Effects of Growth Regulators on Callus Induction and Plant Regeneration from Leaf Explants of Sedum sarmentosum (돌나물 엽절편으로부텨 캘러스 형성 및 식물체 분화에 미치는 생장조절물질의 영향)

  • To establish the system of in uitro plant regeneration, the leaf segments of Sedum sarmentosum were cultured on MS media supplemented with different levels of 2,4-D, NAA and BA. The callus induction and growth showed a good response on MS medium supplemented with 3.0mg/L 2,4-D and 1.0mg/L BA, but a few callus induced on medium containing NAA and BA. In plant regeneration, combination of BA and NAA promoted shoot organogenesis from callus, and the highest frequency was obtained on MS medium supplemented with 0.2mg/L NAA and 3.0mg/L BA. When calli were transferred to the plant regeneration medium containing 0.2mg/L NAA and 3.0mg/L BA, healthy shoots without hyperhydricity were continuously induced (17.2 plantlets per callus) after 50 days of culture. When regenerated plantlets were transferred onto hormone-free MS medium, rooting was easily achieved from all of them.

  • To establish high frequency shoot formation from two cultivars (Cheongyangjaerae and Myungan) of boxthorn (Lycium chinense Mill.), hypocotyl segments with cotyledons from seedlings were used as explants. High frequency adventitious shoot formation (more than 80%) were obtained from hypocotyl segments with cotyledon on MS medium supplemented with 1.0 mg/L zeatin, when precultured for 3 weeks under dark conditions followed by transfer to light conditions. But there was no shoot induction in the explants cultured without preculture under dark conditions. Roots were induced from the shoots when transferred to rooting medium supplemented with 1.0 mg/L IAA for 4 weeks. Regenerated plantlets were grown to normal mature plants in soil.

Effect of carbon source and concentration on in vitro regeneration and propagation in Lycopus lucidus by node culture (쉽싸리의 기내 마디배양 시 탄소급원의 종류와 농도가 식물체 재분화 및 증식에 미치는 영향)

  • We examined the effect of carbon sources on the regeneration and ex vitro acclimatization of Lycopus lucidus Turcz. ex Benth. Plantlets were regenerated on the 1/2MS medium supplemented with different concentrations (3 ~ 10%) of sucrose and glucose. The sucrose concentrations of 3% and 5% that were supplied enhanced shoot multiplication and rooting but hampered high concentration growth (including the length of the shoot and root). During ex vitro acclimatization, the tuberization of the root, the root length, the shoot length and the survival rate of Lycopus lucidus plantlets grown using 3% and 5% sucrose were found to be better than the other carbon sources and concentrations. Thus a sucrose concentration of 3% and 5% in the 1/2MS medium appeared to be better for both in vitro growth and ex vitro acclimatization of Lycopus lucidus.

Comparison of Some Propagation Methods in Paeonia albiflora Pallas and Effective Components of Paeonia albiflora Pallas Root Grafted with Paeonia moutan Sims (작약의 번식방법과 작약 / 목단 접목근의 유효성분 비교)

  • This experiment was carried in order to compare some propagation methods and the effective components in the root of Paeonia albiflora pallas, Paeonia moutan Sims and Paeonia albiflora pallas root grafted with Paeonia mouton sims The optimum seeding time of August 20 to september 10 for increasing germination ratio. The growing and developing of Paeonia albiflora Pall. for root dividing methods was advantageous to practice at Autum than at spring season. The shoot cutting method during the flowering time was proper to increase the rooting ratio with the aid of Ruton, the plant growth regulator but not emerged of apical bud. The grafting stages between the Paeonia albiflora pall. and Paeonia moutan Sims was available to practice during the September for increasing the thickness growth of root. Among the effective components of the paeony the paeoniflorin was shown high, while paeonol in paeony moutan. The paeoniflorin of herbaceous paeony / paeony moutan was increased about 75.9% than herbaceous paeony alone, and the other compoents are similar to those of herbaceous paeony root.

Analysis of Flooding Damage by Heavy Rain on 'Fuji'/M.26 Apple Tree (집중호우에 의한 '후지'/M.26 사과나무의 침수 피해 분석)

  • This survey was carried out to investigate the effect of flooding by heavy rain on growth characteristics and disease incidence of 'Fuji'/M.26 mature apple tree in 1998 at Kyongbuk province. The surveyed regions were Andong, Uisung, and Gunwi area. The six orchards were selected from one area, totally 18 orchards were surveyed. The flooded depth of surveyed orchards was from 70 to 350 cm, and the flooded duration was from 6 to 72 h. Defoliation ratio, number of abnormal budding, and flowering per tree were increased along with the increment of flooded depth and duration. Rooting ratio was decreased rapidly with the increment of flooded duration, and dead root ratio was 16.2% in flooded orchards. Marssonia blotch (Diplocarpon mali), white rot (Botryosphaeria dothidea) of fruit, and phytophthora fruit rot (Phytophthora cactorum) incidence were increased in flooded orchards. The analysis results of pearson correlation coefficient among surveyed items showed that higher relationship of abnormal growth characteristics and increment of disease incidence of 'Fuji'/M.26 apple trees with flooded duration than with flooded depth. With these results, the flooded depth was found to be the main factor for the abnormal growth characteristics and disease incidences of 'Fuji'/M.26 apple trees.

Biofilm Formation and Indole-3-Acetic Acid Production by Two Rhizospheric Unicellular Cyanobacteria

  • Microorganisms that live in the rhizosphere play a pivotal role in the functioning and maintenance of soil ecosystems. The study of rhizospheric cyanobacteria has been hampered by the difficulty to culture and maintain them in the laboratory. The present work investigated the production of the plant hormone indole-3-acetic acid (IAA) and the potential of biofilm formation on the rhizoplane of pea plants by two cyanobacterial strains, isolated from rice rhizosphere. The unicellular cyanobacteria Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 that were isolated from a rice rhizosphere, were investigated. Production of IAA by Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 was measured under experimental conditions (pH and light). The bioactivity of the cyanobacterial auxin was demonstrated through the alteration of the rooting pattern of Pisum sativum seedlings. The increase in the concentration of L-tryptophan and the time that this amino acid was present in the medium resulted in a significant enhancement of the synthesis of IAA (r > 0.900 at p = 0.01). There was also a significant correlation between the concentration of IAA in the supernatant of the cyanobacteria cultures and the root length and number of the pea seedlings. Observations made by confocal laser scanning microscopy revealed the presence of cyanobacteria on the surface of the roots and also provided evidence for the penetration of the cyanobacteria in the endorhizosphere. We show that the synthesis of IAA by Chroococcidiopsis sp. MMG-5 and Synechocystis sp. MMG-8 occurs under different environmental conditions and that the auxin is important for the development of the seedling roots and for establishing an intimate symbiosis between cyanobacteria and host plants.

Effect of Reflective Film Mulching on the Stomatal Features, Transpiration Rate and Photosynthetic Rate of Tomato Plants in Greenhouse Cultivation (반사필름 멀칭이 토마토의 기공특성, 증산속도, 광합성속도에 미치는 영향)

  • These studies were conducted to verify the effect of the supplementary lighting by reflective film mulching and its establishment in the north side of greenhouse on the utility of light at tomato by means of investigating changes of leaf temperatures, stomatal features, transpiration rates and photosynthetic rates. Stomatal density of leaves were high in the reflective film mulching but sizes of stomata were not different. As the osmotic potential in rooting zone was low, the stomatal resistance was high, transpiration rate was low, and leaf temperature was increased by 40.62$^{\circ}C$. And also in the block of reflective film mulching photosynthetic rates were decreased hut chlorophyll contents were not different. Especially, there is an effect of controlling greenhouse whiteflies by treatment of reflective film mulching. It is thought that the reason of high quality or increasing yield at several crops by supplementary lighting, such as reflective film mulching, would be caused by influences of absorption and distribution of nutrients through high transpiration rate and photosynthesis which resulted from increase of stomata.

Pyruvic Acid and Sugar Contents during Storage Duration in Onion (Allium cepa L.) (양파 저장 중의 Pyruvic Acid와 당 함량의 변화)

  • This study was carried out to access the stauts of pyruvic acid and sugars in onion bulbs during storage at room temperature. Pyruvic acid content remained in a steady state until 3 months of storage. However the pyruvic acid content gradually increased as the onions started to root, and rapidly increased after sprouting. There was no difference in dry matter contents during storage.e. Sucrose content increased as storage duration extended, and then gradually decreased right after the onions started to sprout. Fructose content gradually decreased after 45 days of storage, and remained in a steady state after sprouting. Glucose content had a tendency to increase as storage duration extended. Pyruvic acid/total sugar (PA/TS) ratio decreased after 1 month of storage, and then increased after 135 days, and was 83% higher at 150 days of storage than at harvest. Sweetness gradually decreased until 1 month of storage, but thereafter remained steady. Therefore it is desirable that pyruvic acid analysis for sweet onion selection should be conducted before breaking the dormancy.

An Improved Plant Regeneration Protocol using Cotyledonary Explants from Inbred Lines of Chinese Cabbage (Brassica rapa ssp. pekinensis)

  • We studied the effect of genotype, explant, age of explant, medium (plant growth regulators and gelling agents), and standardized an efficient regeneration protocol for inbred lines of Chinese cabbage (Brassica rapa ssp. pekinensis). Of the different concentrations of NAA and BA tested, the combination of $5\;\cal{mg/L}\;BA\;and\;0.5\;\cal{mg/L}$ NAA gave the highest frequency of shoot regeneration. The cotyledonary explants had more shoot regeneration frequency ($\ge40\%$) than the hypocotyl ex-plants. Besides, the cotyledonary explants, excised from the 4-day old seedlings, showed higher shoot regene-ration ($56.7\%$). Of the three gelling agents and their concentrations used, 16 g/L agar was found to be the best for shoot regeneration. Shoot regeneration frequency in-creased significantly by supplementing the medium with $4\;\cal{mg/L}\;of\;AgNO_3$ MS medium devoid of NAA showed higher frequency of rooting in the regenerated shoots than the ones supplemented with NAA. Our improved regeneration protocol will be especially useful for the genetic transformation of Chinese cabbage inbred lines to develop transgenic hybrids.

Factors for high frequency plant regeneration in tissue cultures of Indian mustard (Brassica juncea L.)

  • An efficient system for high frequency plant regeneration was established through investigating various factors such as plant growth regulator combinations, explant types and ages, and addition of $AgNO_3$ influenced on shoot regeneration in Brassica juncea L. cv. BARI sarisha-10. Murashige and Skoog (MS) medium supplemented with 0.1 mg/L NAA (1-naphthaleneacetic acid) and 1 mg/L BA (6-benzyladenine) showed the maximum shoot regeneration frequency (56.67%) among the different combinations of NAA and BA. Explant type, explant age, and addition of $AgNO_3$ also significantly affected shoot regeneration. Of the four type of explants (cotyledon, hypocotyl, root, and leaf explants)- cotyledon explants produced the highest shoot regeneration frequency and hypocotyls explants produced the highest number of shoots per explant, whereas root explants did not produce any shoot. The cotyledonary explants from Four-day-old seedlings showed the maximum shoot regeneration frequency and number of shoots per explant. Shoot regeneration frequency increased significantly by adding $AgNO_3$ to the medium. Two mg/L $AgNO_3$ appeared to be the best for shoot regeneration with the highest shoot regeneration frequency (86.67%) and number of shoots per explant (7.5 shoots). Considerable variation in shoot regeneration from cotyledonay explants was observed within the B. juncea L. genotypes. The shoot regeneration frequency ranged from 47.78% for cv. Shambol to 91.11% for cv. Rai-5. In terms of the number of shoots produced per explant, B. juncea L. cv. Daulot showed the maximum efficiency. MS medium supplemented with 0.1 mg/L NAA showed the highest frequency of rooting. The regenerated plantlets were transferred to pot soil and grown to maturity in the greenhouse. All plants were fertile and morphologically identical with the source plants.
