  • In this study, the anti-allergic activity and mechanim of Salviae radix (SR) were investigated. The ethanol extract of SR showed significant inhibitory effect on degranulation from antigen-stimulated mast cells and it also inhibited the expression and secretion of TNF-${\alpha}$ and IL-4 in antigen-stimulated RBL-2H3 cells. In the mast cell-mediated local animal allergy model, it suppressed the passive cutaneous anaphylaxis in a dose-dependent manner. As its mechanism of action, SR inhibited the activating phosphorylation of Syk, a downstream signaling molecule of Src-family kinase, for the activation of mast cells. The results of the study indicate that the anti-allergic activity of SR is mediated by the inhibition of Src-family kinase in mast cells.

  • Salviae Miltiorrhizae Radix has been used for treatment of cardiovascular diseases in oriental medicine. To investigate the possible involvement of cardiac ion channel in this effect, we examined electrophysiological effects of the extract of Salviae Miltiorrhizae Radix on action potentials and ionic currents in rat ventricular myocytes. The extracts of Salviae Miltiorrhizae Radix were fractionated into nine fractions, and the effect of each fraction on action potential was tested. The fraction containing monomethyl lithospermic acid-A (LSA-A) induced a significant prolongation of action potential duration (APD). LSA-B which is a major component of Salviae Miltiorrhizae Radix, however, did not cause a significant effect. In voltage clamp experiments, the effects of LSA-A on K currents, Ca currents and Na currents were tested. Neither K currents nor L-type Ca currents were affected by LSA-A. On the contrary, LSA-A significantly slowed down the inactivation kinetics of the Na current with no effect on the fast component of the inactivation process. The amplitude of the peak current and the voltage-dependence of activation were not changed by LSA-A. The effect of LSA-A on Na current was abolished when high concentration of $Ca^{2+}$ buffer (10 mM BAPTA) was included in the pipette solution or when Ca2+ current was blocked by nicardipine (1 $\mu$M) in the bath solution.n.

  • Cytotoxicity of glucose oxidase(GO) and cardioprotective effect of Salviae Multiorrhizae Radix(SMR) against GO-induced cardiotoxicity were measured for evaluation of cardiotoxicity on cultured mouse pulmonary endotherial cells(PEC) by MTT assay after PEC were cultured for 8 hours at various concentrations of GO. GO was toxic in a time and dose-dependent manner on cultured PEC after PEC were grown for 8 hours in media containing 1~60mU/ml GO. While, cultures were pretreated with 60 μg/ml SMR for 2 hours increased remarkably cell viability. From the above results, it is suggested that GO is toxic on cultured PEC by the decrease of cell viability, and herb medicine such as SMR is very effective in the prevention of vascular toxicity induced by GO.

  • This study was undertaken to determine whether Salviae-radix (SVR) exraction exerts benefical effect against oxidant-induced inhibition of tetraethylammonium (TEA) uptake which is actively secreted by renal proximal tubules. TEA uptake increased as function of incubation time to 60 min. When renal cortical slices were exposed to 50 mM $H_2O_2$, TEA uptake was significantly inhibited. The inhibition was significantly protected by addition of 0.5% SVR extraction. The benefical effect of SVR was dose-dependent over the concentration range of 0.1-1%; $H_2O_2$ (50 mM)-induced inhibition of TEA uptake was completely protected by 0.5-1% SVR extraction. $H_2O_2$ increased LDH release which was accompanied by an increase in lipid peroxidation in renal cortical slices. These changes were prevented by 0.5% SVR. These results suggest that SVR exerts benefical effect against oxidant-induced impairment of membrane transport function, this effect may be due to by an antioxidant action.

  • To evaluate the neurotoxicity of reactive oxygen species (ROS) in cultured cultured spinal dorsal root(DRG) neurons derived from neonatal mouse, Cytotoxicity was measured by MTS assay after cultured cells were grown for 3 hours in the media containing 1~60 μM hydrogen peroxide (H₂O₂). In addition the neuroprotective effect of Salviae Miltiorrhzae Radix (SMR) was measured in these cultrures. Cell viability was positively decreased in a dose- and time-dependent manner after exposure of cultured mouse DRG neurons to 30 tt M H202 for 3 hours. In the neuroprotective effect of SMR on H₂O₂-mediated toxicity, SMR prevented the H₂O₂-induced neurotoxicity in these cultures. From these results. it suggests that H₂0₂ is toxic in cultured mouse spinal motor neurons and selective herb extract such as Uncariae Ramulus Cum Uncis is effective in prevetion of the neurotoxicity induced by H₂O₂.

  • In order to evaluate the cytotoxic effect of reactive oxygen species(AOS), the cell viability was measured by MTT assay after cultured mouse hippocampal neurons were treated with various concentrations of xanthine oxidase(XO) and hypoxanthine (HX) for 5 hours. And also, the protective effect of Salviae Mutiorrhizae Radix(SMR) on XO/HX-induced neurotoxicity was examined in these cultures. XO/HX significantly decreased cell viability in dose-and time dependent manners when cultured mouse hippocampal neurons were treated with 5~40 mU/ml XO for 5 hours. In the protective effect of SMA, SMR increased cell viability dose-dependently after cultured mouse hippocampal neurons were preincubated with 30~120 ㎍/ml SMR for 2 hours. From these results, it is suggested that XO/HX is toxic on cultured mouse hippocampal neurons, and herbe medicine such as SMR is very effective in blocking the cytotoxicity induced by AOS.
