Porcine Xenotransplantation to Primates

  • Xenotransplantation is a hot topic currently, since the demand for diverse organs is increasing in patients. Among many species, pigs are suitable animals for xenotranplantation as they share many anatomical and physiological characteristics with humans. This review article provides an overview of porcine xenotransplantation and the rejection of pig xenotransplants in primates, and use of genetically modified and cloned pigs in xenotransplantation. It also highlights major target organs in porcine xenotransplantation and virus infection in xenotransplantation.

포화지방산의 피부암 세포주에 대한 항암 및 항산화 효과 (Anticancer and Antioxidant Effects of Saturated Fatty Acid against Skin Cancer Cell Lines)

  • The anticancer and antioxidant effect of different lengths of saturated fatty acids was tested on NIH3T3 fibroblasts and human skin melanoma cellsn in this study. The cell existence rate and antioxidizing capacity and optic reservation of cells were observed. This saturated fatty acid was concentration-dependent. IC50 Concentrations in NIH3T3 fibroblasts, human skin melanoma cells and DPPH radical scavenging activity of fatty acid was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. The reduction in cell number and morphological change in human skin melanoma cells was increasing the order of carbochain length ; caprylic acid < lauric acid < palmitic acid < stearic acid. These results suggest that carbochain length of fatty acid can be used as structure-activity relationships for anticancer and antioxidant.

Cytoprotective Effects of Docosyl Cafferate against tBHP-Induced Oxidative Stress in SH-SY5Y Human Neuroblastoma Cells

  • Neuronal cell death is a common characteristic feature of a variety of neurodegenerative disorders including Alzheimer's disease and Parkinson's disease. However, there have been no effective drugs to successfully prevent neuronal death in those diseases. In the present study, docosyl cafferate (DC), a derivative of caffeic acid, was isolated from Rhus verniciflua and its protective effects on tBHP-induced neuronal cell death were examined in SH-SY5Y human neuroblastoma cells. Pretreatment of DC significantly attenuated tBHP-induced neuronal cell death in a concentration-dependent manner. DC also significantly suppressed tBHP-induced caspase-3 activation. In addition, DC restored tBHP-induced depletion of intracellular Bcl-2, an anti-apoptotic member of the Bcl-2 family. Furthermore, DC significantly suppressed tBHP-induced degradation of IKB, which retains $NF-{\kappa}B$ in the cytoplasm, resulting in the suppression of nuclear translocation of $NF-{\kappa}B$ and its subsequent activation. Taken together, the results clearly demonstrate that DC exerts its neuroprotective activity against tBHP-induced oxidative stress through the suppression of nuclear translocation of $NF-{\kappa}B$.

Technology Innovation, Human Capital and R&D Effects on Economic Growth

  • This paper analyzes the economic effects of the S&T Innovation, R&D, human resources and investment on the economic growth using 18 countries. We have obtained the somewhat mixed results on the existence of unit root roots in variables. While most of Pedroni cointegration tests show that there are no panel cointegration among the variables, Kao cointegration test shows that there is the panel cointegration among the variables such as GDP, human capital, R&D investment and patent. Kao cointegration test result shows that human capital, R&D investment, patent economic growth seem to have the panel cointegration or the long-run relationship among them as a whole. The estimation results of individual OLS and panel estimation show that the human capital, R&D investment and technology innovation or patent had positively significant effects on economic growth or GDP.

인체 혈액응고 9인자 cDNA cloning 및 Escherichia coli 에서의 발현 (Cloning and Expression of Human Clotting Factor 9 cDNA un Escherichia coli)

  • 인체 혈액 응고 9인자는 간에서 생성되며 461개의 아미노산으로 구성된 당 단백질이다. 따라서 인체 혈액 응고 9인자 cDNA를 찾기 위해 태아의 간(fetal liver) cDNA library를 PCR(Polymerase Chain reaction) 방법으로 screening하였으며, 그 결과 ATG개시 코돈으로부터 TAA종료 코돈까지 포함하는 1.4 kb의 9인자 cDNA를 찾았다. 또한 클론된 9인자 cDNA를 박테리아에서 발현시키기 위해 박테리아 발현 벡터인 pGEX-2T 플라스미드에 클로닝하므로써 pGEX-F9 플라스미드를 제조하였다. pGEX-F9로 형질전환된 E. coli에서 PGEX-F9의 발현을 유도하면 73 kDa 크기의 GST-factor9 융합 단백질이 다량생성되며 , 이 단백질이 혈액 응고 9인자 단백질을 함유하는 융합 단잭질임을 혈액 응고 9인자 항체를 이용한 Western blot으로 입증하였다. E. coli에서 발현된 GST-factor 9 융합 단백질은 전체 단백질의 약 20%를 차지하며 GST agarose bead를 이용한 one step purificarion 방법을 통해 GST-factor9 융합 단백질을 쉽게 분리 할 수 있다.

우리나라 과학기술인력의 경력이탈 현황 분석 (An analysis of the deviation from S&T workforce career-path in Korea)

  • 본 논문은 전 생애주기에 걸쳐 나타나는 우리나라 과학기술인력의 경력이탈 규모와 현황을 추정하는 데 일차적인 목적이 있다. 이를 위해 먼저 과학기술인력의 경력이탈에 대한 조작적 정의를 내리고 이를 분석할 수 있는 기본틀을 크게 인재확보 단계와 활용단계로 구분하여 구축하였다. 이 틀에 입각하여 경력이탈 현황을 진단해 본 결과, 우리나라 과학기술인력의 경력이탈은 주로 활용단계에서 나타나고 양적 이탈보다는 창조경제시대를 이끌어갈 우수 인재의 이탈이라는 질적 이탈 문제가 심각한 것으로 나타났다. 마지막으로 생산가능인구 감소시대를 맞이할 경우 이러한 질적 이탈 문제는 더욱 심각해질 수 있으므로, 좋은 일자리 확보와 경력개발 경로 구축 등의 정책을 우선적으로 추진할 필요가 있다는 대안을 제시하고 있다.

Efficacy of Ronidazole for Treatment of Cats Experimentally Infected with a Korean Isolate of Tritrichomonas foetus

  • To evaluate the efficacy of ronidazole for treatment of Tritrichomonas foetus infection, 6 Tritrichomonas-free kittens were experimentally infected with a Korean isolate of T. foetus. The experimental infection was confirmed by direct microscopy, culture, and single-tube nested PCR, and all cats demonstrated trophozoites of T. foetus by day 20 post-infection in the feces. From day 30 after the experimentally induced infection, 3 cats were treated with ronidazole (50 mg/kg twice a day for 14 days) and 3 other cats received placebo. Feces from each cat were tested for the presence of T. foetus by direct smear and culture of rectal swab samples using modified Diamond's medium once a week for 4 weeks. To confirm the culture results, the presence of T. foetus rRNA gene was determined by single-tube nested PCR assay. All 3 cats in the treatment group receiving ronidazole showed negative results for T. foetus infection during 2 weeks of treatment and 4 weeks follow-up by all detection methods used in this study. In contrast, rectal swab samples from cats in the control group were positive for T. foetus continuously throughout the study. The present study indicates that ronidazole is also effective to treat cats infected experimentally with a Korean isolate of T. foetus at a dose of 50 mg/kg twice a day for 14 days.

Antioxidant Effects of Scutellaria baicalensis Georgi Against Hydrogen Peroxide-induced DNA Damage and Apoptosis in HaCaT Human Skin Keratinocytes

  • In this study, we investigated whether S. baicalensis rhizome ethanol extract (SBRE) has antioxidant capacities against oxidative stress induced cellular damage in the HaCaT keratinocytes. Our results revealed that treatment with SBRE prior to hydrogen peroxide ($H_2O_2$) exposure significantly increased the HaCaT cell viability. SBRE also effectively attenuated $H_2O_2$ induced comet tail formation, and inhibited the $H_2O_2$ induced phosphorylation levels of the histone ${\gamma}H2AX$, as well as the number of apoptotic bodies and Annexin V positive cells. In addition, SBRE exhibited scavenging activity against intracellular ROS generation and restored the mitochondria membrane potential loss induced by $H_2O_2$. Moreover, $H_2O_2$ enhanced the cleavage of caspase-3 and degradation of poly (ADP-ribose)-polymerase as well as DNA fragmentation; however, these events were almost totally reversed by pretreatment with SBRE. Furthermore, SBRE increased the levels of HO-1 associated with the induction of Nrf2. Therefore, we believed that SBRE may potentially serve as an agent for the treatment and prevention of neurodegenerative diseases caused by oxidative stress.

경쟁적 효소면역측정법을 이용한 환자 혈청 내 Thyroxine (T$_4$)의 검출 (Studies on the Rapid and Competitive Enzyme-linked Immunosorbent Assay for the Detection of Thyroxine (T$_4$) in Human Sera)

  • 갑상선 질환의 진단에 필요한 갑상선 호르몬의 측정시 thyroxine (T$_4$)농도 측정방법이 가장 흔히 이용되고 있다. 과거에 많이 활용되어온 방사면역측정법 (RH)이 폐기물 처리 및 관리인원 등의 인적, 물적인 문제가 크게 대두되고 있는 점을 감안하면 효소결합면역측정법의 확립은 큰 의미가 있다고 하겠다. 본 연구에서는 thyroxine (T$_4$)에 대한 단클론 항체 (monoclonal antibody)를 이용하여 T$_4$에 horseradish-peroxidase를 화학적으로 결합시킨 microtiter plate에 HRP-conjugate T$_4$와 혈청내의 T$_4$간의 서로 경쟁적인 반응을 이용한 표준곡선을 얻었다. 그리고 이렇게 확립한 기법을 임상적인 갑상선질환 환자의 혈청을 기존의 chemiluminescence 방법을 이용하여 얻은 결과와 비교하여 본 실험에서 확립한 ELISA검사 수치의 유용성을 검토하였으며 이 방법의 임상이용 가능성을 검토하였다.

Two Stages of R&D Spillovers: Technological and Economic Impacts

  • This paper empirically evaluates the effects of regional and industrial R&D on the performances of individual firms in two separated stages: (1) the stage of technological outcome from R&D and (2) the stage of economic outcome from technological outcome. Technological spillovers are separated from negative congestion effects through the stage-specific estimation. The firm-level Korean Innovation Survey data merit in coping with the endogeneity problem inherent in the estimation of spillovers. The estimation results show that: (1) there exist significant R&D spillovers both in regional and industrial dimensions, (2) the hypothesized technological spillovers and economic congestion effects are both in effect, and (3) firms with smaller individual R&D investments show greater spillovers.
