톳(Hizikia fusiforme)과 모자판(Sargassum fulvellum) 에탄올 및 물 추출물의 아질산염 소거작용 (Nitrite Scavenging Activity of The Ethanol and Water Extracts Obtained from Hizikia fusiforme and Sargassum fulvellum)

  • The purpose of this study was to investigate the nitrite scavenging activity of the ethanol and water extracts obtained from Hizikia fusiforme and Sargassum fulvelum using sodium nitrite under different pH conditions. The nitrite scavenging activity was appeared in all the extracts examined and it showed the highest value at pH 1.2 The ethanol extract of Hizikia fusiforme provided higher activity than the water extract of that. Ohterwise, the nitrite scavenging activity of Sargassum fulvelum ethanol extract was similar to that Sargassum fulvelum water extract. There was significant difference among concentration of extracts Hizikia fusiforme and Sargassum fulvelum. Also, nitrite scavenging activity of the extracts Hizikia fusiforme and Sargassum fulvelum decreased with higher pH condition. In addition, the activity was increaed with higher concentration of extracts Hizikia fusiforme and Sargassum fulvelum in the course of reaction time 30 minute to 3 hour.

모자반추출물의 기능성 화장품소재 특성 (Functional Cosmetic Effect of Sargassum Extracts)

  • 전세계적으로 천연화장품에 대한 관심과 수요가 크게 늘어나고 있다. 따라서 기존 화학합성 기능성화장품소재를 대체하기 위한 노력의 일환으로서 천연물에서 기능성화장품소재를 발굴하고자 하였다. 모자반은 우리나라의 해안 전지역에서 쉽게 볼 수 있는 갈조류의 일종으로서 식용으로도 이용되는 만큼 안전성이 확보되어있는 해조류이다. 본 연구에서는 큰잎 모자반(Sargassum coreanum). 짝잎 모자반(Sargassum hemiphyllum), 쌍발이 모자반(Sargassum patens)의 3가지 종류의 모자반으로부터 열수를 이용하여 유효성분을 추출하였으며, 항산화효과, 미백효과, 주름개선효과, 자외선흡수효과 및 항염증효과를 측정하여 기능성 화장품 소재로서의 응용 가능성을 살펴보고자 하였다. 모자반추출물은 세포독성시험결과 안전성이 매우 우수하였고, DPPH radical scavenging activity를 이용한 항산화능 측정결과 우수한 항산화능을 보여주었으며 tyrosinase inhibition을 이용한 미백효과시험에서는 큰잎 모자반(Sargassum coreanum)의 미백효과가 상당히 우수하였다. 그러나 주름개선효과, 자외선흡수효과와 항염증효과는 비교적 낮았다. 결론적으로 큰잎 모자반(Sargassum coreanum)은 세포독성이 적었고, 항산화효과와 미백효과가 우수하여 미백 기능성화장품소재로서 응용가능성이 높음을 알 수 있었다.

해조류 추출물과 페놀성화합물의 in vitro 및 in vivo 간보호활성 (Hepatoprotective Effect of Extracts and Phenolic Compound from Marine Algae in Bromobenzene-treated Rats)

  • The methanol extracts of some marine algae were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rat. The extracts of Enteromorpha compressa, Capsosiphon fulvescens, Gelidium amansii, Hizikia fusiformis, Sargassum siliquastrum and Sargassum thunbergii which decreased the formation of lipid peroxide, inhibited the activity of xanthine and aldehyde oxidases by adding of each extracts. Phloroglucinol isolated from Ecklonia stolonifera reduced bromobenzene-induced hepatic lipid peroxidation. This compound administered daily over one week before intoxication with bromobenzene did not affect the activities of aminopyrine N-demethylase, aniline hydroxylase and glu tathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of phloroglucinol, The results suggest that the bromobenzene-induced hepatic lipid peroxidation by phloroglucinol is reduced by enhancing the activity of epoxide hydrolase.

과산화아질산염과 DPPH 라디칼에 대한 해조추출물의 소거 활성 효과 (Studies on Screening of Seaweed Extracts for Peroxynitrite and DPPH Radical Scavenging Activities)

  • As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.

In vitro Screening of Seaweed Extract on the Proliferation of Mouse Spleen and Thymus Cell

  • A total number of 31 types of seaweed were assessed with regard to their effects on the proliferation of mouse spleen and thymus cells in a culture, using an MTT reduction assay. Acetone:dichloromethane (1:1) extracts of three seaweed plants: Derbesia marina, Sargassum sp., and Hisikia fuziformis, exhibited significantly positive effects on the survival of mouse spleen and thymus cells in vitro. The acetone:dichloromethane (1:1) extracts of Sargassum sp., in particular, much more potent effects on thymus cell activation than did any of the other types of seaweed. However, the methanol extracts of Sargassum ringgoldianium and Chondrus crispus exerted a stimulatory influence only on the proliferation of mouse spleen cells, whereas the methanol extracts of Grateloupia lanceolata exhibited significant cell proliferation properties in both spleen and thymus cells.

큰열매모자반(Sargassum macrocarpum) 추출물의 항산화 효과 및 세포 활성 효과 (Antioxidant activity and cell bioactivity of Sargassum macrocarpum extract)

  • 본 연구에서는 큰열매모자반 추출물의 식품 및 화장품으로서 이용성을 평가하기 위해 항산화능 및 항염능, 항비만효과를 확인하였다. 항산화실험으로서 폴리페놀, 플라보노이드, DPPH, ABTS, NO, FRAP을 실시하였다. 폴리페놀의 경우 30.81±1.12 mg/g으로 나타났다. 플라보노이드의 경우 25.72±0.94 mg/g으로 나타났다. DPPH 실험에서는 6.746 mg ascorbic acid / g extract의 항산화능을 나타내었으며, ABTS 실험에서는 15.59 mg ascorbic acid / g extract의 항산화능을 나타내었으며, NO 실험에서는 6.781 mg ascorbic acid / g extract의 항산화능을 나타내었다. FRAP에서는 큰열매모자반 추출물의 1 mg이 ascorbic acid 4.573±0.097 ㎍의 환원력을 보였다. 한편 세포실험에서는 큰열매모자반 추출물이 세포독성이 없음을 보였으며, NO 생성 억제능의 경우 25.95±0.85%의 염증 억제능을 보였으며, 29.75±2.35%의 지질 축적 억제능을 보여 큰열매모자반 추출물이 항염능, 항비만능을 가진 이너뷰티 제품 원료로서 사용가능함을 보였다. 향후 연구에서는 큰열매모자반이 포함하는 단일 물질들이 항산화, 항염, 항비만에 어떠한 기전으로 영향을 미치는지 연구할 필요가 있다.

부산 연안 해조류 추출물의 항산화 활성 및 Tyrosinase 저해 활성 스크리닝 (Screening for Antioxidizing and Tyrosinase-inhibitory Activities of the Extracts of Marine Algae from Busan Coastal Area)

  • The crude extracts of marine algae were screened for tyrosinase-inhibitory and radical scavenging effects. Among the samples tested, Symphyocladia latiuscula and Gloiopeltis furcata were found to be the most effective in DPPH radical scavenging test while Gigatina tenella, Sargassum thunbergii, and Sargassum sp. were moderately active. For the inhibition against mushroom tyrosinase, Symphyocladia latiuscula and Sargassum confusum showed the strongest inhibition. Codium adhaerens, Corallina pilulifera, Carpopeltis cornea, Halymenia acuminata, Hizikia fuziformis, Porphyra suborbiculata, and Enteromorpha linza exhibited mild inhibitory potency.

바이러스성출혈성패혈증바이러스에 대한 식물 추출물의 항바이러스 효능 탐색 (Antiviral effects of various plant extracts against viral haemorrhagic septicaemia virus (VHSV))

  • Since viral haemorrhagic septicaemia virus (VHSV) was first reported in European rainbow trout (Oncorhynchus mykiss) in the 1930s, it has caused high prices in freshwater and saltwater fish around the world, causing enormous economic damage to the aquaculture industry. We have been seeking required countermeasures against viruses because of economic damage to the aquaculture industry. However, commercial vaccines have the limitations of being costly to use in farms and being effective to only one pathogen. The aquaculture industry these days is taking on new alternatives to vaccines, antibiotics and chemicals. In this study, the suitability of antiviral effects against VHSV was evaluated in vitro for various plant extracts to judge their effectiveness. Atriplex gmelinii, Ixeris repens, Arctium lappa, and Sargassum coreanum were tested to know the correlation between the amount of virus and the concentration of extract investigates if these extracts have antiviral effects. Virus and extracts at various concentrations were inoculated simultaneously as 1:1 ratio into EPC cell lines. There are no antiviral effects with Atriplex gmelinii, Ixeris repens and Arctium lappa. Extract of Sargassum coreanum only has the antiviral activity in a dose-dependent manner. These results show that extract of Sargassum coreanum can be used in aquaculture industry as an antiviral materials.

쌍발이모자반(Sargassum patens) 추출물의 항산화 및 항염효과 (Antioxidant and Anti-inflammatory activity of Sargassum patens extract)

  • 본 연구에서는 쌍발이모자반 추출물의 항산화능 및 항염능을 확인하였다. 항산화능 실험에는 폴리페놀 농도측정, 플라보노이드 농도 측정, DPPH 실험, ABTS 실험, NO 실험, FRAP 실험을 실시하였다. 폴리페놀의 경우 18.99±0.69 mg/g으로 나타났다. 플라보노이드의 경우 11.89±1.16 mg/g으로 나타났다. DPPH 실험에서는 19.78 mg ascorbic acid / g extract의 항산화능을 나타내었으며, ABTS 실험에서는 63.64 mg ascorbic acid / g extract의 항산화능을 나타내었으며, NO 실험에서는 7.966 mg ascorbic acid / g extract의 항산화능을 나타내었다. FRAP에서는 쌍발이모자반 추출물의 1 mg이 ascorbic acid 2.089 ㎍의 환원력을 보였다. 한편 세포실험에서는 세포 독성과 LPS로 유도된 염증에 대한 항염능을 알아보았다. 세포독성의 경우 모든 농도에서 80%이상의 세포 생존률을 보였으며, NO 생성 억제능의 경우 100 ㎍/mL 농도에서 30.64±0.23%의 염증 억제능을 보여 쌍발이모자반 추출물이 항염능을 가진 화장품 원료로서 사용가능함을 보였다.

Polysaccharide Degrading Enzyme을 이용한 참모자반 효소분해 추출물의 생리활성 연구 (Biological Analysis of Enzymatic Extracts from Sargassum fulvellum Using Polysaccharide Degrading Enzyme)

  • SC092 strain, producing a polysaccharide degrading enzyme, was isolated from the seawater. This strain was identified as Microbulbifer sp. using the comparative sequence analysis against known 16S rRNA sequence. A polysaccharide degrading enzyme from this strain was used to acquire the enzymatic extracts of Sargassum fulvellum. DPPH radical scavenging and SOD activity of the enzyme extracts of S. fulvellum were about 61.9% and 82.9% at 2 mg/mL, respectively. Nitrite scavenging activities was 52.5% at 2 mg/mL on pH 1.2. In addition, ${\alpha}$-glucosidase inhibitory activity was also increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. To determine the influence of enzyme extracts of S. fulvellum on alcohol metabolism, the generating activity of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. ADH and ALDH activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}$-glucosidase inhibitory activity of enzyme extracts of S. fulvellum was remarkably increased in a dose-dependent manner and was about 52.7% at 2 mg/mL. These results indicate alcoholizing and ${\alpha}$-glucosidase inhibitory activities can be enhanced by the enzymatic extracts of S. fulvellum.
