Regeneration from Storage Root Disk Culture of Purple Sweet Potato

  • Sweet potato has low regeneration capacity, which is a serious obstacle for the fruitful production of transgenic plants. Simple and rapid regeneration method from storage root explants of purple sweet potato (Ipomoea batatas L.) was investigated. The embryogenic callus was observed from 4 cultivars and its highest rate was induced at 1 μM 2,4-D after 5 weeks of culture. Result revealed that a low concentration of 2,4-D and low light intensity was important factors for embryogenic callus formation. After subculture on medium with 5 μM ABA for 4 days, subsequently, occurred the regeneration of shoots within 4 weeks when these embryogenic callus was transferred onto the MS hormone free medium. Regenerated shoots were developed into platelets, and grown normal plants in the greenhouse. We developed a simple and quickly protocol to regenerate plantlets in storage root explants of purple sweet potato. This regeneration system will facilitate tissue culture and gene transfer research of purple sweet potato.

  • Tissue culture systems to optimize regeneration plant species of Ocimum spp were evaluated as a method to micropropagate individual plants and to better study their biology in vitro. Ocimum species were also evaluated for the production of natural plant products during and following the regeneration process. The primary goal of this project was to enhance the regeneration efficiency of basil. Several factors were examined using different Ocimum species and commercial varieties. The effect of cytokinin combination, activated charcoal, gelling agents, and different carbon sources were investigated. Anthocyanin callus spots were produced only in four varieties among six tested. 'Sweet Dani' showed the best results on anthocyanin accumulation, while 'African beauty', 'Tree basil' and 'Methylcinnamate' produced only a few spots. Shoot regeneration was only achieved from 'Sweet Dani' explants. As the activated charcoal concentration increased, callus formation rate decreased respectively compare to the controls for all varieties. There was a decrease in callus growth with increasing concentration of agar and phytagel.

  • Environmental matters are one of key issues of today's world. At this point, advanced countries are proposing and planning various kinds of policies for sustainable communities. Space designers in those countries are more concerned with regeneration or slow development than development and innovation. As the new government of Korea was started in 2008, it announced 'design Korea project' as key agenda for national competitiveness. But it seems that they are focusing on development of new town than the concept of preservation or regeneration on which most advanced countries put their bases. In these regards, this research starts with the idea that environmental factor in space design is very important and it is directly connected with human survival. With this idea, the researching team created experimental space titled 'regeneration' as one of eco-friendly education program. The team aimed at building up or 'designing one man space-one to one ratio' in deserted place with recycled stuffs. With the idea of 'regeneration' in space, the researcher aims at awakening the sense of environment and putting it into practice in daily living.

Biomaterials-assisted spheroid engineering for regenerative therapy

  • Cell-based therapy is a promising approach in the field of regenerative medicine. As cells are formed into spheroids, their survival, functions, and engraftment in the transplanted site are significantly improved compared to single cell transplantation. To improve the therapeutic effect of cell spheroids even further, various biomaterials (e.g., nano- or microparticles, fibers, and hydrogels) have been developed for spheroid engineering. These biomaterials not only can control the overall spheroid formation (e.g., size, shape, aggregation speed, and degree of compaction), but also can regulate cell-to-cell and cell-to-matrix interactions in spheroids. Therefore, cell spheroids in synergy with biomaterials have recently emerged for cell-based regenerative therapy. Biomaterials-assisted spheroid engineering has been extensively studied for regeneration of bone or/and cartilage defects, critical limb ischemia, and myocardial infarction. Furthermore, it has been expanded to pancreas islets and hair follicle transplantation. This paper comprehensively reviews biomaterials-assisted spheroid engineering for regenerative therapy.

  • This study, the meaning of historical and cultural content utilization in urban regeneration was examined and in the process, the causal relationship between each variable was verified through path analysis among physical, cultural, social, and economic factors. The purpose of this study is to find the main variables of historical and cultural content urban regeneration projects in which the moderating effect of economic factors and demographic characteristics, which are mediating effects, have a significant effect on resident participation and life satisfaction of local residents. Furthermore, by presenting a more integrated and effective urban regeneration policy plan, it is intended to ultimately contribute to the vitalization and direction of urban regeneration.

Assessment of dehydrothermally cross-linked collagen membrane for guided bone regeneration around peri-implant dehiscence defects: a randomized single-blinded clinical trial

  • Purpose: The aim of this study was to determine the clinical feasibility of using dehydrothermally cross-linked collagen membrane (DCM) for bone regeneration around peri-implant dehiscence defects, and compare it with non-cross-linked native collagen membrane (NCM). Methods: Dehiscence defects were investigated in twenty-eight patients. Defect width and height were measured by periodontal probe immediately following implant placement (baseline) and 16 weeks afterward. Membrane manipulation and maintenance were clinically assessed by means of the visual analogue scale score at baseline. Changes in horizontal thickness at 1 mm, 2 mm, and 3 mm below the top of the implant platform and the average bone density were assessed by cone-beam computed tomography at 16 weeks. Degradation of membrane was histologically observed in the soft tissue around the implant prior to re-entry surgery. Results: Five defect sites (two sites in the NCM group and three sites in the DCM group) showed soft-tissue dehiscence defects and membrane exposure during the early healing period, but there were no symptoms or signs of severe complications during the experimental postoperative period. Significant clinical and radiological improvements were found in all parameters with both types of collagen membrane. Partially resorbed membrane leaflets were only observed histologically in the DCM group. Conclusions: These findings suggest that, compared with NCM, DCM has a similar clinical expediency and possesses more stable maintenance properties. Therefore, it could be used effectively in guided bone regeneration around dehiscence-type defects.

The effects of newly formed synthetic peptide on bone regeneration in rat calvarial defects

  • Purpose: Significant interest has emerged in the design of cell scaffolds that incorporate peptide sequences that correspond to known signaling domains in extracellular matrix and bone morphogenetic protein. The purpose of this study was to evaluate the bone regenerative effects of the synthetic peptide in a critical-size rat calvarial defect model. Methods: Eight millimeter diameter standardized, circular, transosseus defects created on the cranium of forty rats were implanted with synthetic peptide, collagen, or both synthetic peptide and collagen. No material was was implanted the control group. The healing of each group was evaluated histologically and histomorphometrically after 2- and 8-week healing intervals. Results: Surgical implantation of the synthetic peptide and collagen resulted in enhanced local bone formation at both 2 and 8 weeks compared to the control group. When the experimental groups were compared to each other, they showed a similar pattern of bone formation. The defect closure and new bone area were significantly different in synthetic peptide and collagen group at 8 weeks. Conclusions: Concerning the advantages of biomaterials, synthetic peptide can be an effective biomaterial for damaged periodontal regeneration.

골 유도 재생술 후 감소된 임플란트 주위 부착 각화 점막 증대를 위한 유리 치은 이식술 증례 (Free gingival graft for the increase of peri-implant attached keratinized mucosa decreased after guided bone regeneration)

  • Purpose: During guided bone regeneration procedures for the augmentation of deficient alveolar ridge, primary closure of flap is necessary. For primary flap closure, flap is repositioned coronally and the zone of attached keratinized mucosa may decreased. The need for attached keratinized mucosa around dental implants is still controversial, but sufficient peri-implant attached keratinized mucosa would be beneficial for functional and esthetic aspects. This case report presents three cases that demonstrated free gingival graft for increasing the zone of peri-implant attached keratinized mucosa which was decreased after guided bone regeneration. Materials and Methods: In first case, maxillary incisors were extracted and guided bone regeneration was performed simultaneously. Because the membrane was exposed at 3 weeks after operation, the membrane was removed and free gingival graft was performed for primary flap closure. Free gingival graft was performed again at implant placement for the increase of attached keratinized mucosa. In second case, guided bone regeneration was performed on lower right first molar area, and implant was placed with free gingival graft. In third case, lower right molar area showed insufficient attached keratinized mucosa after implant placement with guided bone regeneration. When abutments were connected, free gingival graft with apically positioned flap was performed. Result: In these three cases, the zone of attached keratinized mucosa around dental implants was decreased after guided bone regeneration. And the increase of peri-implant attached keratinized mucosa could be obtained effectively by free gingival graft. Conclusion: Free gingival graft could be a effective treatment method increasing the zone of attached keratinized mucosa which was decreased after guided bone regeneration procedures.
