Spontaneous Peripheral Ameloblastic Odontoma in a Male Sprague-Dawley Rat

  • Peripheral ameloblastic odontoma is a rare variant of odontogenic tumor occurring in the extraosseous region. The present report describes a spontaneous tumor in male Sprague-Dawley (SD) rats. The clinically confirmed nodule in the right mandibular region was first observed when the rat was 42 weeks and remained until the terminal sacrifice date when the animal was 48 weeks of age. At necropsy, a well demarcated nodule, approximately $2.5{\times}2.0{\times}2.0cm$, protruded from the ventral area of the right mandible. The nodule was not attached to mandibular bone and was not continuous with the normal teeth. Histopathologically, the tumor was characterized by the simultaneous occurrence of an ameloblastomatous component and composite odontoma-like elements within the same tumor. The epithelial portion formed islands or cords resembling the follicle or plexiform pattern typical of ameloblastoma and was surrounded by mesenchymal tissue. Formation of eosinophilic and basophilic hard tissue matrix (dentin and enamel) resembling odontoma was observed in the center of the tumor. Mitotic figures were rare, and areas of cystic degeneration were present. Immunohistochemically, the epithelial component was positive for cytokeratin AE1/AE3 (CK AE1/AE3), and the mesenchymal component and odontoblast-like cells were positive for vimentin, in the same manner as in normal teeth. On the basis of these findings, the tumor was diagnosed as a peripheral ameloblastic odontoma in an extraosseous mandibular region in a SD rat. In the present study, we report the uncommon spontaneous peripheral ameloblastic odontoma in the SD rat. We also discuss here the morphological characteristics, origin, histochemical, and immunohistochemical features for the diagnosis of this tumor.

Acute Toxicity Study of DA-5018, A Non-narcotic Analgesic Agent (비 마약성 진통제 DA-5018의 급성독성시험)

  • Intravenous and oral acute toxicity tests in ICR mice and SD rats and percutaneous acute toxicity tests in SD rats and NZW rabbits were conducted to evaluate the toxicity of DA-5018 and DA-5018 cream, respectively Clinical signs observed in mice and rats after the administration of DA-5018 were similar regardless of administration route. The observed clinical signs were jumping, wild running, lacrimation, ataxia, reddening of extremities and ears, ventral or lateral recumbency, respiratory distress, cyanosis, convulsion and death. Pulmonary enlargement and hemorrhage were observed in the animals died immediately after the dosing of DA-5018. At terminal necropsy, pulmonary enlargement and hemorrhage, corneal opacity and focal scabbing and depilation around nose were seen. LD$_{50}$ Values of DA-5018 are 11.5 mg/kg (mice, male), 12.6 mg/kg (mice, female), 88.3 mg/kg (rat, male) and 73.2 mg/kg (rat, female) in oral toxicity tests and 11.0 mg/kg (mice, male), 18.7 mg/kg (mice, female), 0.12 mg/kg (rat, male) and 0.32 mg/kg (rat, female) in i.v. toxicity tests. In the percutaneous acute toxicity tests of DA-5018 cream, no deaths occured in all the tested groups during 14-day observation period. There were also no abnormalities in the general conditions, body weight changes and on necropsy findings in all groups. LD$_{50}$ values of 0.1 ~0.9% DA-5018 creams in male and female rats and rabbits are >2000 mg/kg./kg.

The Effect of Shinchulsan on the Rat Model with Allergic Rhinitis (神朮散이 알레르기 비염 유발 흰쥐 모델에 미치는 영향)

  • Objectives : Allergic rhinitis is highly increased in these days. We aimed to investigate therapeutic effect ofShinchulsanby observing changes in blood cells and the protein expression level of IL-4 in spleen of Sprague-Dawley(SD) rats with allergic rhinitis.Methods : Fifteen SD rats were divided into three groups: normal, control, and sample group. Allergic rhinitis was induced in the control and sample group by intraperitional and intranasal sensitization with 0.1% and 0.4% Ovalbumin solution. ThenShinchulsanwas orally administered only to the sample group for 28 days, while the rats in the control group was given normal saline.Results : 1.Shinchulsanshowed significantly decreased eosinophil level on the serum of the rat model.2.Shinchulsanshowed significantly decreased IgE level on the serum of the rat model.3.Shinchulsanshowed significantly decreased the protein expression levels of IL-4 in spleen of the rat model.4.Shinchulsanhad no hepatoxicity, according to AST and ALT on the serum.Conclusion : According to the above results, it is considered thatShinchulsanis helpful in treatment of allergic rhinitis.

Composition and Functionality of Chonma (천마의 일반 성분과 기능성 조사)

  • As the production of chonma became increased by the farmer's cultivation, developments of the processed food such as tea using chonma have been actively pursued. In the present study, the components of chonma and its beneficial effects on health using SD (Sprague-Dawley) rat model were analyzed. The contents of moisture, protein, ash, fat and fiber in dried-chonma were 11.8, 7.6, 3.2, 0.5, and 3.9%(w/w), respectively. The contents of calcium, sodium, iron, phosphorus, magnesium and potassium were 121, 83, 6.2, 170, 69 and 1,278 mg%. When chow diets containing 0, 0.15, 1.5 or 5.0% chonma powder were fed to SD rats for 4 weeks, no significant differences were observed in the composition of the large-intestinal flora, ${\beta}-glucuronidase$ level of the large-intestinal contents and the weight gains of rats. The level of ${\beta}-glucosidase$ was higher and the serum cholesterol level was lower in 5.0% chonma group compared with control group. The highest sedative effect was shown in 0.15% chonma group.

Inhibitory effects of abietic acid in testosterone-induced benign prostatic hyperplastic rats (송진 유래 abietic acid가 전립선 비대증 모델 rat에 미치는 영향)

  • Objectives : Currently, the benign prostatic hyperplasia (BPH) is one of the most common urogenital disorder in old men. We were performed to determine the effects of abietic acid (AC), component of pine resin, in benign prostatic hyperplastic Sprague-Dawley rat (SD rat) induced by testosterone injection (IP). Methods : We monitored body weights in SD rat at start and end date of experiment. After end of experiment, the prostate weights were measured in SD rats. Glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) levels was performed in serum. And we determined the 5-alpha reductase Ⅱ activity, testosterone levels, and dihydrotestosterone levels in prostate tissue and serum using ELISA kit. Results : As results, the prostate wights were increased in BPH group compared to normal group and were decreased in fina, AC30, and AC 50 groups, respectively. Serum GOT levels were decreased in AC50 group compared to BPH group. And Serum GPT levels of AC30 and AC50 groups were lower than BPH group. In addition, the 5-alpha reductase Ⅱ activity, testosterone levels, and dihydrotestosterone levels were decreased the fina, AC10, AC30, and AC 50 groups contrast to the BPH group. Furthermore, 5-alpha reductase Ⅱ activity, testosterone levels, and dihydrotestosterone levels were decreased dose dependent in AC groups compared to BPH group. Conclusion : These results suggest that AC could be used as a potential material for the treatment of BPH by decreasing the androgen levels in benign prostatic hyperplasia model rats.

Studies on Hepatic and Renal Toxicities Possibly Induced by Administration of Farfarae Flos in Rats (관동화 및 관동화 가미방이 흰쥐의 간독성 및 신독성에 미치는 영향)

  • Objectives This study is to investigate whether oral administration of Farfarae flos, JCT (Jeong-cheon-tang), CPT (Cheong-pye-tang) and CPK (Cheong-pye-tang- ga-kwandonghwa) will affect both the levels of serum GOT, GPT, blood urea nitrogen (BUN) and creatinine in SD rats or not, and will change body weight of the rats. Materials and Methods 18 SD rats were randomly divided into 6 groups - including a control group (vehicle 2 ml/rat), Farfarae flos water extract (extract 2 ml/rat), Farfarae flos fine powder (emulsion 2 ml/rat), JCT (extract 2 ml/rat), CPT (extract 2 ml/rat) and CPK (extract 2 ml/rat) group. The drugs were administered to rats for 2 weeks or 30 days (for control and Farfarae flos fine powder group only) and serum GOT/GPT activities and BUN/creatinine concentrations were measured. Also, the changes of body weights of each rat was measured. Results (1) Farfarae flos water extract, Farfarae flos fine powder emulsion, JCT and CPT and CPK did not cause any changes in serum GOT/GPT activities and BUN/creatinine concentrations compared to the ones in control group. (2) There are no significant changes in rats' body weight among the experimental groups during the experimental period. Conclusions Contrary to the reports on human data, 2 weeks or 30 days of oral administrations of Farfarae flos itself and decoctions containing Farfarae flos did not provoke hepatotoxicity and nephrotoxicity.

Toxicity assessment of the palatinose orally administered to Sprague-Dawley rats for 14 consecutive days (팔라티노스의 Sprague-Dawley rats에서 14일 반복 경구투여 독성 평가)

  • In order to examine the toxicity of palatinose-L (Pal-L) bioconverted from sucrose, we performed a 14-consecutive day toxicity study with male and female Sprague-Dawley (SD) rats. We recorded clinical signs of toxicity, body weight, organ weights, hematology, blood biochemical, urinalysis, histological changes in organs, such as the liver and kidneys, and clinical chemistry analysis data for all SD rats. There were no significant changes in food/water consumption, body weight, and organ weights during the experimental period. Although there were some hematologic and urinalysis alterations, these changes were not considered toxicologically significant. In addition, histopathological examination of the liver and kidneys revealed no abnormal or toxicological changes between the control and Pal-L-treated rats of both sexes. Collectively, these results suggest that Pal-L was not indicated to have any toxicity in the SD rats when it was orally administered up to a dose of 1,000 mg/kg/day for 14 days.

Radiation Protection Effects of Dendranthema Zawadskii Var. Latilobum (Maxim.) Kitam. Extracts on Blood Cells, Intestine, and Uterus of Female SD Rats Irradiated with Gamma-Ray 10 Gy (구절초 추출물이 감마선 10 Gy에 조사된 암컷 SD Rat의 혈구 및 소장, 자궁에 미치는 방사선 방호효과)

  • The purpose of this study is to see the radiation protection effect of the oral injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts on the small intestine and uterus of female SD Rat as a natural radiation protection agent. The experimental group was divided into four groups: Normal Control group (NC group), Injected Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts group (DZ group), irradiated group after injecting Dendranthema zawadskii var. latilobum (Maxim.) Kitam. extracts (DZ+IR group). The whole body of SD Rat was irradiated with gamma-ray 10Gy, and the administration of oral Dendranthema zawadskii var. latilobum (Maxim.) Kitam. Extract was 2 cc (71.56 mg/day/kg) once a day for 2 weeks. For this study, chages in blood cell levels, SOD assay, small intestine and uterus were observed. In the 21st white blood cell level, the DZ+IR group recovered to a normal level, and the IR group didn't. The IR group villus length was lower than other groups on Day 1. IR group was partially recovered, and DZ+IR group was recovered like the NC group on Day 21. In the case of the first-day endometrium, the IR group was thin and the boundary was cloudy, and the DZ+IR group was thicker and the boundary was clearer than the IR group. Day 21 IR group still did not recover, and DZ+IR group recovered like NC group. This is believed to have radiation protection effects in the blood cells and small intestine and uterus of the irradiated female SD Rat, and is expected to be useful for the study of natural radiation protection materials.

Re-Examination of Blood Vessels of Rat Tail Using Angiographic and Histological Methods (혈관조형술과 조직학적 방법을 이용한 랫드 꼬리의 혈관구조 규명)

  • Although the tail vessels are frequently used for serial blood sampling in rat, the definition of its vasculature remains in dispute. Herein, we investigated the number of blood vessels in Sprague-Dawley rat tail using angiographic and histological methods. Our results showed rat tail has one dorsal vein, two lateral veins, one ventral artery and one ventral vein. In the conclusion, when deciding which site is best for collecting blood, it is critical to consider the structure and features of the blood vessels to be used. This study will also be helpful for investigators to understand the structure of blood vessels in rat tail.
