Characteristics of the Vegetation Structure of an Abies nephrolepis Forest in the Presence of the Tripterygium regelii Group (미역줄나무 출현에 따른 분비나무림의 식생구조 특성)

  • In this study, the sustainability of Abies nephrolepis in the presence of Tripterygium regelii was analyzed. To accomplish this, a field survey of140 stands was conducted. Based on the presence or absence of T. regelii, as well as the layer in which it was present (where appropriate), the type of T. regelii presence wasclassified as follows: TP type, indicating the presence of T. regelii in the tree and sub-tree layers; SP type, indicating its presence in the shrub layer; and A type, indicatingits absence from all layers. The indicator species analysis revealed T. regelii to be an indicator species of the TP type and SP type. The MRPP test was used to examine the differences among the types. Moreover, the seedling and DBH class analysis indicated there to be a difficulty in terms of the status competition, as the number of young trees generated by A. nephrolepis was the lowest in the TP type. The findings of this study enhanced our understanding of the vegetation structure of A. nephrolepis, thereby contributing to efforts concerningits conservation and management.

Sinomonas terrae sp. nov., Isolated from an Agricultural Soil

  • While searching for the bacteria which are responsible for degradation of pesticide in soybean field soil, a novel bacterial strain, designated 5-5T, was isolated. The cells of the strain were Gram-staining-positive, aerobic and non-motile rods. Growth occurred at 10-42℃ (optimum, 30℃), pH 5.5-9.0 (optimum, pH 7.0-7.5), and 0-2% (w/v) NaCl (optimum, 1%). The predominant fatty acids were C15:0 anteiso, C17:0 anteiso, and summed feature 8 (C18:1 ω7c and/or C18:1 ω6c). The predominant menaquinone was MK-9 (H2). Diphosphatidylglycerol, glycolipids, phosphatidylinositol, and phosphatidylglycerol were the major polar lipids. Phylogenetic analysis of 16S rRNA gene sequences indicated that strain 5-5T is a member of the genus Sinomonas and its closest relative is Sinomonas humi MUSC 117T, sharing a genetic similarity of 98.4%. The draft genome of strain 5-5T was 4,727,205 bp long with an N50 contig of 4,464,284 bp. Genomic DNA G+C content of strain 5-5T was68.0 mol%. The average nucleotide identity (ANI) values between strain 5-5T and its closest strains S. humi MUSC 117T and S. susongensis A31T were 87.0, and 84.3 % respectively. In silico DNA-DNA hybridization values between strain 5-5T and its closest strains S. humi MUSC 117T and S. susongensis A31T were 32.5% and 27.9% respectively. Based on the ANI and in silico DNA-DNA hybridization analyses, the 5-5T strain was considered as novel species belonging to the genus Sinomonas. On the basis of the results from phenotypic, genotypic and chemotaxonomic analyses, strain 5-5T represents a novel speciesof the genus Sinomonas, for which the name Sinomonas terrae sp. nov. is proposed. The type strain is 5-5T (=KCTC 49650T =NBRC 115790T).

Acinetobacter pullorum sp. nov., Isolated from Chicken Meat

  • A bacterial strain, designated B301T and isolated from raw chicken meat obtained from a local market in Korea, was characterized and identified using a polyphasic taxonomic approach. Cells were gram-negative, non-motile, obligate-aerobic coccobacilli that were catalase-positive and oxidase-negative. The optimum growth conditions were 30℃, pH 7.0, and 0% NaCl in tryptic soy broth. Colonies were round, convex, smooth, and cream-colored on tryptic soy agar. Strain B301T has a genome size of 3,102,684 bp, with 2,840 protein-coding genes and 102 RNA genes. The 16S rRNA gene analysis revealed that strain B301T belongs to the genus Acinetobacter and shares highest sequence similarity (97.12%) with A. celticus ANC 4603T and A. sichuanensis WCHAc060041T. The average nucleotide identity and digital DNA-DNA hybridization values for closely related species were below the cutoff values for species delineation (95-96% and 70%, respectively). The DNA G+C content of strain B301T was 37.0%. The major respiratory quinone was Q-9, and the cellular fatty acids were primarily summed feature 3 (C16:1 ω6c/C16:1 ω7c), C16:0, and C18:1 ω9c. The major polar lipids were phosphatidylethanolamine, diphosphatidyl-glycerol, phosphatidylglycerol, and phosphatidyl-serine. The antimicrobial resistance profile of strain B301T revealed the absence of antibiotic-resistance genes. Susceptibility to a wide range of antimicrobials, including imipenem, minocycline, ampicillin, and tetracycline, was also observed. The results of the phenotypic, chemotaxonomic, and phylogenetic analyses indicate that strain B301T represents a novel species of the genus Acinetobacter, for which the name Acinetobacter pullorum sp. nov. is proposed. The type strain is B301T (=KACC 21653T = JCM 33942T).

Isolation and Identification of Antioxidant-producing Marine Bacteria and Medium Optimization. (항산화 물질을 생산하는 해양 미생물의 분리.동정 및 배양 특성 조사)

  • For the research of the natural marine antioxidant, several bacteria were isolated from the coast of jin-Hae in Korea. Among the marine bacteria studied, strain HJ-14, a gram-negative, motile, strait rod, aerobic, and $Na^{+}$ required bacterium showed high activity of 1,1-diphenyl-2-picrylhydrazyl radical scav- enging. The morphological, physiological, and biochemical characteristics of the strain HJ-14 were similar to those of the Alteromonas macleodii ATCC $27126^{T}$ . Thus, it was tentatively identified as Alteromonas sp. HJ-14. The compositions of major fatty acids in cell membrane of Alteromonas sp. HJ-14 were $C_{ 14:0}$ , $ C_{15:0}$ , $C_{16:0}$ and $C_{17:1}$ $_{w8c}$ , which also suggest that it is affiliated with Alteromonas sp. The optimum culture conditions for production of antioxidant materials with Alteromonas sp. HJ-14 were at $25$~$37^{\circ}C$ and pH 6~8. The optimum conditions for the production of antioxidant for carbon, inorganic nitrogen, and sodium chloride sources were 2.5%(w/v) dextrin, 0.5%(w/v) ammonium sulfate, and 2~6%(w/v) sodium chloride, respectively. The hydroxyl radical scavenging ability of Alteromonas sp. HJ-14 broth was 90.03%, which is higher than ascor-bic acid(83.28%) and lower than butylated hydroxyanisole(95.46%) and $\alpha$-tocopherol(97.17%).

S100A8 Induces Secretion of MCP-1, IL-6, and IL-8 via TLR4 in Jurkat T Cells

  • In the pathogenesis of inflammatory diseases such as allergies, S100A8 acts as an important molecule and T lymphocytes are essential cytokine-releasing cells. In this study, we investigated the effect of S100A8 on release of cytokines, specifically MCP-1, IL-6, and IL-8 in T cells, and its associated signaling mechanism. S100A8 increased secretion of MCP-1, IL-6, and IL-8 in a time- and dose-dependent manner. Elevated secretion of MCP-1, IL-6, and IL-8 due to S100A8 was inhibited by the TLR4 inhibitor TLR4i, the PI3K inhibitor LY294002, the $PKC{\delta}$ inhibitor rottlerin, the ERK inhibitor PD98059, the p38 MAPK inhibitor SB202190, the JNK inhibitor SP600125, and the NF-${\kappa}B$ inhibitor BAY-11-7085. S100A8 induced phosphorylation of ERK, p38 MAPK, and JNK in a time-dependent manner, and activation was suppressed by TLR4i, LY294002, and rottlerin. S100A8 induced NF-${\kappa}B$ activation by $I{\kappa}-B{\alpha}$ degradation, and NF-${\kappa}B$ activity was suppressed by PD98059, SB202190, and SP600125. These results indicate that S100A8 induces cytokine release via TLR4. Study of PI3K, $PKC{\delta}$, MAPKs, and NF-${\kappa}B$ will contribute to elucidation of the S100A8-invovled mechanism.

A Study on the Primary Parasites in the Green Peach Aphid, Myzus persicae (Homoptera : Aphididae), and Its Hyperparasites (복숭아흑진딧물에 기생(寄生)하는 진디벌에 관(關)하여)

  • A biological study was done on primary parasites and hyperparasites of green peach aphid, Myzus persicae, collected in Taejeon area during the period from April to September 1985 was. The results of mummification of primary parasites, host selectivity between primary parasites and hyperparasites, and longevity of important species were obtained as follows; 1. Among the 201 mummies collected in the field, adult primary parasites and adult hyperparasites were 38.3% and 44.3%, respectively. 2. Among the primary parasites, Lysiphlebia japonica and Trioxys hokkaidensis were most important species. 3. Among the hyperparasites, Prataphelinus nikalskajae, Eucoila sp. and Ardiles convexa were dominant species. 4. L. japonica was more parasitized by the hyperparasites than T. hokkaidensis. 5. T. hokkaidensis was higher than L. japonica in the ability of parasitism. 6. L. japonica had lived for 3 days by feeding glucose, the other foods had lived shorter than food.

Isolation and Identification of Bacteria Removing Sulfur Compound Odor (황 화합물계 악취 제거 미생물의 분리 및 동정)

  • We isolated 50 strains from sludge of wastewater treatment aeration tank using selective medium for Thio-bacillus sp. by membrane filtration method. They were identified as Thiobacillus neapolitanus (7), T. tepidarius (4), T. dientrificans (5), T. versutus (23), T. intermedius (2) and T. perometabolis (9). We selected Thiobacillus versutus strain KT51, which had the highest removal efficiency (100%) of hydrogen sulfide and the highest removal efficiency (85%) of dimethyl sulfide for 30 min in screen test. Also Thiobacillus versutus strain KT81 had the highest removal efficiency (26%) of dimethyl disulfide for 30 min in screen test. In applification of lab-scale reactor (closed-biological treatment) using Thiobacillus versutus strain KT51, results were 99.8% (<0.02 ppm) removal efficiency of hydrogen sulfide for 15 min.

Halobacillus blutaparonensis sp. nov., a Moderately Halophilic Bacterium Isolated from Blutaparon portulacoides Roots in Brazil

  • A moderately halophilic, Gram-positive, spore-forming bacterium was isolated from the roots of Blutaparon portulacoides, a plant found in sandy soil parallel to the beach line in Restinga de Jurubatiba, Rio de Janeiro, Brazil. The strain, designated $M9^T$, was motile and strictly aerobic with rod-shaped cells. It grew in the absence of NaCl and up to 20% NaCl, and was able to hydrolyze casein and starch. Strain $M9^T$ had a cell-wall peptidoglycan based on L-Orn-D-Asp, the predominant menaquinone present was menaquinone-7 (MK-7), diaminopimelic acid was not found, and anteiso-$C_{15:0}$ and iso-$C_{15:0}$ were the major fatty acids. A phylogenetic analysis based on 16S rRNA gene sequences showed that strain $M9^T$ belonged to the genus Halobacillus and exhibited 16S rRNA gene similarity levels of 97.8-99.4% with the type strains of the other nine Halobacillus species. The DNA-DNA relatedness of strain $M9^T$ with H. trueperi, the closest relative as regards 16S rRNA gene similarity, and H. locisalis was 21% and 18%, respectively. Therefore, on the basis of phenotypic, genotypic, and phylogenetic data, strain $M9^T$ (=ATCC BAA-$1217^T$, =CIP $108771^T$, =KCTC $3980^T$) should be placed in the genus Halobacillus as a member of a novel species, for which the name Halobacillus blutaparonensis sp. nov. is proposed.

Arthrobacter subterraneus sp. nov., Isolated from Deep Subsurface Water of the South Coast of Korea

  • Strain $CH7^T$, a pale yellow-pigmented bacterium and new isolate from deep subsurface water of the South Coast of Korea, was subjected to a polyphasic taxonomic study. $CH7^T$ grew between 5 and $37^{\circ}C$, pH 5.3-10.5, and tolerated up to 13% NaCl. A phylogenetic analysis based on 16S rRNA gene sequences showed that strain $CH7^T$ was associated with the genus Arthrobacter and phylogenetically closely related to the type strains Arthrobacter tumbae (99.4%) and Arthrobacter parietis (99.1%). However, DNA-DNA hybridization experiments revealed 2.1% and 12% between strain $CH7^T$ and Arthrobacter tumbae and Arthrobacter parietis, respectively. Thus, the phenotypic and phylogenetic differences suggested that $CH7^T$ should be placed in the genus Arthrobacter as a novel species, for which the name Arthrobacter subterraneus sp. novo is proposed. In addition, the type strain for the new species is $CH7^T$ ($=KCTC\;9997^T=DSM\;17585^T$).

Isolation and Identification of Lactobacillus kimchicus sp.nov and Bioconversion of Ginseng Saponin

  • Ginseng contained many different kinds of saponin which was the most valuable for people, but its yield cannot satisfy the demand using traditional extract methods. Enzyme transformation is a conformable and highly performed method which was fit for today. A ${\beta}$-glucosidase producing bacterium ($DCY51^T$) was isolated from Korean fermented-vegetable food kimchi. The 16S rRNA gene sequence analysis revealed that the strain $DCY51^T$ belongs to the genus Lactobacillus. The highest sequence similarity was found with Lactobacillus paracollinoides LMG $22473^T$ and Lactobacillus collinoides LMG $9194^T$ with levels of 16S rDNA similarity of 97.4% and 97.3%, respectively. Based on the above results the strain $DCY51^T$ placed in the genus Lactobacillus and proposed a new species, Lactobacillus kimchicus sp. nov. $DCY51^T$ (= KCTC $12976^T$ = JCM $15530^T$). It was culture solution reacted with Red Ginseng extract and $Rb_1$, respectively. The medium of bacteria was the liquid of MRS, the temperatures of growing and reacting between bacteria liquid and saponin were samely $37^{\circ}C$, there spective reacting time were 12 hours and 48 hours. Thus we got different saponins, and TLC and HPLC analysis showed that: enzyme respectively reacted with $Rb_1$ and Red Ginseng extract got the transformed saponin, respectively. The polarity position in TLC was a little higher than Rd; and the polarity position was the same as that of Compound K's, the saponin obtained from HPLC and other experimental results was not Compound K. The constitution of its saponin was hoped to be further confirmed.
