Characterization of Tetracycline Resistant Plasmid in Staphylococcus aureus by Restriction Enzyme Mapping (황색포도상구균에서 테트라사이클린 내성을 나타내는 플라스미드의 동정)

  • The clinical isolate Staphylococcus aureus SA8 was resistant to tetracycline(Tc) and harboured a plasmid pKH1(24.82 kb). pKH1 was shown by curing and by transformation to specify resistance to Tc. The cleavage map of a pKH1 was determined by restricction enzyme mapping techniques. Cleavage map is given for BglII, EcoRI, HpaII, PvuII and SalI. Restriction endonuclease BamHI, BglI, BstEII, HpaI, PstI, and XhoI have no sites on this plasmid. HaeIII, XbaI, and HindIII have 5, 6, 14 sites, respectively.

  • The clinical isolate Staphylococcus aureus SA2 had four kinds of plasmids and was resistant to ampicillin, chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, methicillin, streptomycin, tetracycline and tobramycin. Transformation experiment demonstrated that 4.44 kb plasmid(pKH6) encoded resistance to tetracycline. The cleavage map of pKH6 was determined by restriction enzyme mapping techniques. The cleavage map is given for EcoRV, HindIII, HpaI, HpaII, KpnI and Xbal. Restriction endonucleases BamHl, BglI, BGIII, BstEII, EcoRI, HaellI, PstI, PvuII, SalI, Smal, and Xhol have no site on this plasmid. The restriction map revealed extensive structural homology between pKH6 and pT181.

  • the effect if atmosphere is more important in the West sea of Korea than in other seas because of shallow water and heat storage if the water. The serial oceanographic observation data and coastal station data from NFRID, and the atmosphere data from KMA were used in order to find out the relationship between them The highest water temperature, salinity and weather factor were recorded in Aug, and the lowest of them in Feb. As the water deepens, the maximum time leg in water temperature and the minimum time leg in salinity. Water temperature have the maximum in Oct, the minimum in Apr at 75m of the 311-07 station with 100m depth water temperature (WT)-air temperature, WT-precipitation (Preci.) and salinity (Sal)-wind speed (WS) were in direct proportion, but WT-WS, Sal-AT and Sal-Preci in inverse proportion Water temperature and salinity I-ave time leg at the same depth the maximum had more the delay of $2\~4$ months at a depth if 20 meters than at the surface in all stations except for salinity at 307-05.

  • Variation within the intergenic spacer(IGS) of the ribosomal DNA gene for twenty-two strains of E. oxysporum and its formae speciales was examined by PCR, couped with RELP analysis. The length of the amplified IGS region was about 2.6 kb in all strains except F.oxysporum f. sp. cucumer-inum from Korea and F. oxysporum f. sp. niveum. Those two strains were 2.5 kb long. Restriction digestion of IGS-RELP regions by Eco RI, NruI, HincII, SAlI, SmaI, BalIi, HindIII, XhoI and KpnI gave rise to nine IGS hapoltypes among all strains. Cluster analysis based on the presence of absence of comigrating restriction reagments show the two groups based on 44% genetic similarity. These results demonstrated that analysis of IGS showed some difference within and between F. oxysporum formae speciales.

  • Dioxomolybdenum(VI) complexes, MoO$_2$(H-Sal-R)$_2$, R; arylamines, have been synthesized by reactions of dioxobis (salicylaldehydato) molybdenum(VI) with various primary amines. These complexes have been characterized by electric conductivity and spectroscopic studies. Infrared, uv-vis, and proton nmr spectra show that the complexes are six-coordinated with cis-MoO$_2$ group. And mass spectra indicates that the combining ratios for Mo (VI)-ligand are 1 : 2. They are yellow, stable for a considerably long time in the atmosphere at room temperature and slightly soluble in alcohol, dichloromethane and dimethylformamide but insoluble in benzene, ether and carbon tetrachloride.

  • Cellobiase ($\beta$-glucosidase) is an enzyme of the cellulase system in cellulolytic microor-ganisms. The chromosomal DNA fragment which include cellobiase gene of Cellulomonas biazotea was cloned in Eschericia coli via plasmid pBR 322 vector. Restriction enzyme Sal I was used to obtain adequate size of fragments from C. biazotea. chromosomal DNA. The transformant of E. coli HB101 with recombinant plasmid pBG101 showed cellobiase activity, which is not ordinary in E. coli HB101. The enzyme activity of the transformant was as of 20% lower than that of C. biazotea.

  • In this paper we introduce the concept of $\gamma$-preopen sets in a topological space together with its corresponding $\gamma$-preclosure and $\gamma$-preinterior operators and a new class of topology $\tau_{{\gamma}p}$ which is generated by the class of $\gamma$-preopen sets. Also we introduce $\gamma$-pre $T_i$ spaces(i=0, $\frac{1}{2}$, 1, 2) and study some of its properties and we proved that if $\gamma$ is a regular operation, then$(X,\;{\tau}_{{\gamma}p})$ is a $\gamma$-pre $T\frac{1}{2}$ space. Finally we introduce $(\gamma,\;\beta)$-precontinuous mappings and study some of its properties.

  • The polymeric membrane electrodes based on urea derivative as an ionophore were prepared and studied for the iodide-ion selective electrode. This membrane exhibits a linear stable response over a wide concentration range ($1.0\;{\times}\;10^{-5}\sim1.0\;{\times}\;10^{-2}$) with a slope of -57.7 mV/decade, a detection limit of log[$I^-$] = -5.63, and a selectivity coefficient for iodide against perchlorate anion (log$K^{pot}_{I^-,j}$ = -1.42). The selectivity series of the membrane gives the follow as $I^-$ > $SCN^-$, $Sal^-$ > $ClO_4^-$ > $NO_3^-$ > $Br^-$ > $NO_2^-$ > $Cl^-$ > $F^-$. The proposed electrode showed good selectivity and response for iodide anion over a wide variety of other anions in pH 5.0 buffer solutions.

  • For screening of autoregulator receptor gene from Saccharopolyspora erythraea, PCR was performed with primers of receptor gene designed on the basis of amino acid sequences of autoregulator receptor proteins with known function. PCR products were subcloned into the BamHI site of pUC19 and transformed into the E. coli DH5$\alpha$. The isolated plasmid from transformant contained the fragment of 120 bp, which was detected on 2% gel after BamHI treatment. The insert, 120 bp PCR product, was confirmed as the expected internal segment of gene encoding autoregulator receptor protein by sequencing. Southern and colony hybridization using Saccha. erythraea chromosomal DNA were performed with the insert as probe. The plasmid (pEsg) having 3.2 kbp SacI DNA fragment from Saccha. erythraea is obtained. The 3.2 kbp SacI DNA fragment was sequenced by the dye terminator sequencing. The nucleotide sequence data was analyzed with GENETYX-WIN (ver 3.2) computer program and DNA database. frame analyses of the nucleotide sequence revealed a gene encoding autoregulator receptor protein which is a region including KpnI and SalI sites on 3.2 kbp SacI DNA fragment. The autoregulator receptor protein consisting of 205 amino acid was named EsgR by author. In comparison with known autoregulator receptor proteins, homology of EsgR showed above 30%.
