The Effect of Flavonoid Fraction Extracted from Rhus verniciflua Stokes on Sexual Behavior in SD Male Rats (옻나무 플라보노이드가 수컷 백서의 성행동에 미치는 영향)

  • Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine for a men's sexual enhancement. In this study, we prepared a flavonoid fraction (RWE) from a hot water extract of RVS and its influence on sexual behavior was studied in male rats which were orally administered varying doses of RWE for 2 weeks. All doses of RWE stimulated sexual behavior in male rats such as reducing mounting latency and enhancing mount frequencies. However, intermission numbers were not changed and none of group can observe the ejaculation during behavioral testing time 5 min. Testosterone levels were Increased about 0$\∼$53$\%$ and 92$\∼$164$\%$ by the treatment of RWE for 1 and 2 weeks, respectively. However, estrogen levels in male rats tended to decrease in a dose dependent manner of RWE. At the dose of RWE 200 mg/kg, estrogen level was reduced to 77.5$\%$ and 70.3$\%$ of control after 1 and 2 weeks treatment. These findings suggest that the Rhus flavonoid fraction can stimulate the androgen-dependent male sexual behavior and it can be applied to the material of functional food for enhancing the sexual function.

Protective Mechanism of Flavonoids Isolated from Rhus Vernicifiua on the Paraquat Toxicity Reducing Agent and its Inhibition Mechanism (옻나무 목부로부터 분리한 Favonoids가 Paraquat 독성의 경감기전 검색)

  • In this study, we investigated to elevate the modulatory effect of flavonoid(fustin, sulfuretin, 10 mg/kg) which was isolated from Rhus verniciflua Stokes(RVS) in male Sprague-Dawley rats for 2 weeks on the toxicity of paraquat. In the flavonoids pretreated groups, glutamic oxaloacetic transaminase, glutamic pyruvic transaminase, blood urea nitrogen, creatinine, malondialdehyde and alkaline phosphatase activity in serum and malondialdehyde, alkaline phosphatase activity and collagen in lung tissue which was induced paraquat toxicity were slightly decrease compared to the normal group. In the lung tissue of flavonoids pretreated groups, malodialdehyde value, G-6-phosphatase activity and collagen synthesis were recovered to tile normal values and alkaline phosphatase activity was increased. From these results, we concluded that flavonoids which were isolated from RVS is an effective agent to inhibit the pulmonary and internal toxicities and hence we concluded that acitive components of fustin and sulfuretin which were isolated from RVS might be removed free radicals induced by paraquat.

Reduction of Skin Allergy of Rhus verniciflua Sap Utilizing Radiation Technology (방사선 기술을 이용한 옻나무 수액의 피부 알러지 저감화 효과)

  • Skin contact allergy (SCA) is not life-threatening, but a large number of people have been suffered from the reactions caused by various kinds of chemicals and products. Thus, in this study, radiation technology was employed to improve the traditional herb addition method on the SCA reduction of Rhus verniciflua sap (RVS). Rhus verniciflua has traditionally been used as an herbal medicine plant, but its urushiol derivatives are known as a major allergen for the SCA. The present study was commenced to assess the allergenicity of both gamma-irradiated and non-irradiated RVS by using guinea-pig maximization test (GPMT) in order to probe the mechanism of an SCA. In the acute dermal irritation assays, non-irradiated RVS caused erythema, but the irradiated RVS did not provoke any erythema on the abdominal skin of the guinea pigs. From the result of the GPMT, urushiols, the main chemical components of RVS, were identified as an extreme skin sensitizer, and the removal of urushiols by irradiation extremely reduced the erythema. These results suggest that radiation technology is a novel method to reduce SCA through the removal of urushiols of RVS.

Antioxidant and anti-inflammatory activity of parts of Rhus javanica L. (붉나무의 부위 별 항산화 및 항염증 활성)

  • Rhus javanica L. is Anacardiaceae plant distributed in East Asia. We evaluated the antioxidant activity and antiinflammatory effect of leaf, branch, root of ethyl acetate fraction from R. javanica. To confirm effective each extraction, The antioxidant activity was evaluated using 1,1-Diphenyl-2-picryl-hydrazyl and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) scavenging activity assays, and the anti-inflammatory activity was evaluated based on inhibitory activities on the protein and mRNA expression of iNOS and COX-2 in LPS-induced RAW264.7 cells. The phenolic compounds content of each extract was analyzed with Folin reagents and HPLC/PDA method. The gallic acids were identified and quantified. The roots of R. javanica showed strong antioxidant activity. Its total phenolic compounds content were higher than the orders. In addition, anti-inflammatory activity inhibited the protein and mRNA expression of nitric oxide production factor, following the same pattern as contents of phenolic compounds included gallic acid and its antioxidant activity. In conclusion, R. javanica showed effective antioxidant and anti-inflammatory activity. Especially, the roots were evaluated to be highly valuable as a natural resource for reducing inflammation.

Induction and Proliferation of Callus in Rhus chinensis Mill. and Its Effect on Skin Whitening (붉나무 캘러스 유도 및 그 추출물로부터 피부미백 효과에 관한 연구)

  • The objective of this study was to optimize the condition for induction and proliferation of callus from Rhus chinensis Mill. and investigate the skin-brightening effect of Rhus chinensis callus (RCC). It was confirmed that the most proper plant growth regulator (PGR) for callus induction is 1.0 mg/L of 2,4-dichlorophenoxyacetic acid (2,4-D). The most optimal condition of PGR, medium and additives for callus proliferation were 2,4-D (1.0 mg/L), MS medium and citric acid, respectively. Inhibitory activities of tyrosinase were higher at 50 and 100 ㎍/mL of RCC extracts (41.86 and 75.56%, respectively) than arbutin (27.32%). As the results of measuring melanin inhibition in B16F1 melanocyte and B16F10 melanoma cell, RCC extracts increased its inhibitory activities concentration-dependently, and were found to have higher whitening effect than arbutin at a concentration of 100 ㎍/mL. Therefore, it is suggested that RCC can be used as an effective material for skin-brightening cosmetics.

MicroRNA Expression in Leukemia Cell Line(K562 cell) Using Rhus Verniciflua Stokes (건칠(乾漆)을 이용한 K562 만성 골수성 백혈병 세포주에서의 MicroRNA 발현 규명)

  • Objective : The purpose of the study was to identify expression profiling of miRNAs associated with cancers after treating allergen-removed Rhus Verniciflua Stokes and allergen-removed Rhus Verniciflua Stokes fumigaed Angelica gigas on leukemia cell lines. Methods : miRNA expression has been analyzed using miRNA array method through denaturation and hybridization after isolating the total RNA from leukemic cell line treated with 100 ㎍/㎖ of aRVS and aRVS-A each. Microarray expressions were interpreted as 'significant' on miRNAs when decreased less than 0.5 fold or increased more than 1.5 fold compared with the control group. Results : Among 158 miRNAs in total, 32 miRNAs were significantly presented in miRNAs expression. miRNA has been activated with a variety of genes for predicted targets, and the overexpressed miRNAs were categorized according to proliferation and metastasis of cancer in this study. The findings were reported that seven miRNAs (let-7b, miR-193a-5p, 296-3p, 26a, 22, 124a, 92b) showed significant expressions on proliferation and growth, seven miRNAs (miR-193a-5p, 26a, 200c, 183, 124a, 198, 210) presented meaningful expressions on invasion and metastasis, two miRNAs (let-7b, miR-210) were highly expressed on angiogenesis, five miRNAs (let-7b, miR-26a, 181d, 181c, 296-5p) related with apoptosis, and six miRNAs (let-7b, miR-200c, 183, 370, 124a, 191) were associated with prognosis of cancer and early diagnostic factors for cancer. Conclusion : The mechanism of miRNA takes a role in diagnosis, treatment, and prognotic factors for cancer as well. This study suggested that further detailed research on overexpression of specific miRNA should be carried out continuously in the future.

Rhus Trichocarpa Suppresses IgE-mediated Allergic Response In Vitro and In Vivo (개옻나무 추출물의 IgE 매개성 알레르기 반응 억제 효과 및 기전)

  • Mast cells is the key effector cells for IgE-mediated allergic responses. In this study, we investigated whether Rhus trichocarpa extract (RT) inhibited IgE-mediated allergic responses in mast cells and an allergic animal model. We further tried to find its mechanism of action in mast cells. We found that RT suppressed antigen-stimulated degranulation and production of TNF-${\alpha}$ and IL-4 in rat basophilic leukemia (RBL)-2H3 mast cells and bone marrow-derived mast cells (BMMC), as well as IgE-mediated passive cutaneous anaphylaxis (PCA) in mice. As the mechanism of action of RT, it inhibited the activation of spleen tyrosine kinase (Syk), a pivotal signaling molecule for activation of mast cells and that of LAT, a downstream adaptor molecule of Syk in $Fc{\varepsilon}RI$-mediated signal pathways. RT also suppressed the activation of mitogen-activated protein (MAP) kinases and Akt. The current results demonstrated for the first time that RT has the anti-allergic effect through inhibiting degranulation and secretion of cytokines by suppression of Syk in antigen-stimulated mast cells. Therefore, RT might be useful for allergic diseases.

Successful Outcome of Elderly Patients with Advanced Malignant Melanoma by Standardized Allergen-removed Rhus verniciflua Stokes Extract and Bojungikki-tang: Two Cases

  • Objectives: Advanced malignant melanoma (MM) has a poor prognosis, with an expected 2-year survival rate of 10 to 20%. It has long been recognized as an immunogenic tumor, and is worse for elderly patients. Many studies have suggested that herbal treatments improve immune functions, but few clinical studies have reported on this topic. Patients and History: We present two cases of female patients (72 and 77 years old, respectively) with advanced MM. The 72-year-old female patient was, at first, diagnosed with MM with multiple bone metastases. She received resection of the primary lesion, but refused further chemotherapy. The 77-year-old female patient was diagnosed with cutaneous MM of the left heel, with suspicion of sentinel node lymphadenopathy; however, she also refused any conventional treatment due to old age. Course of Therapy and Results: Both patients were exclusively treated with standardized allergen-removed Rhus verniciflua stokes (aRVS) extract combined with Bojungikki-tang (BT, Bu-Zhong-Yi-Qi-Tang in Chinese or Hochu-ekki-to in Japanese). Both patients are still alive and doing well (Feb. 2014), demonstrating that the 72-year-old patient has lived for 27 months and the 77-year-old patient has lived for 31 months without disease progression since the aRVS and BT administration. Conclusion: We suggest that the combination of aRVS extract and BT could be a candidate for overcoming the cancer's immunoediting process especially for elderly MM patients intolerant of conventional treatment.

Rhus verniciflua Stokes Attenuates Glutamate-induced Neurotoxicity in Primary Cultures of Rat Cortical Cells

  • The methanolic extract of Rhus verniciflua Stokes (RVS-T) and its fractions (RVS-H, RVS-C, RVS-E and RVS-B) showed significant neuroprotective activity against glutamate-induced toxicity in primary cultures of rat cortical cells. RVS-B, which showed the most potent neuroprotective activity, was further fractionated to yield RVS-B5. Treatment of cortical cells with the RVS-T, RVS-B and RVS-B5 reduced the cellular ROS level and restored the reduced activities of glutathione reductase and SOD induced by glutamate. Although, the activity of glutathione peroxidase was not virtually changed by glutamate, RVS-B5 increased the glutathione peroxidase activity. In addition, these three tested fractions significantly restored the content of GSH which was decreased by glutamate insult in our cultures. Taken together, it could be postulated that RVS extract, in particular its fraction RVS-B5, protected neuronal cells against glutamate-induced neurotoxicity through acting on the antioxidative defense system.

Purification and Characterization of Antioxidant Substance from the Stem Bark of Rhus verniciflua (옻나무 껍질에서 항산화물질의 정제와 특성)

  • In order to isolate antioxidant substances from Rhus verniciflua (RV) , the dried stem bark was extracted with water. The crude water extracts was purified by using HPLC method with a DEAE (anionic type) , CN and ODS column. The purified compound remained stable at pH 3.0∼6.0. but unstable above pH 6.5. It was stable at 100$^{\circ}C$ for 4 hours, but still had about 80% of residual activity after treatment at 100$^{\circ}C$ for 5 hours. In antimicrobial test, no inhibition was observed against Gram-positive and negative bacteria. This compound was stronger than that of commercial antioxidant showed that DPPH test, such as BHT, BHC at the same concentration (20 $\mu\textrm{g}$/ml) .
