산화적 스트레스에 대한 섬오갈피 메탄올 추출물의 보호 효과 (Protective Effects of the Methanol Extracts of Acanthopanax koreanum against Oxidative Stress)

  • The protective effects of the aqueous MeOH extracts of stem and root Acanthopanax koreanum Nakai against oxidative stress were investigated. Anti-oxidant activity of the stem extract of A. koreanum was observed in the DPPH free radical scavenging $(IC_{50}=58.7\;{\mu}g/ml)$ and the SOD test $(IC_{50}=17.52\;{\mu}g/ml)$. According to data analysis of cell survival ratio of normal fibroblasts, the skin irritation by both extracts from A. koreanum was concerned. However, in the skin primary parch test, both extracts obtained Grade I, which means that there was no skin irritation. After induction of oxidative irritation, cell survival ratio of normal fibroblasts was also monitored and it turned out that stress-inducing group with both extracts had more increased cell survival ratio. The cell extension of the stress-inducing group treated with the stem extract was most dominant in morphological study. Based on these results, the stem extract of A. koreanum showed the protective effect against oxidative stress on normal fibroblast.

  • 공역리놀레산(CLA)은 항암, 항산화작용, 콜레스테롤저하 등의 효과를 지닌 물질로 알려져 있으며, 일본과 미국에서는 건장보조식품으로 널리 이용되고 있다. 본 연구에서는 CLA 생산능력이 있는 미생물을 김치제조 시에 스타터로 첨가하여 김치의 맛에는 변화를 가져오지 않으면서 CLA를 생산하는 최적조건을 밝히기 위해 혐기성 스타터 비피도박테리아를 배양액상태, 냉동건조된 상태, 캡슐화된 상태 등의 세가지 형태로 접종하였다. 이들 세가지 접종 방법 중 캡슐화된 상태로 스타터를 접종 시에 김치발효 중의 박테리아의 CLA 생산능력이 최적상태로 유지되었다. 김치 제조시에 부재료로 첨가되는 고추에는 상당량의 리놀레산(LA)이 함유되어 있음이 확인되어 추가로 LA를 첨가하지 않았다. 캡슐화된 비피도박테리아 스타터를 접종한 후 김치의 맛과 CLA생산 추이를 살펴본 결과 김치 맛에 영향을 끼치지 않고 CLA를 생산할 수 있는 최적의 접종량은 0.1%(w/w)임이 밝혀졌다.

일부 대학생들의 라면 섭취 관련 식행동과 국물 섭취 실태 조사 (Survey on Dietary Behaviors and Intakes of Instant Noodle (Ramyeon) Soup among College Students)

  • High intakes of sodium may increase the risk of hypertension or cardiovascular diseases. According to the 2010 Korea National Health and Nutrition Survey, the average intake of sodium was 4,878 mg/day with salt, kimchi, soy sauce, fermented soybean paste and Ramyeon being the five main sources of sodium. In order to identify solutions to reduce the intake of sodium, we investigated the intake patterns and eating behaviors of Ramyeon among 347 college students (male 146, female 201) using survey questionnaires. The average age of study subjects was 23.7 years for males and 20.5 years for females. The average Body Mass Index ($kg/m^2$) was 21.9 for males and 20.1 for females. The average frequency of Ramyeon intake was 2.0 times/week. The main reason for eating Ramyeon was convenience (56%), followed by good taste (27%), low price (11%) and other reasons (9%). The criteria for choosing Ramyeon were taste (72%), convenience (14%), price (7%), nutrition (1%), and the other factors (2%). Males' average intake of Ramyeon soup (61%) was higher than that of the females (36%). The estimated intake of Ramyeon soup by survey showed a positive correlation with the measured intake of Ramyeon soup. Sodium contents of Ramyeon were measured separately for the noodles and the soup, which were 1,185 mg/serving and 1,148 mg/serving each. Therefore, the amount of sodium intake can be reduced if students eat less Ramyeon soup. Also, we observed that dietary behaviors and soup intakes of Ramyeon between the sexes were different. Appropriate nutritional education for proper eating habits may help decrease the intake of sodium.

마라소스 주원료인 고추 및 산초 에탄올추출물의 항산화 및 항비만 효과 (Anti-oxidant and Anti-obesity Effects of Red Pepper and Zanthoxylum schinifolium Ethanol Extract, Main Ingredient of Mara Source)

  • 본 연구에서는 in vitro에서 중국 사천지역 고추와 산초 에탄올추출물의 항산화 효능 확인 후, 고지방식이에 의해 유도된 비만 마우스를 이용하여 중국 사천지역 고추, 산초, 고추와 산초복합물 투여 후 체중, 간과 복부 피하지방 무게, 혈중지질 함량을 측정하여 항비만 기능성 소재로서의 가능성 여부를 조사하였다. 실험군은 일반식이군, 고지방식이군, 고지방식이에 고추 및 산초, 고추와 산초복합물, HCA(hydroxyl citric acid)를 경구투여한 군의 6군으로 분류하여 8주간 사육 후, 체중, 간, 피하지방, 혈중지질 함량을 측정한 결과는 다음과 같다. 고지방식이만을 섭취한 군은 일반식이군과 비교하여 체중, 간, 복부 피하지방, 혈장지질 농도가 현저하게 증가하였다. 고지방식이에 고추와 산초 에탄올추출물을 경구투여한 군에서 8주 후 고지방군에 비해 유의적으로 체중을 감소시켰고, 복부 피하지방 양은 고지방군 3.77 g, 고추군, 산초군, 고추와 산초복합군은 각각 0.88 g, 0.9 g, 0.97 g으로 농도 의존적으로 복부 피하지방 양을 감소시켰다(P<0.05). 혈중 중성지질, 총콜레스테롤을 농도 의존적으로 감소시켜 혈액 내 지방의 축적을 저해하였고, HDL 콜레스테롤 농도가 고지방군에 비해 고추, 산초 및 고추와 산초복합군이 농도 의존적으로 증가하여(P<0.05) 조직 중의 콜레스테롤을 간으로 운반하여 콜레스테롤의 분해 및 배설을 촉진하여 조직 중의 콜레스테롤량을 감소시켜, 고추 및 산초에탄올추출물 투여가 마우스의 비만을 억제함을 확인할 수 있었다.

  • 계란의 비린내 강도는 가금류의 종류와 개인의 인식에 따라 다르지만 여러 요인에 의해 발생할 수 있다. 특히 트리메틸아민(TMA)이 원인으로 밝혀졌다. 주목할만한 점은 리보플라빈이 TMA를 무취의 트리메틸아민-N-옥사이드로 전환시키는 역할을 하는 효소인 플라빈 함유 모노옥시게나제 3의 활성을 증가시킬 수 있다는 점이다. 본 연구는 난황의 TMA 함량을 분석하여 비린내에 대한 기여도를 평가하고, 비린내를 방지하는 방법을 개발하는 것을 목적으로 하였다. 고체상 미세추출-기체 크로마토그래피/질량 분석법을 사용하여 리보플라빈이 강화, 보충된 사료를 먹인 암탉의 난황에 있는 휘발성 화합물을 감지하고 정량화했다. 또한 샘플 간 휘발성 물질의 상대적 함량을 비교하기 위해 전자코를 이용하여 상관관계 연구를 수행하였다. TMA는 콜린이 함유된사료를 섭취한 가금류의 난황에서 고농도로 검출되었지만 리보플라빈이 보충된 사료를 섭취한 가금류에서는 검출되지 않았다. 전반적으로, 이 연구는 리보플라빈이 TMA를 포함하여 계란에 존재하는 휘발성 물질의 양과 품질에 영향을 미친다는 것을 시사한다. 이러한 발견이 계란의 비린내를 줄이는 것은 물론 품질 향상에 기여하기를 기대한다.

Fermentation Conditions for the Production of Cell Mass and Comparison of Saccharide Utilization in Bifidobacterium longum and B. breve

  • Saccharide utilizations for the growth by Bifidobacterium longum and Bifidobacterium breve were compared. B. longum fermented glucose more rapidly than lactose as a carbon source whereas B. breve fermented lactose at a rate higher than that of glucose. The highest cell concentration, in the case of B. longum, was obtained when cultivated in a jar fermentor that contained modified MRS medium that half the beef extract was replaced by the same amount of tuna extract, and that pH was controlled at 6.0. B. breve showed the best growth when grown in a jar fermentor containing the MRS medium with lactose instead of glucose, controlled at pH 6.0. The optimal concentration of peptone in MRS medium for the growth of B. breve was 5 g/l.

Isolation and Characterization of Lactate-Tolerant Mutants in Bifidobacterium breve

  • The growth of Bifidobacterium breve strain HP2 was completely inhibited by the addition of lactate higher than 4.0% but not by the addition of acetate. Two kinds of lactate-tolerant mutants were isolated by the nitrosoguanidine treatment, enrichment on a liquid medium with 5% lactate, and selection on agar plates with 5% lactate. The mutants were not only able to grow in the presence of 5% lactate but also improved in viable cell stability in the acidic pH range. In a pH-controlled fermentor, mutant N-1-5 grew at a rate slower than that of the wild type but its growth yield was higher. Notably, mutants were more halotolerant and more osmotolerant than the wild type and they were able to grow in the presence of 3% NaCl or 25% lactose at which the wild type entirely stopped the growth. The enzyme activities involved in the lactose metabolism in B. breve were measured to elucidate the biochemical basis for lactate tolerance. In the mutants, activities of several enzymes including phosphoglucomutase decreased compared to the wild-type, which may explain their lower growth rate. However, the activity of lactate dehydrogenase or its nature of inhibition by lactate was not altered.

Effects of Partial Substitution of Lean Meat with Pork Backfat or Canola Oil on Sensory Properties of Korean Traditional Meat Patties (Tteokgalbi)

  • Korean traditional meat patties (Tteokgalbi) were prepared by replacing part of the lean meat content with either pork backfat or canola oil and the effect of substitution on sensory quality of the meat patties was investigated. Compared to the control patties, pork-loin Tteokgalbi with 10% pork backfat or 10% canola oil had significantly higher overall acceptability and higher perceived intensity of meat flavor, sweetness, umami, and oiliness. The pork-loin patties containing 10% fat also had lower perceived firmness, toughness, and chalkiness of than the control Tteokgalbi. The chicken breast Tteokgalbi with 10% canola oil had the lowest perceived firmness and chalkiness (control > pork backfat > canola oil). No significant difference was noted in the overall acceptability of chicken breast patties with 10% pork backfat and those with 10% canola oil. These results indicate that substituting 10% of lean meat of Tteokgalbi with fat improved the sensory acceptability of the product for Korean customers regardless of the lean meat and/or fat source used in the patties. Lean meat patties formulated with a limited amount of vegetable oil such as canola oil can be a healthy option for Korean consumers by providing desirable fatty acid profiles without sacrificing sensory quality of the product.
