Nrf2-mediated activation of HO-1 is required in the blocking effect of compound K, a ginseng saponin metabolite, against oxidative stress damage in ARPE-19 human retinal pigment epithelial cells

  • Background: The beneficial effects of compound K (CK) on different chronic diseases have been shown to be at least related to antioxidant action. Nevertheless, since its antioxidant activity in human retinal pigment epithelial (RPE) cells is still unknown, here we investigated whether CK alleviates oxidative stress-stimulated damage in RPE ARPE-19 cells. Methods: The cytoprotective consequence of CK in hydrogen peroxide (H2O2)-treated cells was evaluated by cell viability, DNA damage, and apoptosis assays. Fluorescence analysis and immunoblotting were performed to investigate the inhibitory action of CK on reactive oxygen species (ROS) production and mitochondrial dysfunction. Results: H2O2-promoted cytotoxicity, oxidative stress, DNA damage, mitochondrial impairment, and apoptosis were significantly attenuated by CK in ARPE-19 cells. Furthermore, nuclear factor erythroid 2-related factor 2 (Nrf2) phosphorylation level and its shuttling to the nucleus were increased, which was correlated with upregulated activation of heme oxygenase-1 (HO-1). However, zinc protoporphyrin, a blocker of HO-1, significantly abrogated the preventive action of CK in H2O2-treated ARPE-19 cells. Conclusion: This study indicates that activation of Nrf2/HO-1 signaling by CK plays an important role in rescuing ARPE-19 cells from oxidative cellular damage.

THE BONE CHANGES OF THE MIDPALATAL SUTURE AFTER RAPID PALATAL EXPANSION IN YOUNG ADULT DOGS (유성견에서 상악골 급속확장 후 정중구개봉합부에서의 골조직 변화)

  • Rapid palatal expansion(RPE) is a method of inducing the new bone formation in the palate by separation of the midpalatal suture, which can be done conveniently by placing heavy force across the maxillary dental arch. This experiment was undertaken to examine the histologic changes after RPE and during retention period. Four young adult dogs(a control dog, three experimental dogs) aged 4 to 6 months old were used for this experiment. Expansion screw($Hyrax^{\circledR}$, Dentarum Inc.) was delevered to the palate and fumed 180 degrees every morning and evening for 8 days, giving a total expansion of 7.2mm. A control dog was sacrified at the starting point of this study without any treatment and three experimental dogs were sacrified after RPE, 14-day retention, and 28-day retention in each. Thereafter, those samples were observed with hematoxylin-eosin(H-E) stain, ground section(Villanueva stain), alkaline phosphatase(ALP) stain, tartrate-resistant acid phosphatase(TRA) stain. The results were as followings: 1. After RPE, collagen fiber bundles were stretched along the midpalatal suture and few osteoblasts were flattened-inactive state and also, a little osteoid tissues was observed. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. 2. After 14-day retention, collagen fiber bundles were stretched along the midpalatal suture and few osteoblasts which had ALP-positive activity in their cytoplasm were seen. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. 3. After 28-day retention, collagen fiber bundles were arranged like those of control dog and osteoblasts which showed a lot of immature bone formation were cuboidal shape and exhibited ALP-positive activity in their cytoplasm. Few multinucleated osteoclasts which had TRAP-positive activity in their cytoplasm were seen in horizontal section, whereas a few osteoclasts were seen in frontal section, especially in the nasal floor side of palatal bone. According to the above results, the new bone formation after rapid palatal expansion was examined after 14-day retention and significantly increased after 28-day retention.

The Effects of the Intensity of Combined Training on Body Composition, HOMA-IR and HbA1c of Female Students of a Boarding High School (복합운동 강도가 기숙형학교 여고생의 신체조성, HOMA-IR 및 HbA1c에 미치는 영향)

  • Among students of 'K' boarding high school, located in 'B' city, 32 students whose % body fat was 30% or above were divided into three groups - two exercise groups and one control group. They performed Combined Training - a mix of weight training (WT) and step box training (SBT) - for 65 min a day, 3 days a week, for 8 weeks in total. Group A performed WT 70-80%$RM{\times}3$ sets+SBT (RPE 11-13)${\times}1$ set, and group B performed WT 70-80%$RM{\times}1$ set+SBT (RPE 11-13)${\times}3$ sets to yield data on changes of body composition (Soft Lean Mass, SLM), %fat, WHR), HbA1c, and HOMA-IR. Paired t-test was used to process data within each group. Pre- and post experiment differences rates (%diff) were used to perform one-way ANOVA (Duncan test) for group comparisons. The conclusions derived are as follows. Regarding body composition, exercise groups showed an increase in SLM, but there was no such change in the control group. WHR decreased in group A, but increased in the control group. The % body fat decreased in both exercise groups, but increased in the control group. As for the group comparisons, SLM in group A showed a greater increase than in group B and the control group. WHR in groups A and B showed a greater decrease than the control group. The % body fat in groups A and B showed a greater decrease than the control group. The exercise groups showed a significant decrease in HOMA-IR, but the control group showed a significant increase in HOMA-IR. As for the group comparisons, groups A and B showed a greater decrease in HOMA-IR than the control group. The exercise groups showed a significant decrease in HbA1c, however, the control group showed no change in HbA1c. As for the group comparisons, group A showed a greater decrease in HbA1c than the control group. These results confirm that combined training is more effective in improving body composition and metabolic factors when it includes a high proportion of resistance training, rather than aerobic exercise. The results of the study suggest that it is advisable to set a high proportion of WT when deciding the intensity of combined training.

The Formation of Microcrystalline SiGe Film Using a Remote Plasma Enhanced Chemical Vapor Deposition (원격 플라즈마 화학기상 증착법으로 성장된 미세 결정화된 SiGe 박막 형성)

  • SiGe thin films were deposited by remote plasma enhanced chemical vapor deposition (RPE-CVD) at $400^{\circ}C$ using $SiH_4$ or $SiCl_4$ and $GeCl_4$ as the source of Si and Ge, respectively. The growth rate and the degree of crystallinity of the fabricated films were characterized by scanning electron microscopy and Raman analysis, respectively. The optical and electrical properties of SiGe films fabricated using $SiCl_4$ and $SiH_4$ source were comparatively studied. SiGe films deposited using $SiCl_4$ source showed a lower growth rate and higher crystallinity than those deposited using $SiH_4$ source. Ultraviolet and visible spectroscopy measurement showed that the optical band gap of SiGe is in the range of 0.88~1.22 eV.

  • This paper examines the effects on mental fatigue of changes in the intensity of physical activity. A treadmill-equipped instrument and perception tester were used to attain several levels of physical activity In this paper, In order to determine the individual levels of physical activity of subjects, Borg-RPE scale, Heart Rate(HR) and Respiratory Quotient(RQ) were used. Also, an addition test in whole-body activity on treadmill-equipped instrument as an indicator of mental fatigue were performed. In the above experiments, the scores obtained in addition test administered before and after physical activity at each intensity level used. Restricted within the limits of this paper, the results of these tests showed that mental fatigue decreased after physical activity.

Hippo Signaling Circuit and Divergent Tissue Growth in Mammalian Eye

  • Vertebrate organ development is accompanied by demarcation of tissue compartments, which grow coordinately with their neighbors. Hence, perturbing the coordinative growth of neighboring tissue compartments frequently results in organ malformation. The growth of tissue compartments is regulated by multiple intercellular and intracellular signaling pathways, including the Hippo signaling pathway that limits the growth of various organs. In the optic neuroepithelial continuum, which is partitioned into the retina, retinal pigment epithelium (RPE) and ciliary margin (CM) during eye development, the Hippo signaling activity operates differentially, as it does in many tissues. In this review, we summarize recent studies that have explored the relationship between the Hippo signaling pathway and growth of optic neuroepithelial compartments. We will focus particularly on the roles of a tumor suppressor, neurofibromin 2 (NF2), whose expression is not only dependent on compartment-specific transcription factors, but is also subject to regulation by a Hippo-Yap feedback signaling circuit.

Semiquantitative Analysis of metal for Cigarette Product by Inductively Coupled Plasma Mass Spectrometry (유도결합 플라스마 질량분석기를 이용한 담배 제품의 무기물 반정량 분석)

  • Semiquantitative analysis by ICP-MS has proven to be a powerful tool for fast screening, in addition, it does not require the element of interest to be present in the calibration standard, making it especially useful for the analysis of unknown samples. In this study, seven cigarette samples were analyzed by the rapid semi-quantitative analysis method based on the ICP-MS. For each cigarette sample, cut tobacco, cigarette paper, filter (before and after smoking), and smoke condensate were analyzed. The accuracy of the analysis technique was evaluated by comparing results obtained from Calibration Check Standard(CCS) and calibration method. Relative Percentage Error(RPE) value of all elements measured for three CCS showed a stable result of less than ${\pm}20%$. Compared to full quantitative analysis by calibration method, the results for cigarette samples showed average error within ${\pm}15%$.

A Study of Workload Change at Varying Working Posture, Work/Pest Ration and Task Type in an Automobile Assembly Task (자동차 조립작업 시 작업자세, 작업/휴식비율, 작업형태에 따른 작업부하의 변화에 관한 연구)

  • A laboratory experiment was conducted to investigate the effects of working posture. work/rest ration, and task type on the physical workload in automobile assembly jobs. Ten healthy male university students participated as subjects in the experiment. East subject performed 18 experimental session with combination of 3 working posture 3 work/rest ratio and 2 task types. To examine the effect of various working conditions on workload, DMG analysis was performed on 4 different muscles in the neck an arm area, Also RPE scale and heart rate were taken as response variables. The result showed that all working conditions considered in the experiment had significant effects on the workload concluding that labor intensity in auto assembly jobs should be adjusted with respect to different levels of working posture, work/rest ratio, and task type. Detailed analysis are presented in the paper.

Psychophysical Study of Combined Manual Material Handling Tasks (결합된 수동물질취급 작업에서의 인체심리학적 연구)

  • Most psychophysical studies in manual material handling (MMH) have paid more attention to two-handed MMH activities than to one-handed MMH activities. Also, studies are involved only with single MMH activities - lifting, lowering, carrying, holding, pushing or pulling. Very few studies are reported on the determination of workers' capacities for combinations of one-handed MMH activities (lifting a box, then carrying the box, and lowering the box). The objective of this study was to find the psychophysical and physiological responses of human subjects from a simulated industrial task involving one-handed and two-handed combined tasks. The capacities that were determined as the maximum acceptable weight workloads for 1 hr work period for one-handed and two-handed combined tasks - lifting a box from floor to knuckle height, carrying the box for 4.3 m, and lowering the box from knuckle to floor height - were determined psychophysically under three frequencies: six handlings per minute, one handling per minute and one handling per five minutes. In addition to maximum acceptable weight, heart rate and RPE values on wholebody, back, and arm were also measured as responses.

Effect of tool weight and hand posture on the postural tremor of the upper extremity

  • In light-weight hand tools, static posture may result in postural fatigue. Psotural tremor of the upper extremity in a static posture was measured to provide guidelines for hand tool weight. Postural tremor was measured on five levels of tool weitht : no weight, 400g, 800g, 1200g, and 1600g. Three types of camcorder recording postures were selected. For each condition, postural tremor was measured together with ENG of biceps, deltoid and pectoralis major, and Borg's CR-20 ratings of perceved exertion. Results of the experiment are as follows : frequency analysis of tremor revealed increased amplitude of frequency bands of 2-4Hz and 10-14Hz. Postural tremor of the upper extremeity maintained the initial level until fatigue developed. After the development of fatigue, the rate of change of postural tremor significantly increased. Different tool weights and hand postures showed different rates of tremor increase. Time tp fatigue and corresaponding endurance time were positively correlated with Borg's RPE scores.
