  • In the current research for natural products with antimicrobial effects, various extracts of Puerariae thunbergiana and isoflavones against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, Candida albicans was stronger antimicrobial activity than the other microorganisms such as Streptococcus mutans, Staphylococcus epidermidis, and Staphylococcus aureus. The maximum activity was exhibited by methanol extract of the leaves of Puerariae thunbergiana Beth. against Candida albicans(MIC, $400{\mu}g/mL$). These results suggest that methanol extract of Puerariae thunbergiana has a potential antimicrobial activity.

  • Alpinia katsumadai is used in traditional oriental medicine as an antiemetic and for treatment of stomach disorders. This study was to investigate the effect of the 50% methanol extract of Alpinia katsumadai and Puerariae thunbergiana on ethanol metabolism in rat. The administration of 50% methanol extract of Alpinia katsumadai significantly decrease the ethanol concentration in rat. The AUC and Cmax of Alpinia katsumadai was 3.1 and 2.4 times less than the control group, respectively. It was more effective than Pueraria thunbergiana is known to as ethanol detoxicant. Furthermore, we tested on its ethanol metabolizing effect with the preparation containing this herb (Haeju pill) in beagle. The AUC of Haeju pill was about 1.9 times less than the control group. It was estimated that the extract of Alpinia katsumadai had feasibility as a ethanol detoxication agent.

  • Puerarin and daidzin have been isolated from Puerariae thunbergiana Benth. Structures were determined by spectroscopic methods. Compounds showed weak antimicrobial activity against S. mutans, S. epidermidis, S. aureus, and C. albicans (MIC, $800{\mu}g/ml$). However, these compounds were not antioxidative. Puerariae thunbergiana Benth. extracts against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, C. albicans was stronger antimicrobial activity than the other microorganisms. The antioxidative activity of was observed in the etyl acetate extract ($IC_{50},\;119.87{\pm}0.16\;{\mu}g/ml$). The DPPH radical scavenging effect ($IC_{50},\;1,673.3{\pm}0.54\;{\mu}M$) of the puerarin was comparable with that of synthetic antioxidant, BHA $(IC_{50},\;88.39{\pm}1.1){\times}10^{3}{\mu}M$. These results suggest that puerarin and daidzin have a potential antimicrobial activity.

  • This study was carried out to evaluate the ostrogenic/antiandrogenic activity of Puerariae Radix in Sprague-Dawley rats. It has known that diverse phytoestrogen were included in some Puerariae Radix, especially in Pueraria mirifica. The Uterotrophic assay and Hershberger assay were performed to evaluate the ostogenic/antiandrogenic activity of various Puerariae Radix (Pueraria thunbergiana, Pueraria mirifica and Butea superba). In Uterotrophic assay, the extracts of Puerariae Radix were administered subcutaneously to immature female SD rats from 19 to 21 days of age. The wet uterus and vaginal weighs significantly increased in the group only treated with extracts of Pueraria mirifica. But, in Hersh-berger assay, all extracts of Puerariae Radix did not show any effects in the castrated rats. These results suggest that Pueraria mirifica has not undrogenic/antiandrogenic effect but potent estrogenic effect. It is possible that components of Pueraria mirifica may act as endocrine disruptor in human body.

  • The contents of isoflavones and 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging activities were determined in flowers (Puerariae Flos) and roots (Puerariae Radix) of Pueraria thunbergiana to differentiate both pharmacochemical or pharmacognostical usage of both oriental medicinal drugs. The tectorigenin contents in flower before and after acid hydrolysis were shown to be 17.10 ${\mu}mole\;g^{-1}$ and 49.58 ${\mu}mole\;g^{-1}$, respectively, analyzed by HPLC. However, the root displayed much less content of tectorigenin even after acid hydrolysis (6.56 ${\mu}mole\;g^{-1}$) than in the flower. Acid hydrolysis made glycitin and tectoridin of the isoflavone glycosides almost disappear in the flower and root. DPPH assay results demonstrated that acid hydrolysis of both extracts or both glycosides should increase free radical-scavenging activities due to the increase of isoflavone contents. It is also suggested that the flower had much higher concentration of isoflavonoids than in the root.

  • Effective BuOH fraction was extracted from Puerariae radix (Pueraria thunbergiana), and several pharmacological activities were examined. From several pharmacological examinations, it was found that BuOH fraction has an antipyretic activity against typhoid vaccine pyrexia in rabbit, a papaverine-like antispasmodic activity on isolated ileum and jejunum of rat, an analgesic effects on mice treated by 0.7% HAc, and an anti-inflammatory activity of carrageenin edema in rat. It was found that BuOH fraction has detoxication effects on acute and subacute toxic rat induced by alcohol in consequence of biochemical tests of serum. Therefore it was considered that BuOH fraction has preventive and therapeutic effects in alcohol intoxication.

  • Puerariae Flos is a traditional herbal medicine that has long been used as a treatment for colds, diabetes, and hangovers. The herbal medicine contains a wide variety of isoflavones such as kakkalide, tectoridin, and tectorigenin. This study demonstrates that the substances undergo a certain degree of change depending on the storage period by the method of HPLC and $^{13}C-NMR$, and that the HPLC analysis can be used to determine the freshness of Puerariae Flos.

  • We investigated the effect of tectorigenin (1) with hypoglycemic and hypolipidemic effects on Phase I and II enzymes and TF activity to elucidate the action of an immunosuppressive compound (1) in the diabetic rat. Compound 1 was obtained from the hydrolysis of tectoridin easily isolated from the flower of Pueraria thunbergiana(Leguminosae). Puerariae Flos has been used as therapeutics for diabetes mellitus in traditional medicine of Korea. (omitted)

  • In macrophages, nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. In this study, we have examined the inhibition effects of NO production by 85% methanol extract of the flower of Pueraria thunbergiana (PF) in mouse macrophages. Extract of PF (1, 10, 100 ${\mu}g/ml$) inhibited NO production, inducible NO synthase and cyclooxygenase-2 expression in interferon-g and lipopolysaccharide-stimulated mouse peritoneal macrophages and it had no cytotoxicity. These data suggest that 85% methanol extract of PF might be useful in controlling macrophages mediated inflammatory disease.
