In order to reduce the systemic side effects and gastrointestinal irritation after its oral adminitration, ketoprofen was formulated as water-soluble packs. The effects of fatty acids and fatty alcohols on the penetration of ketoprofen through excised rat skins were evaluated. The role of stratum corneum as a protective barrier was also investigated. Fatty acids and fatty alcohols were generally effective in promoting ketoprofen penetration. The flux of ketoprofen through rat skin was maximized when oleic acid or lauryl alcohol was used as an enhancer. As the concentration of fatty acids and fatty alcohols varied from 0% to 10%, the amounts of ketoprofen penetrated were in direct proportion to that of fatty acids but those had no relationship with that of fatty alcohols. The penetration of ketoprofen through stripped skin was enhanced compared to normal skin irrespective of enhancer type, which indicated that the action site of enhancers would be stratum corneum.
Polysaccharide (PS) was fractionated from hot-water extract of Artemisia iwayomogi Kitamura stems. PS showed considerably higher hydroxyl radical scavenging activity than caffeic acid and glutathione. PS showed lower superoxide anion radical scavenging activity than hydroquinone and ascorbic acid. The scavenging activity of PS on the reactive oxygen species (ROS) induced by human neutrophils with zymosan was determined by the lucigenin-enhanced chemiluminescence assay. The scavenging effect of the PS on ROS as determined by the chemiluminescence assay was about 2-fold stronger than that of ascorbic acid at the same concentration. PS significantly decreased protein carbonyl and malonaldehyde contents in UVB irradiated skin homogenates, which was comparable to glutathione at the same concentration. This result suggested that PS derived from A. iwayomogi Kitamura stems may be a potent candidate as functional compound for the protection on UVB induced skin damage in cosmetics.
The skin, the largest organ in the body, undergoes age-related changes influenced by both intrinsic and extrinsic factors. The primary external factor is photoaging which causes hyperpigmentation, uneven skin surface, deep wrinkles, and markedly enlarged capillaries. In the human dermis, it decreases fibroblast function, resulting in a lack of collagen structure and also decreases keratinocyte function, which compromises the strength of the protective barrier. In this study, we found that treatment with γ-aminobutyric acid (GABA) had no toxicity to skin fibroblasts and GABA enhanced their migration ability, which can accelerate skin wound healing. UVB radiation was found to significantly induce the production of matrix metalloproteinase 1 (MMP-1), but treatment with GABA resulted in the inhibition of MMP-1 production. We also investigated the enhancement of filaggrin and aquaporin 3 in keratinocytes after treatment with GABA, showing that GABA can effectively improve skin moisturization. In vivo experiments showed that oral administration of GABA significantly improved skin wrinkles and epidermal thickness. After the intake of GABA, there was a significant decrease observed in the increase of skin thickness measured by calipers and erythema. Additionally, the decrease in skin moisture and elasticity in hairless mice exposed to UVB radiation was also significantly restored. Overall, this study demonstrates the potential of GABA as functional food material for improving skin aging and moisturizing.
본 연구에서 적색의 색소 성분을 함유한 오디 및 자초 추출물의 피부 광노화 보호효과를 조사하였다. 오디 추출물의 총 안토시아닌 함량을 분석한 결과 4.92 mg/g이었고 HPLC 분석을 통한 자초 추출물의 shikonin 함량은 9.58 mg/g으로 나타났다. 오디 및 자초추출물의 전자공여능은 각각 84.32%와 57.46%로 분석되어 오디가 우수하였으며, superoxide radical 소거능은 오디 추출물이 76.34%로 자초추출물 61.55%보다 항산화 활성이 높았다. 오디 및 자초추출물의 ORAC 값은 각각 $545.37{\mu}moles$ TE/g과 $427.18{\mu}moles$ TE/g으로 분석되어 오디 추출물의 항산화 활성이 높음을 알 수 있었다. HS68 세포에서의 UVB 조사군과 비교 시 MMP-1 생산 억제율은 오디 및 자초 추출물 $200{\mu}g/mL$ 첨가 시 각각 68.6%, 32.7%로 분석되었다. 무모쥐를 이용한 UVB 광노화 피부 동물 모델에서 오디 및 자초 추출물의 피부조직의 변화를 조사한 결과, 시료 처리군이 무처리군에 비해 홍반, 주름 두께 및 깊이, 표피 비후, 콜라겐 각질층, 표피층이 감소되었으며 수분함량은 증가하였다. 총 주름 깊이의 경우, 오디 추출물 7% 처리군과 자초 추출물 5% 처리군에서 대조군과 비교 시 30% 가량 억제되어 in vitro에서 뿐만 아니라 in vivo에서도 오디 및 자초 추출물이 UVB에 의한 피부 광노화 보호효과가 있음을 확인하였다.
This study was done to evaluate the antioxidant effect of Crataegi Fructus (CF) extract on the oxidative stress induced by reactive oxygen species (ROS), The human skin fibroblasts (Detroit 551) were cultured with various concentrations of hydrogen peroxide $(H_2O_2)$. The cytotoxicity of $H_2O_2-induced$ oxidative stress was performed by XTT assay for the cell viability according to the dose- and time-dependent treatment. For the protective effect of CF extract on $H_2O_2-mediated$ oxidative stress, cell viability, lactate dehydroganase (LDH) activity, and ferric thiocyanate (FTC) assay for the inhibitive activity of lipid peroxidation on CF extract were carried out. In this study, $H_2O_2-mediated$ oxidative stress was decreased cell viability dose-, and time-dependent manner and increased LDH activity compared with the control in these cultures. In the protective effect, CF extract increased cell viability and decreased LDH activity on $H_2O_2-mediated$ oxidative stress, especially, CF extract has antioxidant effect by the showing the inhibitive activity of lipid peroxidation by FTC assay. From these results, It is suggested that $H_2O_2-mediated$ oxidative stress was highly toxic, and also, CF extract showed the protective effect on $H_2O_2-mediated$ oxidative stress by showing the increased cell viability, decreased LDH activity and lipid peroxidation inhibition in these cultures.
Objectives : The purpose of this study is to investigate the effect of Shinsuwisaengtang (SSWST) extracts on antioxidant activity and inhibition of phototoxicity as a medicine for skin damage due to ultra violet stimulation. Methods : To determine the cytotoxicity of Hs68 cells from SSWST extracts, we investigated cell viability by MTT assay. To determine the protective effect of phototoxicity, we investigated cell viability after UVB radiation. The DPPH radical scavenging activity was examined to determine the antioxidant effect of SSWST extracts. Hoechst 33258 staining was performed to observe the protective effect of SSWST on UVB. Results : The cytotoxicity of SSWST extracts in Hs68 cells was not appeared significantly in all concentration. SSWST extracts significantly increased the viability of UVB-stimulated Hs68 cells at a concentration of 25, 50 and $100{\mu}g/ml$. SSWST extract showed higher DPPH radical scavenging than the control group. It was observed that SSWST extracts inhibited the apoptosis of the UVB-stimulated Hs68 cells through a fluorescence microscope. Conclusions : It was observed that SSWST did not induce cytotoxicity at a constant concentration and had a protective effect on phototoxicity and an antioxidant effect. Thus, it is considered that it maybe used as a medicine to cure skin damage caused by UVB and photoaging changes in the future.

Asterias pectinifera, a species of starfish and cause of concern in the aquaculture industry, was recently identified as a source of non-toxic and highly water-soluble collagen peptides. In this study, we investigated the antioxidant and anti-photoaging functions of compounds formulated using collagen peptides from extracts of Asterias pectinifera and Halocynthia roretzi (AH). Our results showed that AH compounds have various skin protective functions, including antioxidant effects, determined by measuring the scavenging activity of 2,2-diphenyl-1-picrylhydrazyl radicals, as well as anti-melanogenic effects, determined by measuring tyrosinase inhibition activity. To determine whether ethosome-encapsulated AH compounds (E(AH)) exert ultraviolet (UV)-protective effects, human dermal fibroblasts or keratinocytes were incubated with E(AH) before and after exposure to UVA or UVB. E(AH) treatment led to inhibition of photoaging-induced secretion of matrix metalloproteinase-1 and interleukin-6 and -8, which are associated with inflammatory responses during UV irradiation. Finally, the antibacterial effects of AH and E(AH) were confirmed against both gram-negative and gram-positive bacteria. Our results indicate that E(AH) has the potential for use in the development of cosmetics with a range of skin protective functions.
Purpose: This study investigated the knowledge, attitudes, and perceptions of a safe environment, as well as self-reported and observed compliance with the use of personal protective equipment (PPE), among intensive care unit (ICU) nurses. Methods: This study was conducted in October 2021 with 55 nurses working in the medical and surgical ICUs of a general hospital in Seoul. A self-reported questionnaire and an observational tool for compliance with the use of PPE were used to collect data. Results: Except for masks, the observed compliance for all other PPE types was lower than the self-reported compliance. Male nurses showed significantly higher observed compliance than female nurses. Self-reported compliance with PPE use, including "when there is a possibility of contact with objects contaminated with blood or body fluids, mucous membranes, damaged skin, or contaminated skin" (r = .23, p = .015) and "when there is a possibility of contact with blood, body fluids, secretions, or exudates" (r = .27, p = .004) showed significant correlations with knowledge of PPE. In the results for self-reported compliance with PPE use, gown use had a significant correlation with knowledge (r = .24, p < .001) and perceptions of a safe environment (r = .15, p = .016) for PPE, and gloves showed significant correlations with attitudes (r = .14, p = .024) and perceptions of a safe environment (r = .18, p = .003). Conclusion: The observed compliance with PPE use tended to be lower than the self-reported compliance among ICU nurses. It is necessary to develop and apply an effective educational program that can enhance improve actual compliance with PPE use among intensive care unit nurses.
Background: Skin is an organ that protects the human body from various environmental stimuli that can induce immune system activation. Skin aging can be largely divided into two categories: physiological aging, which is caused by the a decreased physiological function of the skin and structural changes with aging, and photoaging, which is caused by the chemical stress induced by external stimuli such as ultraviolet (UV) radiation. Methods and Results: The objective of this study was to investigate the anti-wrinkle and UV protective effect of catechin-rich green tea extract (CGTE) in activated keratinocyte (HaCaT cells) and UV-induced skin damage in hairless mice. The results showed that CGTE inhibits the tumor necrosis factor-alpha interferon-gamma ($TNF-{\alpha}+IFN-{\gamma}$)-induced expression of matrix metalloproteinase (MMP)-1 in HaCaT cells. In addition, the CGTE treatment significantly reduced wrinkle formation, epidermal thickness, collagen deposition, and transepidermal water loss in dorsal skin irradiated with UVB. However, the ${\beta}$-glucosidase activity was significantly increased. The CGTE treatment inhibits mRNA expression and enzyme activity of MMP-2 and MMP-9 in the dorsal skin irradiated with UVB. Conclusions: It is expected that CGTE can be effectively used as a functional food and cosmetic ingredient to improve skin moisture retention and reduce wrinkle formation.
