[$^1H-NMR$ Assignment of HIV Protease Inhibitor, Procyanidin B3 isolated from Rosa rugosa

  • The underground part of Rosa rugosa Thunb. has been used in Korean folk medicine for treating diabetes. The $^{1}H-NMR$ signal of procyanidin B3 isolated from Rosa rugosa was fully assigned by utilizing $^{1}H-^{1}H$ COSY. Procyandin B3 showed a moderate inhibitory activity against HIV-1 protease.

  • A tannin compound has been isolated from the underground part of Rosa rugosa Thunb. and its structure has been established to be methyl gallate $3-O-{\beta}-D-(6'-O-galloyl)-glucopyranoside(1)$ on the basis of chemical and spectroscopic evidences. This compound was the first example in this plant.

쌀과 해당화를 이용한 식초제조 및 품질평가 (Quality Evaluation of Vinegar Manufactured using Rice and $Rosa$ $rugosa$ Thunb)

  • 쌀을 이용한 식초제조에서 증자 공정과 무증자 공정으로 제조된 쌀과 해당화를 혼합한 조성물을 사용하여 식초를 제조하였다. 1단계로 ethanol 발효를 $25^{\circ}C$에서 12일 동안 진행하고, ethanol 함량을 6%로 조정한 후 $Acetobacter$ $aceti$균을 접종하여 $30^{\circ}C$에서 9일 동안 초산 발효하였다. 식초 발효전에 초산을 첨가하는 공정은 발효의 성공여부를 결정하는 요인으로 나타났으며, 첨가되는 초산의 농도에 의존적으로 최종 식초 제품의 유기산 함량이 증가하였다. 무증자법으로 식초를 제조시 유리 아미노산과 GABA 함량이 높게 나타났으며, 주요 아미노산으로는 glutamic acid, arginine 등으로 나타났다. 본 연구결과는 해당화-쌀 식초 제조시 쌀의 열처리 공정여부가 식초의 품질에 큰 영향을 준다는 것을 보여준다.

해당화꽃 추출물의 화장품소재로서 약리활성에 관한 연구 (The Study on Pharmacological Activation as Cosmetic Material of Rosa rugosa Thunb. Flowers Extract)

  • Objectives : In this study, we investigated the effect of ethanol extract from Rosa rugosa Thunb. flowers(ERF) on the activities of antioxidant, antiwrinkle and whitening. Methods : We measured antioxidant efficacy of ERF by using 1,1-diphenyl-2-picrylhydrazyl(DPPH) assay. Also we confirmed the inhibitory effect of ERF on collagenase activity and melanin synthesis by using collagenase assay kit and dihydroxiphenylalanine staining, respectively. To evaluate the anti-inflammatory effects of ERF, we examined the inflammatory mediator IL-6. Results : ERF showed highly efficacy in DPPH radical scavenging activity. ERF dose-dependently suppressed collagenase activity. Ultraviolet-induced production of IL-6 decreased by ERF treatment. In B16F10 cell, ERF significantly reduced tyrosinase activity and melanin synthesis. Conclusions : From the above results, it was indicated that ERF could be utilized as anti-aging and whitening cosmetic ingredients.

해당화(Rosa rugosa Thunb.) 부위별 열수추출물의 항산화 활성 분석 (Antioxidant Activities of Hot Water Extracts from Different Parts of Rugosa rose (Rosa rugosa Thunb.))

  • 본 연구에서는 해당화 부위별 열수추출물에 대한 항산화 활성을 분석하였다. 해당화의 부위별 추출물의 총 페놀성 화합물과 총 플라보노이드 함량은 잎에서 가장 높았으며, 각각 107.29, 24.28 mg/g로 나타났다. DPPH와 ABTS 라디칼 소거능은 꽃, 잎, 씨, 열매, 과육 추출물 순으로 활성이 높았으며, 꽃 추출물의 DPPH, ABTS 라디칼 소거능은 각각 $IC_{50}=0.87$, $0.27mg/m{\ell}$로 나타났다. 항산화 활성이 가장 높았던 꽃과 잎 추출물에서는 gallic acid가 각각 4.51, 0.97 mg/g로 다른 부위별 열수추출물에서보다 높았다. 해당화 부위별 열수추출물을 분획한 결과(A-F), 꽃 추출물의 C 분획물에서 높은 항산화 활성을 확인하였다. 이것은 총 페놀성 화합물과 총 플라보노이드 함량(3305.43, 878.42 mg/g)에 의한 것이며, p-value < 0.001에서 총 페놀성 화합물과 총 플라보노이드의 함량이 증가할수록 라디칼 소거능이 증가하는 상관관계 결과와 일치하였다. 따라서 해당화 열수추출물은 식품, 화장품, 의약품 산업 등에서 천연 항산화제로 사용가능할 것이다.

산딸기와 해당화를 이용하여 제조한 식초의 발효 특성 (Characterization of Vinegar using Rubus crataegifolius and Rosa rugosa Thunb)

  • Production of vinegar by fermentation using Rubus crataegifolius (Korean raspberry) and Rosa rugosa Thunb (Haedanghwa) were investigated. At the first step, ethanol fermentation performed for 12 days on Haedanghwa wine containing $17.6^{\circ}Brix$ sugar concentration, 6.2% ethanol concentration, and pH 2.9, and on Korean raspberry-Haedanghwa wine containing $7.0^{\circ}Brix$ sugar concentration, 11.4% ethanol concentration, and pH 3.4. In the second step, after supplementation of 1% acetic acid to Haedanghwa wine and Korean raspberry-Haedanghwa wine which adjusted their ethanol concentration to 6%, aerotropic acetic acid fermentation was carried out using Acetobacter aceti at $30^{\circ}C$ and 200 rpm for a period of 12 days. Korean raspberry-Haedanghwa vinegar contained a 3.7% concentration of acetic acid. Total phenolic compounds and color in Korean raspberry-vinegar was higher than that in Rosa rugosa Thunb vinegar. This result demonstrates that use of Korean raspberry and Haedangwha mixture for vinegar fermentation improves the color and taste of the final product.

Optimization of Ascorbic Acid Extraction from Rugosa Rose (Rosa rugosa Thunb.) Fruit Using Response Surface Methodology and Validation of the Analytical Method

  • In this study, ultrasound-assisted extraction was performed to extract ascorbic acid from rugosa rose (Rosa rugosa Thunb.) fruit. The optimal conditions were investigated by response surface methodology, using two variable including reaction time (16-44 min) and temperature (16-44℃). The ascorbic acid extraction was sensitive to the reaction time rather than the reaction temperature, and the optimal conditions for ascorbic acid extraction were 25℃ and 30 min. Ascorbic acid and gallic acid in the rugosa rose fruit extract were completely separated by HPLC, with a resolution factor of over 1.5 between the two. The correlation coefficient of the ascorbic acid was 0.999 in a linearity test for 50-150 ㎍/mL concentration of extract. The limit of detection and limit of quantification values were 0.16 ㎍/mL and 29.89 ㎍/mL, respectively. The relative standard deviations (RSD) for repeatability and reproducibility were determined, and each RSD showed good precision at less than 5% (N=6).

Anti-Inflammatory Effect of Rosa rugosa Flower Extract in Lipopolysaccharide-Stimulated RAW264.7 Macrophages

  • Rosa rugosa Thunb, a deciduous shrub of the genus Rosa, has been widely used to treat stomach aches, diarrhoea, pain, and chronic inflammatory disease in eastern Asia. In recent years, our research team has extensively studied the Rosa rugosa flower extract, and specifically undertook pharmacological experiments which have optimized the extraction process. Our methods have yielded a standard extract enriched in phenolic compounds, named PRE. Herein, we expand our efforts and evaluated the anti-inflammatory activity of PRE on lipopolysaccharide (LPS)-induced inflammation in RAW 264.7 macrophages. PRE significantly inhibited production of nitric oxide (NO), prostaglandin $E_2(PGE_2)$, tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6, and interleukin $1{\beta}$ (IL-$1{\beta}$), as well as expression of their synthesizing enzymes, inducible nitric oxide synthase (iNOS) and cyclooxygenase2 (COX-2). Furthermore, PRE inhibited activity of mitogen-activated protein kinases (MAPK) as well as nuclear factor-kappa B (NF-${\kappa}B$) signaling pathway. Our findings are the first to explain the anti-inflammatory mechanism by PRE in LPS-stimulated macrophages. Given these results, we propose that PRE has therapeutic potential in the prevention of inflammatory disorders.
