The effects of exogenous spleen cells on the progesterone and insulin like-growth factor-I (IGF-I) secretions in luteal cells were studied by using in vitro luteal cell culture system in the Hanwoo luteal cells. The corpora lutea(CL) were collected and pooled from the Korean native cattle(Hanwoo) ovaries from a local slaughter house. After enzymatic dissociation, combined large and small luteal cells(LLC and SLC)(1.0$\times$10$^{6}$ cells/$m\ell$) were incubated in D-MEM media containing antibiotics and 10% FCS. Spleen cells (1.0$\times$10$^{6}$ cells/$m\ell$) obtained from castrated adult male Hanwoo were added to luteal cells and co-cultured for 24 h in the absence or presence of luteinizing hormone (LH) (100 ng). Progesterone contents from luteal tissues were increased at CL-3 stage during each stage of estrous cycle. Progesterone secretion from luteal cell culture by the presence of LH (100 ng/$m\ell$) was positively stimulated compared with control. However, progesterone secretion was not changed by the addition of 5, 10 and 20% of spleen cells in the absence of LH. Co-culture of luteal cells with 10% of spleen cells in the presence of LH(l00ng/$m\ell$) significantly. enhanced after 24 h of culture. IGF-Isecretion from in vitro luteal cells co-culture by the addition of spleen cells (5%, 10% and 20%) was not significantly effected. Besides, in the presence of LH (100ng/$m\ell$), IGF-Isecretions from luteal cells by addition of spleen cells were higher than control media. However, LH alone significantly increased IGF-I secretion at 24 h of culture. These data provide the demonstrate that spleen cells can enhance LH action so as to stimulate progesterone secretion from Hanwoo luteal cells but have no effect to stimulate IGF-I secretion.

Minerals is required small amounts among various nutrients, but it has a significant impact on sow longevity and reproduction performance. This study was carried out to see the beneficial effects of marine-derived Ca-Mg complex on the reproductive performance of sows during four-parity periods. Seventy-two gilts ([Yorkshire × Landrace] × Duroc), with an average body weight of 181 kg, were randomly allocated to three groups; CON (basal diet), 0.3LC (CON - MgO - 0.3% limestone + 0.4% Ca-Mg complex), and 0.7LC (CON - MgO - 0.7% limestone + 0.4% Ca-Mg complex). During parity 3 and 4, the expression level of SCD gene was lower in the umbilical cord of piglets born to 0.3LC and 0.7LC sows compared with the CON sows. During parity 2, 3 and 4, SLC2A2 and FABP4 gene expressions were higher in the umbilical cord of piglets born to 0.7LC sows and the placenta of sows from 0.3LC groups, respectively. Ca-Mg complex increased (p < 0.05) Ca and Mg concentrations in sows and their piglets' serum as well as in colostrum regardless of parities. The serum vitamin D concentration was higher (p < 0.05) in their first parity, whereas serum prolactin and estrogen concentrations were higher (p < 0.05) during the fourth and third parity, respectively. The growth hormone concentrations were higher (p < 0.05) in the piglets born to sows during the first and second parity. The fat and immunoglobulin A (IgA) concentrations in colostrum were higher (p < 0.05) during the third and fourth parity, respectively. A reduction (p < 0.05) in salivary cortisol, epinephrine, and norepinephrine concentrations was observed in 0.3LC and 0.7LC sow groups compared with CON after farrowing regardless of parity, however before farrowing, a reduction in norepinephrine was observed. Before farrowing, the epinephrine and norepinephrine concentrations were higher (p < 0.05) during the first and second parity. After farrowing, the concentration of these hormones was higher during the second parity. Taken together, sows' parity and dietary Ca-Mg complex supplementation influenced serum metabolites, colostrum nutrients, stress hormones as well as the gene expressions related to lipid and glucose metabolism.
Genome sequencing of the pig is being accelerated because of its importance as an evolutionary and biomedical model animal as well as a major livestock animal. However, information on expressed porcine genes is insufficient to allow annotation and use of the genomic information. A series of expressed sequence tags of 5' ends of five full-length enriched cDNA libraries (SUSFLECKs) were functionally characterized. SUSFLECKs were constructed from porcine abdominal fat, induced fat cells, loin muscle, liver, and pituitary gland, and were composed of non-normalized and normalized libraries. A total of 55,658 ESTs that were sequenced once from the 5′ ends of clones were produced and assembled into 17,684 unique sequences with 7,736 contigs and 9,948 singletons. In Gene Ontology analysis, two significant biological process leaf nodes were found: gluconeogenesis and translation elongation. In functional domain analysis based on the Pfam database, the beta transducin repeat domain of WD40 protein was the most frequently occurring domain. Twelve genes, including SLC25A6, EEF1G, EEF1A1, COX1, ACTA1, SLA, and ANXA2, were significantly more abundant in fat tissues than in loin muscle, liver, and pituitary gland in the SUSFLECKs. These characteristics of SUSFLECKs determined by EST analysis can provide important insight to discover the functional pathways in gene networks and to expand our understanding of energy metabolism in the pig.

Background and objective: Pravastatin has been shown to have favorable risk-benefit profile when it is administered to hypercholesterolemic subjects to prevent cardiovascular events. However, subjects with impaired OATP1B1 activity may be more susceptible to pravastatin-induced muscle toxicity than subjects with normal OATP1B1 activity. A systematic review was conducted to evaluate the effect of SLCO1B1 genetic polymorphism on pharmacokinetics of pravastatin. Method: Medline$^{(R)}$ and Embase$^{(R)}$ were searched for relevant studies until July 2013. The search terms used were pravastatin AND (SLCO1B1 OR OATP1B1 OR LST1 OR SLC21A6) AND (gene OR $genetic^*$ OR $genomic^*$ OR $pharmacogenet^*$ OR $pharmacogenom^*$ OR $polymorph^*$). Results: A meta-analysis of the area under the concentration-time curve (AUC) of pravastatin in $SLCO1B1^*15$ and $SLCO1B1^*1a/^*1a$ was conducted. Five studies met all the inclusion criteria and methodological requirements. There was no statistically significant difference in the AUC value between $SLCO1B1^*15$ and $SLCO1B1^*1a/^*1a$ (p=0.728). However, $SLCO1B1^*15$ participants exhibited significantly higher AUC values than $SLCO1B1^*1b/^*1b$ carriers (p<0.001). In case of $SLCO1B1^*15^*15$ carriers, they had significantly higher AUC value than $SLCO1B1^*1a/^*1a$ subjects (p=0.002). Lastly, compared with to the subjects of $SLCO1B1^*1a/^*1a$, the carriers of heterozygous $SLCO1B1^*15$ increased the AUC value of pravastatin statistically significantly in Asian population (p=0.014). Conclusion: The present meta-analysis suggests that subjects with $SLCO1B1^*15$ are associated with increased AUC of pravastatin.

It has been reported that Chungkukjang, one of Korean traditional fermented soybean products, may improve hypertension, diabetes, and hyperlipidemia. In this study, we sought to investigate the immunoenhancing effects of Chungkukjang in ovariectomized mice. For the first period, female SLC ddy mice were either sham-operated (Sham; n=27) or ovariectomized (OVX; n=27). As a basal diet, ovariectomized mice were fed low-calcium diet for faster induction of osteoporosis for six weeks, and those in the Sham group were fed AIN-76 diet. For the second period, half of the OVX group (n=9) and the Sham group (n=9) were fed a Chungkukjang-based diet (CKJ); whereas the other half (OVX; n=9/ Sham; n=9) were fed a casein-based diet (CSI) for 8 weeks. After a second period, we collected the blood via heart puncture and measured the splenocytes proliferation, T lymphocyte subsets by flowcytometry, and levels of serum cytokines (IL-2, IL-4, IL-6, IL-10, IFN-${\gamma}$ and TNF-${\alpha}$) by ELISA assay. The OVX+CKJ group showed higher splenocytes proliferation, higher ratio of CD4/CD8, and lower levels of IL-6 and TNF-${\alpha}$ cytokines compared to the OVX+CSI group. The Sham+CKJ group showed cytokine productions, such as higher levels of IL-10 and IFN-${\gamma}$, and lower levels of IL-6 and TNF-${\alpha}$ compared to the Sham+CSI group. The result of this study suggests that Chungkukjang may lower the proinflammatory cytokine levels in both the OVX and Sham groups. In addition, Chungkukjang could make a balance of T cell subset proliferations and enhance the splenocyte proliferations in the OVX group.
To determine the possibility of solidified se wage sludge for use as a soil cover material in reclaimed land, the growth of energy crops and soil chemical properties investigated in each experimental plots during 2 years (2010 and 2011). The experimental plots consisted of the mixing with solidified sewage sludge plot (SS50), the covering with solidified sewage sludge plot (SS100), and the original reclaimed land plot (ORL) on reclaimed land for the intended landfill in Sudokwon Landfill Site Management Corporation (SLC). Plant height, measured in the second year (2011), was highest in the Geodae 1 grown at plots treated with solidified sewage sludge. The growth of energy crops cultivated in both SS50 and SS100 were better than in ORL. The contents of organic matter (OM) and total nitrogen (T-N) at both SS50 and SS100 were considerably higher than that of the ORL over 2 years. However, the soil from ORL showed higher salinity with high contents of exchangeable $Na^+$ cation than that of SS50 and SS100 over 2 years. We consider that soil chemical and physical properties on reclaimed land used in this study could be improved by the application of solidified sewage sludge due to following reasons. Firstly, the application of solidified sewage sludge may provide soil nutrients on reclaimed land i.e. the growth of energy crops better than in ORL, resulted in more OM and T-N contents in SS50 and SS100. Secondly, the top layers mixed or covered with solidified sewage sludge on reclaimed land may be prevented the salinity accumulation due to capillary rise to surface soil, and improved the cultivation layer for effectively propagating the rhizomes of energy crops. Thus the solidified sewage sludge may be a great soil cover materials for cultivation of bioenergy crops in reclaimed land.
This study was carried out to obtain the basic data for selecting the cultivatable bioenergy crops through application of solidified sewage sludge in reclaimed lands. The experimental plots consisted of the mixing with solidified sewage sludge plot (SS50), the covering with solidified sewage sludge plot (SS100), and the original reclaimed land plot (ORL) on reclaimed land for the intended landfill in Sudokwon Landfill Site Management Corporation (SLC). The growth of energy crops (Geodae-Uksae 1, Miscanthus sacchariflorus, and Phragmites australis) were investigated from May to October, 2010 in each experimental plot. The soil from ORL showed higher salinity with high contents of exchangeable $Na^+$ cation than that of SS50 and SS100. Soil properties on reclaimed land used in this study must be improved by increasing the buffering capacity of saline with the treatment of solidified sewage sludge due to the fact that the contents of organic matter (OM) in both of SS50 and SS100 were higher than that of the ORL. Thus the growth of energy crops cultivated in the solidified sewage sludge plots were better than in ORL. Geodae-Uksae 1 which showed an excellent adaptability on reclaimed land treated with the solidified sewage sludge has considerably higher biomass than those of other energy crops (M. sacchariflorus and P. australis). This study suggested that Geodae-Uksae 1 is the most suitable biomass feedstock crop for bioenergy productions, and the solidified sewage sludge may be possible to utilize as a soil cover materials for cultivation of bioenergy crops in reclaimed land.
