Micropropagation of the hybrids of Actinidia deliciosa$\times$A. arguta by tissue culture (참다래$\times$다래 교잡종의 액아배양 및 캘러스 배양에 의한 기내번식)

  • Kiwi (Actinidia deliciosa) is exotic plant and thus susceptible to cold climate in the middle part of Korean peninsular. Several hybrids have recently been developed to enhance cold tolerance by crossing them with domestic species (A. arguta), We have developed an efficient micropropagation technique for the hybrids using both axillary bud and callus culture systems. Shoot proliferation from axillary buds was possible on St medium supplemented with 0.2 mg/L Bh and 3.0 mg/L GA$_3$. In vivo cuttings of the proliferated shoots were more effective for root induction and subsequent survival than in vitro rooting. More than 95% of the plantlets were successfully transferred to field. Effective callus induction was achieved on MS or B$_{5}$ medium with 2,4-D or NAA. Although callus induction could be made from any combinations of media and auxins, shoot regeneration was observed only from the callus induced on medium containing NAA.A.

Effect of Shading Level on the Induction of Inflorescence and Growth of Phalaenopsis Hybrid (차광 수준이 팔레놉시스의 화경 발생과 생육에 미치는 영향)

  • This experiment was carried out to find an optimum shading level for the growth of root and shoot, and to find the effect of shading level after August on the induction and growth of inflorescence of Phalaenopsis hybrid. The shading levels were 50%, 60%, 70%, 80% and 90% of natural light($1200{\mu}molm^{-2}s^{-1})$. The $CO_2$ uptake, transpiration rate, carbohydrate content, fresh weight and dry weight of Phalaenopsis hybrid were higher at 50-60% level than the others. But, it was diminished when the shading level was increased from 70% to 90%. Inflorescence length, the number of inflorescence and flower per plant all increased under 50-60% shading level and the day needed for the flowering after treatment decreased. Especially, the induction of inflorescence was depressed and flowering is not occurred under 90% during experiment period. These results suggest that the optimal shading level for the growth of Phalaenopsis including inflorescence was founded to be 50-60% in the season of light intensity and amount of sunshine decrease after august.

Biologic Response of Human Deciduous Dental Pulp Cells on Newly Developed MTA-like Materials (새로 개발된 MTA 유사 재료에 대한 유치 치수세포의 생물학적 반응)

  • This study compared the in vitro cell viability and differentiation potentials of human deciduous dental pulp cells (DPCs) on mineral trioxide aggregate (MTA)-like products (ProRoot MTA, RetroMTA and Endocem Zr). The experimental materials were prepared as circular discs, which were used to test the effects of the materials on the viability of human DPCs when placed in direct and indirect contact. Furthermore, the pH of the extracted materials was recorded, and their effect on cell differentiation potential was evaluated by evaluating the alkaline phosphatase (ALP) activity and Alizarin Red S staining of DPCs incubated with the test materials. In direct contact, the cell viability of human DPCs was higher with ProRoot MTA and RetroMTA than with Endocem Zr. However, when in indirect contact, the cell viability of human DPCs was generally higher in Endocem Zr than in ProRoot MTA and Retro MTA. With respect to pH, the alkalinity was lower for Endocem Zr than for the other test materials. The ALP activities of the cells were not enhanced by any of the experimental materials. Alizarin Red S staining of the tested human DPCs revealed that their differentiation potential was lower than for cells incubated with osteogenic induction medium. While there were differences in the responses of the human DPCs to the test materials, all displayed degrees of cytotoxicity and were unable to enhance either the viability or differentiation of human DPCs. However, Endocem Zr exhibited better cell viability and was less alkaline than the other test materials.

Production of Ginsenoside in Callus of Ginseng Hairy Roots (인삼 모상근의 캘러스를 이용한 ginsenoside 생산)

  • By the Agrobacterium rhizogenes A$_4$ were induced a transformed callus of ginseng hairy root and examine to find the possibility whether it can produce certain ginsenoside. Investigations for a finding out to optimal culture medium showed that BA application is better than more factorial composition between auxins and cytokinins. For the induction of hairy root callus of ginseng, l/2 MS medium containing 1 to 3 mg of benzyladenine(BA) per liter gave the best result. The growth of ginseng hairy root callus(GHC) cultured with the 1/2MS medium supplemented with 2 mg BA/L was selected for best suspension cultures. The optimum concentration of BA for ginsenosides production was found to be 2 mg/L. Probably the inoculum size of callus plays a role with the ginsenoside production in suspension culture. AS for inoculum size of callus, 50 mg was superior to 150 mg for growth and ginsenoside production. Ginsenoside contents were highest in the suspension culture grown for four weeks under continuous light condition. In fact that continous light treatment promote strongly the synthesis of ginsenoside of the hairy root callus is first result in the world and the numerously induced root hairs of the callus leads a new method for ginsenoside production.

  • Wheat (Triticum aestium L.cv Cho-Kwang) explants were transformed by electrporation. Excised root segments were elechoporated with plasmid DNA of pBI121 and transferred to medium containing 300 mg/L kanamycin. Transformed calli formed within 5-7 days of culture and were selected from electroporated tissue on medium containing kanamycin after 4 weeks. The highest transformation frequency was obtained after electroporation with a pulse of 200 V/800 uF and calli formed at frequencies up to 2.5%. GUS ($\beta$-glucuronidase) assay and dot blot analysis showed that the foreign gene was capable of expressing in root explants subjected to electroporation and calli derived from the explants..

Sigma-1 Receptor Antagonist BD1047 Reduces Allodynia and Spinal ERK Phosphorylation Following Chronic Compression of Dorsal Root Ganglion in Rats

  • Many therapeutic roles have been proposed for sigma-1 receptor (Sig-1R), but the involvement of Sig-1R in neuropathic pain has currently not been well explored. The present study aimed to evaluate the anti-nociceptive effect of Sig-1R antagonist (BD1047) in a rat model of chronic compression of the dorsal root ganglion (CCD), which is a model of human foraminal stenosis and radicular pain. When stainless steel rods were inserted into the intervertebral foramen of lumbar vertebrae 4 and 5, the CCD developed reliable mechanical (from 3 day) and cold allodynia (from 1 day) as compared with the sham operation group. The spinal expressions of Sig-1R and phosphorylation of extracellular signal-regulated kinase (pERK) were significantly increased from day 3 to day 14 after CCD surgery, as is consistent with the manifestation of allodynia. The BD 1047 (10, 30, 100 mg/kg) administered on postoperative days 0~5 dose-dependently suppressed both the induction of allodynia and the elevation of the spinal pERK expression in a manner comparable with that of gabapentin (100 mg/kg). At 7 days post-CCD surgery, BD1047 (10, 30, 100 mg/kg) administration also produced anti-nociceptive effects on the mechanical and cold allodynia similar with those of gabapentin (100 mg/kg). Therefore, this data suggested that Sig-1R may play an important role in both the development and maintenance of CCD-induced neuropathy.

Neuroprotective Effect of the Water-insoluble fraction of Root Barks of Dictamnus dasycarpus 70% Ethanolic Extract on Glutamate-Induced Oxidative Damage in Mouse Hippocampal HT22 Cells (백선피 70% 에탄올 추출물의 비수용성 분획물의 뇌세포 보호 효과)

  • Oxidative stress or accumulation of reactive oxygen species (ROS) leads neuronal cellular death and dysfunction, and it contributes to neuronal degenerative disease such as Alzheimer's disease, Parkinson's disease and stroke. Glutamate is one of the major excitatory neurotransmitter in the central nervous system (CNS). Glutamate contributes to fast synaptic transmission, neuronal plasticity, outgrowth and survival, behavior, learning and memory. In spite of these physiological functions, high concentration of glutamate causes neuronal cell damage, acute insults and chronic neuronal neurodegenerative diseases. Heme oxygenase-1 (HO-1) enzyme plays an important role of cellular antioxidant system against oxidant injury. NNMBS020, the water-insoluble fraction of the 70% EtOH extract of root barks of Dictamnus dasycarpus, showed dominant neuroprotective effects on glutamate-induced neurotoxicity in mouse hippocampal HT22 cells by induced the expression of HO-1 and increased HO activity. In mouse hippocampal HT22 cells, NNMBS020 makes the nuclear accumulation of Nrf2 and stimulates extracellular signal-regulated kinase (ERK) pathway. The ERK MAPK pathway inhibitor significantly reduced NNMBS020-induced HO-1 expression, whereas the JNK and p38 inhibitors did not. In conclusion, the water-insoluble fraction of the 70% EtOH extract of root barks of D. dasycarpus (NNMBS020) significantly protect glutamate-induced oxidative damage by induction of HO-1 via Nrf2 and ERK pathway in mouse hippocampal HT22 cells.

Effects of herbal-aupuncture solution from vitis labrusca root on LPO, RNS and cytochrome P450 (포도근 약침액이 LPO, RNS 및 Cytochrome P450에 미치는 효과)

  • Objectives : In this study, we determinated protective effects of Vitis labrusca Root herbal-acupuncture solution (VLHA) against atherosclerosis and cardiovascular disease induced by oxidative damage and cytochrome P450 (CYP) induction. Methods : Antioxidant capacity of VLHA were evaluated by the ${\beta}-carotene/linoleic$ acid emulsion system and FeCl2-ascorbic acid stimulted lipid peroxidation in rat liver homogenate, respectively. Scavenging effects of VLHA on ONOO- and NO were measured by DHR 123, DAF-2 assay. Furthermore, we measured inhibitory activity of VLHA on CYP 2E1 in rat liver microsome using by Aniline assay. Results : VLHA exhibited potent inhibition rate(74%) on FeCl2-ascorbic acid induced lipid peroxidation, and revealed a strong scavenging effects on ONOO- and NO. In addition, VLHA showed inhibitory effect on CYP 2E1. Conclusions : The present study concludes that our results suggest that VLHA could be used as a good source in the prevention of atherosclerosis and alcoholic liver injury.

Adventitious Root Development and Ginsenoside Production in Panax ginseng, Panax quinquefolium and Panax japonicum

  • This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of $NH_4NO_3$ from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with $NH_4NO_3$. Two-step culture protocol: $NH_4NO_3$-free medium for first two weeks of culture, followed by $NH_4NO_3$ containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.

Evaluation Potential Antidiabetic Effects of Ferula latisecta in Streptozotocin-Induced Diabetic Rats

  • Objectives: The aim of the present work was to evaluate the possible beneficial effects of F. latisecta on blood glucose, lipids, and diabetes-related changes in the liver and kidney of streptozotocin-induced diabetic rats. Methods: Male Wistar rats were randomly allocated into four groups (n = 6): normal control rats, diabetic control rats, diabetic rats treated for 4 weeks with F. latisecta root (400 mg/kg/day), and diabetic rats treated with F. latisecta aerial parts (400 mg/kg/day). Results: Induction of diabetes significantly (p < 0.05) increased the levels of fasting blood glucose (FBG), triglyceride, total cholesterol, low-density lipoprotein (LDL), blood urea nitrogen (BUN), aspartate aminotransferase (AST), and alanine aminotransferase (ALT). Diabetes also increased (p < 0.05) oxidative stress in the kidney and liver (decrease of thiol and increase of superoxide dismutase). The root and aerial parts of F. latisecta significantly reduced the level of LDL (p < 0.05) and restored the content of thiol (p < 0.05) and superoxide dismutase (p < 0.01) in the kidney and liver. F. latisecta had no significant effect on the levels of FBG, BUN, AST, and ALT. The root of F. latisecta also reduced the serum level of total cholesterol (p < 0.05) and prevented the progression of hyperglycemia. Conclusion: These findings suggest that F. latisecta may improve diabetic dyslipidemia by reducing serum LDL. Further studies are needed to confirm our findings.
