Objective: To determine the diagnostic performance of hematuria as a screening test for urinary bladder infiltration in cervical cancer patients with a prospective study design. Materials and Methods: Newly diagnosed cervical cancer patients at Srinagarind hospital from 14 June 2011 to 30 April 2012 were enrolled in this study. We collected midstream urine samples for urinalysis from every patient before routine cystoscopic exam for clinical staging. The presence of 3 or more red blood cells (RBCs) per high power field was defined as positive for hematuria. A two-by-two table was used to determine the diagnostic performance of hematuria to detect urinary bladder mucosal infiltration using cystoscopy and biopsy as the gold standard. Result: A total of 130 were patients included, 54 of which (41.5%) had hematuria. Of these, four patients (3.08%) had pathological report from cystoscopic biopsy confirmed metastatic squamous cell carcinoma. The sensitivity, specificity, PPV, NPV, and accuracy of hematuria as a screening test to detect urinary bladder mucosal infiltration of cervical cancer were 100%, 60.3%, 7.4%, 100%, and 61.5%, respectively. There was no single case of urinary bladder mucosal infiltration in patients initially staged less than stage III. Conclusions: Hematuria can be used as a screening test to detect urinary bladder mucosal infiltration of cervical cancer. This can reduce the number of cervical cancer patients who really need to undergo cystoscopy as a staging procedure to less than half and to less than 20% if stage III or more were included without missing a single case of urinary bladder mucosal infiltration.

Background: The accumulation of unpaired α-globin chains in patients with β-thalassemia major may clinically create ineffective erythropoiesis, hemolysis, and chronic anemia. Multiple blood transfusions and iron overload cause cellular oxidative damage. However, α-tocopherol, an antioxidant, is a potent scavenger of lipid radicals in the membranes of red blood cells (RBCs) of patients with β-thalassemia major. Purpose: To evaluate the effects of α-tocopherol on hemolysis and oxidative stress markers on the RBC membranes of patients with β-thalassemia major. Methods: Forty subjects included in this randomized controlled trial were allocated to the placebo and α-tocopherol groups. Doses of α-tocopherol were based on Institute of Medicine recommendations: 4-8 years old, 200 mg/day; 9-13 years old, 400 mg/day; 14-18 years old, 600 mg/day. Hemolysis, oxidative stress, and antioxidant variables were evaluated before and after 4-week α-tocopherol or placebo treatment, performed before blood transfusions. Results: Significant enhancements in plasma haptoglobin were noted in the α-tocopherol group (3.01 mg/dL; range, 0.60-42.42 mg/dL; P=0.021). However, there was no significant intergroup difference in osmotic fragility test results; hemopexin, malondialdehyde, reduced glutathione (GSH), or oxidized glutathione (GSSG) levels; or GSH/GSSG ratio. Conclusion: Use of α-tocopherol could indirectly improve hemolysis and haptoglobin levels. However, it played no significant role in oxidative stress or as an endogen antioxidant marker in β-thalassemia major.
Radish (Raphanus sativus L.) is a cruciferous vegetable, and its leaves have antioxidant and anticancer properties. This study was conducted to evaluate the effects of ethyl acetate extracts from radish leaves on hypertension in 11-week-old spontaneously hypertensive rats (SHRs). The SHRs were randomly divided into 3 groups of 6 rats each on the basis of initial systolic blood pressure (SBP) and were treated with oral administration of radish leaf extract (0, 30, or 90 mg/kg body weight [bw], respectively) for 5 weeks. Six Wistar rats were used as normotensive controls. The amount of the radish leaf extract had no effect on body weight. The SBP of the SHRs showed a decreasing trend with the consumption of the radish leaf extract. In the third week, the SBP of the group fed 90 mg extract/kg bw reduced from 214 mmHg to 166 mmHg and was significantly lower than that of the normotensive and hypertensive controls. The extract did not show a significant effect on the angiotensin-converting enzyme (ACE) activity in the serum, kidney, and lung. The extract increased the concentration of NO in serum and the activities of antioxidant enzymes such as glutathione peroxidase and catalase in red blood cells (RBCs). The serum concentrations of $Na^+$ and $K^+$ were not significantly different between all groups. However, the fecal concentrations of $Na^+$ and $K^+$ increased; the fecal concentrations of $Na^+$ and$K^+$for the normotensive and hypertensive controls were not different. Urinary excretion of $Na^+$ was higher in the normotensive Wistar rats than in the SHRs, while that of $K^+$ was not significantly different. These findings indicate that consumption of radish leaves might have had antihypertensive effects in SHRs by increasing the serum concentration of NO and fecal concentration of $Na^+$ and enhancing antioxidant activities.
Background: Reduced deformability of red blood cells (RBCs) may play an important role on the pathogenesis of chronic vascular complications of diabetes mellitus. However, available techniques for measuring RBC deformability often require washing process after each measurement, which is not optimal for dayto-day clinical use at point of care. The objectives of the present study are to develop a device and to delineate the correlation of impaired RBC deformability with diabetic nephropathy. Methods: We developed a disposable ektacytometry to measure RBC deformability, which adopted a laser diffraction technique and slit rheometry. The essential features of this design are its simplicity (ease of operation and no moving parts) and a disposable element which is in contact with the blood sample. We studied adult diabetic patients divided into three groups according to diabetic complications. Group I comprised 57 diabetic patients with normal renal function. Group II comprised 26 diabetic patients with chronic renal failure (CRF). Group III consisted of 30 diabetic subjects with end-stage renal disease (ESRD) on hemo-dialysis. According to the renal function for the diabetic groups, matched non-diabetic groups were served as control. Results: We found substantially impaired red blood cell deformability in those with normal renal function (group I) compared to non-diabetic control (P = 0.0005). As renal function decreases, an increased impairment in RBC deformability was found. Diabetic patients with chronic renal failure (group II) when compared to non-diabetic controls (CRF) had an apparently greater impairment in RBC deformability (P = 0.07). The non-diabetic cohort (CRF), on the other hand, manifested significant impairment in red blood cell deformability compared to healthy: control (P = 0.0001). Conclusions: The newly developed slit ektacytometer can measure the RBC deformability with ease and accuracy. In addition, progressive impairment in cell deformability is associated with renal function loss in all patients regardless of the presence or absence of diabetes. In diabetic patients, early impairment in RBC deformability appears in patients with normal renal function.
This study was conducted to investigate changes in blood properties of high potassium (HK) phenotype Jindo dogs (15kg$\pm$2kg) after daily oral administration with water celery extracts (10 ml/kg) for 7 days. Blood samples were collected for three days in a row before administration of water celery extracts. After water celery extracts administration, blood samples were collected at 3h, 6h, 9h and then on daily basis until day 10 post administration (PA). At day 15, final sample was collected. Blood samples were analyzed on the basis of red blood cell (RBC), white blood cell (WBC), packed cell volume (PCV), hemoglobin (Hb) concentration, mean corpuscular volume(MCV), mean corpuscular hemoglobin concentration(MCHC), gluthathione concentration(GSH) and met-hemoglobin(Met-Hb) concentration. The significant changes (p<0.01, p<0.05) of RBCs were shown at 3 h to day 5, and days 7 and 9 after administration. PCV values were decreased form 3 h to day 10 after administration. Mean Hb concentration showed significant increase as 3 h to day 3, and day 6 to day 9 after administration. The significant changes (p<0.05) of WBCs were shown at 9 h and day 1 after administration. The increased numbers of MCV were detected at days 6 to 9 after administration (p<0.05, p<0.01). The significant changes of MCHC were shown at 9h and day 1 after administration. The significant increases (p<0.01, p<0.05) of GSH concentration were detected at days 1, 6 and 7 after administration. In Met-Hb concentration, the significant increases (p<0.05) occurred at only 9h and day 7 after administration, The significant increases (p<0.01, p<0.05) of reticulocyte were detected at days 2, 4, 5, 6 and 7. Data from blood samples collected at day 15 after administration showed that all of blood analysis results returned to normal level, compared to controls.
A total number of 668 apparently healthy fish were obtained from farm to study the effect of two heavy metals in a form of (Copper sulfate and Mercuric chloride) on some hematological and biochemical parameters of blood. The $LC_{50}$ /96 hr. of Cu and Hg were estimated and fish exposed to $\text\tiny{^1/_2}$$LC_{50}$ for 7 days and for $1/_{10}$$LC_{50}$ for 8 weeks from each product separately. Results showed decrease in RBCs count, PCV% and Hb in acute and chronic mercury while a significant increase was shown in acute and chronic copper toxicity, total leucocytic count showed decrease in acute mercury toxicity and increase in the chronic case, while in copper toxicity non-significant decrease in acute and significant decrease in chronic toxicity was noticed. Elevated serum urea and creatinine in both acute and chronic mercury and copper toxicity was detected. No changes in total bilirubin in the acute mercury and chronic copper toxicity while significant increase in chronic mercury and acute copper. Elevation of serum AST and ALT in some days of acute toxicity of mercury and copper while in chronic mercury toxicity a significant elevation of both serums AST and ALT were detected .while in chronic copper toxicity serum AST was fluctuated and ALT showed no significant changes. CK study revealed significant decrease in acute mercury with fluctuation in the chronic toxicity while in copper toxicity it showed fluctuation in acute and significant decrease in chronic toxicity. Glucose value decreased in acute and chronic mercury toxicity while in copper toxicity it showed significant increase in the acute and increase followed by significant decrease in the chronic copper toxicity.
Anaplasmosis is a tick-borne disease of large and small ruminants, causing losses through mortality, abortion, weight loss and reduced milk production. In one dairy farm, for example, 250 of a total of 800 imported goats were diagnosed with a mysterious type of anemia during the summer and autumn of 1992. The etiologic agent was identified as Anaplasma spp by acridine orange and ultrastructure by electron microscopy. In order to monitor variations in blood biochemical and hematological parameters associated with the disease, blood samples were collected by jugular venipuncture from 50 goats at 3 month intervals between the period of February and October, 1993. The levels of RBCs, HB and HCT decreased from $18.48{\pm}1.96$ to $13.47{\pm}2.48X10^6/mm^3$, $12.25{\pm}1.41$ to $9.54{\pm}1.77g/dl$, and $43.09{\pm}4.75$ to $30.93{\pm}5.78%$, respectively. The values of MCH(Mean corpuscular hemoglobin), MCHC(Mean corpuscular hemoglobin concentration) and PLT(Platelet) were elevated from $6.58{\pm}0.30$ to $7.05{\pm}0.47pg$, $28.40{\pm}1.20$ to $30.82{\pm}1.85g/dl$ and $1688.34{\pm}750$ to $2046.82{\pm}783X10^3/mm^3$, respectively. Percent parasitized erythrocytes(PPE) increased from $0.61{\pm}0.5$ to $2.22{\pm}1.9%$, clinical biochemical parameters aspartate aminotransferase and alanine aminotransferase were $66.64{\pm}23.1K.U$ and $14.90{\pm}6.59K.U$, respectively and persisted at high levels throughout the observation period. The level of albumin(2.46)0.52 g/dl) was decreased corresponding to an elevated globulin and a reduced albumin/globulin ratio in October as compared with the values in February. It is concluded that caprine anaplasmosis may be an important cause of anemia and hepatic malfunction in goats.

In this study, the feasibility of spectral domain optical Doppler tomography for measuring blood flow characteristics in a micro-tube was demonstrated through several experiments. The use of an SD-ODT system in blood flow measurement can provide high resolution images (5 microns resolution). We prepared three capillary tubes to reveal the effect of different concentrations of hematocrit ratio (HR). One tube serves as the control. The two other tubes contained different concentrations of HR (5%, 25%). Three different capillary tube inlet flow velocities were tested in the present study. The Reynolds number (Re) which is based on the capillary tube inner diameter ranges from Re=6 to 48. We calculated a Doppler shift of the power spectrum of the temporal interference fringes with Kasai autocorrelation function to achieve the velocity profile of the flow. As a result, SD-ODT systems could not detect the cell depletion layer in the present study due to the limitation of spatial resolution. Nevertheless, these systems were proven to be capable of observing the RBCs of blood.

The micro device, coil, and channel for the biosensor integrated with the GMR-SV device based on the antiferromagnetic IrMn layer was fabricated by the light lithography process. When RBCs coupled with several magnetic beads with a diameter of $1{\mu}m$ passed on the micro channel, the movement of $RBC+{\mu}Beads$ is controlled by the electrical AC input signal. The $RBC+{\mu}Beads$ having a micro-magnetic field captured above the GMR-SV device is changed as the output signals for detection status. From these results, the GMR-SV device having the width magnitude of a few micron size can be applied as the biosensor for the analysis of a new magnetic property as the membrane's deformation of RBC coupled to magnetic beads.
Objectives : Angelicae gigas, A. sinensis and A. acutiloba are three types of plants used as Angelicae Radix (Dang-Gui). Many doctors of Korean medicine want to know the difference in clinical use of these three species. This study aimed to compare the hemoglobin-related activity of the extracts of Angelicae gigas, A. sinensis and A. acutiloba roots by measuring the intensity of binding oxygen to hemoglobin using Raman spectroscopy. Methods : Hemoglobin activity was measured by chemical analysis and Raman spectroscopy to compare the pharmaceutical efficacy of three Angelica root extracts. The oxygenated hemoglobin intensity, blood decursinol and acetylcholinestrase(AChE) concentration in mice were measured. In addition, the effects of three Angelica root extracts on oxygenated hemoglobin intensity, decursinol and AChE concentration in red blood cells (RBC) from human were also investigated. Results : The contents of decursin, decursinol and decursinol angelate, which affected physiological activity and RBC properties, were higher in the extract of A. gigas root than in those of A. sinensis and A. acutiloba roots. Moreover, oxygenated hemoglobin intensity in the A. gigas extract was higher than that of other two species in the blood of mice and human RBCs. Also, the blood decursinol and AChE concentrations of A. gigas root extract were higher than that of A. sinensis and A. acutiloba roots. Conclusions : These results suggest that A. gigas is more effective in treating disease related oxygen deficiency in RBC deformation under oxidative stress.
