삼색싸리 메탄올 추출물의 3T3-L1지방세포와 db/db 마우스에서의 PPARγ 작용제와 인슐린 유사효과를 통한 혈당조절 개선효과 (Methanol extract of Lespedeza maximowiczii var. tricolor Nakai improves glucose metabolism through PPARγ agonist and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice)

  • 이 연구의 목적은 당 대사에 대한 삼색싸리(Lespedeza maximowiczii var. tricolor Nakai; LMTN)의 효과를 조사하는 것이다. LMTN 추출물은 대조군과 비교하여 3T3-L1 지방 세포에서 당 섭취능 및 지질축적을 유의하게 향상시켰다. 또한, 3T3-L1 지방 세포에서 LMTN 추출물은 퍼옥시좀 증식제 활성화 수용체(PPAR)γ, 인슐린수용체기질-1 (IRS-1) 및 포도당수송체(GLUT)4의 단백질 발현을 유의하게 증가시켰다. LMTM 추출물의 당 섭취능 또는 인슐린 신호 전달계의 조절 효과는 양성 대조물질인 트로글리타존 또는 피니톨보다 낮았지만 PPARγ 단백 활성화는 증가하였다. 또한, LMTM 추출물은 인슐린 유사효과를 나타냈다. db/db 마우스에서, LMTN 추출물(250 mg/kg BW)은 물과 식이 섭취량, 혈당, 중성지방과 총 콜레스테롤 함량을 유의적으로 감소시켰다. 더불어 지방과 근육조직에서의 PPARγ 및 GLUT4 mRNA의 발현도 LMTN 추출물 투여군에서 유의적으로 증가되었다. 따라서, 본 연구의 결과는 LMTN 추출물이 3T3-L1 지방세포 및 db/db 마우스에서 PPARγ 및 인슐린 유사효과를 통해 당 대사를 조절하는 것으로 밝혀졌다.

  • CRP (cAMP receptor protein)은 cAMP와 결합하여 cAMP-CRP 복합체를 형성하여 전사조절의 조절인자로서 작용한다. crp 유전자에 변이를 도입하여 cAMP의 비존재 상태에서 cAMP-CRP와 비슷한 기능을 가진 crp 유전자가 도입된 대장균 MK2001 (crp, cya::km)을 숙주로 사용하여 cAMP 혹은 cGMP의 비존재하에서도 mal 유전자의 발현을 촉진시키는 유전자 sfs (sugar fermentation stimulation) 수 종을 클로닝 하였다. 본 실험에서는 이미 밝혀진 nlp (Ner like protein) 유전자와 같이, sfs의 새로운 유전자를 탐색하여, 그 중 sfs4의 2126 bp 전 염기배열을 결정하고, 잠정적인 sfs4의 promoter 영역에는 CRP 단백질과의 결합 DNA 공통 염기배열(5' AAT TGTGA ACACCA TCACC CGT 3')이 존재함을 확인했다. lacZ 융합 유전자를 작성하여 TP2010R1 MK2001의 균주에서 cAMP를 첨가할 경우 각각 2.3배, 1.8배의 ${\beta}-galactosidase$ 활성이 증가하는 것으로 보아 sfs4는 cAMP-CRP에 의해 발현 조절을 받는 것으로 나타났다.

유류 유출 사고 해역에 서식하는 패류의 다환방향족탄화수소(PAHs) 위해도 평가 (Polycyclic Aromatic Hydrocarbons Hazard Assessment of Shellfish due to Oil Spill Accidents)

  • On 7 December 2007, about 12,547 kL of crude oil spilled from the Hong Kong registered tanker Hebei Spirit along the west coast of the Republic of Korea, including Taean-gun, Chungcheongnamdo Province. This study evaluated the safety of seafood collected from the coastal area polluted by the crude oil. The range of total polycyclic aromatic hydrocarbons (${\sum}PAHs$) at 22 stations was 3.9-37.1 ng/g. The concentration of ${\sum}PAHs$ was higher in oysters, Crassoatrea gias, than that in short-necked clams, Ruditapes philippinarum. Benzo(a)pyrene, a highly toxic PAH, ranged from 0.07-1.47 ng/g, which did not exceed the European Union regulatory limit for benzo(a)pyrene. The toxicity equivalent of benzo(a)pyrene in oysters and short-necked clams was 0.49-1.70 and 0.09-1.01 ng/g, respectively. The estimated life time cancer risk was very low, i.e., $1.31{\times}10^{-8}$ for the oysters and $6.9{\times}10^{-9}$ for the short-necked clams. The body burden of PAHs in bivalves originated mostly from petroleum contamination, but the levels was not sufficiently high to harm human health.

음식 용기로 사용하는 각섬암의 암석-광물학적 특성: 그 효용성과 안정성에 대한 고찰 (Petrological and Mineralogical Characteristics of Amphibolite Used as Rock Bowl and Pot: Implications for Its Utility and Stability)

  • 우리나라에서 암석의 사회적 이용측면에서 대중화된 것 중 하나가 음식점에서 흔히 사용하는 돌그릇이다. 이 돌그릇은 대부분이 Mg과 Ca 함량이 높은 각섬암으로 제작되었으며, 주구성광물은 각섬석, 양기석, 투각섬석, 사장석, 투휘석이다. 각섬석은 대부분 주상으로 산출되며, Mg-각섬석 내지는 쳐마카이트의 광물조성을 보인다. 양기석과 투각섬석은 단경/장경 비가 $0.10{\sim}0.13$인 침상의 조직을 보이며, Mg/(Mg+Fe) 비가 약 0.65-0.90의 범위를 보인다. 이런 침상의 양기석과 투각섬석은 비석면형에 속한다. 이 광물들을 함유한 암석을 음식 용기로 사용하기 위해서는 구성광물의 종류와 결정형태와 같은 암석학적 또는 광물학적 자료를 바탕으로 그 안정성과 효용성에 대한 품질기준이 필요하다고 판단된다. 또한 음식용기 제작에 사용되는 원석과 상업적으로 유통되고 있는 곱돌의 산출상태와 암석-광물 화학성분의 상이성은 현재 유통되고 있는 대부분의 곱돌 음식 용기들이 원석 채취지역이 불분명한 암석으로 제작되었음을 지시한다. 이것은 음식용기로 부적합한 각섬암이 음식 용기로 사용될 잠재적 위험성이 있으며, 따라서 인간의 생활/건강과 밀접하게 연관되어 사용하는 암석의 지질학생물학적인 연구와 체계적인 관리와 조사가 요구된다.

서울시 중앙버스전용차로 도입의 부가적인 대기오염 영향성 평가 (Impact of the Exclusive Median Bus Lane System on Air Pollution Concentrations in Seoul, Korea)

  • Since many previous studies reported the health effect of air pollution and indicated traffic as a major pollution source, significant policy efforts have been made to control traffic to reduce air pollution. However, there have been few studies that evaluated such policy implementation. In Seoul, Korea, the exclusive median bus lane system was implemented in 2004, and the metropolitan government applied air pollution reduction policies such as conversion of diesel buses to compressed natural gas buses and installation of emission control devices. This paper aimed to investigate the impact of the exclusive median bus lane system on air pollution reduction. Using hourly concentrations of particulate matter ($PM_{10}$) and nitrogen dioxide ($NO_2$) measured at 131 regulatory monitoring sites in Seoul and Gyeonggi-do for 2001-2014, we calculated annual and daily average concentrations at each site. We assessed the impact of the policy using differences-in-differences analysis by annual and daily average models after adjusting for geographic and/or meteorological variables. This method divides population into treatment and control groups with and without policy application, and compares the difference between the two time periods before and after the policy implementation in the treatment group with the difference in the control group. We classified all monitoring sites into treatment and control groups using two definitions: 1) Seoul vs. Gyeonggi-do; 2) within vs. outside 300 meters from the median bus lane. Pre- and post-policy periods were defined as 2001-2005 and 2006-2014, and 2004 and 2014 in the annual and daily models, respectively. The decrease in $PM_{10}$ concentrations between the two periods across monitoring sites in the treatment group was larger by $1.73-5.88{\mu}g/m^3$ than in the control group. $NO_2$ also showed the decrease without statistical significance. Our findings suggest that an efficient public transport policy combined with pollution abatement policies can contribute to reduction in air pollution.

2-deoxy-D-glucose와 quercetin이 방사선조사 MC3T3-E1 골모세포주의 분화시 bone sialoprotein과 osteocalcin 유전자의 발현에 미치는 영향 (Effects of 2-deoxy-D-glucose and quercetin on the gene expression of bone sialoprotein and osteocalcin during the differentiation in irradiated MC3T3-E1 osteoblastic cells)

  • Purpose : To investigate the effects of 2-deoxy-D-glucose (2-DG) and quercetin (QCT) on gene expression of bone sialoprotein (BSP) and osteocalcin (OC) during the differentiation in irradiated MC3T3-E1 osteoblastic cells. Materials and Methods : When MC3T3-E1 osteoblastic cells had reached 70-80% confluence, cultures were transferred to a differentiating medium supplemented with 5 mM 2-DG or $10{\mu}M$ QCT, and then irradiated with 2, 4, 6, and 8 Gy. At various times after irradiation, the cells were analyzed for the synthesis of type I collagen, and expression of BSP and OC. Results : The synthesis of type I collagen in cells exposed to 2 Gy of radiation in the presence of 2-DG or QCT showed no significant difference compared with the control group within 15 days post-irradiation. When the cells were irradiated with 8 Gy, 2-DG facilitated the irradiation mediated decrease of type I collagen synthesis, whereas such decrease was inhibited by treating with QCT. During MC3T3-E1 osteoblastic cell differentiation, the mRNA expression of BSP and OC showed the peak value at 14 days and 21 days, respectively. 2-DG or QCT treatment alone decreased the level of BSP mRNA, but increased the OC mRNA level only at early time of differentiation (day 7). In the cells irradiated with 2, 4, 8 Gy, the mRNA expression of BSP and OC decreased at 7 days after the irradiation. The cells were treated with various dose of radiation in the presence of 2-DG or QCT, the mRNA level of both BSP and OC increased although this increase was observed at low dose of radiation (2 Gy) and at the early stage of differentiation. However, when the cells were exposed to 4, 6, or 8 Gy, the increase of BSP and OC mRNAs was detected only in cells co-incubated with QCT. Conclusion : This study demonstrates that 2-DG and QCT affect differently the expression of bone formation related factors, type I collagen, BSP, and OC in the irradiated MC3T3-E1 osteoblasic cells, according to the dose of radiation and the times of differentiation. Overall, the present findings suggest that 2-DG and QCT could have the regulatory roles as radiation-sensitizer and -protector, respectively.

고삼추출물이 치은섬유아세포의 세포주기 조절단백질 발현에 미치는 영향 (Effects of Sophorae Radix Extracts on the Expression ofcell cycle regulatory porteins in Human Gingival Fibroblasts)

  • Fibroblasts are major cellular components of gingiva and periodontal ligament. They regulate the healing process after surgery or injury. Recently, many natural medicines, whose advantages are less side effects and possibility of long-term use, have been studied for their capacity, their anti-bacterial and anti-inflammatory effects and regenerative potential of periodontal tissues. Sophorae radix have been traditionally used as an anti-bacterial and antiinflammatory drug in oriental medicine. The purpose of present study was to investigate the effects of Sophorae radix extract on cell cycle progression and its molecular mechanism in human gingival fibroblasts. Sophorae radix extracts($100{\mu}g/ml$) notably increased cell proliferation and cell activity in the human gingival fibroblasts as compared to non-supplemented controls. There was an increase in the S phase and a decrease in the G1 phase in $100{\mu}g/ml$ of Sophorae radix extracts group as compared to non-supplemented controls. The level of cyclin E and cdk 2 protein in test group was higher than that of control groups. But that of cyclin D, cdk 4, and cdk 6 was not distinguished from controls. The level of p53 protein in test group was lower than that of controls, whereas that of p21 was not different. The level of pRB protein in test group was higher than that of controls, whereas that of p16 was lower. These results indicate that the increase of cell proliferation by Sophorae radix extracts may be due to the increased expression of cyclin E and cdk 2, and the decreased expression of p53 and p16 in human gingival fibroblasts.

중국의 최근 대기오염 규제 및 관리 정책에 대한 고찰 (An Overview on China's Recent Air Pollution Regulation and Management Policy)

  • 중국의 고속 경제성장과 도시화 산업화는 생활환경 전반을 급격히 악화시키고 있으며, 특히 대기오염물질은 중국 국내 뿐 아니라 인접국가인 한국의 대기환경 악화의 주요 외부요인으로 작용하고 있다. 최근 중국 정부는 석탄연소로 인한 대기오염에 대한 방지 관리와 대기환경 개선 정책을 대대적으로 시행해 왔다. 본 연구에서는 석탄연소에 따른 대기오염과 석탄화력 발전 대기오염물질 배출에 대한 중국정부의 규제관리 동향을 살펴봄으로써 중국 대기환경 정책의 주요 제도적 배열의 흐름과 상세 현황을 파악하였다. 그 결과, 석탄화력발전 대기오염물질 배출에 대한 중국 정부당국의 규제는 권역별 규제와 발전설비 유형별 규제 실시 등을 통해 이전보다 세분화된 것을 확인하였고, 대기오염물질 배출허용 기준 역시 강화되고 있으며, 13차 5개년 계획 기간의 목표년도인 2020년에 맞춰 직접적인 규제 및 방지 관리 등이 정책제도 측면에서 더욱 강화될 것으로 파악되었다. 하지만, 중국 대기질 개선 정책의 실질적인 효과와 실행 가능성 여부는 과학적 근거에 기반을 두고 신중하게 평가되어야 하며, 한국 등 인접국가에 미치는 영향이 충분히 고려되지 않은 한계점에는 개선이 요구된다. 이를 위해서는 지속가능한 한 중 환경협력 네트워크를 구축하고 강화시켜 나가는 것이 필수적으로 전제되어야 하며, 비단 중국의 대기오염뿐만 아니라 에너지정책, 산업 기술, 글로벌 환경거버넌스 등 다양한 요인을 고려한 통합적 접근과 중장기적인 연구가 필요하다.

  • We have established three immortal bovine muscular epithelial (BME) cell lines, one spontaneously immortalized (BMES), the second SV40LT-mediated (BMEV) and the third hTERT-mediated (BMET). The morphology of the three immortal cell lines was similar to that of early passage primary BME cells. Each of the immortal cell lines made cytokeratin, a typical epithelial marker. BMET grew faster than the other immortal lines and the BME cells, in 10% FBS-DMEM medium, whereas neither the primary cells nor the three immortal cell lines grew in 0.5% FBS-DMEM. The primary BME cells and the immortal cell lines, with the exception of BMES, made increasing amounts of p53 protein when treated with doxorubicin, a DNA damaging agent. On the other hand, almost half of the cells in populations of the three immortal cell lines may lack $p16^{INK4a}$ regulatory function, compared to primary BME cells that were growth arrested by enforced expression of $p16^{INK4a}$. In soft-agar assays, the primary cells and immortal cell lines proved to be less transformed in phenotype than HeLa cells. The three immortal epithelial-type cell lines reported here are the first cell lines established from muscle tissue of bovine or other species.

체외배양 중인 생쥐 난소에서 초기난포 조절인자의 발현 (mRNA Expression of the Regulatory Factors for the Early Folliculogenesis in vitro)

  • Objective: To understand the crucial requirement for the normal early folliculogenesis, we evaluated molecular as well as physiological differences during in vitro ovarian culture. Among the important regulators for follicle development, anti-Müllerian hormone (AMH) and FSH Receptor (FSHR) have been known to be expressed in the cuboidal granulosa cells. Meanwhile, it is known that c-kit is germ cell-specific and GDF-9 is also oocyte-specific regulator. To evaluate the functional requirement for the competence of normal follicular development, we investigated the differential mRNA expression of several factors secreted from granulosa cells and oocytes between in vivo and in vitro developed ovaries. Materials and Methods: Ovaries from ICR neonates (the day of birth) were cultured for 4 days (for primordial to primary transition) or 8 days (for secondary follicle formation) in ${\alpha}$-MEM glutamax supplemented with 3 mg/ml BSA without serum or growth factors. The mRNA levels of the several factors were investigated by quantitative real-time PCR analysis. Freshly isolated 0-, 4-, and 8-day-old ovaries were used as control. Results: The mRNA of AMH and FSHR as granulosa cell factors was highly increased according to the ovarian development in both of 4- and 8-day-old control. However, the mRNA expression was not induced in both of 4- and 8-day in vitro cultured ovaries. The mRNA expression of GDF-9 known to regulate follicle growth as an oocyte factor was different between in vivo and in vitro developed ovaries. In addition, the transcript of GDF-9 was expressed in the primordial follicles of mouse ovaries. The mRNA expression of c-kit was not significantly different during the early folliculogenesis in vitro. Conclusion: This is the first report regarding endogenous AMH and FSHR expression during the early folliculogenesis in vitro. In conclusion, it will be very valuable to evaluate cuboidal granulosa cell factors as functional marker(s) for normal early folliculogenesis in vitro.
