Yeungnam University type drive-through (YU-Thru) coronavirus disease 2019 (COVID-19) screening system: a rapid and safe screening system

  • Active and prompt scale-up screening tests are essential to efficiently control the coronavirus disease 2019 (COVID-19) outbreak. The goal of this work was to identify shortcomings in the conventional screening system (CSS) implemented in the beginning of the outbreak. To overcome these shortcomings, we then introduced a novel, independently developed system called the Yeungnam University type drive-through (YU-Thru), and distributed it nationwide in Korea. This system is similar to the drive-throughs utilized by fast food restaurants. YU-Thru system has shortened the time taken to test a single person to 2-4 minutes, by completely eliminating the time required to clean and ventilate the specimen collection room. This time requirement was a major drawback of the CSS. YU-Thru system also reduced the risk of subjects and medical staff infecting one another by using a separate and closed examination system. On average, 50 to 60 tests were conducted per day when using the CSS, while now up to 350 tests per day are conducted with the YU-Thru system. We believe that the YU-Thru system has made an important contribution to the rapid detection of COVID-19 in Daegu, South Korea. Here, we will describe the YU-Thru system in detail so that other countries experiencing COVID-19 outbreaks can take advantage of this system.

  • Recent technological innovations in the drug discovery process such as combinatorial synthesis and high throughput screening have led to the identification of an increasingly large number of compounds at the hits-to-leads stage. Therefore, rapid and precise pharmacokinetic/metabolic screening is essential to enhance the tractability of selected leads and to minimize the risk of failure in the later stages of drug development.   • To determination of antioxidant substance homoorientin, from Phyllostachys bambusoides leaves, the ultrasonic extraction and HPLC on-line $ABTS^+$ screening method were empolyed. Also, the various experimental variables such as the frequency and time of ultrasonic system were investigated and homoorientin was extracted efficiently at the low frequency 35 kHz and the extraction time 60 min. The values were positive peak 1574.71 (relative area, 23.67%) and negative peak 6924.34 (relative area, 1.23%), respectively. This HPLC on-line $ABTS^+$ screening method was rapid and efficient to search for antioxidants from natural products. These results will provide a database for investigating the constituents of natural products and the resources of pharmaceutical and cosmetic products.

Rapid Screening of Naturally Occurring Radioactive Nuclides (238U, 232Th) in Raw Materials and By-Products Samples Using XRF

  • Background: As new legislation has come into force implementing radiation safety management for the use of naturally occurring radioactive materials (NORM), it is necessary to establish a rapid and accurate measurement technique. Measurement of $^{238}U$ and $^{232}Th$ using conventional methods encounter the most significant difficulties for pretreatment (e.g., purification, speciation, and dilution/enrichment) or require time-consuming processes. Therefore, in this study, the applicability of ED-XRF as a non-destructive and rapid screening method was validated for raw materials and by-product samples. Materials and Methods: A series of experiments was conducted to test the applicability for rapid screening of XRF measurement to determine activity of $^{238}U$ and $^{232}Th$ based on certified reference materials (e.g., soil, rock, phosphorus rock, bauxite, zircon, and coal ash) and NORM samples commercially used in Korea. Statistical methods were used to compare the analytical results of ED-XRF to those of certified values of certified reference materials (CRM) and inductively coupled plasma mass spectrometry (ICP-MS). Results and Discussion: Results of the XRF measurement for $^{238}U$ and $^{232}Th$ showed under 20% relative error and standard deviation. The results of the U-test were statistically significant except for the case of U in coal fly ash samples. In addition, analytical results of $^{238}U$ and $^{232}Th$ in the raw material and by-product samples using XRF and the analytical results of those using ICP-MS ($R^2{\geq}0.95$) were consistent with each other. Thus, the analytical results rapidly derived using ED-XRF were fairly reliable. Conclusion: Based on the validation results, it can be concluded that the ED-XRF analysis may be applied to rapid screening of radioactivities ($^{238}U$ and $^{232}Th$) in NORM samples.

  • Trigonopsis variabilis ATCC10679 (TW)의 변이균주를 선별하기 위하여 쉽고 간편한 균주 선별방법을 개발하였다. 변이 균주(T26)의 D-Amino acid oxidase (D-AAO)는 야생균주(TW)의 D-AAO 효소보다 cephalosporin C에 대한 기질특이성이 약 30% 높았다. 이들 두 균주에서 D-AAO 유전자를 클로닝하여 각 유전자를 비교해본 결과 811번 우치의 T가 C로 변경되었으며 이에 따른 아미노산은 Phe-258이 Ser-258로 바뀌었다.

국내 건물의 폭발물 테러 위험도 요인 분석 (Analysis on the Risk of Explosive Terror in Domestic Buildings)

  • 2002년부터 2010년까지 세계적으로 발생한 테러 현황을 조사 및 분석한 결과 전체 1만 9천 946건의 테러유형 중 51.8%인 1만 333건이 폭파에 의한 테러로 나타났으며, 사용무기에 있어서도 폭파와 관련된 폭발물이 약 52.2%인 1만 431건으로 높게 나타나고 있다. 이에 본 논문에서는 FEMA-rapid visual screening을 통해 국내의 건물을 높이별로 테러위험도를 분석하였다. 그 결과 건물의 높이가 높아질수록 테러에 대한 위험도가 높게 나타나고 있어 전체위험도는 건물의 높이와 비례하여 증가하는 것을 확인하였다. 높이 100m 이상 건물의 경우 Threat 항목이 전체위험도에 가장 크게 영향을 미치며, 폭발과 관련된 시나리오에 따른 위험도 분석결과 내부 폭발 항목에서 높게 나타나고 있다.

Effect of rapid screening parameters on seismic performance of RC buildings

  • This study investigates the effects of soft story, short columns, heavy overhangs, pounding, and construction and workmanship quality parameters on seismic response of reinforced concrete buildings through nonlinear static and dynamic procedures. The accounted parameters are selected for their common use in rapid screening of RC buildings. The 4- and 7-story buildings designed according to pre-modern codes are used to reflect majority of the existing building stock. The relative penalty scores are employed in this study to evaluate relative importance of certain irregularities in the existing rapid seismic assessment procedures. Comparison of relative scores for the irregularities considered in this study show that the overall trend is similar. The relatively small differences may be accounted for regional construction practices. It is concluded that initial-phase seismic assessment procedures based on architectural features yield in somewhat similar results independent of their bases. However, the differences in the scores emphasize the proper selection of the method based on the regional structure characteristics.

A Rapid and Effective Colony PCR Procedure for Screening Transformants in Several Common Mushrooms

  • In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant's tissue ($1-10{\mu}g$) to $20{\mu}l$ 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at $34^{\circ}C$ for 15 min. Finally, $2{\mu}l$ of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.

효모 탐색을 위한 Pyrolysis Mass Spectrometry의 활용 (Application of Pyrolysis Mass Spectrometry on Yeast Screening)

  • To develop the effective microbial screening method, pyrolysis mass spectrometry (PyMS) fingerprinting was evaluated as a tool that discriminate various yeast strains. The target yeast strains were isolated from industrial wastewater. Seventeen environmental isolated yeast strains were examined by pyrolysis mass spectrometry and sequencing analysis of the large subunit rRNA gene D1/D2 region. The PyMS results were compared with those of sequencing analysis. Taxonomic correlations were observed between the PyMS data and the sequencing results. It was concluded that PyMS provides a rapid, reliable and cost-reducing method for discrimination of the yeast strains.
