Identification of Glutathione Conjugates of 2, 3-Dibromopropene in Male ICR Mice

  • Hepatotoxic potential of 2, 3-dibromopropene (2, 3-DBPE) and its conjugation with glutathione (GSH) were investigated in male ICR mice. Treatment of mice with 20, 50, and 100 mg/kg of 2, 3-DBPE for 24 h caused elevation of serum alanine aminotransferase and aspartate aminotransferase activities. The hepatic content of GSH was not changed by 2, 3-DBPE. Meanwhile, the GSH content was slightly reduced when mice were treated with 2, 3-DBPE for 6 h and significantly increased 12 h after the treatment. Subsequently, a possible formation of GSH conjugate of 2, 3-DBPE was investigated in vivo. After the animals were treated orally with 20, 50, and 100 mg/kg of 2, 3-DBPE, the animals were subjected to necropsy 6, 12, and 24 h later. A conjugate of S-2-bromopropenyl GSH was identified in liver and serum treated with 100 mg/kg of 2, 3-DBPE by using liquid chromatography-electrospray ionization tandem mass spectrometry. The protonated molecular ions $[M+H]^+$ of S-2-bromopropenyl GSH were observed at m/z 425.9 and 428.1 in the positive ESI spectrum with a retention time of 6.35 and 6.39 min, respectively. In a time-course study in livers following an oral treatment of mice with 100 mg/kg of 2, 3-DBPE for 6, 12, and 24 h, the 2, 3-DBPE GSH conjugate was detected maximally 6 h after the treatment. The present results suggested that 2, 3-DBPE-induced hepatotoxicity might be related with the production of its GSH conjugate.

  • Protein malnutrition of children is one of the most serious nutritional deficiencies in developing country. Urea nitrogen excretion in ureotelic animals is the function most sensitive to dietary protein. The 24 hours excretion of creatinine in the urine of a given subject is remarkably constant from day to day. The creatinine excretion of different individuals of the same age and sex is also quite constant. Low ratios of urinary urea to creatinine are found children low protein intake. The foregiving world-wide investigations indicate that the urea nitrogen/creatinine ratios seems to be a good biochemical indicator to distinguish among group with different levels of protein intake. The purpose of this study is to evluate an indicator of protein intake on the elementary school children ranged from 6 to 8 years of age living in rural and urban areas. Each child measured for height and weight of body. weight measured by means of a plate from scale and height by a vertical measuring rod. Biochemical test were taken from a finger-tip and urine. Hemoglobin level in the blood was measured by cyanomethemoglobin method. From the urine samples, urea nitrogen and urea creatinie were determined by Folin-Wu method and: calculate the ratio. The following result were obtained: 1) Mean of the body weight and height in urban children(Seoul) was higher and heavier than rural children(Kyunggi, Kangwon). And 12% of boys, 18% of girls in Kyunggi and 25% of boys, 22% of girls in Kangwon area weight less than 80% of Korean Physical Standard weight level. 2) The mean hemoglobin values of boys and girls in Seoul are children were 13. 3g/100ml, 13.1g/100ml and the mean of hemoglobin values in Kyunggi 12.9g/100ml of boys, 12.4g/100ml of girls, and 12.4g/100ml of boys, 12.9g/100ml of girls in Kangwon children. It is found that 22% to 24% children inrural area (Kyunggi, Kangwon) had hemoglobin level less than 12g/100ml which means anemia. 3) The mean of hematocrit level of Seoul, boys and girls children were 33.5%, 34.1% and 33.4%, 33.1%, in Kyunggi area and 33.1%, 32.9% in Kangwon area. 4) Urea nitrogen/creatinine ratios in Seoul children were 9. 0, 10. 0 of boys and girls, the ratio were 8.2, 8.0 in Kyunggi boys and girls children, and 7.5 and 7.4 in Kangwon boys, girls children. Low-income rural and upper-income urban background large differences between two groups in the urea nitrogen/creatinine ratio(Seoul: Kangwon in male, female children. p<0.05, p<0.001). The urea nitrogen/creatinine ratio definetly seems to be a good indicator of the quantity of the protein intake. However, whether or not it is an indicator of the quality of the ingested protein ramains to be seen.

유산균 및 홍국균 발효 꽃송이버섯 추출물과 잔사의 식이섬유와 베타 글루칸의 함량 (Dietary Fiber and β-Glucan Contents of Sparassis crispa Fruit Fermented with Lactobacillus brevis and Monascus pilosus)

  • 본 연구에서는 식이섬유와 ${\beta}$-glucan이 풍부한 꽃송이버섯을 발효한 후 추출하여 이들 성분의 함량을 보다 증가시킨 추출물과 잔사를 제조하고자 하였다. 이를 위해 열풍 건조한 꽃송이버섯 자실체의 분말을 유산균(L. brevis)과 홍국균(M. pilosus)으로 각각 발효한 후 열수와 수용성 에탄올(50, 70, 90%, v/v)로 추출하여 추출물과 잔사를 제조하고 이들의 수용성, 불용성 및 총 식이섬유와 ${\beta}$-glucan의 함량을 버섯원물의 추출물과 잔사의 각 성분의 함량과 비교하였다. 홍국균 발효 버섯의 총 식이섬유 함량은 74.4 g/100 g으로 버섯원물(64.4 g/100 g)과 유산균 발효 버섯(66.1 g/100 g)의 총 식이섬유 함량에 비해서 유의적으로(p<0.05) 높았다. 버섯원물, 유산균 발효 버섯, 홍국균 발효 버섯의 ${\beta}$-glucan 함량(21.9~24.4 g/100 g) 간에는 유의적인 차이가 없었다. 추출물의 경우, 홍국균 발효 버섯의 열수 추출물에서 총 식이섬유(21.6 g/100 g)와 수용성 식이섬유 함량(19.3 g/100 g)이 가장 높았으며 버섯 원물의 열수 추출물의 총 식이섬유(16.4 g/100 g)와 수용성 식이섬유 함량(14.6 g/100 g)보다도 유의적으로 (p<0.05) 높았다. 홍국균 발효 버섯을 열수 추출하고 남은 잔사의 총 식이섬유 함량은 90.5 g/100 g이었고 이들의 대부분이 불용성 식이섬유로 구성되어 있었으며, ${\beta}$-glucan 함량은 버섯 원물이나 홍국균 발효 버섯보다 높은 31.0 g/100 g이었다. 따라서 본 연구에서 제조한 홍국균 발효 꽃송이버섯의 열수 추출물과 잔사는 각각 액상과 분말 형태의 건강기능식품 및 가공식품 소재로 개발될 수 있을 것으로 기대된다.

식물공장 내 광주기 및 광도가 흰민들레의 생육과 수량에 미치는 영향 (Effects of LED(Light Emitting Diode) Photoperiod and Light Intensity on Growth and Yield of Taraxacum coreanum Nakai in a Plant Factory)

  • 본 연구는 완전 인공광형 식물공장 내에서 RBW LED(red:blue:white = 2:1:1)의 광주기와 광도가 흰민들레 생육에 미치는 영향을 구명하고자 수행하였다. 완전 인공광형 식물공장에서 각각 광주기 3수준과 광도 4수준으로 설정하였고, 분무수경으로 광주기 실험은 270일, 광도 실험은 120일 동안 재배하였다. 광주기별 전 재배기간 동안 8회 수확한 1주당 수확 총엽수는 16/8시간 처리구에서 주당 224매로 12/12시간 처리구의 220매와 비슷하게 많았고 8/16시간 처리구에서 151매로 가장 적어 대체로 명기가 길수록 수확엽수도 많아지는 경향이었다. 지상부 생체중은 16/8h 처리구에서 125g으로 가장 높았고 다음으로 12/12h 처리구 91g, 8/16h 처리구 56g 순이었다. 광도별전 재배기간 동안 총 4회 수확한 엽수는 $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구에서 123매로 가장 많았으며 다음으로 107매인 $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구와 95매인 $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구 이었고, $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구에서 56매로 가장 적었다. 1주당 지상부 총 생체중은 $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구에서 43.6g으로 가장 높았고 다음으로 $100{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$$200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구로써 각각 34.6g과 32.2g이었으며 $50{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 처리구에서 18.2g으로 가장 낮았다. 이상의 결과 흰민들레 최적 생장을 위한 완전 인공광형 식물공장의 RBW LED(red:blue:white = 2:1:1) 조명의 광조건은 광주기 16/8h, 광도 $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$이었다.

도시호텔에 나타난 공용부문의 공간 구성적 특성에 관한 연구 - 서울시 특급 호텔을 중심으로 - (A Study on the Spatial Characteristics for the Public Area in the City Hotels Lobby - Focused on First Class Hotels in Seoul -)

  • This study is mentioning about basic information and guide related block types, through a comparison and an analysis of floor types which is shown as function in block types and public interior spaces. For the range of this survey, it is defind in the beginning of 1970' s to the end of 1990' s which was rapidly developed in hotel industry and also is chosen by 12 places in the first class hotels. On the purpose of this study is written by designing the basic information of architectures, new hotels which are built and managed in urban, refer to the survey and analysis of the characteristic in hotel. The following is conclusion by based on pre-survey of the analysis of a plan and a site in these hotels. First, as a result of analysis in block type, there are equally distributed in three types (in-type, combination type, close-type), except a atrium type exception, in 12 hotels(100%). Second, as a result of analysis of function type in public interior space, Con-course type is consist of 7(58%), Hall type is 5(42%) in the type of room in public interior space, in 12 hotels(100%). Third, the type in lobby on space plan are equally shown by the type of lobby in center(4PL.), of lounge in center(4PL.), and a compromising type(4PL.).

Flexible 마이크로시스템을 위한 압전 박막 공진기의 설계 및 제작 (Design and fabrication of film Bulk Acoustic Resonator for flexible Microsystems)

  • This paper reports on the air-gap type thin film bulk acoustic wave resonator(FBAR) using ultra thin wafer with thickness of 50$\mu\textrm{m}$. It was fabricated to realize a small size devices and integrated objects using MEMS technology for flexible microsystems. To reduce a error of experiment, MATLAB simulation was executed using material characteristic coefficient. Fabricated thin FBAR consisted of piezoelectric film sandwiched between metal electrodes. Used piezoelectric film was the aluminum nitride(AlN) and electrode was the molybdenum(Mo). Thin wafer was fabricated by wet etching and dry etching, and then handling wafer was used to prevent damage of FBAR. The series resonance frequency and the parallel frequency measured were 2.447㎓ and 2.487㎓, respectively. Active area is 100${\times}$100$\mu\textrm{m}$$^2$.Q-factor was 996.68 and K$^2$$\_$eff/ was 3.91%.

참깨박 에탄올 추출물의 항산화 및 α-amylase및 α-glucosidase 저해 활성 (Antioxidant, α-amylase and α-glucosidase inhibitory ability effects of sesame meal ethanol extract)

  • 기름을 착유하고 얻어지는 부산물인 참깨박의 활용과 이용가치를 높이기 위해 일반성분(수분, 탄수화물, 조단백질, 조지방, 조회분), 무기질을 측정하였고 탈지 처리한 후 70% 에탄올 추출하여 항산화 활성 및 항당뇨 활성을 분석하였다. 참깨박의 일반성분 함량은 수분 6.51%, 탄수화물 16.22%, 조단백질 46.30%, 조회분 9.88%, 조지방 21.09%로 이었고, 무기질은 K 1128.08 mg/100 g, Ca 1356.27 mg/100 g, Fe 12.29 mg/100 g, P 2022.14 mg/100 g, Cu 2.08 mg/100 g, Mg 643.40 mg/100 g, Na 7.29 mg/100 g 함량을 나타내었다. 탈지 참깨박의 총 폴리페놀과 플라보노이드 함량은 각각 184.98 mg GAE/g, 27.63 mg QE/g였으며, DPPH 및 ABTS 라디칼 소거활성 IC50을 측정한 결과 각각 91.84, 340.09 ㎍/mL, 환원력은 0.87 OD으로 측정되었다. 탈지 참깨박의 α-amylase 및 α-glucosidase 저해 활성 IC50의 측정 결과는 각각 74.31, 16.73 mg/mL을 나타내었다. 따라서 70% 에탄올로 추출한 탈지 참깨박은 우수한 항산화 활성과 항당뇨 활성을 나타내 기능적 가치를 높일 수 있을 것으로 기대된다. 또한 참깨를 착유하고 남은 참깨박을 사용하게 되므로 환경보호와 자원순환 측면에 활용가치가 있을 뿐만 아니라 건강기능식품소재로 이용가치가 있을 것이라 판단된다.

The Bioinformatics and Molecular Biology Approaches for Vascular Cell Signaling by Advanced Glycation Endproducts Receptor and Small Ubiquitin-Related Modifier

  • The advanced glycation endproducts receptor (AGE-R) is a signal transduction receptor for multiligand such as S100b and AGEs. S100b has been demonstrated to activate various cells with important links to atherosclerosis initiation and progression including endothelial cells, and smooth muscle cells via AGE-R, triggering activation of multiple signaling cascades through its cytoplasmic domain. Many studies have suggested AGE-R might even participate in the cardiovascular complications involved in the pathogenesis of type I diabetes. Recently, Small Ubiquitin-Related Modifier 1 (SURM-1 also known as SUMO-1) has been recognized as a protein that plays an important role in cellular post-translational modifications in a variety of cellular processes, such as transport, transcriptional, apoptosis and stability. Computer Database search with SUMOplot Analysis program identified the five potential SURMylation sites in human AGE-R: K43, K44, K123, and K273 reside within the extracellular domain of AGE-R, and lastly K374 resides with the cytosolic domain of AGE-R. The presence of the consensus yKXE motif in the AGE-R strongly suggests that AGE-R may be regulated by SURMylation process. To test this, we decided to determine if AGE-R is SURMylated in living vascular cell system. S100b-stimulated murine aortic vascular smooth muscle cells were used for western blot analysis with relevant antibodies. Taken together, bioinformatics database search and molecular biological approaches suggested AGE-R is SURMylated in living cardiovascular cell system. Whilst SURMylation and AGE-R undoubtedly plays an important role in the cardiovascular biology, it remains unclear as to the exact nature of this contribution under both physiological and pathological conditions.

오대산 국립공원 진고개 Eco-bridge 식재계획 (Eco-Bridge Planting Plan in Chingogae of Odaesan National Park)

  • 본 연구의 목적은 개발로 인해 고립된 생태계를 잇는 Eco-bridge의 식재계획을 제시하는데 있다. Eco-bridge는 생물의 이동을 위해 생태계를 연결하는 통로로써 양쪽의 식생을 연결하기 위해 진고개길 양쪽으로 6개 조사지를 선정하고 16개 조사구(각 조사구는 100m$^{2}$)를 조사하였다. Eco-bridge 식재계획을 위한 조사, 분석결과 진고개지역식생은 신갈나무에서 서어나무, 까치박달, 물푸레나무, 피나무 등으로 천이가 진행될 것이다. 유사도 분석에서는 인접한 조사지간의 유사도가 높아 식생연속성이 뚜렷하였다. 종수 및 개체수분석에서 식재계획 최종단계 목표는 교목층 100m$^{2}$당 목본 약 15종내외로 교목층 10~12주, 아교목층 17~18주, 관목층 100여주의 밀도를 유지해야 할 것이다. 식재수종은 주변 출현수종을 중심으로 까치박달, 층층나무, 신갈나무 등 교목 12종, 개회나무, 딱총나무, 당단풍 등 아교목 및 관목 12종을 선발하였다.

육류 및 수산물을 이용한 조리식품에 함유된 수용성 비타민 함량 (Water-soluble Vitamin Content in Dishes Containing Meat and Seafood)

  • In this study, the content of water-soluble vitamins such as vitamin B1 (thiamin), B2 (riboflavin), B3 (niacin), B5 (pantothenic acid), B6 (pyridoxine), B7 (biotin), B12 (cyanocobalamin), and C (ascorbic acid) in dishes containing meat and seafood consumed in Korea were determined by high-performance liquid chromatography (HPLC) with ultraviolet and fluorescence detection. All analyses were performed under strict quality control of vitamins B1, B2, B3, B5, B6, B7, B12, and C. The highest content of vitamin B1 was observed in Bugeo-gangjeong (1.373 mg/100 g) and the highest level of vitamin B2 (5.162 mg/100 g) was found in pig liver. Bugeo-gangjeong showed the highest content of vitamin B3 (21.676 mg/100 g), and kkomak-muchim contained considerable amounts (43.310 mg/100 g) of vitamin B5. Vitamin B6 was not detected in most seafood dishes except for yangnyeom myeongran-jeot (0.274 mg/100 g) and was present at low levels or not present at all in meat dishes. The highest content of vitamin B7 was 6.506 ㎍/100 g in saeu-jeon and kkomak-muchim showed the highest content (21.132 ㎍/100 g) of vitamin B12. The highest content of vitamin C was in yangnyeom myeongran-jeot (84.508 mg/100 g). In addition, the analysis methods of each water-soluble vitamin were verified. These results showed that seafood-based ingredients in several dishes could be a good source of water-soluble vitamins.
