Rapid Determining for Subtypes and Pandemic Type of Swine Influenza Virus by Diagnostic One-step RT-PCR (진단용 one-step RT-PCR을 통한 돼지 인플루엔자 바이러스의 아형 및 pandemic 유형에 대 한 신속한 결정)

  • Swine influenza virus (SIV) causes one of the most common diseases of the pig population, and its subtypes are determined by hemagglutinin (HA) and neuraminidase (NA). Recently, the SIV subtype diagnosis has been developed. The method using antigen-antibody reaction rather than PCR was mainly used because of the large change in the ribonucleotide sequences of SIV. Here, we have developed 10 diagnostic primer sets through multi-nucleotide sequences alignment of spreaded SIV since 2008 in Korea and then optimized the reaction of the one-step RT-PCR for rapid determination of SIV subtype. In addition, specific primers were designed to early determine the pandemic SIV by detecting unique M sequences proven in highly infectious and virulent subtypes of the influenza H1N1 (pH1N1). Here, some of the SIVs spread in Korea from 2008 to 2014 have been tested to determine the subtypes and pandemic potential of SIV. All diagnostic primer sets were found to be able to accurately determine the SIV subtype and to detect the pandemic SIV. In conclusion, it was confirmed that the optimized one-step RT-PCR analysis using these primer sets is useful for rapid diagnosis of SIV subtypes. These results can be used for development of SIV subtype diagnostic kit to early detect before virulent SIV spreads do.

  • The SIV(Static InVerter) of the electric coach mainly consists of GTO type inverter. But, it is the drawback that GTO type inverter must have the complicated driving circuits. Recently, appearing the IGBT of high speed switching frequency, simple driving circuits, relative large capacity, GTO type inverter is changed IGBT type inverter. In this paper, IGBT type SIV substituting for the existing GTO type SIV is designed and producted. The experimental results are presented to verify the performance of the designed IGBT type SIV.

  • 표면영상유속계(SIV, Surface Image Velcimetry)는 수표면의 영상분석을 통해 유속을 측정하는 도구이다. SIV는 하천의 유속을 매우 간편하게 측정할 수 있기 때문에, 야외 실험이나 하천의 유속 측정에 매우 유용한 장비이다. 그러나, SIV를 이용하여 유량을 산정하고자 할 경우, 하천 표면의 평면 측량 자료와 하천의 단면 측량 자료가 반드시 필요하다. 그러나, 측량 작업 특히 강우 중의 측량 작업은 매우 번거롭기 때문에, SIV의 간편성과 유용성에도 불구하고, 이용자들로 하여금 SIV를 쉽게 이용하기 어렵다는 그릇된 인식을 줄 수 있다. 만일 번거로운 측량을 거치지 않고도 효율적이고 간편하게 하천의 평면을 추정할 수 있다면, SIV를 마치 일반적인 프로펠러 유속계처럼 쉽게 이용할 수 있을 것이며, 그 적용성도 크게 증진될 것이다. 본 연구는 카메라 영상 모형을 유도하는 것이다. 이 카메라 모형을 이용하여 번거로운 측량 작업이 없이 평면의 좌표점을 추정할 수 있게 되며, 유속장 측정을 자동화할 수 있을 것이다. 이를 위하여 평면 좌표와 참조점을 관련짓는 사영 변환과 회전 변환 등 사진 측정 기법을 도입하였다.

Outcomes of endoscopic retrograde cholangiopancreatography in patients with situs inversus viscerum

  • Background/Aims: Situs inversus viscerum (SIV) is a congenital condition defined by left-to-right transposition of all visceral organs. This anatomical variant has caused technical challenges in endoscopic retrograde cholangiopancreatography (ERCP). Data on ERCP in patients with SIV are limited to case reports of unknown clinical and technical success rates. This study aimed to evaluate the clinical and technical success rates of ERCP in patients with SIV. Methods: Data from patients with SIV who underwent ERCP were retrospectively reviewed. The data were collected by querying the nationwide Veterans Affairs Health System database for patients diagnosed with SIV who underwent ERCP. Patient demographics and procedural characteristics were collected. Results: Eight patients with SIV who underwent ERCP were included. Choledocholithiasis was the most common indication for ERCP (62.5%). The technical success rate was 63%. Subsequent ERCP with interventional radiology-assisted rendezvous has increased the technical success rate to 100%. Clinical success was achieved in 63% of cases. Among cases of subsequent rendezvous ERCP after conventional ERCP failure, clinical success was achieved in 100%. Conclusions: The clinical and technical success rates of ERCP in patients with SIV were both 63%. In patients with SIV in whom ERCP fails, interventional radiology-assisted rendezvous ERCP can be considered.

  • Surface Image Velocity (SIV) is a technique which measures the surface velocity of river by using the principle of Paticle Image Velocimetry (PIV). The technique is economical and efficient way to measure velocity in rivers. The present paper aims to apply the technique to three rivers in Korea. It uses pairs of river surface images taken with two digital-cameras and reference points and cross section data which were acquired through plane survey. The performance of SIV was verified with automatic cart on an experimental flume. The test revealed that average error was less than 10 %, which assures that SIV can be used to measure velocity accurately. When it was applied to rivers with low water levels or rough weather condition, however, it showed the error about 20 %. If the problems of SIV technique are settled down, it can be one of the most convenient and economical ways to measure water discharge anytime and anywhere. And then it would be helpful to river management as developing a real-time river information system.

Optimalization of ELISA using Recombinant p27 Protein of SIV for Detection of Anti-SIV (SIV의 p27 재조합 단백질을 이용한 SIV 항체 검출을 위한 ELISA의 최적 조건)

  • The p27 coding region of the SIVmac239 isolate was amplified by PCR and cloned into an expression vector, pMAL-cri, which expressed high levels of the p27 protein from Escherichia coli. The purified p27 protein was used for detection of anti-SIV antibodies with the sera from 11 macaques and 21 marmosets by immunoblot assay of which one macaque was suspicious for the SIV infection. The optimum conditions of ELISA was studied by the check board system with the recombinant purified p27 protein. For the plate coating, 200ng/well of the purified p27 was satisfactory. The conjugate was diluted 1:1000. The sera from the 32 monkeys were negative for the anti-SIV by ELISA.

  • Seven transmembrane segment (7TMS) receptors for chemokines and related molecules have been demonstrated to be essential, in addition to CD4, for HIV and SIV infection. The beta-chemokine receptor CCR5 is the primary, perhaps sole, coreceptor for HIV-1 during the early and chronic phases of infection, and supports infection by most primary HIV-1 and many SIV isolates. Late-stage primary and laboratory-adapted HIV-1, HIV-2, and SIV isolates can use other 7TMS receptors. CXCR4 appears especially important in late-stage HIV infection; several related receptors can also be used. The specificity of SIV viruses is similar. Commonalities among these receptors, combined with analyses of mutated molecules, indicate that discrete, conformationally-depenclent sites on the chemokine receptors determine their association with the third variable and conserved regions of viral envelope glycoproteins. These studies are useful for elucidating the mechanism and molecular determinants of HIV-1 entry, and of inhibitors to that entry.

The prevalence of swine influenza viral antigens and serum antibodiesin Piglets in Jeju (제주지역 돼지에서 Influenza 바이러스 항원 및 혈중 항체 조사)

  • Ninety pigs under the age of 120-day-old requested at the diagnostic laboratory of animal diseases in Cheju National University were evaluated for the prevalence of tissue antigen and serum antibody to swine influenza virus (SIV). For histopathologic examination there was sampled at the consolidated area in cranioventral or dorsocaudal lobes of lungs. Lung tissues from all pigs were tested for the antigen of SIV type A by immunohistochemistry (IHC). Sera from 56 pigs were used for the antibody detection to SIV type A (subtype H1N1 and H3N2) by haemagglutinin inhibition test. Pneumonic lesions were observed in 72 cases (80%) of 90 pigs. Broncho-interstitial or interstitial pneumonia were more prevalent than suppurative or fibrinous bronchopneumonia. According to HI test, 46.4% of the tested sera showed seropositive. Positive sera were consisted with 5.3% for SIV H1N1, 28.6% for SIV H3N2, and 12.5% for both subtype to be tested, respectively. SIV antigens were detected in 51 cases(56.6%) of 90 pigs. Most SIV antigens were presented in the epithelium of the bronchi and bronchiole. Necrotizing bronchitis or bronchiolitis were observed in 28(31.1%) cases of all inspected pigs. These results suggested that SIV might be an important role to induce swine pneumonia in Jeju. Also IHC was very useful for the detection of SIV in the lung.

Sero-prevalence of swine influenza virus (H1N1, H3N2) In Korea (국내의 돼지 인플루엔자 바이러스(H1N1, H3N2)의 혈청학적 조사)

  • Swine influenza is an acute, infectious respiratory disease caused by type A influenza viruses in pigs. In the previous studies, serological surveys have indicated the presence of H3N2 swine influenza virus (SIV) since 1995 in Korea. And the percentage of the antibody-positive rate was 39.12% in the survey determining the prevalence of H1N1 SIV antibodies in 2002. The purpose of this study was therefore to investigate the sero-prevalence of SIV regard to the age of the pig and the season between June 2004 and May 2005. In this study, a total of 932 sera were used. These sera were randomly selected from blood samples, which were submitted to Department of Veterinary Pathology, Kangwon National University and Department of Veterinary Virology, Seoul National University from June 2004 to May 2005. These sera have been tested by ELISA test kit (IDEXX Lab, USA) for the SIV H3N2, H1N1 respectively. SAS version 9.1 was used for the statistical analysis based on the age of the pig and the season. The overall sero-prevalence of the antibody against H3N2 SIV was 20.82% (194/932). The overall sero-prevalence of the antibody against H1N1 SIV was 37.23% (347/932). The overall dual sero-prevalence of the antibody against H3N2 and H1N1 SIV was 10.62% (99/932). H3N2 has significant difference in statistically regarding the age of the pig and the season (p<0.0001). H1N1 has significant difference in statistically regarding the age of the pig (p<0.0001) but has not significant difference in statistically regarding the season (p=0.5882).

Comparative Analysis of Day and Night Time Video Accuracy to Calculate the Flood Runoff Using Surface Image Velocimeter (SIV) (표면영상유속계(SIV)를 활용한 홍수유출량 산정 시 주·야간영상의 정확도 비교분석)

  • This study analyzed the velocimetry of runoff and measured the flood discharge by applying the SIV (Surface Image Velocimetrer) to the daytime and nighttime flow image data with special reference to Seong-eup Bridge at Cheonmi stream of Jeju during the flow by the severe rainstorm on May 27, 2013. A 1000W lighting apparatus with more than 150 lux was installed in order to collect proper nighttime flow image applied to the SIV. Its value was compared and analyzed with the velocity value of the fixed electromagnetic wave surface velocimetry (Kalesto) at the same point to check the accuracy and applicability of the measured velocity of flow. As a result, determination coefficient $R^2$ values were 0.891 and 0.848 respectively in line with the velocity distribution of the daytime and nighttime image and the flow volume measured with Kalesto was approximately 18.2% larger than the value measured with the SIV.
