Effects of Extrasct of Pueraria radix on Lipid Metabolism in Rats Fed High Fat Diet (갈근추출물이 고지방식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • This study was designed to investigate the effect of Pueraria radix extract on lipid metabolism in the rats fed high fat diet. Male sprague dawley rats were given 5% Pueraria radix extract and fed diet containing 20 weight % fat. The results obtained were summarized as follows. Serum ALT and AST activities were reduced by Pueraria radix extract. Levels of total lipid, total cholesterol and triglyceride in the serum were higher in the HF group than in the control, but showed no significant differences between control and HF Pue group. Liver total lipid, total cholesterol and triglyceride contents were increased by high fat diet, but were lower in the HF Pue group than in the HF group. Fat absorption rate was increased with high fat diet feeding and higher in the HF Pue group than in the HF group.

Antioxidative Property and Whitening Effect of the Pueraria Radix, Poria Cocos and Coptidis Rhizoma (갈근, 복령 및 황련의 항산화성 및 미백효과)

  • In order to fine antioxidant and whitening agent source from nature, the comparisons of antioxidative activity and tyrosinase inhibitory activity were carried out for various ethanol extract on Pueraria Radix, Poria Cocos, and Coptidis Rhizoma. Comparing for three ethanol extracts, the highest electron donating ability was found at Poria Cocos (81.2%), but, the highest SOD-like ability, at the Coptidis Rhizoma(58%). Xanthine oxidase experiment exhibited 23.3% of hindrance effect in Pueraria Radix, and 79.3% in Poria Cocos. To evaluate at the whitening effect, tyrosinase inhibitory activity was conducted. Tyrosinase inhibitory activity was detected at 17.9% in the Puerariae Radix, 5.2% in the Poria Cocos and 83.3% in the Coptidis Rhizoma. From these results, we suggest that the ethanol extracts from Pueraria Radix, Poria Cocos, and Coptidis Rhizoma can be used for cosmetic ingredients.

Effects of Extract of Pueraria radix on Hematological Properties and Lead Level of the Tissues of the Pb-administered Rats (갈근추출물이 납 중독된 흰쥐의 혈액성분 및 조직의 납 축적에 미치는 영향)

  • This study was designed to investigate the effects of Korean Pueraria radix extract in Pb administered rats. Pb exposed rats were given 1% Pb in the diet. $\delta$-Adminolevulinic acid(DALA) and urinary glucose levels were increased with Pb administration and were lower in the Pb group than in the group administered Pb alone. Hematocrit value was decreased with Pb administration and was higher in the Pb group than in the C-Pb grou. $\delta$-Aminolevulinic acid dehydratase (DALAD) activity was decreased in the Pb group. ALT and AST were increased by Pb added and were lower in the Pb group than in the C-Pb group. Serum Pb content was higher in the Pb exposed rats than in the control groups, and no significant difference was found due to extract of Pueraria radix supplementation. Levels of liver, kidney and femur lead were reduced by Pueraria radix. Lead contents in feces and urine were higher in the Pb added groups than in the control group, and level of feces lead was increased by extract of Pueraria radix.

Antioxidative Components of Pueraria Root (칡뿌리의 항산화 성분)

  • As a search for natural antioxidants, antioxidative fractions in pueraria root were extracted and identified using column chromatography, thin layer chromatography and high performance liquid chromatography. Components which have most effective antioxidative activities were futher identified by IR and GC/MS. The strongest antioxidative component of pueraria root methanol extract was identified as puerarin. Puerarin obtained from pueraria root was practically effective as antioxidant at the level of 100 ppm. Antioxidative activity of the puerarin was higher in linoleic acid-water system than in a linoleic acid substrate. Puerarin, daidzin and daidzein contents in pueraria root juice were 0.39%, 0.45% and 0,03%, respectively.

  • This study was designed to investigate the effect of Pueraria radix extract on lipid peroxidation in ethanol administered rats. Male sprague dawley rats were given 25% ethanol(2.5g per Kg body weight; E), 10% pueraria radix extract(CP), 25% ethanol and 10% pueraria radix extract(EP). The activity of hepatic superoxide dismutase was increased by ethanol and was lower in the EP group than in the E group. Hepatic catalase activity was increased by ethanol, but decreased by Pueraria radix extract. E group rats had significantly higher liver glutathione peroxidase activity. Activity of hepatic glutathione S transferase was higher in the CP group than in the other groups. No significant dif ferences was found in liver glutathione and lipid peroxide contents between control and EP group. These data indicate that the peroxidative damage associated with chronic ethanol consumption might be decreased by Pueraria radix extract.

  • Recently, along with the remarkable increase in interest in natural products, the application of phytoestrogens in anti-aging products has become very important. In the focus, we developed Pueraria mirifica (PM), in this in vivo study on skin elasticity and wrinkles, and investigated the anti-aging properties. Thirty healthy women volunteers, between the ages of 30 and 60, applied the cream formula with $4\% Pueraria mirifica or placebo, on the face area twice a day for 16 weeks. The evaluation was made by silicon replica analysis, Cutometer SEM 575 and Comeometer followed by statistical treatment using the student's t-test. Results of skin elasticity measurement showed that topical application on the skin increased the total surface of skin elasticity in comparison with placebo group (p<0.05). However, PM cream does not have prominent effect on the depth of wrinkles and skin moisturization. This in vivo study demonstrates that, PM exhibits a significant effect on skin elasticity therefore, is of great interest in anti-aging and firmness skin care products.

In Vitro Antioxidant and Repaired Effects of the Extracts from Pueraria thunbergiana Roots (갈근 추출물의 항산화와 수복효과에 관한 연구)

  • The antioxidative activity was observed in the ethyl acetate extract $(IC_{50},\;119{\pm}0.16\;{\mu}g/mL)$ from Pueraria thunbergiana. The DPPH radical scavenging effect of this extract was comparable with that of synthetic antioxdant, BHA $(IC_{50},\;88.39{\pm}1.1\;{\mu}g/mL){\times}\;10^{-3}\;{\mu}M$. Pueraria thunbergiana extracts against microorganisms were evaluated in terms of the minimum inhibitory concentrations (MIC). In general, C. albicans was stronger antimicrobial activity than the other microorganisms. in vitro the antitoxic activity of extracts of Pueraria thunbergiana on NIH 373 fibroblasts was evaluated by the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide) method. Generally, detoxification effect by the extracts of Pueraria thunbergiana increased in proportion to the concentrations. When $10\;{\mu}g/mL$ of the hexane extract of Pueraria thunbergiana was treated it showed the highest antitoxic effects (p<0.01). These results suggest that the crude extract of P. thunbergiana has a potential therapeutic activity.

Anti-inflammatory Activity of Complex Saponin Separated from Pueraria flos and Adzuki Beans Mixture Extract

  • Pueraria flos and Adzuki Beans contains several bioactive compounds, such as saponin, oleanolic acid, and flavone. Pueraria flos and Adzuki Beans has traditionally been used to treat disorders of antioxidant activity, diabetes and liver detoxication, and it has antinociceptive and anti-inflammatory properties. However, complex saponin were validity of the anti-inflammatory activity has not been scientifically investigated. In this study, to determine anti-inflammatory activity of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract on nitric oxide and prostaglandinE2 assay. The anti-inflammatory activities of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract were evaluated for inhibitory activities against lipopolysacchride induced nitric oxide and prostaglandinE2 production protein expressions in RAW264.7 cell lines. The complex saponin separated from Pueraria flos and Adzuki Beans mixture extract inhibitory activity for both tests with protein high depressions(%) values showed in the ranges of 50~100 ㎍/ml. Overall, prostaglandinE2 tests had a higher inhibitory effect on inflammation than nitricoxide tests. Theseis result suggest a potential role of complex saponin separated from Pueraria flos and Adzuki Beans mixture extract as source of anti-inflammation agent.

  • Acute toxicity on the polysaccharide fraction of Pueraria lobata was examined using male and female Sprague-Dawley rats. The polysaccharide fraction of Pueraria lobata was orally administered at a dose of 5 mg/kg, 50 mg/kg, 500 mg/kg, 2,000 mg/kg and 5,000 mg/kg and observed for two weeks. No mortality and abnormal clinical signs were observed at the doses used. There were not any significant differences in parameters of blood biochemical values and urinalysis by the treatment of test material. All rats were appeared to be healthy and normal throughout the observation period. Also there was no difference in net body weight gain and gross pathological findings among the groups rats treated with different doses of the polysaccharide fraction with Pueraria lobata.

Downregulatory Effect of Extracts from Mistletoe (Viscum album) and Pueraria Root (Pueraria radix) on Cellular NF-κB Activation and heir Antioxidant Activity (겨우살이(Viscum album)와 칡뿌리(Pueraria radix) 추출물의 NF-κB활성 억제 및 항산화 효과)

  • Effects of mistletoe (Viscum album) extract and pueraria (Pueraria radix) extract on cellular NF-$textsc{k}$B activity were evaluated in human malignant keratinocytes (SCC-13) to elucidate the possible correlation of NF-$textsc{k}$B with antioxidant activity. The antioxidant activities of these natural extracts were examined in four different evaluation methods, i.e., lipid peroxidation value (POV) evaluation test, I,l-diphenyl-2-picrylhydrazyl radical (DPPH), nitric oxide (NO) scavenging test, and reducing power assay. Pueraria extract (0.5 mg) and mistletoe extract (5 mg) downregulated the cellular NF-$textsc{k}$B activation up to 35% and 10% compared to the control, respectively, although their effects were lower than the known NF-$textsc{k}$B downregulator, vitamin C (8.8 mg, 53%) in a cell-based NF-$textsc{k}$B activity assay system. In the POV test, relative antioxidant activities of mistletoe extract (86%) and pueraria extract (75%) were significantly higher than the known antioxidant, vitamin C (48%) at the same concentration (10 mg) and the degree of activity increased in a dose-dependent manner. Pueraria extract showed more potential radical scavenging activities than those of mistletoe extract evaluated in both DPPH and NO test. Especially, the NO radical scavenging activity of pueraria extract ($SC_{50}$/, 88 $\mu$g) was comparable to that of vitamin C ($SC_{50}$/, 77 $\mu$g). Even pueraria extract possessed a much less reducing power compared to vitamin C, it also revealed higher reducing power than that of mistletoe extract. These results indicate that mistletoe extract and pueraria extract may serve as an useful natural antioxidant agents, and led to suggest the hypothesis that compounds having an antioxidant activity, i.e., radical scavenging activity or reducing power may be correlated with the downregulation of NF-$textsc{k}$B activation in human keratinocytes.
