Processing Optimization and Physicochemical Characteristics of Collagen from Scales of Yellowfin Tuna (Thunnus albacares)

  • This study was conducted to investigate the optimal conditions of collagen extraction from scales of yellowfin tuna (Thunnus albacares) using surface response methodology. Four independent variables of NaOH concentration and pretreatment fime in alkali pretreatment and enzyme concentration and treatment time in enzyme hydrolysis were used to predict a model equation for the collagen yield. The determinant coefficient ($R^2$) for the equation was 0.906. The values of the independent variables for the maximum yield were 0.32 N NaOH, 16.38 h alkali pretreatment time, 0.18% enzyme concentration, and 31.02 h enzyme treatment time. In the physicochemical properties of tuna scale collagen, sodium dodecyl sulfate-polyacrylamide gel electrophoresis of tuna scale collagen showed the same migration distances as that of calf skin collagen. The amide A, I, II, and III regions of tuna scale collagen in Fourier transform infrared measurements were shown in the peaks of 3,414 $cm^{-1}$, 1,645 $cm^{-1}$, 1,553 $cm^{-1}$, and 1,247 $cm^{-1}$, respectively. The amount of imino acids in tuna scale collagen was 18.97% and the collagen denaturation temperature was $33^{\circ}C$. The collagen solubility as a function of NaCl concentration decreased to 4% NaCl (w/v) and the collagen solubility as a function of pH was high at pH 2-4 and sharply decreased from pH 4 to pH 7. Viscosity of the collagen solution decreased continuously until $30^{\circ}C$ and this decreasing rate slowed in the temperature range of $35-50^{\circ}C$.

  • Purpose : The aim of this study is to develope the Magnetic Resonance Spectroscopy(MRS) data processing S/W which plays an important role as a diagnostic tool in clinical field. Materials and methods : Post-processing software of MRS based on graphical user interface(GUI) under windows operating system of personal computer(PC) was developed using MATLAB(Mathwork, U.S.A.). This tool contains many functions to increase the quality of spectrum data such as DC correction, zero filling, line broadening, Gauss-Lorentzian filtering, phase correction, etc. And we obtained the normal human brain $^1H$ MRS data from parietal white matter, basal ganglia and occipital grey matter region using 1.5T Gyroscan ACS-NT R6 (philips, Amsterdam, Netherland) MRS package. The analysis of the MRS peaks were performed by obtaining the ratio of peak area. Results : The peak ratios of NAA/Cr, Cho/Cr, MI/Cr for the different MRS machines have a little different values. But these peak ratios were not significantly different between different echo time MRS peak ratios in the same machine (p<0.05). Conclusion : MRS post-processing S/W based on GUI using PC was developed and applied to the analysis of normal human brain $^1H$ MRS. This independent MRS processing job increases the performance and throughput of patient scan of main console. Finally, we suggest that the database for normal in-yivo human MRS data should be obtained before clinical applications.

Determination of N-nitrosamines in Water by Gas Chromatography Coupled with Electron Impact Ionization Tandem Mass Spectrometry (EI-GC/MS/MS를 이용한 니트로사민류의 수질분석)

  • This study assessed analysis of N-nitrosamines by separation, identification, and quantification using a gas chromatography (GC) mass spectrometer (MS) with electron impact (EI) mode. Samples were pretreated by a automated solid phase extraction (SPE) and a nitrogen concentration technique to detect low concentration ranges. The analysis results by EI-GC/MS (SIM) and EI-GC/MS/MS (MRM) on standard samples with no pretreatment exhibited similar results. On the other hand, the analysis of pretreated samples at low concentrations (i.e. ng/L levels) were not reliable with a EI-GC/MS due to the interferences from impurity peaks. The method detection limits of eight (8) N-nitrosamines by EI-GC/MS/MS analysis ranged from 0.76 to 2.09 ng/L, and the limits of quantification ranged from 2.41 to 6.65 ng/L. The precision and accuracy of the method were evaluated using spiked samples at concentrations of 10, 20 and 100 ng/L. The precision were 1.2~13.6%, and the accuracy were 80.4~121.8%. The $R^2$ of the calibration curves were greater than 0.999. The recovery rates for various environmental samples were evaluated with a surrogate material (NDPA-$d_{14}$) and ranged 86.2~122.3%. Thus, this method can be used to determine low (ng/L) levels of N-nitrosamines in water samples.

Antibacterial Activity and Component Analysis of Persicaria perfoliata Extracts (며느리배꼽 추출물의 항균 작용과 성분 분석)

  • In this study, the antibacterial activity and component analysis of Persicaria perfoliata extracts were investigated. MIC (Minimum Inhibitory Concentration) values of ethyl acetate fraction from P. perfoliata on Escherichia coli, Propionibacterium acnes, and Staphylococcus aureus were 0.25%, equally. The results showed that the antibacterial activity of the ethyl acetate fraction was the highest in the S. aureus and P. acnes. Ethyl acetate fraction from P. perfoliata extracts showed 5 bands in TLC and 8 peaks in HPLC (320nm). In HPLC chromatogram of ethyl acetate fraction, first main peak 2 (24.99%) and peak 3 (22.65%) were identified as quercetin-3-o-glucuronide and isoquercitrin in the order of elution time. Also, in HPLC chromatogram of deglycosylated fraction, main peak 2 (76.93%) was identified as quercetin in the order of elution time. And these components are very effective as antioxidants. Thus, these results indicate that extract/fractions of P. perfoliata can be applicable to new functional cosmetics for antioxidant, antibacterial activity.

Sedimentological and Hydromechanical Characteristics of Bed Deposits for the Cultivation of Manila clam, Ruditapes philippinarum in Gomso Tidal Flat (곰소만 조간대 바지락 양식장 저질의 퇴적학적 및 수리역학적 특성)

  • To investigate the effects of hydromechanical and textural characteristics of sediment deposits on the cultivation of Manila clam, Ruditapes philippinarum surface and sub-surface core sediments were collected seasonally in Gomso tidal flat. Grain size distribution were analyzed to investigate the annual variation of sediment texture. In winter unimodal distribution of grain size with the peak at $5\phi$ is dominant However, during the summer sediment texture become a little bit coarser and grain size distribution shows the peaks at $4\~5 \phi$. Optimum sediment texture for the cultivation of manila clam, R. philippinarum was found to be sandy silt in which mean Brain size was between 4 and $5 \phi$ with the sand content less than $50\%$ and clay content of $5\~10\%$. Mechanical and hydrological characteristics of sediment deposits were also studied in the laboratory and the results were applied to the numerical simulation for the behavior of surface sediment subjected to the cyclic loading from sea-water level change. Results of numerical simulation illustrate that the permeability of sediment had to be maintained in the range of $10^{-11}\sim10^{-12}m^2$ to ensure the proper sedimentological environment for the cultivation of manila clam, R. philippinarum. The deposits of virtually impermeable mud layer, with the threshold thickness of 4 cm, would be very hazardous to clam habitat.

Identification of the Sectional Distribution of Sound Source in a Wide Duct (넓은 덕트 단면내의 음원 분포 규명)

  • If one identifies the detailed distribution of pressure and axial velocity at a source plane, the position and strength of major noise sources can be known, and the propagation characteristics in axial direction can be well understood to be used for the low noise design. Conventional techniques are usually limited in considering the constant source characteristics specified on the whole source surface; then, the source activity cannot be known in detail. In this work, a method to estimate the pressure and velocity field distribution on the source surface with high spatial resolution is studied. The matrix formulation including the evanescent modes is given, and the nearfield measurement method is proposed. Validation experiment is conducted on a wide duct system, at which a part of the source plane is excited by an acoustic driver in the absence of airflow. Increasing the number of evanescent modes, the prediction of pressure spectrum becomes further precise, and it has less than -25 dB error with 26 converged evanescent modes within the Helmholtz number range of interest. By using the converged modal amplitudes, the source parameter distribution is restored, and the position of the driver is clearly identified at kR = 1. By applying the regularization technique to the restored result, the unphysical minor peaks at the source plane can be effectively suppressed with the filtering of the over-estimated pure radial modes.

Subspecies Classifying and Characterizing the Two Groups of Antagonistic Sorangium cellulosum against Botrytis cinerea and Colletotrichum acutatum (Botrytis cinerea와 Colletotrichum acutatum에 항균활성을 갖는 점액세균 Sorangium cellulosum에 대한 아종 분류 및 길항 특성 연구)

  • We classified the previously reported antagonistic strains of Sorangium cellulosum into 5 subspecies (A-E). Four strains were antagonistic to Botrytis cinerea (AB group) and two strains were antagonistic to Colletotrichum acutatum (AC group). According to the genetic and sequential analyses with standard genes, xynB1, bglA2, groEL1 for grouping, all strains of AB group were belonged to subspecies C and all strains of AC group were belonged to subspecies D. In addition, high pressure liquid chromatography with the culture filtrates confirmed the genetic results, because AB group had peaks with retention time at 20-22.5 minutes, whereas AC group had no peak. There was positive relationship ($R^2=0.9652$) between the control values of infecting B. cinerea on cherry tomatoes and the main peak areas of chromatograms among the four isolates of AB group. From the subspecies results of AB group, the main peak of KYC 3270 was expected to be epothilone D. However the retention times of the standard of commercial epothilone D and the main peak of KYC 3270 culture filtrate were different as 9.9 and 11.581 min., respectively. Finally, the antagonistic metabolite of AB group was inferred as 7-ketone epothilone D.

Molecular analysis of peptide toxins secreted by various Pseudomonas tolaasii strains (다양한 Pseudomonas tolaasii 균주에 의해 분비되는 펩티드 독소의 분석)

  • Pseudomonas tolaasii is a pathogen causing brown blotch disease in cultivated mushrooms. In previous study, various strains of P. tolaasii were isolated from the mushrooms with disease symptoms and they were further divided into Ptα, Ptβ, and Ptγ subtypes according to the 16S rRNA gene analysis. To investigate the secretion of peptide toxins, tolaasin and its analog peptides, culture extracts of Pt group strains were analyzed by gel permeation chromatography. Those of Ptα subtype strains contained two chromatographic peaks, band A and B. Meanwhile, those of Ptβ and Ptγ subtype strains contained mainly band A component and a little of band B. Molecular weights of toxic peptides of culture extracts were measured by MALDI-TOF mass spectrometry. In Ptα subtype strains, the peptide compositions of band A and B were same including tolaasin I (1,987 Da), tolaasin II (1,943 Da), and its two analog peptides, 1,973 Da and 2,005 Da. The strains of Ptβ and Ptγ subtype secreted many components of MW 1,100-1,200 Da, but they did not synthesize any tolaasin-like peptides. These results suggest that the only Ptα subtype strains secrete tolaasin and its analog peptide toxins and the strains of Ptβ and Ptγ subtypes have different pathogenic characters causing brown blotch disease.

Construction and Evaluation of Cohort Based Model for Predicting Population Dynamics of Riptortus pedestris (Fabricicus) (Hemiptera: Alydidae) Using DYMEX (상용소프트웨어(DYMEX)를 이용한 톱다리개미허리노린재(Riptortus pedestris) 밀도 변동 양상 예측 모델 구축 및 평가)

  • A Cohort based model for temperature-dependent population dynacmics of Riptortus pedestris was constructed by using a commercial software (DYMEX) and seasonal occurrence along with pesticide treatments effect was simulated and validated with pheromone trap data. Ten modules of DYMEX software were used to construct the model and Lifecycle module was consisted of seven developmental stages (egg, 1 - 5 nymphal instars, and adult) of R. pedestirs. Simulated peaks of adult populations occurred three or four times after the peak of overwintered populations which was similar to those observed from pheromone trap catch. Estimated dates for the second peak were quite similar (1-2 day difference) with those observed with pheromone trap. However, the estimated dates for the first population peak were 9-16 days later than the observed dates by pheromone trap and the estimated dates for the last peak were 17-23 days earlier than the observed dates. When insecticide treatments were included in the simulation, the biggest decrease in R. pedestris adult density occurred when insecticide was applied on July 1 for the first peak population: the estimated adult density of the second peak was 3% of untreated population density. When insecticide was assumed to be applied on August 30 for the second peak population, the estimated adult density of the following generation was about 25% of untreated population and the peak density of the following generation reached about two weeks later than untreated population. These results can be used for the efficient management strategies for the populations of R. pedestris.

Characterization of Dissolved Organic Matter in Stream and Industrial Waste Waters of Lake Sihwa Watershed by Fluorescence 3D-EEMs Analysis (형광 3D-EEMs를 이용한 시화호유역 하천 및 공단폐수의 유기물 특성 분석)

  • This study is conducted to examine spatial variations of Dissolved Organic Matter (DOM) in stream and waste waters of the different watershed areas (agricultural, residential, and industrial complex area) by using fluorescence 3D-EEMs (3 Dimensional Excitation Emission Matrix Spectroscopy). Furthermore, the research investigates the changes of DOM characterization by synchronous and 3D-EEMs during a rainfall event. The characterizations of DOM obtained by 3D-EEMs show two noticeable peaks at humic and protein-like regions. Humic-like substances (HLS) are found in rural and urban areas, and humic and protein-like substances (PLS) are shown in industrial area. According to the fluorescence peak $T_1:C_1$ ratios, it is observed that high amount of HLS was discharged from Banweol Industrial Complex (3TG). Additionally, linear relationships (Regression rate, $r^2$=0.65, $r^2$=0.66) have been shown between PLS (peak $T_1,\;B_1$) and biochemical oxygen demand (BOD), which indicates the impact of sewage. For the rainfall event (30 mm), no remarkable difference of DOM was found at rural area except increment of fluorescence intensity comparing dry period. In contrast, HLS at urban area is highly discharged within 30 minutes from the beginning of rainfall. Also, there are high influences of HLS and PLS within 20 minutes at industrial complex (4TG). Fluorescence 3D-EEMs has not only verifies a watershed of DOM origination but also monitors diffuse and point source impacts.
