PFA와 CDT 알고리즘을 이용한 2차원 ISAR 영상 생성 (2D ISAR Imaging using PFA and CDT Algorithms)

  • 레이다 데이터로부터 ISAR(Inverse Synthetic Aperture Radar) 영상을 생성 할 때 가장 보편적으로 사용되는 것이 FFT 알고리즘이다. FFT 알고리즘을 넓은 방위각 데이터에 적용할 때는 MTRC(Moving Through Resolution Cell) 현상으로 영상의 번짐이 발생하는데 이를 보정하기 위하여 polar formatting 기법을 적용한다. 그러나 polar formatting도 적용할 수 있는 각도의 한계가 존재하며 만약 지나치게 넓을 경우 polar formatting을 적용하여도 원하는 영상을 얻을 수 없게 된다. 이 논문에서는 ISAR 영상 생성시 내삽법을 사용하는 polar formatting기법과 polar formatting의 단점인 내삽법을 사용하지 않는 CDT(Coherent Doppler Tomography) 기법을 비교하여 각각의 장단점을 분석하였다.

Cell Growth Inhibitory Effect of Tissue Cultured Root of Wild Panax ginseng C.A. Mayer Extract on Various Cancer Cell Lines

  • This study was performed to investigate the cell growth inhibitory effect of tissue cultured root of wild Panax ginseng C.A. Mayer (tcwPG). The human stomach carcinoma cell line, MKN 74, was incubated with 70% EtOH extract of tcwPG or Panax ginseng C.A. Mayer (PG) for 24 hrs. tcwPG inhibited cell growth at a concentration of $250{\mu}g/ml$. However, Panax ginseng extract did not inhibit cell growth at the same concentration. We also tested the ethyl acetate and $H_2O$ fractions of tcwPG. The inhibitory effect of the ethyl acetate fraction on cell proliferation in MKN 74 cells was more potent than that of the crude extract, and the inhibitory effect of the $H_2O$ fraction was less than that of the ethyl acetate fraction. When we separated tcwPG into polar and non-polar saponin fractions and then measured cell growth inhibition, the non-polar saponin in tcwPG exhibited cytotoxicity. To compare the effects of tcwPG on various cancer cell lines, we measured cytotoxicity in MKN 74 (stomach cancer cell line), SW 620 (colon cancer cell line) and PC 3 (prostate cancer cell line). All three cell lines showed cell growth inhibition, and the cell growth inhibitory effects were not quite different in the various cell lines. The non-polar saponins of tcwPG arrested PC 3 cells at G1-phase as did Panax ginseng.

A Modified Cryopreservation Method of Psychrophilic Chlorophyta Pyramimonas sp. from Antarctica

  • Polar psychrophiles which thrive under extreme conditions such as cold temperature, high salinity, and high dose ultraviolet light, emerge as novel targets for biotechnology. To prevent genetic drift and the possibility of contamination by subculturing, cryopreservation was employed for two psychrophilic microalgae, Porosira sp. (KOPRI AnM0008) and Pyramimonas sp. (KOPRI AnM0046), which have anti-freeze activities. Five cryoprotectants (dimethyl sulphoxide, ethylene glycol, glycerol, methanol and propylene glycol) showed toxicity at 20-30% (v/v). The optimal cryoprotectant concentration and equilibration time were less than 20% and 10 min, respectively. Cryopreservation was carried out in the presence of cryoprotectants either by direct freezing in liquid nitrogen ($LN_2$) or controlled freezing using a controlled rate freezer followed by storage in the $LN_2$ tank. As a result, Pyramimonas sp. (KOPRI AnM0046), a psychrophilic chlorophyta was revived. Cryopreserved Porosira sp. was not revived from either freezing protocols probably due to the silicic cell wall and its relatively large cell size. In the case of Pyramimonas sp. (KOPRI AnM0046), the controlled freezing method showed higher revival yield than the direct freezing method.

Statistical optimization of phytol and polyunsaturated fatty acid production in the Antarctic microalga Micractinium variabile KSF0031

  • Polar microorganisms produce physiologically active substances to adapt to harsh environments, and these substances can be used as biomedical compounds. The green microalga Micractinium variabile KSF0031, which was isolated from Antarctica, produced phytol, a natural antimicrobial agent. Furthermore, several polyunsaturated fatty acids (PUFAs), including omega-3, exhibit antioxidant properties. Here statistical methods (Plackett-Burman design and Box-Behnken design) were used to optimize the culture medium of KSF0031 to improve biomass production, and K2HPO4, MgSO4·7H 2O, and ammonium ferric citrate green (AFCg) were selected as significant components of the culture medium. Changes in the concentration of K2HPO4 and MgSO4·7H 2O as positive factors and AFCg as a negative factor affected cell growth to a remarkable degree. The biomass production in a 100 L culture using the optimized medium for 24 d at 18℃ was improved by 37.5% compared to that obtained using the original BG-11 medium. The quantities of PUFAs and phytol obtained were 13 mg g-1 dry cell weight (DCW) and 10.98 mg g-1 DCW, which represent improved yields of 11.70% and 48.78%, respectively. The results of this study could contribute to an improved production of phytol and fatty acids from Antarctic microalgae in the biomedical industry.

Optimization of Culture Medium for the Production of an Exopolysaccharide (p-CY02) with Cryoprotective Activity by Pseudoalteromonas sp. RosPo-2 from the Antarctic Sea

  • When cells are exposed to freezing temperatures, high concentrations of cryoprotective agents (CPA) prevent ice crystal formation, thus enhancing cell survival. However, high concentrations of CPAs can also cause cell toxicity. Exopolysaccharides (EPSs) from polar marine environments exhibit lower toxicity and display effects similar to traditional CPA. In this study, we sought to address these issues by i) selecting strains that produce EPS with novel cryoprotective activity, and ii) optimizing culture conditions for EPS production. Sixty-six bacteria producing mucous substances were isolated from the Ross Sea (Antarctic Ocean) using solid marine agar plates. Among them, Pseudoalteromonas sp. RosPo-2 was ultimately selected based on the rheological properties of the produced EPS (p-CY02). Cryoprotective activity experiments demonstrated that p-CY02 exhibited significantly cryoprotective activity at a concentration of 0.8% (w/v) on mammalian cells (HaCaT). This activity was further improved when combined with various concentrations of dimethyl sulfoxide (DMSO) compared to using DMSO alone. Moreover, the survival rate of HaCaT cells treated with 5% (v/v) DMSO and 0.8% (w/v) p-CY02 was measured at 87.9 ± 2.8% after freezing treatment. This suggests that p-CY02 may be developed as a more effective, less toxic, and novel non-permeating CPA. To enhance the production of EPS with cryoprotective activity, Response Surface Methodology (RSM) was implemented, resulting in a 1.64-fold increase in production of EPS with cryoprotective activity.

Comparison of Cell Lysis Techniques via Q-TOF LC/MS

  • Untargeted metabolomics is a useful tool for drug development focusing on novel chemotherapeutic and chemopreventative agents against cancer cells. In recent years, quadrupole time of flight liquid chromatography-mass spectrometry (Q-TOF LC/MS)-based untargeted metabolomic approaches have gained importance to evaluate the effect of these agents at the molecular level. The researchers working on cell culture studies still do not apply standardized methodologies on sample preparation for untargeted metabolomics approaches. In this study, the rough and wet lysis techniques performed on MCF-7 breast cancer cells were compared with each other via the Q-TOF LC/MS-based metabolomic approach. The C18 and hydrophilic interaction liquid chromatography (HILIC) columns were used for the separation of the metabolites in MCF-7 cell lysates. 505 peaks were detected through the HILIC column and 551 peaks were found through the C18 column for the wet lysis technique. This situation supported by the base peak chromatograms showed that the wet lysis technique allowed us to extract higher number of non-polar metabolites. Almost equal number of metabolites was found for the C18 and HILIC columns (697 peaks for the HILIC column and 695 peaks for the C18 column) when the rough lysis technique was used. However, the intensities of polar metabolites were higher for the rough lysis technique on base peak chromatograms for both the HILIC and C18 columns. Although cell lysis technique, which is the first step in the sample preparation for cell culture studies, does not cause dramatic differences in the number of the detected metabolite peaks, it affects the polar and non-polar metabolite ratio significantly. Therefore, it must be considered carefully especially for in vitro drug development studies.

  • The non-polar optical phonon scattering in the valence band depends on the masses, ratios, and types of mixtures of constituent atoms. Therefore, the random distribution of atoms in alloy semiconductors should be considered in the analysis of scattering mechanisms. For this purpose, the force equations of n atoms in a unit cell are expressed in a n x n matrix form to obtain the angular frequencies due to the acoustic and non-polar optical phonons. And, n is then assumed to be infinity. When this work is compared with other results published elsewhere, it is concluded that the independence of atomic displacement or amplitude of oscillation as ell as the infinite number of atoms in a unit cell must be taken into account for the random distribution of atoms in alloy semiconductors.

Populus 잎절편의 극성분화시 내생 Polyamine의 함량과 Polyamine 생합성 효소의 활성도 변화 (Changes in Endogenous Polyamine Levels during Polar Regeneration from Populus Leaf Segments)

  • Polyamine titers and the activities of arginine decarboxylase(ADC) and ornithine decarboxylase (ODC), enzymes which catalyze rate-limiting steps in polyamine biosynthesis, were investigated during polar regeneration of Populus leaf segments. The polar regeneration occurred at the basal cut end of Populus leaf segments through cell division around the vascular bundle. In the process of polar regeneration, the titers of putrescine and spermidine increased rapidly but the content of spermine remained constant. The leaf segments were then divided into three separte part ; the proximal, middle and distal. Spermidine titers showed an increase mainly in the proximal parts where polar regeneration occurred. On the other hand, putrescine titers showed an increase in the other two parts. In the course of polar regeneration, the activities of ADC and ODC increased, the ADC activities being higher than those of ODC. However, ODC activity was higher in the proximal part. Therefore, the spermidine contents and ODC activities are suggested to be related to polar regeneration in Populus leaf segments.

Non-Polar Myxococcus fulvus KYC4048 Metabolites Exert Anti-Proliferative Effects via Inhibition of Wnt/β-Catenin Signaling in MCF-7 Breast Cancer Cells

  • The Wnt/β-catenin signaling pathway is involved in breast cancer and Myxococcus fulvus KYC4048 is a myxobacterial strain that can produce a variety of bioactive secondary metabolites. Although a previous study revealed that KYC4048 metabolites exhibit anti-proliferative effects on breast cancer, the biochemical mechanism involved in their effects remains unclear. In the present study, KYC4048 metabolites were separated into polar and non-polar (ethyl acetate and n-hexane) fractions via liquid-liquid extraction. The effects of these polar and non-polar KYC4048 metabolites on the viability of breast cancer cells were then determined by MTT assay. Expression levels of Wnt/β-catenin pathway proteins were determined by Western blot analysis. Cell cycle and apoptosis were measured via fluorescence-activated cell sorting (FACS). The results revealed that non-polar KYC4048 metabolites induced cell death of breast cancer cells and decreased expression levels of WNT2B, β-catenin, and Wnt target genes (c-Myc and cyclin D1). Moreover, the n-hexane fraction of non-polar KYC4048 metabolites was found most effective in inducing apoptosis, necrosis, and cell cycle arrest, leading us to conclude that it can induce apoptosis of breast cancer cells through the Wnt/β-catenin pathway. These findings provide evidence that the n-hexane fraction of non-polar KYC4048 metabolites can be developed as a potential therapeutic agent for breast cancer via inhibition of the Wnt/β-catenin pathway.

Cellular growth and fatty acid content of Arctic chlamydomonadalean

  • Arctic microalgae thrive and support primary production in extremely cold environment. Three Arctic green microalgal strains collected from freshwater near Dasan Station in Ny-Alesund, Svalbard, Arctic, were analyzed to evaluate the optimal growth conditions and contents of fatty acids. The optimal growth temperature for KNF0022, KNF0024, and KNF0032 was between 4 and 8℃. Among the three microalgal strains, KNF0032 showed the maximal cell number of 1.6 × 107 cells mL-1 at 4℃. The contents of fatty acids in microalgae biomass of KNF0022, KNF0024, and KNF0032 cultured for 75 days were 37.34, 73.25, and 144.35 mg g-1 dry cell weight, respectively. The common fatty acid methyl esters (FAMEs) analyzed from Arctic green microalgae consisted of palmitic acid methyl ester (C16:0), 5,8,11-heptadecatrienoic acid methyl ester (C17:3), oleic acid methyl ester (C18:1), linoleic acid methyl ester (C18:2), and α-linolenic acid methyl ester (C18:3). KNF0022 had high levels of heptadecanoic acid methyl ester (26.58%) and heptadecatrienoic acid methyl ester (22.17% of the total FAMEs). In KNF0024 and KNF0032, more than 72.09% of the total FAMEs consisted of mono- and polyunsaturated fatty acids. Oleic acid methyl ester from KNF0032 was detected at a high level of 20.13% of the FAMEs. Arctic freshwater microalgae are able to increase the levels of polyunsaturated fatty acids under a wide range of growth temperatures and can also be used to produce valuable industrial materials.
