Yield characteristics of Pleurotus ostreatus according to the use of spent mushroom substrate with high nitrogen content (질소원이 증진된 수확후배지를 이용한 느타리버섯 수량 특성)

  • The aim of this study was to re-use spent mushroom substrate (SMS) with increased total nitrogen (T-N) and amino acid content and reduce the amount of cottonseed meal used as nutrient supplement in Pleurotus ostreatus cultivation. Bacteria used for improvement of the T-N content were GM20-4(Bacillus sp.) and Rhodobacter sphaeroides (RS). GM20-4 was isolated from the SMS of P. ostreatus and RS was obtained from Gwangjusi agricultural technology center. SMS in T1, T2, and T3 was reused as substrate after drying and the T-N content of dried SMS (D-SMS) was increased by 0.34% by treatment with the bacteria. T1 with 8% D-SMS and T2 with 18% D-SMS had higher rates of primordia formation compared with T3 and the control. The biological efficiency of the control and of treatment with 8%, 18%, and 26% D-SMS was 110%, 114%, 112%, and 79%, respectively. Considering the economic cost, yield, and biological efficiency, T2 with 18% D-SMS as the culture substrate for P. ostreatus was shown to be the most effective for cultivation.

Characteristics of Lactate Dehydrogenase Produced from Lactobacillus sp. FFy111-1 as a Ruminant Probiotic (반추동물용 활성제로서 Lactobacillus sp. FFy111-1이 생산한 Lactate Dehydrogenase의 특성에 관한 연구)

  • The objective of this experiment is to study the possibility of lactate dehydrogenase(LDH) enzyme to prevent lactate accumulation in the rumen, For understanding capacity of bacterial LDH in rumen environments, this study was conducted to explore the effects of temperature, pH, VFAs and metal ions on Lactobacillus sp. FFy111-1's LDH activity, and the LDH activation in rumen fluid accumulated lactate. The optimum pH and temperature of LDH were pH 7.5 and 40$^{\circ}C$, respectively. The LDH activity had a good thennostability at range from 30 to 50$^{\circ}C$. The highest pH stability of the enzyme was at ranges from pH 7.0 to 8.0 and the enzyme activities showed above 64% level of non-treated one at pH 6.0 and 6.5. The LDH was inactivated by VFAs treatments but was enhanced by metal ion treatments without NaCl and $CuSO_4$ Especially, the LDH activity was increased to 127% and 124% of its original activity by 2 mM of $BaCl_2$ and $MnSO_4$, addition, respectively. When the acidic rumen fluid was treated by LDH enzyme of Lactobacillus sp. FFy111-1, the lactate concentration in the rumen fluid was lower compared with non-treated rumen fluid(P<0.05). This lactate reduction was resulted from an action of LDH. It was proved by result of purified D,L-LDH addition that showed the lowest lactate concentration among the treatments(P<0.05). Although further investigation of microbial LDH and ruminal lactate is needed, these findings suggest that the bacterial LDH has the potential capability to decrease the lactate accumulated in an acidic rumen fluid. Also, screening of super LDH producing bacteria and technical development for improving enzyme activity in rumen environment are essential keys for practical application.

  • This paper presents a 1.8 GHz six-port-based impedance modulator using CMOS technology, which can select an arbitrary load impedance with switch control. The proposed 1.8-GHz impedance modulator comprises a Wilkinson power divider, three quadrature hybrid couplers, and four SP3T switches for each load impedance selection. The measured insertion loss of -13 dB and the input/output return losses of >10 dB are achieved in the range of 1.4~2.2 GHz. The low drop output regulator for a stable 3.3 V DC power and the serial peripheral interface(SPI) for an easy digital control are integrated. The chip size, including the pads, is $1.7{\times}1.8mm^2$.

Antifungal activity of N-[1-(benzotriazol-1-yl)aryl]arylamine derivatives and quntitative structure-activity relationships(QSAR) (N-[1-(benzotriazol-1-yl)aryl]arylamine 유도체의 항균성과 정량적 구조활성 관계(QSAR))

  • A series of new N-[1-(benzotriazol-1-yl)aryl]arylamine derivatives were synthesized and their antifungal activities $(pI_{50})$ in vitro against Pyricularia oryzae, Fusarium oxysporum f. sp. sesami, Valsa ceratosperma and Botrytis cinerea were dertermined by the agar medium dilution method. From the results of the quantitative structure-activity relationships $(QSAR_S)$ analysis, $hydrophobicity({\pi})$, $electronic({\Sigma\sigma})$ and molar $refractivity({\Sigma}M_R)$ parameter of X & Y-substituents on the phenyl group were also shown to be important factor in determining the variation in the antifungal activity. 4-Bromo group substituents (1d & 2b) were the most effective compounds and the $half-life(T_{1/2})$ on the hydrolysis of X(1) at netural pH was about 1.5 day. Molecular orbital(MO) functions of substrate compound, linear free energy relationships$(LFER_S)$ on the antifungal reactivity arid the results of molecular design were also discussed.

UA Study on the Polarization Selective Antenna for UHF RFID System (UHF RFID 시스템을 위한 Polarization selective 안테나 연구)

  • TIn this paper, it is designed the polarization selective antenna for UHF RFID system. The proposed antenna is consist of microstrip patch antenna with dual feeding and two SPDT switches and a SP4T switch and 3dB hybrid coupler. Through control of voltage of switches, the proposed reader antenna can select horizontally linear polarization, vertically linear polarization, left-hand circular polarization (LHCP) and right hand circular polarization (RHCP). The proposed reader antenna satisfied 2:1 VSWR at 902MHz~928MHz. and it has under 3dB AR(axial ratio). Peak gain of antenna is 7.71dBi, 7.55dBi with linear polarization and 7.31dBic, 7.81dBic with circular polarization at x-y plane. Also Axial ratio of antenna is 2.01~2.83dB and 2.02~2.60dB respectively. It is satisfied 3dB axial ratio.

Genome sequence of Caballeronia sordidicola strain PAMC 26577 isolated from Cladonia sp., an Arctic lichen species (북극 지의류 Cladonia종에서 분리한 Caballeronia sordidicola균주 PAMC 26577의 유전체 서열 분석)

  • Caballeronia sordidicola strain PAMC 26577 was isolated from Cladonia sp., a lichen collected from Svalbard Archipelago in the Arctic Ocean. Draft genomic sequences of PAMC 26577 were determined using Illumina and 182 contigs were submitted to GenBank and N50 value was 159,226. The genome of PAMC 26577 was comprised of 8,334,211 base pairs and %G+C content was 59.4. The genome included 8 ribosomal RNA genes and 51 tRNA genes as non-coding sequences. Protein-coding genes were 8,065 in number and they included central metabolism genes as well as butanol/butyrate biosynthesis, polyhydroxybutyrate metabolism, serine cycle methylotrophy genes, and glycogen metabolism. Membrane transporters were more than two-hundreds in number, but sugar phosphotransferase system and TRAP transporters were lacking. PAMC 26577 lacked CRISPR-associated sequences and proteins. No transposable elements were observed and there were only limited number of phage remnant regions with 11 phage-related genes.

Neocucurbitaria chlamydospora sp. nov.: A Novel Species of the Family Cucurbitariaceae Isolated from a Stink Bug in Korea

  • The fungal strain KNUF-22-18B, belonging to Cucurbitariaceae, was discovered from a stink bug (Hygia lativentris) during the investigation of insect microbiota in Chungnam Province, South Korea. The colonies of the strain KNUF-22-18B were wooly floccose, white to brown in the center on oatmeal agar (OA), and the colonies were buff, margin even, and colorless, reverse white to yellowish toward the center on malt extract agar (MEA). The strain KNUF-22-18B produced pycnidia after 60 days of culturing on potato dextrose agar, but pycnidia were not observed on OA. On the contrary, N. keratinophila CBS 121759T abundantly formed superficial pycnidia on OA and MEA after a few days. The strain KNUF-22-18B produced chlamydospores subglobose to globose, mainly in the chain, with a small diameter of 4.4-8.8 ㎛. At the same time, N. keratinophila CBS 121759T displayed a globose terminal with a diameter of 8-10 ㎛. A multilocus phylogeny using the internal transcribed spacer regions, 28S rDNA large subunit, b-tubulin, and RNA polymerase II large subunit genes further validated the uniqueness of the strain. The detailed description and illustration of the proposed species as Neocucurbitaria chlamydospora sp. nov. from Korea was strongly supported by molecular phylogeny.

Multi-component kinetics for the growth of the cyanobacterium Synechocystis sp. PCC6803

  • The growth kinetics of phototrophic microorganisms can be controlled by the light irradiance, the concentration of an inorganic nutrient, or both. A multi-component kinetic model is proposed and tested in novel batch experiments that allow the kinetic parameters for each factor to be estimated independently. For the cyanobacterium Synechocystis sp. PCC6803, the estimated parameters are maximum specific growth rate $({\mu}_{max})=2.8/d$, half-maximum-rate light irradiance $(K_L)=11W/m^2$, half-inhibition-rate light irradiance $(K_{L,I})=39W/m^2$, and half-maximum-rate concentration for inorganic carbon $(K_{S,Ci})=0.5mgC/L$, half-maximum-rate concentration for inorganic nitrogen $(K_{S,Ni})=1.4mgN/L$, and half-maximum-rate concentration for inorganic phosphorus $(K_{S,Pi})=0.06mgP/L$. Compared to other phototrophs having ${\mu}max$ estimates, PCC6803 is a fast-growing r-strategist relying on reaction rate. Its half-maximum-rate and half-inhibition rate values identify the ranges of light irradiance and nutrient concentrations that PCC6803 needs to achieve a high specific growth rate to be a sustainable bioenergy source. To gain the advantages of its high maximum specific growth rate, PCC6803 needs to have moderate light illumination ($7-62W/m^2$ for ${\mu}_{syn}{\geq}1/d$) and relatively high nutrient concentrations: $N_i{\geq}2.3 mgN/L$, $P_i{\geq}0.1mgP/L$, and $C_i{\geq}1.0mgC/L$.

IN VITRO MICRO-SHEAR BOND STRENGTH OF FIVE COMPOSITE RESINS TO DENTIN WITH FIVE DIFFERENT DENTIN ADHESIVES (미세-전단 결합 강도 시험을 이용한 상아질 접착제와 수복용 복합 레진의 호환성에 관한 연구)

  • The purpose of this study was to compare and to evaluate the combination use of 5 kinds of dentin adhesive systems and 5 kinds of composite resins using micro-shear bond test. Five adhesive systems (Prime & Bond NT (PBN). Onecoat bond (OC), Excite (EX), Syntac (SY), Clearfil SE bond (CS)) and five composite resins (Spectrum (SP), Synergy Compact (SC), Tetric Ceram (TC), Clearfil AP-X (CA), Z100 (Z1)) were used for this study ($5{\;}{\times}{\;}5{\;}={\;}25group$, n =14/group). The slices of horizontally sectioned human tooth were bonded with each bonding system and each composite resin, and tested by a micro-shear bond strength test. These results were analyzed statistically. The mean micro-shear bond strength of dentin adhesive systems were in order of CS (22.642 MPa), SY (18.368 MPa), EX (14.599 MPa). OC (13.702 MPa). PBN (12.762 MPa). The mean bond strength of self-etching primer system group (CS, SY) in dentin was higher than that of self-priming adhesive system groups (PBN, EX, OC) significantly (P<0.05). The mean bond strength of composite resins was in order of SP (19.008 MPa), CA (17.532 MPa). SC (15.787 MPa), TC (15.068 MPa). Z1 (14.678 MPa). Micro-shear bond strength of SP was stronger than those of other composite resins significantly (P < 0.05). And those of TC and Z1 were weaker than other composite resins significantly (P < 0.05). No difference was found in micro-shear bond strength of composite resin in self-etching primer adhesive system groups (CS, SY) statistically. However, there was significant difference of micro-shear bond strength of composite resin groups in self-priming adhesive systems group (PBN, EX, OC). The combination of composite resin and dentin adhesive system recommended by manufacturer did not represent positive correlation. It didn't seem to be a significant factor.

Sargassum sp. Attenuates Oxidative Stress and Suppresses Lipid Accumulation in vitro (모자반추출물의 항산화활성 및 지방세포 생성억제 효과)

  • Oxidative stress causes tissue damage and facilitates the progression of metabolic diseases, including diabetes, cardiovascular heart diseases, and obesity. Lipid accumulation and obesity-related complications have been observed in the presence of extensive oxidative stress. As part of an ongoing study to develop therapeutic supplements, Sargassum sp. were tested for their ability to scavenge free radicals and intracellular reactive oxygen species (ROS), as well as to suppress lipid accumulation. Three species, S. hemiphyllum, S. thunbergii, and Sargassum horneri, were shown to scavenge free radicals in a di(phenyl)-(2,4,6-trinitrophenyl)iminoazanium (DPPH) assay. In addition, Sargassum sp. was shown to scavenge intracellular ROS and to decrease nitric oxide (NO) production in $H_2O_2$ and lipopolysaccharide (LPS)-induced in RAW264.7 mouse macrophages, respectively. Taken together, the results suggest that Sargassum sp. possess huge potential to relieve oxidative stress and related complications, as well as lipid-induced oxidation. They indicate that S. hemiphyllum, S. thunbergii, and S. horneri are potent functional supplements that can produce beneficial health effects through antioxidant and antiobesity activities, with S. hemiphyllum being the most potent among the Sargassum sp. tested. A potential mechanism for the effect of Sargassum sp. on the suppression of lipid accumulation in differentiating 3T3-L1 mouse preadipocytes through deactivation of the peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) is presented.
