Somaclonal variation was observed in the field on doubled haploid plants derived from single pollen of a rice cultivar "Hwaseongbyeo". The variations of seven quantitative traits including plant height and one qualitative trait (pubescence) in 436 lines ($R_2$ generation) were analyzed. The number of lines which fell beyond the boundaries of the 95% confidence intervals of the check variety, Hwaseongbyeo was checked for each quantitative trait, and of those fertility showed the highest variation frequency (85.6%), followed by plant height (77.5%), flag leaf length (66.5%), grains per panicle (42.2%), days to heading (34.5%), panicle length (30.7%) and panicles per hill (22.7%). And the variations of quantitative traits except days to flowering appeared to move in the negative direction compared to "Hwaseongbyeo". Variability within lines was also observed for quantitative and qualitative traits. Twenty-nine $R_2$ lines (7%) segregated for pubescence and 130 $R_2$ lines (30%) showed variation with regard to fertility. This suggests that mutations usually occur before diploidization. Twenty-nine $R_2$ lines representing a wide spectrum of variation were chosen for RAPD analysis. The number of lines showing DNA polymorphism compared to Hwaseongbyeo ranged 0 from to 10 according to the primer used and this seems to indicate that specific loci have highly mutable genomic site.utable genomic site.

This study was conducted to investigate the specific expression genes in the cloned bovine tissues. Donor cells, cloned tissues were analysed by RAPD-RFLP method. The results were detected three genes (CH-U7B, CH-U7M and CH-U7P) in the cloned fetus. It was found a single copy genes by southern hybridization. Sequence analysis of CH-U7M gene was shown 99% homology to a previously reported EST from a cloned bovine fetus. The putative ORF was encode a protein of hydrophobicity index 0.03. Semi-quantitative RT-PCR by using the CH-LS001 specific primer was remarkably detected in the lung tissue of cloned fetus. Further investigation of these genes may provide one of the key information to explain the early death, abnormal fetus, large off-spring and the low pregnancy rate in the production of cloned bovine.
Recently there has been an increasing amount of foreign livestock products distributed in the domestic market due to the market opening. Some vicious dealers sell the foreign beef in the trade name of the native beef during the final distribution step to arouse the social criticism frequently. In this report, we investigated a method to distinguish the native beef from the foreign one scientifically using the PCR-RAPD, a recent gene technique. Hygienical safety was also examined using a microbiological test for toxicity of Escherichia coli 0157:H7 and the food poisoning bacteria. The conditions of DNA amplification for the PCR analysis were $1{\times}Taq$ polymerase buffer, 1.5 mM $MgCl_2,\;50\;\mu\textrm{M}$ dNTP, 100 ng primers, 2.5 unit Taq polymerase and 5~20 ng template DNA, with the fmal volume of $50\;\mu\textrm{\ell}$. The size of the amplified product was detected mostly in the range of 0.5~2.0 kbp. The size of DNA, gene marking factor, which could be a criterion distinguishing the native beef from the foreign one, appeared approximately 1.2 kbp. The native beef was distinguished from the foreign beef with more than 90% of confidence by the gene marking factor. This method was expected to be useful in the breed discrimination between the native beef and the foreign one. The hygienical test results showed that, fortunately, neither Salmonella spp. and Listeria monocytogenes which form a principal cause of the food poisoning nor Enterohemorrhagic Escherichia coli : EHEC which have provoked a recent social disturbance, were detected at all.
The aim of this study was to described the genetic structure of Eleutherococcus senticosus in Korea. We investigated 10 patches, which are eight Korean patches and two foreign patches come from Russia and China growing at Korean habitat, using ISSR(inter-simple sequence repeats) markers. In ISSR PCR, the overall percentage of polymorphic ISSR amplicons was 76% and the mean number of amplicons per ISSR primer was 11.5, which were higher than the RAPD results for the some cultivars collected in Korea(Kim et al., 1998) ; 57% and 5.7, respectively. So ISSR markers provide more powerful tool than RAPD markers for the investigation of genetic variation in E. senticosus. There are relatively high genetic variation among patches as 62.8%, but low variation within eight Korean patches. Such pattern of genetic variation, which is not ordinary in other tree species, may be result from the narrow and limited habitats and the asexual reproduction of this species at the natural stands in Korea. Although the small sample size in this study seemed to be resulted in the high genetic variation among patches, the overall genetic interpretation of this study might not be much affected on the basis of the characteristics of the distribution and the reproduction system of E. senticosus. Analysis of genetic distance between all pairs of the patches did not reveal any trends with regard to geographic distance, which was confirmed by the results obtained from AMOVA(analysis of molecular variance) and PCA(principal component analysis). These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of patches with small number of individuals per patch is more effective for the ex situ conservation and for maintaining the genetic diversity of E. senticosus in Korea than smaller patches with large number of individuals.
A new cultivar "Dan Bi" of Pleurotus eryngii was developed by the method of mono-mono crossing between monokaryotic strains derived from KNR2312 and KNR2596. The parental strains, KNR2312 and KNR2596, are characterized by the property of high quality and a small number of primordia formation, respectively. The optimum temperature of mycelial growth was 25 and that of fruiting body development was $15{\sim}16^{\circ}C$. The period of harvesting including primordia formation was 0.7~1.3 days later than that of control strain Knneutari No. 3 in the culling cultivation. The color of pileus and stipe surface was neutral-brown and pure white, respectively. The shape of pileus was dome and has a scale like as cobweb. The yield was $93{\pm}9.7$ g per 850 cc of plastic bottle. Analysis of the genetic characteristics of the new commercial variety "Dan Bi" showed a different profile as that of the control strain, Knneutari No. 3, when RAPD (Random Amplified Polymorphic DNA) primer #8005 was used. This new variety "Dan Bi" of Pleurotus eryngii is characterized by a small number of primordia formation after scratching.
Paenibacillus polymyxa is a plant growth-promoting rhizobacterium (PGPR) which can be used for biological control of plant diseases. Several bacterial strains were isolated from rotten roots of Korean ginseng (Panax ginseng C. A. Meyer) that were in storage. These strains were identified as P. polymyxa, based on a RAPD analysis using a P. polymyxa-specific primer, cultural and physiological characteristics, an analysis utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME), and the 16S rDNA sequence analysis. These strains were found to cause the rot in stored ginseng roots. Twenty-six P. polymyxa strains, including twenty GBR strains, were phylogenetically classified into two groups according to the ERIC and BOX-PCR analyses and 16S rDNA sequencing, and the resulting groupings systematized to the degrees of virulence of each strain in causing root rot. In particular, highly virulent GBR strains clustered together, and this group may be considered as subspecies or biovar. The virulence of the strains seemed to be related to their starch hydrolysis enzyme activity, but not their cellulase or hemicellulase activity, since strains with reduced or no starch-hydrolytic activity showed little or no virulence. Artificial inoculation of the highly virulent strain GBR-1 onto the root surfaces of Korean ginseng resulted in small brown lesions which were sunken and confined to the outer portion of the root. Ginseng root discs inoculated in vitro or two-year-old roots grown in soil drenched with the inoculum developed significant rot only when the inoculum density was $10^{6}-10^{7}$ or more colony-forming units (CFU) per ml. These results suggest that P. polymyxa might induce ginseng root rot if their population levels are high. Based on these results, it is recommended that the concentration of P. polymyxa should be monitored, when it is used as a biocontrol agent of ginseng, especially in the treatment of stored roots.
A new commercial strain of oyster mushroom (was developed by hyphal anastomosis, and was improved byhybridization between a monokaryotic strain derived from Pleurotus ostreatus ASI 0635 (Gonji 7ho) and a dikaryotic strain derived from P. ostreatus ASI 0666 (Mongdol). The optimum temperatures for mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $12{\sim}18^{\circ}C$, respectively. When PDA (potato dextrose agar medium) and MCM (mushroom complete medium) were compared, mycelial growth was faster in MCM. Similar results were observed with the control strain P. ostreatus ASI 2504 (Suhan 1ho). Analysis of the genetic characteristics of the new cultivar ('Gosol') showed a different DNA profile from that of the control ASI 2504 strain, when RAPD (raurpDNA) primers URP1, 2, 3, and 7 were used. Fruiting body production per bottle was approximately116 g based on a production performance test. In addition, yields from a farm field trial were stably achieved in an inadequate production enviro. The color of the pileus was blackish gray, and the stipe was long and thick. Therefore, we expect that this new strain will satisfy consumer demand for high quality mushrooms.

When two different media including PDA (potato dextrose agar medium) and MCM (mushroom complete medium) were compared, the mycelial growth of this mushroom was faster in MCM than in PDA. Similar result was observed with the control strain P. ostreatus ASI 2504. Analysis of the genetic characteristics of the new cultivar "Mongdol" showed a different DNA profile as that of the control strain ASI 2504, when RAPD (Random Amplified Polymorphic DNA) primer URP3 and URP6 were used. Fruiting body production per bottle was about 106 g using demonstration farms. The color of pileus was blackish gray and the stipe was long. Therefore, we expect that this new strain "Mongdol" will satisfy the consumer's demand for high quality mushrooms.
To develop a new cultivar of King oyster mushroom(Pleurotus eryngii), G09-21 as parental strain was selected by the method of Di-mon crossing between monokaryotic strains derived from ASI 2824(Keunneutari No.2) and dikaryotic strain ASI 2887(Aeryni 3). The Pe21-51($G09-21-10{\times}2844-11$) was shown the best cultural characteristics, selected to be a new cultivar and named as 'Song-A'. The 'Song-A' was formed incompatibility line distinctly in the confrontation growth of parental strains Keunneutari No.2, Aeryni 3 and ASI 2844. The optimum temperature for mycelial growth, fruiting body development and pH arrange were $25{\sim}30^{\circ}C$, $14{\sim}16^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about $94.7{\pm}29.5$ g which is almost 106% quantity compared to that of other cultuvar Keunneutari No.2. Analysis of the genetic characteristics of the new cultivar 'Song-A' showed a different DNA profile as that of the control strains, Keunneutari No.2, Aeryni 3 and ASI 2844, when RAPD(Random Amplified Polymorphic DNA) primers URP4 and 7 were used. This new cultivar 'Song-A' of Pleurotus eryngii is characterized by a small number of primordia formation and the stip is thick and long. Therefore, we expect that this new strain will save of labor and cost by without culling work.

Analysis of the genetic characteristics of the new variety 'Dagul' showed different DNA bands as that of the control strains, Suhan No.3 and ASI 2782, when RAPD(Random Amplified Polymorphic DNA) primers URP7 and Rcb1 were used. This new variety 'Dagul' of oyster mushroom is characterized by multiple of individual generation and the stipe is long. We therefore expect that this new strain will increase of the income by cultivation of field.
