Objective: To evaluate the effectiveness of three different caries-preventing agents on artificial caries in a Streptococcus mutans-based caries model. Methods: Sixty-five caries-free human molar enamel blocks were treated with a demineralization solution and a remineralization solution. The specimens were assigned to the following groups according to the caries-protective product applied: group A, chlorhexidine varnish; group B, fluoride-releasing chemically cured sealant; group C, fluoride-releasing lightcured sealant; group D, positive control (specimens that were subjected to de- and remineralization cycles without treatment with any caries-protective agents); and group E, negative control (specimens that were not subjected to de- and remineralization cycles). Samples in groups A-D were stored in demineralization solution with S. mutans and thereafter in artificial saliva. This procedure was performed for 30 days. Average fluorescence loss (${\Delta}F$) and surface size of the lesions were measured using quantitative light-induced fluorescence at baseline and on the 7th, 14th, and 30th days. Results: After 30 days, group A demonstrated a significant increase in ΔF and the surface size of the lesions, no significant difference in comparison with the positive control group, and a significant difference in comparison with the negative control group. Group B showed no significant changes in both parameters at any of the measurement points. While group C showed increased ${\Delta}F$ after 14 days, no significant fluorescence change was observed after 30 days. Conclusions: Both fluoride-releasing sealants (chemically or light-cured) show anti-cariogenic effects, but the use of chlorhexidine varnish for the purpose of caries protection needs to be reconsidered.

Laboratory facilities for biology are designed as biosafety level 1, biosafety level 2, biosafety level 3, and biosafety level 4. Biosafety level designations are based on a composite of the design features, construction, containment facilities, equipment, practice and operation procedures required for working with agents from the various risk groups. Generally, biosafety level 3 means the facility that is appropriate for the experiments using pathogens which can cause serious diseases by aerosol transmission. The biosafety level assigned for the specific work to be done is driven by professional judgement based on a risk assessment, rather than by automatic assignment according to the particular risk group designation of the pathogenic agents to be used. In this paper, we introduced the biosafety level 3 facility operated in ADD(Agency for defense development). It contains the overview of facility, microbiological experiment, animal experiment, decontamination and waste disposal. Biosafety level 3 laboratory in ADD has served the vital role in the research of biological agents and antidote development.

Whitening agents are eagerly demanded especially by oriental women who often suffers from the pigmentary disorders such as melasma and solar lentigines. As these pigmentary disorders are exacerbated by ultraviolet (UV), the whitening agents could exert its effect not only by inhibiting melanin synthesis but also by inhibiting UV activated signals. Eumelanin protects UV-induced DNA damages so that the chemicals which could reduce UV-induced DNA damages might be the ideal lightening agents. The effect of newly synthesized antioxidants, a-tocopheryl ferulate, on protective effect for UV-induced DNA damages as well as inhibiting melanin synthesis are briefly shown. For clinical evaluation, our results of the efficacy of lightening agents on treating pigment macules in combination with chemical peeling are shown. Furthermore, newly developed facial image analyzers to quantitatively evaluate the improvement of pigment macules are introduced.
This study was aimed to investigate factors that affected the status of wearing personal protective equipment (PPE) for handling poultry carcasses with potential exposure to virulent avian infectious agents. A large outbreak of high pathogenic avian influenza (HPAI) occurred in South Korea in 2014. Many public officers participated for euthanizing and handling livestock carcass. However, several safety issues with using PPE were revealed. Therefore, a questionnaire survey was conducted for 340 people who participated euthanasia and carcass disposal in the place where HPAI mainly occurred in 2014. It was found that 31.8% of the respondents had ever taken off their protective equipment during operations because of its inconvenience. The most inconvenient PPE was goggles (54.6%), followed by mask (20.2%), latex gloves (11.6%), shoe covers (5.9%) and protective clothing (5.3%). The main complaints about this individual PPE was unclear sight, damp emitted toward eye, sweating, tearing easily and sweating, respectively. Considering such problems of PPE, new possible directions for improvement of gloves and goggles were suggested. With newly developed rubber coating fabric gloves and conventionally using latex and fabric gloves, H3N2 influenza virus transmission experiment was conducted. Rubber coating fabric gloves showed similar efficiency for blocking virus transmission with latex laboratory gloves and were not easily torn by sharp claws of chicken. In addition, air flow control safety eyewear was suggested to minimize moisture formation. The air flow control system efficiently suppressed moisture formation inside the goggles. Therefore our study will provide more specific directions about new PPE development for safety protection of actual wearers.
The bioassay-guided fractionation of protective agents against sepsis-induced lethality from the root cortex of Paeonia suffruticosa ANDREWS (Ranunculaceae) led to the isolation of eight known compounds: paeonol (1), 2,5-dihydroxy-4-methoxyacetophenone (2), acetovanillone (3), paeonoside (4), paeoniflorin (5), oxypaeoniflorin (6), apiopaeonoside (7), and methyl 3-hydroxy-4-methoxybenzoate (8). Among them, 3 showed the highest survival rate (100% with a dose of 30 mg/kg versus 17% for the control experiment) and reduced alanine aminotransferase level to be a half of the control value on the sepsis model induced by lipopolysaccharide/D-galactosamine.
Cis-dichlorodiammineplatinum(II)(cisplatin) is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. To evaluate the effect or radical scavengers on cisplatin nephrotoxicity in rats, cisplatin and Vitamin C were given intraperitoneally. Remarkable protective effects of Vitamin C against nephrotoxicity of cisplatin were observed when Vitamin C was administered to rats 1hr before cisplatin injection. hepatotoxicity induced by combination treament of cisplatin and Vitamin C was evaluated by measuring serum glutamic pyruvate transmainase(sGPT) and serum glutamic oxalate transminase(sGOT). Combination treatment did not affect the levels of sGPT and sGOT, and any combination treatment did not induce metallothionein biosynthesis in kidny, Vitamin C which has radical scavenging effect induce metallothionein biosynthesis in kidney. Vitamin C which has radical scavenging effect directly reduced nephrotoxicity of cisplatin in vivo. Thus, it seems that free radical is the cause of cisplatin nepthrotoxicity. Also, combination treatment did not reduce anticancer activity of cisplatin. The present results indicate that Vitamin C, when it is given with cisplatin, may provide protection against cisplatin nephrotoxicity without reducing anticancer activity.
cis-Dichlorodiammineplatinum(cisplatin) is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. So, to evaluate the effects of 2(3)-tert-butyl-4-hydroxyanisole(BHA) on cisplatin nephrotoxicity in rats, both compounds were given intraperitoneally. Remarkable protective effects of BHA against nephrotoxicity of cisplatin were observed when BHA was administered to rats 1hr after cisplatin injection. On the other hand pretreatment with BHA 1hr prior to cisplatin did not reduce weight loss, blood urea nitrogen and creatinine levels. Hepatotoxicity induced by combination treatment of cisplatin and BHA was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of SGPT and sGOT except 1hr pretreatment. The present results indicate that BHA may provide protection against cisplatin nephrotoxicity, when it is given 1hr after cisplatin.
Green tea catechins (GTC) and its major component, (-)-epigallocatechin gallate (EGCG) were studied for their protective effects against reactive oxygen species (ROS)-induced oxidative stress. GTC and EGCG skewed the strong antioxidative effects on the lipid peroxidation of ethyl linolate with Fenton's reagent and free radical scavenging effect to DPPH radical generation. They also protected $H_2O$$_2$- or KO$_2$-induced cytotoxicity in CHL cells or mouse splenocytes. These results indicate that GTC and EGCG are capable of protecting the lipid peroxidation, flee radical generation and cytotoxicity induced by ROS. The mechanism of inhibition in ROS-induced cytotoxicity may be due to their antiofidative and free radical scavenging properties. Therefore, GTC and EGCG may be useful chemopreventive agents by protecting the free radical generation which are involved in cancer and aging.
Cisplatin is one of the most effective antitumor agents currently available for cancer therapy. However, its clinical use has been limited by its severe side effects, especially nephrotoxicity. Therefore, brazilin, which has a radical scavenging effect, was given intraperitoneally to evaluate the effect on cisplatin nephrotoxicity in rats. Remarkable protective effects against nephrotoxicity of cisplatin were observed when brazilin was administered to rats simultaneously with cisplatin. Hepatotoxicity induced by combination treatment of cisplatin and brazilin was evaluated by measuring serum glutamic pyruvate transaminase and serum glutamic oxalate transaminase. Combination treatment did not affect the levels of sGPT and SGOT, and any combination treatment did not induce metallothionein in kidney. Brazilin which has radical scavenging effect directly reduced nephrotoxicity of wisplatin in vivo. Thus, it seems that nephrotoxicity of cisplatin was caused by free radicals. The present results Indicate that brazilin, when it is given with cisplatin, may provide protection against cisplatin nephrotoxicity in rats.
Five liglans, sauchinone (1), di-0-methyltetrahydrofuriguaiacin B (2), manassantin A (3), manassantin B (4) and saucerneol B (5), have been isolated from the MeOH extract of Saururus chinesis roots. The evaluation for protective effect of compounds 1-5 against tacrine-induced cytotoxicity in human liver-derived Hep G2 cells was conducted. Compounds 1,2, and 5 showed significant protective effects with the EC$_{50}$ values of74.2${\pm}$0.9, 111.3${\pm}$0.8,64.3${\pm}$0.8 ${\mu}$M, respectively. Silybin, one of the well-known hepatoprotective agents, used as a positive control, and also showed protective effect with an EC$_{50}$ value of 86.2${\pm}$0.5 ${\mu}$M.
