A Bisdesmosidic Cholestane Glycoside from the Rhizomes of Polygonatum sibiricum

  • A bisdesmosidic cholestane glycoside was isolated from the rhizomes of Polygonatum sibiricum and the structure was elucidated by spectroscopic methods and acid hydrolysis as (22S)-cholest-5-ene-$1{\beta}$,$3{\beta}$,$16{\beta}$,22-tetrol 1-O-${\alpha}$-L-rhamnopyranosyl 16-O-${\beta}$-D-glucopyranoside. This compound exhibited weak cytotoxic activity with the $IC_{50}$ value, $63.6\;{\mu}M$ in human MCF-7 breast cancer line, whereas it failed to show agonistic activity at $100{\mu}M$ in TGR5 assay with Chinese hamster ovary (CHO) cells. This is the first report of a bisdesmosidic cholestane glycoside from Polygonatum species and the full assignments of $^1H$, $^{13}C$ NMR by HMBC, TOCSY and NOESY experiments were provided.

Neuroprotective Effects of Some Plant Extracts Against Dopamine-induced Oxidative Stress on Neuronal Cell (Dopamine에 의해 산화적 스트레스를 받은 Neuronal Cell에 뇌 보호 효과를 가지는 수종 생약추출물의 검색)

  • Parkinson's disease (PD) is the second most common neurodegenerative disorder after Alzhemier's disease. Neuropathologically, PD is characterized by the selective loss of dopaminergic neurons. The neuronal toxicity of cytosolic excess dopamine (DA) has been described in many studies using several cell lines. In dopaminergic neurons, cytosolic excess DA is easily oxidized via monoamine oxidase (MAO)-B, tyrosinase or by auto-oxidation to produce neurotoxic metabolites such as DA quinone. So, in the present study, we induced cell death by treatment of DA ($600{\mu}M$) in human neuroblastoma SH-SY5Y cell which was treated samples before 24 hr, and cell viability was measured by fluorescence activated cell sorter (FACs) analysis. Of those tested, the extracts of Poria cocos (赤茯笭)(whole), Gastrodia elata (rhizomes), Eucommia ulmoides (炒)(barks), Syneilesis palmata (whole), Acorus gramineus (rhizomes), Ligustrum japonicum (leaves) showed neuroprotective effects in dose dependent manner.

  • A new species, Viola breviflora Jungsim Lee & M. Kim is named and described from Mt. Seogbyeongsan, Gangwondo Province, Korea. The Viola breviflora shares several characteristics (acaulescent leaves, white flowers, pubescent leaf blades and petioles, etc.) with its related species V. keiskei Miq., but is distinct from V. keiskei which has cordate leaf shapes, deep-cordate leaf bases, long pedicels, few flowers, shapeless rhizomes, green fruits, and low altitude habitats by having ovate leaf shapes, shallow-cordate leaf bases, short pedicels, many flowers, Lshape rhizomes, purplish green fruits, and high altitude habitats.

Anti-osteoporotic and Antioxidant Activities by Rhizomes of Kaempferia parviflora Wall. ex Baker

  • In this report, we investigated the antioxidant (peroxyl radical-scavenging and reducing capacities) and anti-osteoporotic activities of extracts and isolated constituents (1 - 16) from the rhizomes of Kaempferia parviflora Wall. ex Baker on pre-osteoclastic RAW 264.7 cells. Compound 5 exhibited significant peroxyl radical-scavenging capacity, with TE value of $8.47{\pm}0.52{\mu}M$, while compound 13 showed significant reducing capacity, with CUPRAC value of $5.66{\pm}0.26{\mu}M$, at $10.0{\mu}M$. In addition, flavonoid compounds 2, 4, 6, 8, 10, 12, and terpene compound 15 showed significant inhibition of tartrate-resistant acid phosphatase (TRAP) in NF-${\kappa}B$ ligand-induced osteoclastic RAW 264.7 cells, with values ranging from $16.97{\pm}1.02$ to $64.67{\pm}2.76%$. These results indicated that K. parviflora could be excellent sources for the antioxidant and anti-osteoporotic traditional medicinal plants.

Protective Effects of Sinomenium acutum Rhizome and Its Alkaloids against Dexamethasone-induced Atrophy in C2C12 Myoblasts (방기 뿌리 (Sinomenium acutum rhizome) 추출물 및 그 알칼로이드 성분의 항근위축 효과)

  • Skeletal muscle is an organ that regulates biological metabolic energy. Its dysfunction causes decline of body functions and disability, thus deteriorating the overall quality of life. Various materials are being developed with an anti-sarcolytic effect. However, anti-sarcolytic effect of Sinomenium acutum rhizomes extract (SAE) remains unclear. Therefore, this study aimed to investigate anti-muscle atrophy effects of SAE and its alkaloids, including sinomenine (SIN), magnoflorine (MF), acutumine (ACU), and N-ferultyramine (NFT) isolated from SAE, on dexamethasone (Dex)-induced myotubules. C2C12 myogenic cells differentiated for 6 days were treated with 1 mM Dex for 24 hours. Induction of muscular atrophy was confirmed by a decrease in myogenin expression. We found that Dex increased expression levels of muscle-specific ubiquitin ligases MuRF1 and MAFbx/atrogin-1. However, mRNA and protein levels of these muscle-specific ubiquitin ligases were significantly reduced by cotreatment with SIN, MF, and NFT in myotubes. Glucose uptake reduced by Dex in myotubules were also restored by SIN, MF, and NFT treatments. These results suggest that SIN, MF, and NFT can reduce muscle wasting and enhance glucose uptake in Dex-treated myotubes, highlighting their potential as therapeutic agents to prevent muscle atrophy.

Establishment of Herbicide Screening Methods for Reed (Phragmites communis Trin.) Control - I. Propagation of Reed (갈대(Reed, Phragmites communis Trin.)의 방제를 위한 제초제 스크리닝방법의 확립 - I. 갈대의 육묘)

  • This experiment was conducted to find out an effective propagation method for reed(Phragmites communis Trin.), ensuring a continuous herbicide screening for reed control. Reed propagation methods were compared under a greenhouse condition using tour different materials; seeds, rhizomes, depressed stolons of P. japonica Steud., and stem cuttings. Although reed seeds were easy to harvest and store, their germination rate(${\leq}$5%) was very low and seedling growth from the seeds was slow. Rhizomes were difficult to harvest and their harvest time was limited from November to March. Furthermore, reed propagation using rhizomes had problems of a relatively low germination rate(46%), no uniformity in size and shape, individual differences at the early stage of growth, and difficulties in material storage. Rate of reed growth from rhizomes was higher in commercial soil mix(Boo Nong soil) than in sand or in sand+upland soil(1:1). Depressed stolons of P. japonica had a moderate germination rate(65%) and were relatively easy to harvest. However, their harvest time was limited only from August to September. Propagation method using stem cuttings had several advantages over the above methods using other materials. Reed plants could uniformly be propagated from the stem cuttings with a relatively high germination rate(75%). Stem cuttings of central nodes showed a higher germination rate compared to those of upper or lower nodes. Stem cuttings from the field should be used immediately after harvest, since their germination rate decreased rapidly when they were stored under a wet- or a dry-refrigerated condition. Furthermore, the germination of stem cuttings tended to decrease when they were collected from the field after August. This indicates that there is a limitation of harvest time for stem cuttings. However, a year-round propagation of reed using stem cuttings is possible if parent plants are grown in a greenhouse, and thus herbicide screening for reed control could continuously be performed.

Alpha-glucosidase Inhibition Activity of Methanol Extracts Obtained from Nine Pteridophyte Species Native to Korea (자생 양치식물 9종의 성엽 및 근경 추출물의 α-glucosidase 억제 활성)

  • This study was conducted not only to analyze ${\alpha}$-glucosidase inhibition activity with fronds and rhizomes of nine Pteridophyte species, but also to select the plant materials suitable for natural ${\alpha}$-glucosidase inhibitor. Harvested rhizomes and fronds were washed, freeze-dried and grinded. After conducting ultrasonification extraction for 30 minutes in ultrasonic water tank with 100% methanol solvent, and vacuum filtration, ${\alpha}$-glucosidase inhibition activity was measured. Acarbose was used as the positive control. After mixing $100{\mu}L$ of 0.7 unit ${\alpha}$-glucosidase enzyme solution into $50{\mu}L$ of extract and reacting them at $37^{\circ}C$ for 10 minutes, $50{\mu}L$ of 1.5 mM ${\rho}$-NPG solution was taken and reacted at $37^{\circ}C$ for 20 minutes. The reaction was stopped with 1 mL of 1 M $Na_2CO_3$ and absorbance was measured in 405 nm. With the regression analysis, the content of solubility solids (the value of $IC_{50}$) which can inhibit 50% of 0.7 unit ${\alpha}$-glucosidase solution's activity was investigated. The frond ($IC_{50}=14.00{\sim}913.33{\mu}g{\cdot}mL^{-1}$) and rhizome extracts ($IC_{50}=12.93{\sim}205.84{\mu}g{\cdot}mL^{-1}$) of nine Pteridophyte species showed higher ${\alpha}$-glucosidase inhibition activity in comparison with acarbose ($IC_{50}=1413.70{\mu}g{\cdot}mL^{-1}$). The extracts of fronds and rhizomes showed higher value than acarbose by 1.55~100.98 and 6.87~109.33 times each. Especially, ${\alpha}$-glucosidase inhibition activities of Pyrrosia lingua in fronds and Osmunda cinnamomea var. fokiensis in rhizomes were the highest. The necessary biomass of fronds and rhizomes for inhibiting 50% of ${\alpha}$-glucosidase activity showed the lowest value, 0.35, 0.27 mg each, in O. cinnamomea var. fokiensis. $IC_{50}$ value of P. lingua was the highest among fronds of nine Pteridophyte species, but content of soluble solids was 2.4 times less than O. cinnamomea var. fokiensis. So frond of O. cinnamomea var. fokiensis is more economic in comparison with P. lingua. As the result of this study, O. cinnamomea var. fokiensis showed high ${\alpha}$-glucosidase inhibition activity even with small biomass. Therefore it was considered to be high-valued economic material as natural ${\alpha}$-glucosidase inhibitor.

Growth Characteristics and Available Component of Saururus chinensis Baill in Different Soil Texture (토성에 따른 삼백초 생육특성과 유효성분 함량)

  • This study was carried to investigate the effect of soil texture on the growth and the contents of quercetin-glycoside and lignans, and to improve the qualities of Saururus chinensis Baill. Soil texture resulted no significant effects on the number of nodes, the number of leaves, the number of branches and dry matter ratio. However, the shoot dry weight was higher in sandy loam, loam, silt loam and sand soil in that order. Although the weight of rhizomes of below 5 mm in diameter was not significantly different among soil textures, the weight of rhizomes between 5.1 and 10.9 mm and the weight of rhizomes of above 11 mm in diameter ranged $437{\sim}465\;g$ and was larger in clay loam than in other soil textures. No significance difference was showed in rhizome dry ratio ranging from 19.1 to 20.8%. The amount of quercetin-glycoside in leaves was higher in loam and sandy loam and ranged from 219.3 to 222.4 mg/100 g of quercetin-glycoside quercitrin, rutin, isoquercitrin and hyperin were higher in that order. On the other hand, quercetin-glycoside contents in stem were 14.8 mg/100 g and 12.4 mg/100 g in sandy and sandy loam, respectively, and were higher than in other soil textures of quercetin-glycoside constituents, the content of rutin was the highest. The content of lignans was increased in clay loam, loam, sandy loam, and sandy in that order of lignans, the manassatin B was the highest.

Regrowth Patterns of the Perennial Weeds Treated with Sulfonylureas and Starch Degradation in the Rhizome (Sulfonylurea계 제초제가 처리된 다년생 잡초의 재생양상과 저장전분량의 변화)

  • Characteristics of regrowth and starch degradation in perennial weed rhizomes were investigated in a greenhouse. Cyperus serotinus started regrowth at 24 days after treatment of 1.25 g ai/ha of pyrazosulfuron-ethyl. The regrowth of Sagittaria trifolia, Eleocharis kuroguwai, and S. pygmaea required $30{\sim}39$ days, and Potamogeton distinctus regrew at 55 days after treatment of 1.25 g ai/ha of pyrazosulfuron-ethyl. However, all of 5 perennial weeds hardly regrew until 45 days after treatment more than 5 g ai/ha of pyrazosulfuron-ethyl. Regrowth of C. serotinus 4-node rhizomes was 2 times higher than 2-node rhizomes, and $1{\sim}1.5g$ of E. kuroguwai large tubers regrew faster than $0.3{\sim}0.5g$ of small tubers treated with bensulfuron-methyl. Regrowth of C. serotinus was enhanced with delayed application of bensulfuron-methyl, however, 2-leaf stage of E. kuroguwai plants regrew better than 3 leaf stage of plants when treated with equal volume of bensulfuron-methyl. The critical periods of S. trifolia and E. kuroguwai growth depending upon the rhizome detachment were 12 and 18 days after sprouting, respectively. The starch stacked in the S. trifolia and E. kuroguwai tubers were exhausted at 18 and 27 days after sprouting, respectively. The highest level of sugar contents was showed at 9 days after sprouting in S. trifolia, and 18 days after sprouting in E. kuroguwai.

Estimation of Aboveground Biomass and Belowground Nutrient Contents for a Phyllostachys pubescens stand (맹종죽(孟宗竹) (Phyllostachys pubescens) 임분(林分) 내(內) 지상부(地上部) 생체량(生體量) 및 지하부(地下部) 양분(養分) 함량(含量) 추정(推定))

  • Above and belowground biomass for Phyllostachys pubescens was determined in Jinju, Gyeongsangnam-do. Regression analyses of dry weights of culms, foliages, and twigs versus diameter at breast height were used to calculate regression equations of the form of log Y = a + blogX. Total aboveground biomass for Phyllostachys pubescens was 69.7 ton/ha and rhizomes and roots biomass were 13.7 ton/ha and 7.5 ton/ha, respectively. Culms account for about 60% of total aboveground biomass. The aboveground biomass of each component was decreased in the order of culms>foliages>twigs. As diameter at breast height grew thicker, the proportion of culms to total aboveground biomass increased. The proportion of dry weight of culms to green weight gradually increased with height in a bamboo tree and ages. Nutrients (kg/ha) of litter layer were distributed as follows: N(45.1), Ca(17.3), K(6.1), Mg(3.6), P(3.5) and Na(0.7). Nitrogen and K were given much weight in total nutrients of rhizomes and roots. These results will be useful in measuring carbon stock and drawing up management plan to increase it for Phyllostachys pubescens stand.
