Emitted CO2 is an attractive material for microbial electrochemical CO2 reduction. Microbial electrochemical CO2 reduction (i.e., microbial electrosynthesis, MES) using biocatalysts has advantages compared to conventional CO2 reduction using electrocatalysts. However, MES has several challenges, including electrode performance, biocatalysts, and reactor optimization. In this study, an MES system was investigated for optimizing reactor types, counter electrode materials, and CO2-converting microorganisms to achieve effective CO2 upcycling. In autotrophic cultivation (supplementation of CO2 and H2), CO2 consumption of Rhodobacter sphaeroides was observed to be four times higher than that with heterotrophic cultivation (supplementation of succinic acid). The bacterial growth in an MES reactor with a single-chambered shape was two times higher than that with a double chamber (H-type MES reactor). Moreover, a single-chambered MES reactor equipped with titanium mesh as the counter electrode (anode) showed markedly increased current density in the graphite felt as a working electrode (cathode) compared to that with a graphite felt counter electrode (anode). These results demonstrate that the optimized conditions of a single chamber and titanium mesh for the counter electrode have a positive effect on microbial electrochemical CO2 reduction.
역삼투 해수담수화(SWRO)공정의 큰 문제점 중 하나인 biofouling 현상을 초기에 감지하기 위한 센서 개발의 선행 연구로써, 본 연구는 역삼투막에 바이오필름을 형성하는 문제성 있는 박테리아를 센서의 타겟 박테리아로 제시하는 것에 중점을 두었다. 문헌조사와 실제 해수담수화 공정에서 사용된 해수 원수와 오염된 역삼투막에 존재하는 박테리아를 계통발생학적으로 분석한 결과를 토대로 Bacillus sp., Flavobacterium sp., Mycobacterium sp., P. aeruginosa, P. fluorescens, 그리고 Rhodobacter sp.의 여섯종의 모델 박테리아를 선정하였고, 선정된 모델 박테리아 중, 막 오염 잠재력을 가진 종을 찾아내기 위해 각각 박테리아의 역삼투막 부착 능력, 고압내성, 그리고 소수성을 비교 분석하였다. 그 결과, 역삼투막 부착능력은 Rhodobacter sp.와 Mycobacterium sp.가 뛰어났으며 소수성이거나 역삼투막(접촉각 약 $63^{\circ}$)과 비슷한 접촉각을 가진 박테리아가 역삼투막에 잘 부착하였다. 800 psi의 고압을 적용 한 후, Rhodobacter sp.는 여섯 종류의 모델 박테리아 중 59-73%의 가장 큰 개체수의 감소를 보였고, P. fluorescens는 1-29%로 가장 높은 고압내성을 보였다. 부착, 소수성, 고압내성 특성을 통한 역삼투막에 biofouling을 유발하는 영향력 있는 박테리아 선정 실험 결과, 여섯 종류의 모델 박테리아 중 Mycobacterium sp.가 부착 능력이 뛰어나고 높은 소수성 특성을 가지며, 800 psi의 고압에서도 50% 이상의 cell의 생물학적 활성능력을 가지고 있어, 막 오염을 유발시키는 가장 잠재력 있는 박테리아로 분석되었다.
이 연구에서는 글라이코 캘릭스 형성 박테리아를 이용한 콘크리트 구조체의 표면 코팅기술의 실제 적용의 중요성과 문제점을파악하기 위하여 유사 하수환경에서 Mock-up 실험을 수행하였다. 황산염 열화의 조건에서 원주형 콘크리트시험체의 압축강도와 질량의 변화를 평가하였다. 박테리아의 생장성 및 글라이코 캘릭스 생성량은 하수관에서 채취한 시료로부터 평가되었다. 이와 함께 유사 하수환경에서 Rhodobacter capsualtus의 적용이 주변 생태환경에 미치는 유해성을 평가하기 위해 차세대 염기서열 분석이 수행되었다. Mock-up 실험결과 개발된 코팅재가 적용된 콘크리트 시험체는 황산염 열화에 의한 콘크리트의 압축강도 저하 및 질량감소는 발생하지 않았으며, 하수 환경에서 우수한 열화저항성능을 갖는 것으로 평가되었다. 재령 91일에서 경화된 코팅재에서 분리된 박테리아의 생균수 및 글라이코 캘릭스 생성량은 각각 1.4×104cell/mL 및 67.5mg/㎤이었다. 더불어 글라이코 캘릭스 코팅 콘크리트 하수관이 설치된 하수에서는 유해균주가 발견되지 않았다. 이는 코팅 재료로서 활용된 Rhodobacter capsulatus가 하수 환경에서 주변 생태환경에 미치는 영향이 없음을 의미한다.
Rhodobacter capsulatus 유래의 hemA 유전자를 항시발현 벡터(pHCEIIB vector)에 클로닝한 후 과발현시킨 Escherichia coli BLR(DE3) 균주를 사용하여, 5-aminolevulinic acid (ALA) 생산을 위한 적정 배양조건을 조사하였다. ALA 생산을 위한 배양온도는 $37^{\circ}C$보다는 $30^{\circ}C$에서 더 나은 결과를 나타내었다. Glycine 농도가 세포생장에 미치는 영향이 컸으며, 세포생장을 저해하지 않고ALA를 고농도로 생산하기 위한 적정 glycine 농도는 5-10 g/L 수준이었다. Succinic acid의 적정 첨가수준은 10-20 g/L이었으며, succinic acid의 일부를 glutamate로 대체할 경우에 ALA의 생산량을 향상시키는 효과가 있었다. 한편, 배지 중의 glucose 첨가는 ALA 생산성을 저하시키는 결과를 초래하였다. 이상과 같은 배양 조건 최적화를 통한 jar-fermentor 발효실험을 통하여, 고가의 유도제(IPTG)나 ALA dehydratase inhibitor를 첨가하지 않고도 8.2 g/L 수준의 고농도 ALA를 얻을 수 있었다.
광합성세균을 이용한 고농도 유기폐수인 두부공업폐수의 정화처리와 동시에 두부공업폐수를 생물제초제인 ${\delta}-aminolevulinic$ acid (ALA) 생산용 대체기질로 이용할 목적으로 Rhodobacter capsulatus KK-10을 사용하여 COD 제거율과 ALA 생산능을 검토하였다. 사용된 두부공업폐수의 성상은 pH 4.2, COD/BOD와 BOD/N의 비율은 0.98 및 17.2, BOD : N : P의 함유비율은 10 : 6 : 4, 젖산 함유량 1,080 ppm 등으로 광합성세균의 생육에 적당한 조건이었다. 두부공업폐수에서 본 균주를 4일 동안 배양한 결과 약 94% COD 값의 감소와 1.2 g/l 균체생육을 보였으며 대수기 중기에서 ALA 탈수효소 저해제인 levulinic acid(LA)를 15 mM 첨가한 결과 55 mg/l의 ALA 생산량을 나타내었다. ALA의 전구물질인 succinate 및 glycine과 LA를 각각 15 mM의 농도로 동시 첨가시 최대 114 mg/l의 생산량을 보였다. 상기의 물질들을 각각 15 mM 농도로 연속 첨가한 후 ALA 생성량을 검토한 결과 LA 1회와 glycine과 succinate를 3회에 걸쳐 첨가하였을 때 120 mg/l로 가장 효과적이었으며 COD 제거율은 92%를 나타내었다.
Purple non-sulfur bacteria, Rhodobacter sphaeroides KD131 grew to reach the maximum cell concentration in 45 hrs of incubation in the synthetic media containing (NH4)2SO4, L-aspartic acid and succinic acid as the carbon and nitrogen sources, respectively, at 30oC under 8 klux irradiance using halogen lamp. The strain produced hydrogen from the middle of the logarithmic growth phase and continued until the cell growth leveled out. The strain grew and produced hydrogen under the irradiance of 3-30 klux, but cell growth was inhibited over 100 klux. In addition, anaerobic/light culture condition was better than the aerobic/dark on the hydrogen production. Among various photo-bioreactors examined, the flat-vertical reactor manufactured using clear acrylic plastic material showed the best hydrogen production rate at the given culture condition.
With the aim of increasing the $CoQ_{10}$ production in mass culture, the effect of aeration-agitation on the $CoQ_{10}$ production using Rhodobactor sphaeroides was investigated in a l-L bioreactor. The maximum $CoQ_{10}$ production was 1.69 mg/g of dry cell weight under conditions of 50 Lux, $30^{\circ}C$, 300 rpm, and 5-vvm aeration. The $CoQ_{10}$ production was improved to produce 2.91 mg/g of dry cell weight under reduced conditions of agitation speed (200 rpm) and aeration rate (0.2 vvm). When R. sphaeroides was cultivated under more reduced DO levels during the exponential phase of the cell, the $CoQ_{10}$ production yield of 3.88-mg/g dry cell weight was the maximum obtained. Therefore, poorer conditions of aeration-agitation resulted in higher production of $CoQ_{10}$, and thus DO content was a crucial factor in the production of $CoQ_{10}$. Accordingly, it was necessary to control the DO concentration in order to enhance the $CoQ_{10}$ biosynthesis within a large-scale production.
