1D, 2D interpretation of stream flooding by HEC-RAS and TELEAMC-2D (HEC-RAS, TELEMAC-2D 모형을 이용한 1, 2차원 하천 범람 해석)

  • 급격히 변화하고 있는 산업화와 도시화로 지구 온난화 현상으로 기상이변의 발생빈도가 높아졌고 기후가 불안정하여 예전보다 많은 집중호우가 발생하면서, 홍수로 인한 제내지 침수가 발생되기도 한다. 기후변화로 인한 수재해에 대응하기 위하여 하천 호소 수리 예측 모형의 개선이 필요한 실정이다. 하지만, 자연하천 유역의 강우-유출 상관관계와 지표면 유출현상 및 하도 수리 특성을 자연현상의 복잡성, 강우발생의 시간적 공간적인 발생과정의 임의성, 정확한 해석방법 및 확률 분석에 따르는 불확실성 들을 토대로 단순한 이론과 제한적인 경험공식 등에 의해서 해석, 재현 및 평가를 한다는 것은 매우 어려운 문제이다. 최근 IT 기술의 발전과 더불어, 많은 연구자, 엔지니어들이 기존 수리 수문학적 지식과 IT기술을 융합하여 복잡 다단한 수자원 환경 문제를 해결하고자 한다. 이와 같은 최근 연구 동향에 의거하여, 본 연구에서는 HEC-RAS, TELEMAC-2D 1, 2차원 수리 모형을 연계하여 하천 흐름 분석 및 홍수 범람 해석에 적용하였다. 본 연구에서는 HEC-RAS, TELEMAC 모형을 적용하여 2012년 태풍 '산바(SANBA)'로 인해 홍수 피해를 입은 고령군에 위치한 낙동강 본류 회천 유역(상류 회천교 ~ 하류 도진교)의 하도 내 흐름 분석과 하천 인근 제내지 홍수범람을 예측하였다. 범람해석에 필요한 지형자료를 기초로 하여 각 지형의 조건에 맞게 수치자료를 이용하여 작성하였고, 수자원 정보를 이용하여 유랑, 수위 등 시계열자료를 지류 및 상 하류의 경계조건으로 설정하고, 조도계수 등 하천 기본정보들을 입력하였다. HEC-RAS 모형은 회천교부터 도진교까지 전구간에 대한 종단면과 횡단면별 홍수침수범위 및 홍수위 크기 등 거시적인 1차원 수리해석에 적용하였고, TELEMAC 모형은 HEC-RAS 시뮬레이션 결과를 바탕으로 HEC-RAS에서 나타내기 힘든 2차원 흐름특성, 침수현상 등 일부 범람 구간에 대해 수리해석에 적용하였다. HEC-RAS 시스템은 수공구조물들의 영향과 하천의 영향을 종 횡단면으로 다양한 홍수침수 범위를 1차원으로 나타 낼 수 있으며, TELEMAC 시스템 수리 모의를 통해 얻어진 결과는 유속, 유량, 수심, 하상고 높이 등 2차원으로 나타낼 수 있다. TELEMAC 시스템을 활용한 2차원 분석은 실측자료와 비교적 유사하고 시각, 공간적으로 이해하기 쉽게 표현되므로, 모형 적용성이 우수한 것으로 판단된다. 향후 유역 해석을 위한 수치데이터, 수위, 유량자료를 확보하여 HEC-RAS, TELEMAC 1, 2차원 연계 모형을 적용 한다면, 하천 준설, 하천 구조물 설치, 홍수피해 등 전반적인 하천관리 계획에 활용할 수 있을 것이라 판단된다.

Combination Therapy of the Active KRAS-Targeting Antibody inRas37 and a PI3K Inhibitor in Pancreatic Cancer

  • KRAS activating mutations, which are present in more than 90% of pancreatic cancers, drive tumor dependency on the RAS/mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K)/AKT signaling pathways. Therefore, combined targeting of RAS/MAPK and PI3K/AKT signaling pathways may be required for optimal therapeutic effect in pancreatic cancer. However, the therapeutic efficacy of combined MAPK and PI3K/AKT signaling target inhibitors is unsatisfactory in pancreatic cancer treatment, because it is often accompanied by MAPK pathway reactivation by PI3K/AKT inhibitor. Therefore, we developed an inRas37 antibody, which directly targets the intra-cellularly activated GTP-bound form of oncogenic RAS mutation and investigated its synergistic effect in the presence of the PI3K inhibitor BEZ-235 in pancreatic cancer. In this study, inRas37 remarkably increased the drug response of BEZ-235 to pancreatic cancer cells by inhibiting MAPK reactivation. Moreover, the co-treatment synergistically inhibited cell proliferation, migration, and invasion and exhibited synergistic anticancer activity by inhibiting the MAPK and PI3K pathways. The combined administration of inRas37and BEZ-235 significantly inhibited tumor growth in mouse models. Our results demonstrated that inRas37 synergistically increased the antitumor activity of BEZ-235 by inhibiting MAPK reactivation, suggesting that inRas37 and BEZ-235 co-treatment could be a potential treatment approach for pancreatic cancer patients with KRAS mutations.

Dam Break Analysis with HEC-HMS and HEC-RAS (HEC-HMS와 HEC-RAS를 이용한 댐 붕괴 해석)

  • This study simulates the dam break situation by a probable maximum precipitation of Soyang-River Dam using HEC-HMS model and HEC-RAS model and compares the simulated results. The probable maximum precipitation was calculated using the flood event of the typhoon Rusa occurred in 2002 and using the mean areal precipitation of the Gangreung region and the moisture maximization method. The estimated probable maximum precipitations were compared for the duration of 6, 12, 18, and 24 hrs and were used as input data for the HEC-HMS model. Moreover, the inflow data calculated by HEC-HMS were utilized as ones for HEC-RAS, and then unsteady flow analysis was conducted. The two models were used for the dam break analysis with the same conditions and the peak flow estimated by HEC-HMS was larger than that of the HEC-RAS model. The applicability of two models was performed from the dam break analysis then we found that we could simulate more realistic peak flow by HEC-RAS than HEC-HMS. However, when we need more fast simulation results we could use HEC-HMS. Therefore, we may need the guidelines for the different utilizations with different purposes of two models. Furthermore, since the two models still include uncertainties, it is important to establish more detailed topographical factors and data reflecting actual rivers.

Overexpression of GAP Causes the Delay of NGF-induced Neuronal Differentiation and the Inhibition of Tyrosine Phosphorylation of SNT in PC12 Cells

  • The GTPase activating protein (GAP) can function both as a negative regulator and an effector of $p21^{ras}$. Overexpression of GAP in NIH-3T3 cells has been shown to inhibit transformation by ms or src. To investigate the function of GAP in a differentiative system, we overexpressed this protein in the nerve growth factor (NGF)-responsive PC12 cell line. Two-fold overexpression of GAP caused a delay of several days in the onset of NGF- but not FGF-induced neuronal differentiation of PC12 cells. However, the NGF-induced activation or tyrosine phosphorylation of upstream (Trk, PLC-${\gamma}1$, SHC) and downstream (B-Raf and $p44^{mapk/erk1}$) components of $p21^{ras}$, signalling cascade was not altered by GAP overexpression. Therefore, the change of phenotype induced by GAP was probably not due to GAP functioning as a negative regulator of $p21^{ras}$. Rather, we found that NGF-induced tyrosine phosphorylation of SNT, a specific target of neurotrophin-induced tyrosine kinase activity, was inhibited by GAP overexpression. SNT is thought to function upstream or independent of $p21^{ras}$. Thus in PC12 cells, overexpressed GAP may control the rate of neuronal differentiation through a pathway involving SNT rather than the $p21^{ras}$ signalling pathway.

Screening of Inhibitory Activity of Plant Extracts against Farnesyl Protein Transferase (식물추출물의 파네실 전달효소 저해활성 검색)

  • Ras proteins play an important role in intracellular signal transduction pathways involved in cell growth and the mutated twas genes have been found in thirty percent of human cancers. Ras proteins (H-, K- and N-Ras) are small guanine nucleotide binding proteins that undergo a series of posttranslational modifications including the farnesylation onto cysteine 186 at C-terminal of Ras by farnesyl protein transferase (FPTase). This is a mandatory process for retention of transforming ability. Therefore, inhibitors of FPTase have a promising to be effective antitumor agents. In our screening program for FPTase inhibitors, the methanol extracts of 193 plants were screened for the inhibitory activity against FPTase partially purified from the rat brain. Extracts of 7species plants including Areca catechu, Saururus chinensis, Curcuma longa, Artemisa princeps, Paeonia suffruticosa, Spatholobus suberectus, Cinnamomum cassia, Cinnamomum japonicum inhibited more than 60% of FPTase activity at a concentration of $100\;{\mu}g/ml$.

Design of Closed Seawater Recirculating Aquaculture System for Korean Rockfish Sebastes schlegeli Culture

  • Recirculating aquaculture system (RAS) consists of different treatment compartments that maintain water quality within the ranges commonly recommended for fish cultures. However, common RASs still exert considerable environmental impact since concentrations of organic matter and nutrients in their effluents are high. Compared with the traditional RAS, the model RAS developed here use a sedimentation basin for digestion purposes and then use the released volatile organic matter to stimulate a denitrification process. Different treatment compartments for solids, total ammonia nitrogen, and nitrate removal have been reviewed. This paper provides the basic information on designing different treatment compartments as well as the engineering criteria in closed seawater RAS, consisting of circular tanks for fish cultures; dual drain systems, sedimentation basins and foam fractionators for removal of solids; nitrification biofilters for TAN removal; denitrification biofilters for nitrate removal; and aerators for aeration. The main purpose is to outline a common procedure in designing of closed RAS for marine fish culture with an emphasis on easy management and low expense, as well as reduction of the environmental impact.

A Study to comparison of 1-D Model on dam break flood analysis (댐 붕괴 홍수류해석을 위한 1차원 모형의 비교 연구)

  • '96년, '98년 임진강 유역과 경기북부 지역 집중호우를 시작으로 '02년 태풍 "루사", '03년 "매미", '04년 "메기" 등의 영향으로 대규모 수공 구조물의 설계빈도를 초과하는 폭우를 동반하여 전국적으로 사상 유례없는 막대한 홍수피해를 발생시켰다. 이러한 과정에서 "연천댐", "장현, 동막저수지" 붕괴 등 댐 붕괴로 인하여 댐 하류지역의 홍수피해가 크게 발생하여 이에 대한 대처계획 수립 필요성 및 이를 위한 댐 붕괴로 인한 홍수류 해석에 대한 연구가 절실히 필요하다. 본 연구에서는 비상대처계획(Emergency Action Plan, EAP)수립 대상 국가시설물 중 하나인 양양양수발전소 하부댐에 대한 댐 붕괴 해석을 실시하였다. DAMBRK를 통하여 댐 지점의 붕괴유출수문곡선을 산정하였으며, 댐 하류하천 수위분석은 1차원 DAMBRK와 HEC-RAS모형을 이용비교 분석하였고 이를 통하여 댐붕괴로 인한 하류하천 홍수류 해석시 HEC-RAS모형의 적용성을 검토하였다. 검토결과 HEC-RAS모형 계산치가 DAMBRK 모형에 의한 결과 보다 과대하게 산정 되었다. 향후 댐 붕괴로 인한 홍수류 해석을 위한 HEC-RAS모형 모의시 주변지형의 홍수터화 등의 기법을 첨가하여 홍수위 분석이 필요한 것으로 사료된다.

  • Mutated forms of the ras oncogenes are associated with about 30% of human tumors. The ras genes encode 21KDa proteins, called p21 or Ras, that are associated with the plasma membrane. FPTase is a dimeric enzyme that catalyses the transfer of the farnesyl group from farnesyl pyrophosphate onto cysteine 186 at the C-terminus of the Ras protein. This is mandatory process for triggering ras oncogene toward tumor formation. Therefore, selective inhibitors of FPTase have the potential to be used as antitumorgenic agents.

Analysis of Flood Reduction Effect of Washland using Hydraulic Experiment (수리실험을 이용한 강변저류지의 홍수조절효과 분석)

  • Recently, we have a growing interest in the washland construction for the function of flood control. Therefore this study analyzed flood reduction effect of washland using hydraulic experiment. We used open-channel unit for flood reduction effect of washland. And we used side weir to lateral structure. The result of hydraulic experiment showed a decrease of the average velocity and discharge according to the installation location of side weir. Also, we compared the results of HEC-RAS model and hydraulic experiment for the review of the result of hydraulic experiment. As the result, hydraulic experiment and HEC-RAS model have showed the similar results.

  • For the effective operation of irrigation reservoirs, a general and systematic policy is suggested to make balance of the conflicting purposes between water conservation and flood control. In this study, the flood effective analysis system was developed through the integration of long-term water budget analysis model, GIS-based HEC-HMS model and HEC-RAS model. The system structure consists of long-term water budget model using modified TANK theory, flood runoff and flood effects analysis model using HEC-GeoHMS, HEC-HMS and HEC-RAS models. The flood effects analysis system simulated the flood runoff from the upstream, downstream flood and long-term runoff of the watershed using the observed data collected from 1998 to 2002 of Seongju dam. The simulated results were reasonably good compared with the observed data. The optimal management method of the reservoir during the whole season is suggested in this study, and the flood analysis system can be a useful tool to evaluate a reservoir operation quantitatively for the mitigation of flood damages of reservoir basin.
