg) of fresh fruit-body were 24 g and 45,000 g, respectively.

Comparison of Growth Characteristics of Ganoderma lucidum Based on Media and Environmental Conditions (배지 및 환경조건에 따른 영지버섯의 생육특성 비교)

  • In order to determine the possibility of artificial cultivation of Ganoderma lucidum, a study was carried out to optimize the medium composition used for bed culture. Sawdust spawn of medium composition for optimal growth was found to be the combination of 75% oak-sawdust, 10% cotton-seed meal, and 15% beet pulp. The optimal conditions for the formation of fruiting bodies were shown to be a temperature of $28-30^{\circ}C$, light intensity of 1,000-1,500 lux, and $CO_2$ concentration of 1%.

  • The recycling method of enokitake cultural waste and the potentiality of second flush for enokitake were determined, because this fungus is not as prolific as the more commonly cultivated white rot fungi in the conversion of sawdust to mycelial mass. The mycelial growth of F. velutipes on several substrates, variously treated with rice bran was promoted at ratios of 10-20% (w/w) on all substrates, but suppressed at above ratios, although some difference was there. The mycelial densities generally increased correlated to the supplementation contents of rice bran. It could be concluded that F. velutipes preferred mild acidic to acidic conditions for mycelial growth, considering that the mycelial growth rate was highest on waste of pH 6.01, treated with 0.1 % Ca(OH)$_2$ and on populus mixed waste of pH 6.02, non treated. The ranges of substrate bulk densities, which was pertinent for mycelial linear growth were from B.D. (g/cc) 0.17 to 0.23 on waste and populus mixed waste all. The pertinent contents of rice bran supplementation in bottle cultivation was from 20 to 30% on waste and 20% on populus mixed waste, considering the requried duration for pinheading and fruiting yields. Standard bulk density for filling and utilizing the waste and populus mixed waste for commercial f. velutipes cultivation were B.D.(g/cc) 0.19 ~ 0.23, and 0.23~ 0.25, which could be conversed to 510~ 540g/900m1 and 520~ 570g/900m1, respectively, The second flush of F. velutipes was tried and the re-inoculation by sawdust and liquid spawn showed somewhat good results, indicating the potentiality of second crop and suggesting further research for it.

Cultural Characteristics and Fruitbody Formation of Phellinus gilvus (Phellinus gilvus의 배양적 특성과 자실체 형성)

  • For artificial cultivation of Phellinus gilvus (Schw.) Pat we have conducted a study on cultural characteristics and condition of fruitbody formation. The optimum temperature was about $30^{\circ}C$ at pH $6.0{\sim}7.0$ for mycelial growth. Optimum sawdust media were oak sawdust+willow sawdust(5:5, V/V), oak sawdust+willow sawdust+rice bran (4.5:4.5:1, V/V) and oak sawdust+pine sawdust+rice bran(4.5:4.5:1, V/V) and, the spawn incubation period was about $25{\sim}26$ days. Mycelial growth in the inner portion of oak log was 200 mm after 60 days and duration for the first fruitbody primordia were formed about 90 days after inoculation.
