Optimization of bio-$H_{2}$ production from acid pretreated microalgal biomass (미세조류로부터 바이오 수소 생산을 위한 산(acid) 전처리의 최적화)

  • In this study, dark fermentative hydrogen production (DFHP) from acid pretreated microalgal biomass was optimized with via statistical experimental design. Acid concentration and reaction time were varied from 0.1 to 3% (v/w) and 10 to 60 min with substrate concentration of 76 g dry cell weight (dcw)/L and initial pH of 7.4, respectively. During the fermentation, pH was not controlled. The optimal condition was found that at $H_{2}$ yield reached to 37.3 mL $H_{2}/g$ dcw at 1.2% HCl and 48 min. Through regression analysis, it was found that $H_{2}$ yield was well fitted by a quadratic polynomial equation ($R^{2}$=0.95). HCl concentration was the most significant factor influencing DFHP. The results of ANOVA verify that HCl concentration was the most significant factor influencing DFHP.

Optimization of the Molecular Press Dehydration Method for Ginger Using Response Surface Methodology (반응표면분석에 의한 생강의 분자압축탈수 방법 최적화)

  • The purpose of this study was to determine the optimal mixing ratios for three different concentrations of maltodextrin, dehydration time, and cutting thickness to prepare gingers using molecular press dehydration (MPD) through response surface methodology (RSM) based on the dehydration rate, rehydration rate, and a sensory evaluation. As maltodextrin concentration increased, dehydration rate, rehydration rate, external, flavor, texture, overall acceptability of the gingers tended to be high, and the color tended to be low. As the dehydration time and cutting thickness increased, dehydration rate, rehydration rate and the sensory evaluation of the gingers tended to be high. The maltodextrin concentrations were 70-82%, dehydration time was 5.2-9.2 hr, and cutting thickness levels were 1.0-1.2 mm. The optimal mixing ratios, maltodextrin concentration, dehydration time, and cutting thickness for manufacturing the best quality of gingers using molecular press dehydration were 76%, 7.2 hr, and 1.1 mm, respectively.

Optimization of Extraction Conditions of Polyphenolic Compounds from Apple Pomace by Response Surface Methodology (반응표면분석에 의한 사과 pomace로부터의 폴리페놀 추출조건 최적화)

  • This study examined the optimization of alcohol extraction conditions for maximizing the total polyphenols derived from apple pomace, by response surface methodology (RSM). The effects of four independent variables, including $X_1$ (ratio of solvent to sample content), $X_2$ (dipping time), $X_3$ (extraction time), and $X_4$(extraction temperature), were investigated at five levels using central composite design (CCD). $Y_1$ (yield) and $Y_2$ (total polyphenols) were chosen as dependent variables. The coefficients of determination, $R^2$, were greater than 0.900 (0.9042 and 0.9555). The results showed that the model fit was very significant (p<0.001). The optimum extraction conditions were as follows: 13.00 mL/g for the ratio of solvent to sample content, 89.02 min for dipping time, 180 min for extraction time, and $70^{\circ}C$ for extraction temperature. At these conditions, the predicted total polyphenol content was 29.68 mg catechin equiv./g.

  • This study was to investigate the physicochemical and sensory characteristics of snack with various amounts Cham chwi and oil. This result was used to determine the optimum condition of adding levels of Cham chwi and oil. Flavor, crispness, greasiness, oil absorption, expansion rate were selected as the physicochemical and sensory characteristics to determine the optimum conditions of the amount of Cham chwi and oil. The predicted values were obtained by the regression method of RSM(response surface methodology). Conditions were standardized with maximum range of expansion rate when expected value of crispness was more than five and expected value of greasiness was less five. The optimum conditions of Cham chwi snack was established as the 8.6% of Cham chwi and 7.6% of oil.

The Quality Characteristics of Beef Sausage with the Addition of Gynura procumbens (명월초 가루를 첨가한 우육 소시지의 최적화 연구)

  • The aims of this study were to determine the optimal mixing condition for two different amounts of Gynura procumbens powder and olive oil for the preparation of beef sausage. The experiments were designed according to the central composite design of response surface methodology, which showed 10 experimental points including two replicates of Gynura procumbens powder and olive oil. The physicochemical and mechanical characteristics as well as the sensory properties were measured, and these values were applied to the mathematical optimization models. The results of the physiochemical and mechanical analysis of each sample, including chewiness, cohesiveness, color a, color b, moisture content, salinity and heating loss showed significant differences. The sensory measurements were significantly different in color, flavor, tenderness, texture and overall quality. The optimum formulation, which was calculated using the numerical and graphical method, was determined to be 2.1 g Gynura procumbens powder and 7.06 g of olive oil.

Optimization of Ethanol Extraction Conditions for Functional Components from Lespedeza cuneata Using Response Surface Methodology (반응표면분석법을 이용한 야관문(Lespedeza cuneata) 기능성분의 에탄올 추출조건 최적화)

  • This study was conducted to monitor the quality characteristics of Lespedeza cuneata ethanolic extracts, by a response surface methodology. The independent variables were the extraction temperature ($35{\sim}95^{\circ}C}$), extraction time (2~10 hr), and ethanol concentration (10~90%). The coefficients of the determinations (R2) were 0.8562 (p<0.1), 0.9787 (p<0.01), and 0.8344 (p<0.1) in total polyphenol, total flavonoid, and electron donating ability, respectively. The electron donating ability and nitrite scavenging effect were improved with an increase of ethanol concentration, rather than the extraction temperature. ORAC (Oxygen Radical Absorbance Capacity) with extraction conditions was 1,636.47~2,696.99 ${\mu}moles$ TE/g, and was increased with the increase of extraction temperature and 40~60% of ethanol concentration. Estimated conditions for the maximized extraction including the yield, total polyphenol, electron donating ability, nitrite scavenging effect, and ORAC, were $60{\sim}68^{\circ}C$ in extraction temperature, 6~7 hr in extraction time, and 38~60% in ethanol concentration.

Optimization of Germinated Brown Rice Cookie with Added Spinach Powder (시금치 가루를 첨가한 발아현미쿠키의 최적화)

  • Germinated brown rice is a useful material that contains natural nutrients and biologically active substances. This study was conducted to develop a optimal composite recipe for functional germinated brown rice cookie with added spinach (spinacia oleracea L) powder and with high preference in all age groups. The experimental design utilized herein was based on the Central composite design methodology of response surface, which included 16 experimental points, including 2 replicates for spinach, sugar, and butter. The physical, mechanical, and sensory properties of the test were measured, and these values were applied to the mathematical models. A canonical form and perturbation plot showed the influence of each ingredient on the mixed final product. The results of spread ratio decreased significantly with increases in spinach powder, sugar, and butter(p<0.01). L value shows that lightness decreased significantly with an increase in sugar(p<0.05), but the a and b values were not significant. Sensory evaluation found significant values for color(p<0.05), flavor(p<0.05), texture(p<0.05) and overall quality(p<0.05) in the predicted model. The optimum formulation by the numerical and graphical methods was calculated as follows: spinach powder 3 g, sugar 35.5 g butter 54.4 g.

Development of climate change uncertainty assessment method for projecting the water resources (기후변화에 따른 수자원 전망의 불확실성 평가기법 개발)

  • It is expected that water resources will be changed spatially and temporally due to the global climate change. The quantitative assessment of change in water availability and appropriate water resources management measures are needed for corresponding adaptation. However, there are large uncertainties in climate change impact assessment on water resources. For this reason, development of technology to evaluate the uncertainties quantitatively is required. The objectives of this study are to develop the climate change uncertainty assessment method and to apply it. The 5 RCMs (HadGEM3-RA, RegCM4, MM5, WRF, and RSM), 5 statistical post-processing methods (SPP) and 2 hydrological models (HYM) were applied for evaluation. The results of the uncertainty analysis showed that the RCM was the largest sources of uncertainty in Spring, Summer, Autumn (29.3~68.9%), the hydrological model was the largest source of uncertainty in Winter (46.5%). This method can be possible to analyze the changes in the total uncertainty according to the specific RCM, SPP, HYM model. And then it is expected to provide the method to reduce the total uncertainty.

Processing Optimization and Antioxidant Activity of Chocolate Added with Mulberry (오디 초콜릿의 제조 최적화 및 항산화 활성)

  • The purpose of this study was to determine the optimal mixing conditions for two different amounts of added mulberry powder and fresh cream to prepare functional chocolate with added mulberry powder. The experiment was designed according to the central composite response surface design, which showed 10 experimental points, and included two replicates for mulberry powder and fresh cream. The physiochemical, mechanical, and sensory properties of the test were measured, and these values were applied to the mathematical models. The results of the physiochemical and mechanical analyses of each sample, including pH, moisture content, total phenolic content, DPPH free radical scavenging activity, color L, color b, hardness, gumminess, and cohesiveness showed significant differences. The sensory characteristics of the samples tested were significantly different in flavor, texture, sourness, bitterness, and overall acceptability. The optimum formulation calculated by numerical and graphical methods was 25.76 g mulberry powder and 72.21 g fresh cream.

Optimization of Calcium Acetate Preparation from Littleneck Clam (Ruditapes philippinarum) Shell Powder and Its Properties (바지락(Ruditapes philippinarum) 패각분말로부터 초산칼슘 제조 및 특성)

  • The optimal condition for preparation of powdered calcium acetate (LCCA) which has high solubility, from calcined powder (LCCP) of the littleneck clam shell by response surface methodology (RSM) was examined. Increased molar ratio of LCCP led to reduced solubility, yield, color values, and overall quality. The critical values of multiple response optimization of independent variables were 2.57 M of acetic acid and 1.57 M of LCCP. The actual values (pH 7.0, 96.1% for solubility, and 220.9% for yield) under the optimized condition were similar to the predicted values. LCCA showed strong buffering capacity between pH 4.89 and 4.92 on addition of ~2 mL of 1 N HCl. The calcium content and solubility of LCCA were 21.9-23.0 g/100 g and 96.1-100.1%, respectively. The FT-IR and XRD patterns of LCCA were identified as calcium acetate monohydrate, and FESEM images revealed an irregular and rod-like microstructure.
