INTERCALATIVE NANOENCAPSULATION OF COSMETIC INGREDIENTS INTO SKIN-FRIENDLY INORGNICS

  • Functional cosmetic ingredients such as L-ascorbic acid, retinoic acid, indole-3-acetic acid, salicylic acid, acidic dye(indigo carmine) are intercalatively encapsulated by skin-friendly metal hydroxides and oxides matrices. Such functional organic-inorganic nanohybrids are realized via chemical coprecipitation and surface coating reactions. The hetero-structural nature of these nanohybrids, their particle morphology and textural characterizations are mainly discussed on the basis of powder X-ray diffraction, electron microscopies, and high performance liquid chromatographic analyses. The cosmetic ingredients encapsulated in inorganics show greatly improved storage stability, sustained releasing property as well as higher transdermal transfer efficiency.

Effect of True Partition Coefficient on the Determination of Extraction Coefficient of Ion-Pair Complexes (이온대 화합물의 참분배계수가 축출정수 결정에 미치는 영향)

  • Whether ($AB_{w}$) may be neglected against ($A_{w}^{+}$) in the calculation of the extraction coefficient of ion-pairs was criticized by both experiments and theoretical consideration, where ($AB_{w}$) and ($A_{w}^{+}$) mean the molar concentration of ion-pair AB and cation $A^{+}$ in the aqueous phase. Ion-pair complexes were partitioned between phosphate buffer (pH 7.4) and n-octanol. Tetrabutylammonium, isopropamide and methylene blue were selected as cations and benzoic acid, p-toluenesulfonic acid, salicylic acid and taurodeoxycholic acid were selected as counter ions (anions). As a result, conventional methods which assume no existence of ($AB_{w}$) were proven to lack generality. The equation proposed in my earlier report was confirmed to be valid as a general method.

  • The endothelial cell of brain capillary called the blood-brain barrier (BBB) has carrier-mediated transport systems for nutrients and drugs. The mechanism of the BBB transport of basic and acidic drugs has been reviewed and examined for endogenous transport systems in BBB in WKY and SHRSP. Acidic drugs such as salicylic acid and basic drugs such as eperisone are taken up in a carrier mediated manner through the BBB via the monocarboxylic acid and amine transport systems. The specific dysfunction for the choline transport at the BBB in SHRSP would affect the function of the brain endothelial cell and brain parenchymal cell. The utilization of the endogenous transport systems of monocarboxylic acid and amine could be promising strategy for the effective drug delivery to the brain.

Simplified the Screening and In Vitro Appraisal of Antioxidant, Cytotoxic, Thrombolytic, Antimicrobial and Membrane Stabilizing Activities of Lablab Purpures at a Time

  • To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.

  • Cinnamyl alcohol dehydrogenase (CAD, EC 1.1.1.95), catalyzes the reduction of hydroxycinnamaldehydes to give hydroxycinnamyl alcohols, or "monolignols," the monomeric precursors of lignin. Lignins are important components of cell walls and lignified secondary cell walls play crucial roles in long distance transport of water and nutrients during plant growth and development and in plant defense against biotic and abiotic stresses. Here a cDNA clone containing a CAD gene, named as PgCAD, was isolated from a commercial medicinal plant Panax ginseng. PgCAD is predicted to encode a precursor protein of 177 amino acid residues, and its sequence shares high homology with a number of other plant CADS. The expression of PgCAD in adventitious roots and hairy roots of P. ginseng was analyzed using reverse transcriptase (RT)-PCR under various abiotic stresses such as salt, salicylic acid, wounding and chilling treatment that triggered a significant induction of PgCAD at different time points within 2-48 h post-treatment. This study revealed that PgCAD may help the plants to survive against various abiotic stresses.

Synthesis of 8-HQR and 8-HQRS Chelate Resins and It's Ion Exchange Properties (8-HQR 및 8-HQRS 킬레이트 수지의 합성과 그의 이온교환 성질)

  • 8-Hydroxyquinoline-resorcinol(8-HQR) and 8-hydroxyquinoline-resorcinol-salicylic acid (8-HQRS) chelate resins were prepared by the condensation reaction of 8-hydroxyquinoline, or 8-hydroxyquinoline-salicylic acid, in the presence of resorcinol with formaldehyde. The ion exchange capacities of 8-HQR and 8-HQRS resins were 4.1 meq/g and 5.4 meq/g, respectively. The adsorption and distribution coefficient of metal ions, such as Fe(III), Cu(II), Pb(II), Co(II) and Ni(II) on these resins were discussed. The adsorption of metal ions on these chelate resins showed that the maximum adsorption condition is pH 7. And the distribution coefficient of metal ions on these resins was increased with decreasing of hydrochloric acid concentration.

Optimization of Extraction Conditions for Functional Components of Roasted Pleurotus eryngii by Microwave-Assisted Extraction (볶음 새송이버섯 기능성분의 마이크로웨이브 추출조건 최적화)

  • Response surface methodology was employed to optimize extraction conditions for finding the maximizing the functional properties of roasted Pleurotus eryngii. Based on the central composite design, independent variables were ethanol concentration ($0{\sim}100%$), extraction time ($1{\sim}9$ min) and microwave power ($25{\sim}125$ W). Soluble solid content, electron donating ability and nitrite-scavenging ability were mainly affected by ethanol concentration, but ACE inhibition activity was largely affected by extraction time. The optimum ranges of extraction conditions resulting from superimposing the response surface were predicted to be ethanol concentration ($25{\sim}50%$), extraction time ($3{\sim}9$ min) and microwave power ($80{\sim}125$ W). Total protein and total phenolic compound content of optimal extracts were 45.80 mg/g and 7.42 mg/g, respectively. In phenolic compounds of roasted Pleurotus eryngii extracts, protocatechuic acid was the highest concentration at 1226.32 ${\mu}g/g$, followed by salicylic acid, catechin, p-hydroxybenzoic acid, caffeic acid, coumaric acid and hesperidin.

Synergic Antimicrobial Activity of Scutellariae Radix, Coptidis Rhizoma and Salicylic Acid Combination against Escherichia coli and Pseudomonas aeruginosa (대장균과 녹농균에 대항하는 황금과 황련 및 살리실산 조성물의 항균상승효과)

  • Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Listeria. monocytogenes and Bacillus cereus are pathogenic bacteria that should not be detected in cosmetics and foodstuffs. Therefore, we first investigated the antimicrobial activities of extracts of Scutellariae Radix(SR), Coptidis Rhizoma (CR) and salicylic acid(SA) in these pathogenic microorganisms. Although SA has been known to exhibit anti-inflammation and antimicrobial activity against pathogenic microorganisms, a high concentration of SA may cause serious side effects such as skin redness, skin burning, peeling or tissue damage. Hence, we focused on diminishing side effects followed by treatment of a high concentration of SA and investigated whether the combinations of SA with various concentrations(25-400 mg/mL), SR and CR with a concentration(100 mg/mL) which did not show antimicrobial activity against E. coli and P. aeruginosa exhibited meaningful antimicrobial effect against both strains. In our results, the combinations of SA with the lowest concentration(25 mg/mL), SR(100 mg/mL) and CR(100 mg/mL) exhibited significant antimicrobial activity against E.coli in comparison to SA alone(25 mg/mL) showing no antimicrobial activity. Moreover, the combinations of SA (100 mg/mL), SR and CR showed seven times higher antimicrobial activity against E. coli than SA alone(100 mg/mL) and exhibited a significant antimicrobial activity in comparison to ampicilin (p<0.05). The combinations of SA(100 mg/mL), SR and CR showed two times higher antimicrobial activity against P. aeruginosa than SA alone. Therefore, these results indicated that the combinations of SR, CR and SA with low concentration expressed the synergistic antimicrobial effect against E. coli and P. aeruginosa and showed great potential as an antimicrobial agent.

The Effect of Salicylic Acid on the Accumulation of $Cd^{2+}$ and Nonprotein-SH Synthesis in Roots and Epidermal Strips of Commelina Communis L. (닭의장풀에 $Cd^{2+}$처리 시 $Cd^{2+}$ 흡수와 nonprotein-SH 합성에 미치는 살리실릭산의 영향)

  • Three weeks old Commelina was cultivated in Hoagland solution with or without $(\pm{100\mu{M}\;Cd^{2+}$ and with $100\mu{M}\;Cd^{2+}+100\mu{M\; SA)$ for 4 days. The contents of $Cd^{2+}$ and nonprotein-SH synthesis in roots and isolated epidermal strips were measured. In the treatment of $Cd^{2+}$, the concentration of $Cd^{2+}$ was $98\mu{M}\;Cd^{2+}$/g.fr.wt and it was 6 times higher than that of control. $Cd^{2+}$+SA increased 1.2 times higher concentration of $Cd^{2+}$ than that of the treatment ofTEX>$Cd^{2+}$ alone. In the treatment of TEX>$Cd^{2+}$, nonprotein-SH also increased 2 times higher than that of the control. TEX>$Cd^{2+}$ + SA increased 2 times higher concentration of nonprotein-SH than that of the treatment of TEX>$Cd^{2+}$ as well. In case of isolated epidermal strips, the response was similar with the roots. Therefore, it suggest that the effect of salicylic acid on the accumulation of TEX>$Cd^{2+}$ and nonprotein-SH synthesis in roots, and isolated epidermal strips was related with an increase of the membrane permeability by SA and that SA may be positively involved in the process of nonprotein- SH synthesis.

Effect of Sorbitol and Salicylic Acid on Quality and Functional Food Contents of Tomato Fruit (Solanum lycopersicum) (솔비톨 및 Salicylic Acid 처리가 토마토 과실의 품질 및 기능성 물질 함량에 미치는 영향)

  • In this study, we aimed to understand the effect of sorbitol and salicylic acid (SA) on quality and functional food contents of tomato. Sole or combined application of sorbitol and SA in different concentrations (0.5 mM and 1.0 mM) greatly improved the characteristics of tomato, such as red color tone, soluble solid content, hardness and weight as well as increasing the functional food contents such as of lycopene, anthocyanins and vitamins in comparison with the control. The cyanidin content increased approximately 52-61% and delphinidin contents also increased 20-24% and 39-41% in tomato plants treated with SA alone and sorbitol with SA, respectively. Furthermore, the sole or combined application of sorbitol and SA also increased the vitamin $B_1$ and C contents. In particular, vitamin C content was increased 1.2-fold by treatment with sorbitol alone and was increased 2-fold by SA treatment compared to the control. In the sorbitol with SA treatments, vitamin C content was approximately 4-fold increased relative to control plants.
