Identification of Quantitative Trait Loci for Resistance to Soybean Cyst Nematode Race 5 (콩 Cyst 선충 Race 5에 대한 저항성 QTL 탐색)

  • The objectives of this study were; (1) to identify and localize QTLs for resistance to soybean cyst nematode(SCN) race 5 on RAPD map, (2) to idntify the magnitude and mode of inheritance for each QTL, and (3) to identify the best combinations of QTLs for resistance to SCN race 5. Based on the univariate regression analysis, we detected 26 markers(22 RAPD and 4 RFLP) which showed significant association(P<0.05) with resistance to SCN race 5. From MAPMAKER /QTL analysis, we identified two regions (LGC-20 and Group 2) for resistance to SCN race 5. The QTL that was localized at 8.0 cM from pK418C on LGC-20 showed a recessive mode of inheritance and the QTL that was localized between W03 and E02$^3$ on Group 2 showed a dominant mode of inheritance. Two pairs of flanking markers (E02$^3$ and W03, pK418C and pK418E$_1$) and one unlinked RAPD marker, G10$^1$ were used for multiple regression analysis. Marker combination which was composed of 4 markers, E02$^3$, G10$^1$, W03, and pK418E$_1$, explained the highest amount of phenotypic variation by SCN (35.2%). Further research for the identification of QTLs for resistance to SCN race 5 to explain larger portion of phenotypic variation is needed.

  • The analyses of relationship between specific genes and economic traits were performed on the basis of the genetic characterization of Korean native chicken in this study. Three generation reference pedigree was established from the several lines of Korean native chicken and cornish breed for the linkage analysis and many economic traits were collected for QTL(quantitative trait loci) detection. The experimental design were planed to detect QTLs of meat quality and laying performance from those two distinctive breeds.

Identification of Quantitative Trait Loci(QTL) for Meat Color Trait on Chromosome 7 in Pig (돼지 7번 염색체에서 육색 연관 QTL 확인)

  • The objective of this study was to identify the quantitative traits loci(QTL) for meat quality traits in pigs. Three-generation resource population was constructed from a cross between Korean native boars and Landrace sows. The resource population including founders, $F_1$ and $F_2$ was genotyped for 23 microsatellite markers on chromosome 7. The sex average total length of linkage map on chromosome 7 was estimated 154.6 cM. Meat quality traits including meat pH, meat color, drip loss, shear force, heating loss, crude fat, crude protein, crude ash and water content in muscle were collected from $F_2$ animals. For the QTL mapping, we used $F_2$ QTL Analysis Servlet of QTL express for web-based QTL mapping tools(http://qtl.cap.ed.ac.uk/). The QTLs for CIE-a and CIE-b on SSC7 were significantly detected at 1% and 5% chromosome-wide level, respectively.

Analysis of Quantitative Trait Loci (QTL) Associated with the Gel Consistency in Rice (쌀의 호응집성에 대한 QTLs 분석)

  • The purpose of this study was to identify the quantitative trait loci (QTL) associated with gel consistency (GC), and to evaluate the relationships between GC and physico-chemical properties related to eating quality in rice. A total of 120 doubled haploid (DH) lines derived from a cross between 'Samgang' and 'Nagdong' and 26 varieties were used for this study. The 120 DH lines were divided into four groups, soft (61~100 mm), medium (41~60 mm), hard (26~40 mm) and whole population, according to the gel length. A negative correlation between GC and amylose content was observed only in the soft group. The GC negatively correlated with lipid content in the medium, and the whole population, but positively correlated with lipid content in the soft group. The positive correlation between GC and protein content was observed in the soft group, while negative correlations were observed in the medium group and the whole population. Two significant QTLs regarding GC were detected on chromosomes 4 and 11. They collectively explained 23% of phenotypic variation with LOD score of 3.1 and 2.9, respectively. The DNA markers of S4026 and RM287 were tightly linked to GC in the DH population and 26 varieties, respectively.

  • 한우 17번 염색체 유전자 지도에서 QTL (quantitative trait loci) 분석을 실시하여 선별된 Loci 값들을 순열검정(Permutation Test)을 이용하여 유의성 검정을 실시하였다. 한편, 우수 경제형질 DNA marker들을 K-평균 군집법을 실시 파악하였다. 또한, 부스트랩 방법을 이용하여 선별된 Locus의 DNA Marker들의 신뢰구간을 구하였다. 이들 QTL과 K-평균법, 부스트랩 방법에 의해 한우의 염색체 17번 BMS941의 우수 DNA Marker 85, 105번을 선별하였다.

  • 한우 6번 염색체 유전자 지도에서 QTL (quantitative trait loci) 분석을 실시하여 선별된Locus 값을 순열검정(Permutation Test)을 이용하여 유의성 검정을 실시하였다. 한편, 우수경제형질 DNA marker들을 K-평균 군집법을 실시 파악하였다. 이들 QTL과 K-평균법에 의해 한우의 염색체 6번 ILSTS035의 우수 DNA marker 235번을 선별하였다. 선별된 DNA Marker 235번을 출품우에 적용하여 Bootstrap 방법을 이용하여 신뢰구간을 구하여 검정하였다.

Mapping QTL for Grain Quality Traits Using an Introgression Line Population from a Cross between Ilpumbyeo and Moroberekan in Rice (일품벼/모로베레칸 이입계통을 이용한 미질특성 관련 QTL 분석)

  • We conducted a QTL analysis of grain quality traits using 117 $BC_3F_4$ and $BC_3F_5$ lines developed from a cross between Ilpumbyeo and Moroberekan. Genotypes of 117 $BC_3F_5$ lines were determined using 134 simple sequence repeat (SSR) markers. A linkage map constructed using 134 SSR markers was employed to characterize quantitative trait loci (QTL). The 117 $BC_3F_4$ and $BC_3F_5$ lines were evaluated for eleven grain quality traits in 2005 and 2006. A total of 18 QTLs were identified for eleven traits, and the phenotypic variance explained by each QTL ranged from 9.9% to 35.2%. Moroberekan alleles contributed positive effects in the Ilpumbyeo background at two QTL loci for 1,000 grain weight. Four QTLs, two for chalky rice and one each for 1,000 grain weight and head rice were consistently detected in two consecutive years indicating that these QTLs are stable. Clusters of QTLs were observed in three chromosome regions. One cluster harboring five QTLs including head rice and brown rice ratio near SSR markers RM190 and RM314 was detected on chromosome 6. Another cluster harboring grain weight and white belly was detected on chromosome 2. Increase in white belly at this locus might be due to the increase in grain weight due to the presence of the Moroberekan allele. The Moroberekan alleles at two QTL loci, gw3 and gw4 associated with increased grain weight might be useful in breeding programs to develop high-yielding cultivars.

QTL Mapping of Resistance to Gray Leaf Spot in Ryegrass: Consistency of QTL between Two Mapping Populations

  • Gray leaf spot (GLS) is a serious fungal disease caused by Pyricularia oryzae Cavara, recently reported on the important turf and forage species, perennial ryegrass (Lolium perenneL.). This fungus also causes rice blast, which is usually controlled by host resistance, but durability of resistance is a problem. Few instances of GLS resistance have been reported in perennial ryegrass. However, two major QTL for GLS resistance have been detected on linkage groups 3 and 6 in an Italian x perennial ryegrass mapping population. To confirm that those QTL are still detectable in the next generation and can function in a different genetic background, a resistant segregant from this population has been crossed with an unrelated susceptible perennial clone, to form a new mapping population segregating for GLS resistance. QTL analysis has been performed in the new population, using two different ryegrass field isolates and RAPD, RFLP, and SSR marker-based linkage maps for each parent. Results indicate the previously identified QTL on linkage group 3 is still significant in the new population, with LOD and percent of phenotypic variance explained ranging from 2.0 to 3.5 and 5% to 10%, respectively. Also two QTL were detected in the susceptible parent, with similar LOD and phenotypic variance explained. Although the linkage group 6 QTL was not detected, the major QTL on linkage group 3 appears to beconfirmed. These results will add to our understanding of the genetic architecture of GLS resistance in ryegrass, which will facilitate its use in perennial ryegrass breeding programs.

QTL Analysis of Seed and Growth Traits using RIL Population in Soybean (콩 종실 및 생육형질 연관 분자표지 탐색)

  • An RIL population from a Shinpaldalkong2/GC83006 cross was employed to identify quantitative trait loci (QTL) associated with agronomic traits in soybean. The genetic map consisted of 127 loci which covered about 3,000cM and were assigned into 20 linkage groups. Phenotypic data were collected for the following traits; plant height, leaf area, flowering time, pubescence color, seed coat color and hilum color in 2005. Seed weight was evaluated using seeds collected in 2003 to 2005 at Suwon and in 2005 at Pyeongchang and Miryang sites. Three QTLs were associated with 100-seed weight in the combined analysis across three years. Among the three QTLs related to seed weight, all GC83006 alleles on LG O ($R^2\;=\;12.5$), LG A1 ($R^2\;=\;10.1$) and LG C2 ($R^2\;=\;11.5$) increased the seed weight. A QTL conditioning plant height was linked to markers including Satt134 (LG C2, $R^2\;=\;25.4$), and the GC83006 allele increased plant height at this QTL locus. For two QTLs related to leaf area, 1aM on LG M ($R^2\;=\;10.0$) and laL on LG L ($R^2\;=\;8.6$), the Shinpaldalkong2 alleles had positive effect to increase the leaf area. Satt134 on LG C2 ($R^2\;=\;41.0$) was associated with QTL for days to flowering. Satt134 (LG C2) showed a linkage to a gene for pubescence color. Satt363 (LG C2) and Satt354 (LG I) were linked to the hilum color gene, and Sat077 (LG D1a) was linked to the seed coat color. The QTL conditioning plant height was in the similar genomic location as the QTLs for days to flowering in this population, indicating pleiotropic effect of one gene or the tight linkage of several genes. These linked markers would be useful in marker assisted selection for these traits in a soybean breeding program.

Introgression of Oryza minuta into Rice, Oryza sativa (벼 Oryza sativa x O. minuta 여교배 계통에서 이입 염색체단편 검정)

  • An introgression line, WH79006 was produced from a single plant from $BC_5F_3$ families from a cross between Hwaseongbyeo used as a recurrent parent and O. minuta (BBCC, Ace. No. 101154) as a donor parent, which was subsequently self-pollinated for three generations. WH79006 resembled the O. sativa parent, Hwaseongbyeo. However it differed from Hwaseongbyeo in several traits including days to heading, culm length, grain size, spikelets per panicle and fertility. These differences in the traits between WH79006 and Hwaseongbyeo can be attributed to the O. minuta introgressions. To detect the introgressions, 294 SSR markers of known chromosomal position have been used. At least, 28 introgressed chromosomal segments have been identified using SSR markers and they map to all chromosomes except chromosome 2. The size of the introgressed segments ranged from 4 to 35cM. A QTL related to culm length was detected using 75 $F_2$ plants from the Hwaseongbyeo/WH79006 cross. This QTL, cl6 located on chromosome 6 explained $9.6\%$ of the total phenotypic variation in the population. This QTL has not been detected in the previous QTL studies between Oryza sativa cultivars, indicating potentially novel alleles from O. minutan.
