Rapid Screening of Apple mosaic virus in Cultivated Apples by RT-PCR

  • The coat protein (CP) gene of Apple mosaic virus (ApMV), a member of the genus Ilarvirus, was selected for the design of virus-specific primers for amplification and molecular detection of the virus in cultivated apple. A combined assay of reverse transcription and polymerase chain reaction (RT-PCR) was performed with a single pair of ApMV-specific primers and crude nucleic acid extracts from virus-infected apple for rapid detection of the virus. The PCR product was verified by restriction mapping analysis and by sequence determination. The lowest concentration of template viral RNA required for detection was 100 fg. This indicates that the RT-PCR for detection of the virus is a 10$^3$times more sensitive, reproducible and time-saving method than the enzyme-linked immunosorbent assay. The specificity of the primers was verified using other unrelated viral RNAs. No PCR product was observed when Cucumber mosaic virus (Cucumovirus) or a crude extract of healthy apple was used as a template in RT-PCR with the same primers. The PCR product (669 bp) of the CP gene of the virus was cloned into the plasmid vector and result-ant recombinant (pAPCP1) was selected for molecule of apple transformation to breed virus-resistant transgenic apple plants as the next step. This method can be useful for early stage screening of in vitro plantlet and genetic resources of resistant cultivar of apple plants.

  • As a life cycle of product bas shortend, Rapid prototypiug has come into the spotlight to shortened the time of producint a prototype. But It followed in laminated directivity and of the prototype production which is decided the interference which Beccuase of invisible rough surface, some inferences have occured during an assembly hour. Consequently, This study on rapid prototyping has parameter to determine surface roughness and surface form precision of total shape. In order to improve Surface form precision, it can control parameters which are Laminate Height, Layer Hatch Overcue, Fill Cure Depth. In addition, we found the parameter which have a deep effect on surface roughness among various parameters, and then, tried to reduce the interference that can be occurred by surface roughness during assembly.

  • Reverse engineering enables a fast creation of CAD model from an existing part without any technical drawing or digital data. This method of data acquisition is necessary in several industrial applications when the modification of an existing product design has to be performed and verified afterwards. In this paper, the surface of an existing ship propeller was measured by a tactile measuring system and the acquired point data of the surface were used for modeling of a solid model in CAD system. By using LOM and casting processes, an aluminum propeller for functional test was manufactured and then its accuracy was measured by a laser scanner and compared with the origin릴 CAD data. As the results show, the use of LOM as a rapid prototyping process and casting as a secondary process delivers a functional prototype propeller which can be used for test purposes.

  • Recently, the lead-time of a product is to be shortened in order to satisfy consumer's demand. It is thus important to reduce the manufacturing time and the cost of 3D-shaped microstructures. Micro-Electro-Mechanical Systems (MEMS) and devices are usually fabricated by lithography-based methods. Above method is not flexible for the rapid manufacture of 3D-shaped microstructures because it depends on work's experiences and requires excessive cost and time for making many masks. In this paper, the effective laser micrornachining is developed to fabricate UV sensitive polymer microstructures using laser ablation. The proposed process, named by laser microRP, is a very useful method on rapid manufacturing for 3D-shaped microstructures.

  • With the rapid growth of the B2B electronic commerce, digital catalogs for expressing product information effectively and exchanging it easily between companies become increasingly important. In this paper, we refer a product expression model to effectively integrate product information, and implement the model using XDK(XML Development Kit), which is given by Oracle 8i. We will first derive an XML DTD reflecting the model. We will then develop a system that stores and retrieves product information on/from Oracle 8i databases, which is written in XML, complying the XML DTD structure. Finally, We will give an experiment on the system.

  • Better understanding and sharing information are getting important to manage interdisciplinary product development team in a globally-distributed company. This study proposes a solution to implement RPD(Rapid Product Development) system for the distributed development teams using SOAP(Simple Object Access Protocol). And a new approach is introduced for the better understanding of product geometry among the development members in different place and the easy sharing of product information. An application example shows that SOAP operates in distributed environment more efficiently than other RPC(Remote Procedure Call) techniques and it does not respond sensitively to firewall. And SOAP is an excellent RPC and messaging technique to exchange structured data. Procedures developed with use of SOAP are worked together with web, and users can use remote services as an application program in their local computer.

  • To achieve rapid developments and cost savings in manufacturing industries including automotive die shops, new paradigm and its supporting systems of information managements through total product life cycle are needed for concurrent and collaborative engineering. For manufacturing of automotive press dies, integrated and efficient managements of PPR information including product, manufacturing process and resource are essential. In this paper, we introduce a PLM approach to achieve engineering collaborations in product development and production of automotive dies. To prove concepts and benefits of PPR information managements, we implement new business workflow and detail procedures, PPR information management system and other related applications. By PPR information managements in PLM, improvements in quality of engineering results and savings in time from design to production of dies are possible.

  • A new directed product graph(DPG) is proposed from the product graph for electrical networks. By introducing the direction of an dege and the concept of a loop to product graph, it is more easy and rapid to obtain topologically the denominator of Mason's formula without relation of the sign rule and without arising terms cancelled. Also the constraints of tree selection at a given network-graph can be removed.

The Quality of the Information Sharing between the Korean Apparel Manufatures and the Contractors (국내 의류 제조업체와 생산 협력업체간 정보 공유 특성)

  • As apparel manufacturers have increased their outsourcing garment making to cope with the rapid changing market, the information sharing has been the most important factor for the Korean apparel industry. The purpose of this research was to study the present state and the needs of the information sharing between the apparel manufacturers and the contractors; and the relationship between the product property (basic/fashion product) and the information sharing property. A total of 86 Korean apparel manufacturers were surveyed for the study. The results are: 1) the information sharing level on manufacturing capacity, manufacturing processing condition, and product quality were a bit higher than other surveyed information, but the level on inventories and sales were lower than others. in addition, the information sharing needs by online system of manufacturing capacity, manufacturing processing condition, and product quality were greater than other information; 2) In the case of fashion product, the information sharing needs of product quality by online system were much greater than in the case of basic product. This study is expected to help apparel manufacturers to construct their information sharing system which is apt for their product property and needs.
