A triptolide-lysozyme (TP-LZM) conjugate was synthesized to achieve renal specific delivery and to reduce the side effects of triptolide. Triptolide was coupled to lysozyme through succinic via an ester bond with an average coupling degree of 1 mol triptolide per 1 mol lysozyme. The lysozyme can specifically accumulate in the proximal tubular cells of the kidney, making it a potential carrier for targeting drugs to the kidney. The structure of triptolide succinate (TPS) was confirmed by IR, $^{1}H-NMR$, MS and UV. The concentrations of triptolide in various samples were determined by reversed-phase high-performance liquid chromatography (HPLC). In this study, the physicochemical and stability profiles of TP-LZM under various conditions were investgated the stability and releasing profiles of triptolide-lysozyme (TP-LZM) under various conditions. In vitro release trails showed triptolide-lysozyme was relatively stable in plasma (less than 30% of free triptolide released) and could release triptolide quickly in lysosome (more than 80% of free triptolide released) at $37^{\circ}C$ for 24 h. In addition, the biological activities of the conjugate on normal rat kidney proximal tubular cells (NRK52E) were also tested. The conjugate can effectively reduce NO production in the medium of NRK52E induced by lipopolysaccharide (LPS) but with much lower toxicity. These studies suggest the possibility to promote curative effect and reduce its extra-renal toxicity of triptolide by TP-LZM conjugate.
Objective: To investigate the effect of short-term vibration frequencies on muscle force generation capabilities. Method: Six healthy participants were recruited for this study and only their dominant leg was tested. The subjects were tested under five conditions of vibration frequencies with constant amplitude: 0 Hz (no vibration), 30 Hz, 60 Hz, and 90 Hz, and the vibration amplitude was 10 mm for all frequency conditions. The vibration was applied to the rectus femoris (RF). The subjects were then instructed to maintain a steady-state isometric knee joint torque (100 Nm) for the first 6 s. After the steady-state torque production, the subjects were required to produce isometric knee joint torque by leg extension as hard as possible with a start signal within the next 3 s. The vibration was applied for ~4 s starting from 1 s before initiation of the change in the steady-state knee joint torque. Results: The results showed that the maximum voluntary torque (MVT) of the knee joint increased with the vibration frequencies. On average, the MVTs were 756.47 Nm for 0 Hz (no vibration) and 809.61 Nm for 90 Hz. There was a significant positive correlation (r = 0.71) between the MVTs and integrated electromyograms (iEMGs). Further, the co-contraction indices (CCIs) were computed, which represent the ratio of the iEMGs of the antagonist muscle to the iEMGs of all involved muscles. There was a significant negative correlation (r = 0.62) between the CCIs and MVTs, which was accompanied by a significant positive correlation (r = 0.69) between the iEMGs of the vibrated muscle (RF). There was no significant correlation between the MVTs and iEMGs of the antagonist muscle. Conclusion: The results of this study suggest that the short-term vibration on the muscle increases the level of muscle activation possibly owing to the increased Ia afferent activities, which enhances the muscle force generation capability.
This study deals with reconfigurable modular robot with respect to the compact and capability of representing the various actions for promoting creativity through education and play. Generally modular robot can be designed as a suitable robot that is transformed to various structure by reconstructing each cells, However, there are only few research on the education and play using those robots in the world and still nothing domestically. Unlike the existing modular robots only having a repeating motion, the proposed modular works by individual module such as sound is produced by sound module, wheel is driven by wheel module, LED module controls the visual expression, power is supplied by battery module, bluetooth module for communication, and dynamic motion realization is possible by using joint module. By manipulating the abilities endowed by individual modules, diversity of creative activities is possible and thus made an easy access for children. This study deals with the design of modular robotic by using the variety of different modules to endowed the learning and playing ability. And the study showed the utility of the operating behavior over the actual production and testing.
The metabolic response of Labeo rohita to thermal acclimation was assessed. Advanced fingerlings of L. rohita (average weight $31{\pm}1.4g$) were acclimated to 31, 33 and $36^{\circ}C$ compared with ambient temperatures ($26^{\circ}C$) for 30 days and different enzymes associated with stress response were estimated. Glycolytic enzyme-Lactate dehydrogenase, (LDH, E.C.1.1.1.27), TCA cycle enzyme-Malate dehydrogenase (MDH, E.C.1.1.1.37), Protein metabolizing enzymes-Aspartate amino transferase (AST, E.C.2.6.1.1) and Alanine amino transferase (ALT, E.C.2.6.1.2) of liver, gill and muscle, Gluconeogenic enzymes-Fructose 1,6 Bi phosphatase (FBPase, E.C. 3.1.3.11) and Glucose 6 phosphatase (G6Pase, E.C. 3.1.3.9) of liver and kidney were significantly (p<0.05) different with increasing acclimation temperatures. Heat Shock Protein-70 (HSP-70) was expressed in increasing intensity at 31, 33 and $36^{\circ}C$ but was not expressed at $26^{\circ}C$. Results suggest that higher acclimation temperatures enhance metabolism and L. rohita maintains homeostasis between $26-36^{\circ}C$ via an acclimation episode. Such adaptation appears to be facilitated by resorting to gluconeogenic and glycogenolytic pathways for energy mobilization and induction of HSPs.
Methyl paraben (MP), which is used as a preservative in pharmaceutical and cosmetic (shampoo) products, foods and beverages, enters into the aquatic environment and can pose a potential fish health hazard. In this experiment, effects of MP were evaluated in adult male common carp (Cyprinus carpio) by exposing them to fractions (1/$143^{rd}$ to 1/29th) of the $LC_{50}$ dose with every change of water for 28 days. Vitellogenin induction, metabolic enzymes, somatic indices and bioaccumulation were studied at weekly intervals. The $96^{th}$ h $LC_{50}$ of MP in fingerlings was 120 mg/L. Compared to the control, except for increases (p<0.01) in alkaline phosphatase (EC 3.1.3.1), alanine aminotransferase (EC 2.6.1.2) and liver size, there were decreases (p<0.01) in activity of acid phosphatase (EC 3.1.3.2), aspartate aminotransferase (EC 2.6.1.1), and testiculosomatic index following exposure to any dose of MP. Vitellogenin induction was significantly higher (p<0.01) in exposed than unexposed (control) fish. The bioaccumulation of MP in testis, liver, brain, gills and muscle tissues of fish increased significantly (p<0.01) with increase of dose from 0.84 ppm to 1.68 ppm. Dose and duration of exposure (p<0.01) indicated that an exposure period of 1 to 2 weeks was sufficient to cause changes in the quantifiable parameters studied. Fish exposed to 4.2 ppm MP became lethargic after the $26^{th}$ d. Histologically, degeneration, vacuolization and focal necrotic changes in liver and fibrosis-like changes in testicular tissue were noted.

Two experiments in the sequential order were conducted to determine the effects of different dietary lipid sources on the growth and fatty acid composition of rohu (Labeo rohita) and to examine the viability of a return fish oil finisher diet in restoring the human cardio-protective fatty acid profile. In the first experiment, fish were fed either with coconut oil (D1), olive oil (D2), sunflower oil (D3), linseed oil (D4) and fish oil (D5) as the main lipid source in the isonitrogenous diet for 90 days. No significant differences in growth were observed. Among the experimental diets moisture content of fish varied significantly (p<0.05) between the groups. Dietary lipid sources had a profound influence on the fatty acid profile of the muscle and liver as tissue fatty acid profile reflected the dietary fatty acid composition. Increased amounts of eicosapentaenoic acid and docosahexaenoic acid were observed in tissue of fish fed D4 and arachidonic acid was observed in the tissue of fish fed D3. We have also detected the metabolites of n-3 and n-6 pathway in D4 and D3 groups respectively, which prompted us to conclude that rohu, can desaturate and elongate $C_{18}$ essential fatty acids to $C_{20}$ and $C_{22}$ HUFA. A second feeding trial was conducted using the animals from the five different treatment groups for the duration of 30 days with fish oil rich diet (D5). Feeding with fish-oil rich washout diet resulted in the near equalization of all the other treatment groups tissue fatty acid profiles to that of fish oil (D5) fed group. These results indicate that a finishing fish oil diet can be effectively used to restore the human cardioprotective fatty acid profile in rohu fed with vegetable oils as lipid source.

White spot syndrome virus (WSSV) is one of the major viral pathogens affecting shrimp aquaculture. Four proteins, WSSV199, WSSV 222, WSSV 249 and WSSV 403, from WSSV are predicted to encode a RING-H2 domain, which in presence of ubiquitin conjugating enzyme (E2) in shrimp can function as viral E3 ligase and modulate the host ubiquitin proteasome pathway. Modulation of host ubiquitin proteasome pathway by viral proteins is implicated in viral pathogenesis. In the present study, a time course expression profile analysis of WSSV Open Reading Frame (ORF) 199 and Penaeus monodon ubiquitin conjugating enzyme (PmUbc) was carried out at 0, 3, 6, 12, 24, 48 and 72 h post WSSV challenge by semi-quantitative RT-PCR as well as Real Time PCR. EF1${\alpha}$ was used as reference control to normalize the expression levels. A significant increase in PmUbc expression at 24 h post infection (h.p.i) was observed followed by a decline till 72 h.p.i. Expression of WSSV199 was observed at 24 h.p.i in WSSV infected P. monodon. Since the up-regulation of PmUbc was observed at 24 h.p.i where WSSV199 expression was detected, it can be speculated that these proteins might interact with host ubiquitination pathway for viral pathogenesis. However, further studies need to be carried out to unfold the molecular mechanism of interaction between host and virus to devise efficient control strategies for this chaos in the shrimp culture industry.
